Abstract:ABSTRACT: Poor physicochemical properties like solubility, dissolution profile, stability, etc. of drugs remain as an obstacle to formulation development. In the last two decades, pharmaceutical cocrystal gaining lots of interest from researchers as these physicochemical, mechanical, and pharmacokinetic properties can be altered without compromising the therapeutic effect. The multi-component systems wherein one component is essentially the Active Pharmaceutical Ingredient and another may be either one more drug or coformer are termed as pharmaceutical cocrystal. Cocrystal is a novel, promising, and emerging approach to encountering such problems and improving the performance of pharmaceuticals. The present article describes the types, rationale of design, synthesis methods, and characterization of pharmaceutical cocrystals. In addition to this, challenges before and after synthesizing cocrystal along with currently approved formulations based on the pharmaceutical cocrystal approach and cocrystals under clinical investigation are highlighted.

Abstract:Pharmaceutical cocrystals are multicomponent systems in which at least one component is an active pharmaceutical ingredient and the others are pharmaceutically acceptable ingredients. Cocrystallization of a drug substance with a coformer is a promising and emerging approach to improve the performance of pharmaceuticals, such as solubility, dissolution profile, pharmacokinetics and stability. This review article presents a comprehensive overview of pharmaceutical cocrystals, including preparation methods, physicochemical properties, and applications. Furthermore, some examples of drug cocrystals are highlighted to illustrate the effect of crystal structures on the various aspects of active pharmaceutical ingredients, such as physical stability, chemical stability, mechanical properties, optical properties, bioavailability, sustained release and therapeutic effect. This review will provide guidance for more efficient design and manufacture of pharmaceutical cocrystals with desired physicochemical properties and applications. Abstract:Pharmaceutical co-crystals are novel class of pharmaceutical substances, which possess an apparent probability of advancement of polished physical properties offering stable and patentable solid forms. These multi-component crystalline forms influence pertinent physicochemical parameters like solubility, dissolution rate, chemical stability, physical stability, etc. which in turn result in the materials with superior properties to those of the free drug. Co-crystallization is a process by which the molecular interactions can be altered to optimize the drug properties. Co-crystals comprise a multicomponent system of active pharmaceutical ingredient (API) with a stoichiometric amount of a pharmaceutically acceptable coformer incorporated in the crystal lattice. By manufacturing pharmaceutical co-crystals, the physicochemical properties of a drug can be improved thus multicomponent crystalline materials have received renewed interest in the current scenario due to the easy administration in the pharmaceutical industry. There is an immense amount of literature available on co-crystals. However, there is a lack of an exhaustive review on a selection of coformers and regulations on co-crystals. The review has made an attempt to bridge this gap. The review also describes the methods used to prepare co-crystals with their characterization. Brief description on the pharmaceutical applications of co-crystals has also been incorporated here. Efforts are made to include reported works on co-crystals, which further help to understand the concept of co-crystals in depth.

Abstract:Most of the Active Pharmaceutical Ingredients (APIs) are typically formulated and administered to patients in oral solid dosage forms due to ease of administration, patient compliance and cost effectiveness. Poor water solubility, low permeability and low bioavailability of APIs are major hurdles in development of oral solid dosage forms. In recent years, cocrystal development has evolved as a feasible approach for enhancing the solubility and bioavailability of poorly soluble drugs. Crystal engineering strategies have been asserted to enhance the likelihood of discovering new solid forms of an API. A pharmaceutical cocrystal is made up of two basic components, an API and a harmless material known as a coformer in stoichiometric ratio. Cocrystallization of an API with a pharmaceutically acceptable coformer can improve the physical characteristics of the API, such as solubility, hygroscopicity, and compaction behavior, without affecting the API's pharmacological efficacy. This review article offers a comprehensive overview of pharmaceutical cocrystals, their physiochemical characteristics, and methods of preparation, with an emphasis on cocrystal screening and cocrystal characterization. The review also included recent FDA and EMA guidance on pharmaceutical cocrystals as well as an outline of multidrug cocrystals. Abstract:Pharmaceutical cocrystals are still gaining the interest of the researchers due to their potential to alter physicochemical, mechanical, and pharmacokinetic properties of active pharmaceutical ingredients without negotiating therapeutic action. The diverse new applications of cocrystals, like taste masking, reduced toxicity, patenting opportunities, commercial potential, etc. act as driving force to the rising interest of the pharmaceutical industries. Initially, cocrystals from the view of regulatory authorities, design strategies, cocrystal preparation in brief with special emphasis on scalable and solvent-free hot melt extrusion method, and practical guide to characterization have been provided. The special focus has been given to the biopharmaceutical attributes of the cocrystal. Finally, challenges before and after cocrystal preparation are presented in this review along with some commercial examples of the cocrystals.

