Abstract:ConspectusEnzymes are essential for life, especially in the development of disease and on drug effects, but as we cannot yet directly observe the inside interactions and only partially observe biochemical outcomes, tools “translating” these processes into readable information are essential for better understanding of enzymes as well as for developing effective tools to fight against diseases. Abstract:Overexpression of β-galactosidase (β-gal) in tumor cells may serve as a valuable biomarker for the early diagnosis of some cancers (such as ovarian cancer). In addition, abnormal accumulation of β-gal is also considered an essential marker of cell senescence. Therefore, it is important to construct fluorescent probes with excellent fluorescence properties to visualize β-gal in biological systems. Here, we designed and screened a novel fluorescent probe XM for the detection of β-gal. Spectral data show that the probe has a good affinity (Km = 2.6 μM) for β-gal, large stokes shift (190 nm), fast response speed (stable within 20 min), and low detection limit (6.7 × 10−3 U/mL). Based on the above advantages, XM can not only detect β-gal content in cancer cells but also track the changes of β-gal content in zebrafish at different developmental period. We believe XM will become a powerful tool for the early cancer diagnosis and cellular senescence.

Abstract:As a kind of toxic gas, carbon monoxide (CO) can hinder uptake of oxygen in humans. However, more and more studies have shown that CO is an important gaseous messenger in the body and playing an indispensable role in intracellular signaling pathways. So, it is necessary to develop an analytical method to study CO in living organisms. Although there are many CO-responsive probes, most of them have the disadvantages of a small Stokes shift or short emission wavelength. In order to address the above issue, a novel probe (FDX-CO) with a large Stokes shift (190 nm) and long emission wavelength (770 nm) was firstly synthesized to detect CO. The probe shows high sensitivity and superior selectivity toward CO. Moreover, the probe was successfully used for visualizing exogenous and endogenous CO in cells by fluorescence imaging, 3D quantification analysis and flow cytometric analysis. More importantly, FDX-CO could excellently detect CO in mice, which suggests that this probe has the potential ability to image CO in vivo. This probe can be viewed as a useful tool in the research of CO.

Abstract:Carbon monoxide (CO) is an important gas signaling molecule and has been widely involved in regulating important life processes. Effective monitoring of CO in living systems is critical. Combined with the accuracy of ratio detection and the advantages of two-photon imaging, a simple ratiometric two-photon fluorescent probe RTFP was rationally designed and synthesized using 7-(diethylamino)-4-hydroxycoumarin as a two-photon fluorophore and allyl carbonate as the reactive unit. Probe RTFP exhibited excellent selectivity and sensitivity towards CO, and was successfully applied to image endogenous CO in living cells and zebrafish.

Abstract:Carbon monoxide (CO) is an important gas transporter and is closely associated with a series of physiological and pathological processes. So far, it is clear that plenty of fluorescent probes have been prepared, but most of them still have some disadvantages and is unsuitable for imaging in vivo. Herein, a novel fluorecent probe named HPQ-BI-CO is synthesized and used to detect CO. In this probe, the benindole group is introduced to extend the wavelength. This probe possess a variety of advantages such as long emission wavelength, high sensitivity and selectivity, and rapid response capability. Moreover, the probe possesses a low cytotoxicity, which make it apply in the imaging of exogenous and endogenous CO in living 4T1 cells. Furthermore, this probe can be utilized as a promising tool for fluorescent bioimaging of CO in vivo like zebrafish.

Abstract:Herein, we present the synthesis and application of a fluorogenic, large Stokes-shift (>100 nm), bioorthogonally conjugatable, membrane-permeable tetrazine probe, which can be excited at common laser line 488 nm and detected at around 600 nm. The applied design enabled improved fluorogenicity in the orange/red emission range, thus efficient suppression of background and autofluorescence upon imaging biological samples. Moreover, unlike our previous advanced probes, it does not require the presence of special target platforms or microenvironments to achieve similar fluorogenicity and can be generally applied, e.g., on translationally bioorthogonalized proteins. Live-cell labeling schemes revealed that the fluorogenic probe is suitable for specific labeling of intracellular proteins, site-specifically modified with a cyclooctynylated, non-canonical amino acid, even under no-wash conditions. Furthermore, the probe was found to be applicable in stimulated emission depletion (STED) super-resolution microscopy imaging using a 660 nm depletion laser. Probably the most salient feature of this new probe is that the large Stokes-shift allows dual-color labeling schemes of cellular structures using distinct excitation and the same detection wavelengths for the combined probes, which circumvents chromatic aberration related problems.

Abstract:In vivo measurement of abnormal β-galactosidase (β-gal) in living cells and tissues is urgently required for early diagnosis and treatment of primary ovarian cancer. Herein, a FRET-based ratiometric two-photon fluorescent probe FTR-βgal with a large emission shift, rapid response and low cytotoxicity was developed for sensitively detecting β-gal activity in vivo. FTR-βgal was designed with a naphthalene derivative as the energy donor and a rhodol derivative as the energy acceptor, which provided an efficient FRET strategy. Remarkably, FTR-βgal was successfully applied in TP imaging of β-gal with the ratio signal rapidly transform from 0.15 to 5.9 (∼40-fold) between 540 nm and 450 nm. Furthermore, FTR-βgal was used to visualize β-gal levels in tumor upon tissue penetration of 30–150 μm achieved. Therefore, FTR-βgal was applicable for clinical detection in living cells and tissues as a promising strategy for diagnosis of various associated cancers.

