Abstract:Nitric oxide (NO)-an endogenous diatomic molecule-plays key roles in various physiological and pathological processes, including smooth muscle relaxation in blood vessels, immune response, neurotransmission, respiration, and cell apoptosis.The biological functions of this molecule greatly depend on the location, timing, and dosage at which it is released.It is important to develop NO-delivery platforms capable of holding NO stably during storage and subsequently release optimal amounts of NO spatiotemporally at the desired location and time.In this review, recent advancements in the preparation of new exogenous NO donors including diazeniumdiolates, S-nitrosothiols, nitrobenzene, and metal-nitrosyl complexes are discussed.The integration of these NO donors with various nanoplatforms for controlled NO delivery and their potential applications in the biomedical field are highlighted.

Abstract:Nitric oxide (NO) is an essential signaling molecule that participates in various important physiological and pathological processes, such as angiogenesis, neuronal transmission, immunomodulation, and tumor growth. The multifunctional roles of NO in biological processes have substantially increased interest in research regarding effective strategies in delivering exogenous NO for biomedical applications. The development of various NO prodrugs, such as organic nitrates, diazeniumdiolates, S -nitrosothiols, furoxan nitrogen oxides, and metal-nitrosyls, continue to undergo innovative modifications to improve NO payload and to prolong NO release. However, the clinical application of these low-molecular-weight NO donors is limited due to problems associated with burst release and untargeted delivery. The tunable administration of NO by biomaterial-based carriers offers an attractive and efficient strategy to achieve a controlled and targeted delivery of NO to specialized tissues or organs. In this review, we summarize the progress made in the development of NO donors and NO delivery systems and highlight potential biomedical applications aiming to inspire new perspectives in the field of biomaterial-based NO delivery systems.

Abstract:Nitric oxide (NO) has been recognized as a ubiquitous gaseous transmitter and the therapeutic potential has nowadays received increasing interest. However, NO cannot be easily directly administered due to its high reactivity in air and high concentration-dependent physiological roles. As such, a plethora of NO donors have been developed that can reversibly store and release NO under specific conditions. To enhance the stability and modulate the NO release profiles, small molecule-based NO donors were covalently linked to polymeric scaffolds, rendering them with multifunctional integration, prolonged release durations, and optimized therapeutic outcomes. In this minireview, we highlight the recent achievements of NO-releasing macromolecules in terms of chemical design and biomedical applications. We hope that more efforts could be devoted to this emerging yet promising field.

Abstract:Nitric oxide (NO) , an important biological second messenger and functional mole- cule, involves in numerous physiological and pathological processes. Several studies have shown that NO is of pleiotropic mechanisms against carcinomas. A few NO-donor drugs have been confirmed to have favorable anticancer activity and the potential for the further development of anticancer therapy. One of such potentials is to make a specific tissue-targeted delivery of NO-donor drugs to have tumor cells apop-tosis. This article presents advances of NO-donor drugs with anticancer activities, including NO-nonste-roidal anti-inflammatory drugs (NO-NSAIDs) and novel target NO-donors delivery.

Abstract:Nitric oxide (NO) plays a crucial role as a messenger or effector in the body, yet it presents a dual impact on cardio-cerebrovascular health. Under normal physiological conditions, NO exhibits vasodilatory effects, regulates blood pressure, inhibits platelet aggregation, and offers neuroprotective actions. However, in pathological situations, excessive NO production contributes to or worsens inflammation within the body. Moreover, NO may combine with reactive oxygen species (ROS), generating harmful substances that intensify physical harm. This paper succinctly reviews pertinent literature to clarify the in vivo and in vitro origins of NO, its regulatory function in the cardio-cerebrovascular system, and the advantages and disadvantages associated with NO donor drugs, NO delivery systems, and vascular stent materials for treating cardio-cerebrovascular disease. The findings provide a theoretical foundation for the application of NO in cardio-cerebrovascular diseases.

Abstract:Nitric oxide (NO), a gasotransmitter, is known for its wide range of effects in vasodilation, cardiac relaxation, and angiogenesis. This diatomic free radical also plays a pivotal role in reducing the risk of platelet aggregation and thrombosis. Furthermore, NO demonstrates promising potential in cancer therapy as well as in antibacterial and antibiofilm activities at higher concentrations. To leverage their biomedical activities, numerous NO donors have been developed. Among these, N-nitrosamines are emerging as a notable class, capable of releasing NO under suitable photoirradiation and finding a broad range of therapeutic applications. This review discusses the design, synthesis, and biological applications of polymeric N-nitrosamines, highlighting their advantages over small molecular NO donors in terms of stability, NO payload, and target-specific delivery. Additionally, various small-molecule N-nitrosamines are explored to provide a comprehensive overview of this burgeoning field. We anticipate that this review will aid in developing next-generation polymeric N-nitrosamines with improved physicochemical properties.