Abstract:As a low-risk, low-cost, but high-reward route, cocrystallization of drugs with appropriate coformers is applied to improve the physiochemical and biopharmaceutical properties of drugs. Currently, most researchers concentrate their efforts on the preparation, characterization, and improvement of physicochemical properties of pharmaceutical cocrystals. On the contrary, the biological study of pharmaceutical cocrystals has not attracted wide attention of researchers. In this review, we have focused on recent advances reporting the biological studies of pharmaceutical cocrystals. The covered areas consist of the solubility and permeability, the pharmacokinetics study, metabolism and distribution, pharmacodynamics research, and the toxicological evaluation of pharmaceutical cocrystals. Besides, discussions have been made on the in vivo-in vitro correlations for pharmaceutical cocrystals, the enhancement of efficiency and reduction of toxicity for pharmaceutical cocrystals, and the interaction between APIs and coformers in pharmaceutical cocrystals and marketed pharmaceutical cocrystals as well as their biological studies. At the same time, some problems such as the amount of animal samples, the number and distribution of blood sampling points, investigation on the pharmacokinetics of physical mixtures containing APIs and coformers, and the consideration of species differences should be taken into account. Although pharmaceutical cocrystals face some challenges in clarifying the characteristics of metabolism and distribution, revealing potential pharmacological mechanism, and evaluating safety, cocrystal engineering is still considered a green and promising approach to developing valuable new drugs.

Abstract:The majority of the active pharmaceutical components have issues with poor solubility, bioavailability, chemical stability, and moisture absorption. Pharmaceutical crystals are a reliable way to change the aforementioned physicochemical properties of drugs without changing their pharmacological behaviour. However, the success of these approaches depends on the physical and chemical properties of the molecules being developed. The development of drug products with superior physicochemical characteristics, such as melting point, tablet ability, solubility, stability, bioavailability, and permeability, while preserving the pharmacological properties of the active pharmaceutical ingredient is greatly facilitated by co-crystallization of drug substances. All the components of a cocrystal, which is a stoichiometric homogeneous multicomponent system connected by non-covalent interactions, are solid under ambient conditions.

Abstract:Background: The poor water solubility of an active pharmaceutical ingredient leads to a lower dissolution profile that in turn results in poor bioavailability of drugs. Various approaches like solid dispersion, nano-technology, complexation, and micronization techniques, etc. are frequently used by pharmaceutical researchers to overcome these issues. In this context, crystal engineering emerges as a viable technique. Objective: This review endeavors to cover the latest developments in the field of solubility enhancement using crystal engineering techniques. Methods: Extensive literature survey was conducted in order to gain information on the past and present developments in the field of crystal engineering. Results: In the co-crystallization process, the API and coformer interact with each other in a fixed stoichiometric ratio. The backbone of co-crystals is structurally repeating units called supramolecular synthons. These synthons provide the flexibility of transfer from one co-crystal system to another, making crystal engineering a viable approach for physicochemical property modification. Further, the availability of a large number of food and drug grade coformers with a diverse functional group and a range of preparation methods provide an excellent opportunity for tuning up desired physicochemical properties of an API. Conclusion: This review focuses on the latest developments in the field of crystal engineering in the context of screening, preparation methods, characterization, and their application in the pharmaceutical field. Also, the concern over scale-up and regulatory guidelines are covered.