Abstract:To realize sensing and labeling biomarkers is quite challenging in terms of designing multimodal imaging probes. In this study, we developed a novel β-galactosidase (β-gal) activated bimodal imaging probe that combines near-infrared (NIR) fluorescence and magnetic resonance imaging (MRI) to enable real-time visualization of activity in living organisms. Upon β-gal activation, Gal-Cy-Gd-1 exhibits a remarkable 42-fold increase in NIR fluorescence intensity at 717 nm, allowing covalent labeling of adjacent target enzymes or proteins and avoiding molecular escape to promote probe accumulation at the tumor site. This fluorescence reaction enhances the longitudinal relaxivity by approximately 1.9 times, facilitating high-resolution MRI. The unique features of Gal-Cy-Gd-1 enable real-time and precise visualization of β-gal activity in live tumor cells and mice. The probe's utilization aids in identifying in situ ovarian tumors, offering valuable assistance in the precise removal of tumor tissue during surgical procedures in mice. The fusion of NIR fluorescence and MRI activation through self-immobilizing target enzymes or proteins provides a robust approach for visualizing β-gal activity. Moreover, this approach sets the groundwork for developing other activatable bimodal probes, allowing real-time in vivo imaging of enzyme activity and localization.

Abstract:A small fluorescence ratiometric probe consisting of a single dye species, N-methyl-6-hydroxyquinolinium (MHQ), and coupled enzymatic substrates, exhibits a dramatic colour change (deep blue to red) and possesses a huge response ratio (over 2000 fold) upon specific recognition of target enzymes. Such dramatic responses are attributed to the excited-state proton transfer processes of MHQ molecules in water. Here the detection of beta-galactosidase and porcine pancreatic lipase is successfully demonstrated and this class of molecules has the potential to be developed as a "naked-eye" probe in vitro.

Abstract:Glycosidases play important roles in modulating the structural and functional integrity of glycoproteins and glycolipids, and thus are promising biomarkers for disease diagnosis. While current approaches for glycosidase detection mainly rely on an enhancement of the UV-vis absorbance or fluorescence emission of glycosyl indicators, here we develop a ratiometric fluorescent probe for the sensitive and selective detection of glycosidase activity based on the combined mechanisms of excited-state intramolecular proton transfer (ESIPT) and solid-state luminescence en-hancement (SSLE). The probe behaves like a typical SSLE when glycosylated, and exhibits a ~140 nm red-shift in fluorescence owing to activation of ESIPT after deglycosylation. Such a large Stokes shift may facilitate the unbiased analysis of glycosidase activities when used in diagnostic and drug-screening assays.

Abstract:Probe 1 has been synthesized as the first ratiometric fluorescent cobalt probe, and it exhibits several highly favorable features, in particular a remarkable ratiometric fluorescence response at two wavelengths. Importantly, the electronic energy transfer (EET) efficiency of the probe is modulated by the energy acceptor molar absorptivity variations on cobalt binding, which then transforms to a large ratiometric fluorescence response at two wavelengths. This signaling mechanism should open a new avenue for the development of powerful ratiometric probes with large emission signal ratios at two wavelengths for exciting applications in many fields. The first ratiometric fluorescent cobalt probe has been synthesized, and it exhibits several highly favorable features, in particular a large ratiometric fluorescence response at two wavelengths. Abstract:Long-wavelength emission with high contrast toward species of interest is highly desired for fluorescence imaging in living objects. Herein, we proposed an exquisite enzyme-regulated "aggregation-induced emission (AIE) and intramolecular charge transfer (ICT)" strategy to optimize the signal-to-noise ratio for highly sensitive detection of enzymes. Benefiting from the synergistic effect, probes ABT-Gal-1 and ABT-Gal-2 exhibited faint background signals but dramatically enhanced near-infrared fluorescence upon exposure toward β-galactosidase (β-gal). Notably, both probes could be accessible in aqueous media. Preferably, the fluorescence intensity of ABT-Gal-1 exhibited a satisfactory linear relationship in the wide concentration range of 0.5–30 U/L with a very low detection limit to 0.24 U/L. Further, by monitoring endogenous β-gal in ovarian cancer cells, a precious merit of resistance to multiple washes was identified, which was facilitated by the nanoaggregate formation in situ upon activation. This work proposes a guiding strategy for manipulating various lighting mechanisms to develop attractive probes and, equally importantly, it provides an ideal practical tool for elucidating the biological role of β-gal in biological scenarios.

Abstract:Developing a ratiometric H2S fluorescent probe with fast response is of great importance for studying the H2S physiology. Herein, two hemicyanine-based H2S probes were constructed; the one with a propanoic acid group (CouPa) showed poor sensitivity while the other one with theN,N-diethylpropionamide moiety (CouDE) exhibited distinctly improved performance. CouDE showed the ability to detect mitochondrial H2S level fluctuation, which was triggered by alteration of CBS enzyme activity. Moreover, endogenous H2S change in solid tumours was monitored using CouDE.

Abstract:We have developed a novel colorimetric and ratiometric fluorescence probe for the selective and sensitive monitoring of hydrogen sulfide based on a dicyanoisophorone platform. An excellent linear relationship of fluorescence intensity ratio (I-637/I-558) (R-2 = 0.9867) versus hydrogen sulfide concentration in the range of 1-12 mu M was obtained. This probe exhibited a remarkable fluorescence response to hydrogen sulfide over other physiological thiols or biological species, which fluoresces in the red region with a large Stokes shift (172 nm). This probe was successfully utilized to monitor H2S under in vitro physiological conditions and for imaging H2S in living cells and living zebrafish in vivo.