Abstract:Nitric oxide (NO) serves a multitude of functions in human physiology and pathology, and therapeutic NO‐releasing materials are vigorously developed for biomedical applications. Owing to its complex functions and its potential systemic effects, controllable NO release is desired for clinical applications. This review summarizes recent developments in the last five years in designing controllable NO‐delivering platforms for treating diseases such as cancer, bacterial infection, cardiovascular diseases, wounds, and eye diseases, with a focus on the most commonly used NO donors, including N‐diazeniumdiolates, S‐nitrosothiols, arginine, and N‐nitrosoamines. The progress and success of NO delivery strategies are highlighted, and their limitations are also included. Moreover, the therapeutic mechanisms of NO in the diseases are summarized, along with the corresponding means of controlling NO release. Finally, the prospects and potential challenges regarding the development of NO gas therapy are described, with the hope of encouraging new research in this area.

Abstract:Nitric oxide (NO), as an essential signaling molecule, participates in various physiological processes such as cardiovascular homeostasis, neuronal transmission, immunomodulation, and tumor growth. The multiple role of NO in physiology and pathophysiology has triggered a massive interest in the strategies of delivering exogenous NO for biomedical applications. Hence, different kinds of NO prodrugs have been developed up to date, including diazeniumdiolates, S-nitrosothiol, metal-nitrosyl, nitrobenzene, and so on. However, the clinical application of these low molecular weight NO donors has been restricted due to the problems of burst release, low payloads, and untargeted delivery. The delivery of NO by biomaterial-based carrier offers a beneficial strategy to realize the controlled and sustained delivery of NO to the targeted tissues or organs. In detail, NO-donor prodrugs have been attached and loaded to diverse biomaterials to fabricate nanoparticles, hydrogels, and coating platforms by means of physical, chemical, or supramolecular techniques. These NO-releasing biomaterials hold promise for a number of biomedical applications ranging from therapy of the ischemic disease and several types of cancer to cardiovascular devices and wound dressing. First, surface coating with NO-releasing biomaterials could mimic the physiological function of vascular endothelium, therefore promoting vascularization and improving the patency of cardiovascular implants. Next, because NO also mediates many important processes that take place after cutaneous injury, NO-releasing biomaterials could serve as ideal wound dressing to accelerate tissue regeneration. Finally, biomaterials enable localized delivery of high dose of NO to tumors in a sustained manner, thus generating potent tumoricidal effect. In this review, we will summarize the progress of different NO-releasing biomaterials, and highlight their biomedical applications with a hope to inspire new perspectives in the area of biomaterial-based NO-delivery systems.

Abstract:NO donors have found wide medical use long before the biorelevance of nitric oxide (NO) was unveiled. From glyceryl trinitrate, amyl nitrite and nitroprusside, to nitrosothiols, NONOates and more, a wide variety of chemicals have been employed as nitric oxide donors for treating pathological conditions and investigating the NO biology. Yet, the release kinetics of these classic NO donors are hard-to-define and hard-to-monitor in biological milieu. Novel NO donors with robustly defined NO release profiles are sought after. These endeavours have introduced a plethora of novel NO donors, including stimulus-triggered NO donors, fluorophore-labeled NO donors. The field also sees a recent addition, i. e. photo-triggered and photo-calibrated NO donors, whose NO release is photo-controlled and further accompanied by a fluorescence turn-on as a built-in calibration mechanism of NO release. This review provides an overview of the NO biochemistry, as a roadmap for the chemistry of NO donors. The design rationale and the release mechanism of different classes of NO donors were discussed. Some interesting and notable applications were briefly discussed, e. g. super-resolution localization microscopic imaging of cell structures, and photo-acoustic probes for disease diagnosis. With joint-force, these NO donors constitutes a group of indispensable molecular tools of chemical biology and medicine in real applications.

Abstract:Nitric oxide (NO) is an important pharmaceutical agent of considerable therapeutic interest ascribed to its vasodilative, tumoricidal and antibacterial effects. Rapid development of functional nanomaterials has provided opportunities for us to achieve controllable exogenous delivery of NO. In the current review, a variety of functionalized colloidal nanovehicles that have been developed to date for nitric oxide delivery are reported. Specifically, we focus on inorganic nanomaterials such as semiconductor quantum dots, silica nanoparticles, upconversion nanomaterials, carbon/graphene nanodots, gold nanoparticles, iron oxide nanoparticles as the functional or/and supporting materials to carry NO donors. N-diazeniumdiolates, S-nitrosothiols, nitrosyl metal complexes and organic nitrates as main types of NO donors have their own unique properties and molecular structures. Conjugating the NO donors of different forms with appropriate nanomaterials results in NO delivery nanovehicles capable of releasing NO in a dose-controllable or/and on-demand manner. We also consider the therapeutic applications of those NO delivery nanovehicles, especially their applications for cancer therapy. In the end, we discuss possible future directions for developing exogenous NO delivery systems with more desired structure and improved performance. This review aims to offer the readers an overall view of the advances in functionalized colloidal nanovehicles for NO delivery. It will be attractive to scientists and researchers in the areas of material science, nanotechnology, biomedical engineering, chemical biology, etc.