Abstract:Abstract A growing number of co‐crystals in the literature are proof of how significant the co‐crystallization concept has become. Co‐crystallization enhances physicochemical properties through the formation of intermolecular interactions between a drug and a co‐former. A co‐crystal is a single crystalline material consisting of at least two molecular components solid at room temperature and present in a definite stoichiometric ratio. Pharmaceutical co‐crystals consist of the active pharmaceutical ingredient and the co‐former selected from generally regarded as safe (GRAS) list of the United State Food and Drug Administration. Co‐crystal formation requires an understanding of a drug target, a proper choice of a co‐former and is only achieved experimentally after several trials. Other beneficial co‐crystallization outcomes include binary eutectics, solid dispersions, amorphous forms, etc. Several key issues including design strategies, co‐former selection, and co‐crystallization methods; tradition and newly synthetic methods that are more efficient and suitable for large scale have been briefly described. The co‐crystal preference is demonstrated with a particular emphasis on multidrug co‐crystals and their contribution to the drug combination strategies used for the treatment and management of drug resistance and adverse side effects in serious medical conditions that require the administration of high doses such as HIV/AIDS, tuberculosis, and others.

Abstract:BACKGROUND The development of solid drug dosage form and food ingredients is constrained by their low solubility, low dissolution, low bioavailability and poor physicochemical properties. Formation of cocrystal is a novel and promising method to enhance and improve the properties of materials without breaking the covalent bonds. METHODS The goal of this review is to summarize the cocrystals and their applications in the field of Active Pharmaceutical Ingredients (APIs) and food ingredients (AFIs), mainly on the effective improvements of APIs' and AFIs' pharmacokinetic, physicochemical and mechanical properties by the formation of cocrystals. RESULTS After years of research and development on cocrystals in the area of pharmaceutical and food industries, significant progress has been made. Formation of cocrystal is an efficient method for improving the solubility, dissolution rate, permeability and in vivo bioavailability of APIs and AFIs, as well as for enhancing stability and mechanical properties. CONCLUSION Cocrystals exhibit complex structures which can conspicuously affect the physical and chemical properties of original substance, with good clinical performance and outstanding stability during processing and storage.

Abstract:Cocrystals are long known yet recently applied molecular entities. In the past decade, a significant potential has been demonstrated by these novel solids in terms of modifying the physicochemical and pharmacokinetic properties of drugs. Thus far, publications have outlined various aspects of cocrystals at the molecular level concentrating on their design, growing techniques, and physicochemical characterizations. However, to take cocrystals from bench to bedside, they have to be incorporated into suitable formulations notwithstanding that the attention paid to cocrystal formulations is so diminutive. This is the first systematic review based exclusively on cocrystals formulation. In this contribution, the impact of cocrystals on essential pharmaceutical properties is glanced at before shining the spotlight on cocrystals formulation. The findings suggest that preformulation characteristics play a significant role, however, after which a number of approaches are desired in order to develop successful cocrystal formulations. It further highlights the main hurdles encountered with cocrystals formulation and other challenges to the transformation of cocrystals into viable medicines to have the full picture. There are marketed cocrystal products now, and it can be said it is only a matter of time before cocrystals are added to the main selection toolbox alongside salts for developing medicinal products.

Abstract:Cocrystals, a long known but understudied class of crystalline solids, have attracted interest from crystal engineers and pharmaceutical scientists in the past decade and are now an integral part of the preformulation stage of drug development. This is largely because cocrystals that contain a drug molecule, pharmaceutical cocrystals, can modify physicochemical properties without the need for covalent modification of the drug molecule. This review presents a brief history of cocrystals before addressing recent advances in design, discovery and development of pharmaceutical cocrystals that have occurred since an earlier review published in 2004. We address four aspects of cocrystals: nomenclature; design using hydrogen-bonded supramolecular synthons; methods of discovery and synthesis; development of pharmaceutical cocrystals as drug products. Cocrystals can be classified into molecular cocrystals (MCCs) that contain only neutral components (coformers) and ionic cocrystals (ICCs), which are comprised of at least one ionic coformer that is a salt. That cocrystals, especially ICCs, offer much greater diversity in terms of composition and properties than single component crystal forms and are amenable to design makes them of continuing interest. Seven recent case studies that illustrate how pharmaceutical cocrystals can improve physicochemical properties and clinical performance of drug substances, including a recently approved drug product based upon an ICC, are presented.