Abstract:Nitric oxide (NO) is a versatile endogenous molecule, involved in various physiological processes and implicated in the progression of many pathological conditions. Therefore, NO donors are valuable tools in NO related basic and applied applications. The traditional spontaneous NO donors are limited in scenarios where flux, localization, and dose of NO could be monitored. This has promoted the development of novel NO donors, whose NO release is not only under control, but also self-calibrated. Herein, we reported a phototriggered and photocalibrated NO donor (NOD565) with an N-nitroso group on a rhodamine dye. NOD565 is nonfluorescent and could release NO efficiently upon irradiation by green light. A bright rhodamine dye is generated as a side-product and its fluorescence can be used to monitor the NO release. The potentials of NOD565 in practical applications are showcased in in vitro studies, e.g., platelet aggregation inhibition and fungi growth suppression.

Abstract:The shortened is as follows: Therapeutic gas nitric oxide (NO) has demonstrated the unique advances in biomedical applications due to its prominent role in regulating physiological/pathophysiological activities in terms of vasodilation, angiogenesis, chemosensitizing effect, and bactericidal effect. However, it is challenging to deliver NO, due to its short half-life (<5 s) and short diffusion distances (20-160 mu m). To address these, various polymeric NO delivery nanoplatforms (PNODNPs) have been developed for cancer therapy, antimicrobial and cardiovascular therapeutics, because of the important advantages of polymeric delivery nanoplatforms in terms of controlled release of therapeutics and the extremely versatile nature. This reviews highlights the recent significant advances made in PNODNPs for NO storing and targeting delivery. The ideal and unique criteria that are required for PNODNPs for treating cancer, cardiovascular diseases and infection, respectively, are summarized. Hopefully, effective storage and targeted delivery of NO in a controlled manner using PNODNPs could pave the way for NO-sensitized synergistic therapy in clinical practice for treating the leading death-causing diseases.

Abstract:Nitric oxide (NO) acts a pivotal role in regulating various physiological processes of vasodilation, platelet aggregation, and vascular smooth muscle cell mitogenesis and proliferation. This makes NO a promising candidate for the treatment of cardiovascular problems like hypertension and vascular stenosis. However, the high reactivity of NO poses an issue for effective NO delivery. To overcome this limitation, recent developments on three-dimensional (3D) materials have been explored with either physical or chemical incorporation of NO releasing donors, to provide spatiotemporal control over NO-signaling pathways in blood vessels. Here, we offer an overview on the current efforts, and propose future perspectives for precise regulation on NO delivery in advanced 3D materials toward proper vascular functionality.

Abstract:Nitric oxide (NO) donors are valuable tools to probe the profound implications of NO in health and disease. The elusive nature of NO bio-relevance has largely limited the use of spontaneous NO donors and promoted the development of next generation NO donors, whose NO release is not only stimulated by a trigger, but also readily monitored via a judiciously built-in self-calibration mechanism. Light is without a doubt the most sensitive, versatile and biocompatible method of choice for both triggering and monitoring, for applications in complex biological matrices. Herein, we designed and synthesized an N-nitroso rhodamine derivative (NOD560) as a photo-triggered and photo-calibrated NO donor to address this need. NOD560 is essentially non-fluorescent. Upon irradiation by green light (532 nm), it efficiently release NO and a rhodamine dye, the dramatic fluorescence turn-on from which could be harnessed to conveniently monitor the localization, flux, and dose of NO release. The potentials of NOD560 for in vitro biological applications were also exemplified in in vitro biological models, i.e. mesenchymal stem cell (MSC) migration suppression. NOD560 is expected to complement the existing NO donors and find widespread applications in chemical biological studies.

Abstract:Nitric oxide (NO) is a relatively short-lived inorganic free radical, which can be produced by different types of cells in multi-cellular organisms. This diffusible messenger functions as either an effector or a second messenger in many intercellular communications or intracellular signaling pathways. NO becomes noxious if it is produced in excess. These effects are mainly mediated by the reactivity of NO with various reactive oxygen species, which can be countered by antioxidant enzymes. In addition, NO can directly modify biological molecules via S-nitrosylation and lead to altered signaling responses. Accumulating evidence suggests that NO has a double-edged role in a dose-dependent, cell-type specific, and biological milieu-dependent way. In the present review, we summarized the synthesis and signaling pathway of NO, and especially focused on its involvement in biological processes, such as endoplasmic reticulum stress, apoptosis and autophagy. Besides, we discussed the functions of NO in the nervous system and its potential role in neurodegenerative diseases. We proposed the target on NO may shed light on the treatment of the related diseases.

Abstract:The increasing understanding of the role of nitric oxide (NO) in cancer biology has generated significant progress in the use of NO donor-based therapy to fight cancer. These advances strongly suggest the potential adoption of NO donor-based therapy in clinical practice, and this has been supported by several clinical studies in the past decade. In this review, we first highlight several types of important NO donors, including recently developed NO donors bearing a dinitroazetidine skeleton, represented by RRx-001, with potential utility in cancer therapy. Special emphasis is then given to the combination of NO donor(s) with other therapies to achieve synergy and to the hybridization of NO donor(s) with an anticancer drug/agent/fragment to enhance the activity or specificity or to reduce toxicity. In addition, we briefly describe inducible NO synthase gene therapy and nanotechnology, which have recently entered the field of NO donor therapy.