Abstract:Cocrystallization is an emerging approach for improving physico-chemical characteristics of an active pharmaceutical ingredient (API) for instance dissolution rate, solubility, stability in addition to mechanical properties without affecting their therapeutic activity. It is of great importance when other approaches like salt or polymorph formation do not encounter the estimated targets. In this review article, an outline of pharmaceutical cocrystals will be presented, with highlighting on factors affecting cocrystallization which include ∆pKa, donors and acceptors hydrogen bonds, molecular recognition point, synthon forming functional groups flexibility, dicarboxylic acid coformers carbon chain length and solvent effect, as well as and the methods for cocrystal preparation. Abstract:Pharmaceutical cocrystals, owing to their manifold applications, are acting as bridge between drug discovery and pharmaceutical product development. The ability to scale up pharmaceutical cocrystals through continuous manufacturing approaches offers superior and economic pharmaceutical products. Moreover, cocrystals can be an aid for the nanoparticulate systems to solve the issues related to scale-up and cost. Cocrystals grabbed attention of academic researchers and pharmaceutical scientist due to their potential to target various diseases like cancer. The present review is mainly focussed on the diverse and comprehensive applications of pharmaceutical cocrystals in drug delivery including solubility and dissolution enhancement, improvement of bioavailability of drug, mechanical and flow properties of active pharmaceutical ingredients, controlled/sustained release and colour tuning of API. Besides, phytochemical based cocrystals, multi-drug cocrystals and cocrystals for tumour therapy have been discussed in this review. Numerous approaches like nanotechnology, solid dispersion technique, micronization techniques, etc were aimed by scientists in the past to resolve this issue, but still not enough to get the desired outcomes. : Key focus of this review is the study of the advantages and disadvantages of cocrystallization, nanotechnology and solid dispersions drug delivery techniques and the benefits of using cocrystallization techniques over the above-mentioned techniques. : Various parameters including pharmaceutical, pharmacological and toxicological effects related to these mentioned drug delivery systems have been compared. Their advantages and disadvantages have been elaborated. : For drug delivery purpose, the cocrystallization process has numerous advantages over nanotechnology and solid dispersions drug delivery techniques discussed in the text. Cocrystallization is a newer technique that can modify the physicochemical and pharmaceutical properties of active pharmaceutical ingredients (API) with low solubility, low stability or sensitivity toward environmental hazards like temperature, moisture or photostability issues. During cocrystallization, the drug and the coformer interact with each other non-covalently in a fixed stoichiometric ratio. Abstract:Drug stability plays a significant role in the pharmaceutical industry from early-phase drug discovery to product registration as well as the entire life cycle of a product. Various formulation approaches have been employed to overcome drug stability issues. These approaches are sometimes time-consuming which ultimately affect the timeline of the product launch and may further require formulation optimization steps, affecting the overall cost. Pharmaceutical cocrystal is a well-established route to fine tune the biopharmaceutical properties of drugs without covalent modification. This article highlights the role of cocrystallization in mitigating the stability issues of challenging drug molecules. Representative case studies wherein the drug stability issue is addressed through pharmaceutical cocrystals have been discussed briefly and are summarized in tabular form. The emphasis has been made on the structural information of cocrystals and understanding the mechanism that improves the stability of the parent drug through cocrystallization. Besides, a guided strategy has been proposed to modulate the stability of drug molecules through cocrystallization approach. Finally, the stability concern of fixed-dose or drug combinations and the challenges associated with cocrystals are also touched.

Abstract:Many active ingredients from traditional Chinese medicine (TCM) exhibit poor pharmaceutical properties (e.g., low aqueous solubility, slow dissolution, low bioavailability, poor stability, and high hygroscopicity, etc.), which are closely associated with various formulation performance issues. Cocrystals or coamorphous systems, incorporating drug and guest molecules into a homogeneous single crystalline or amorphous phase, have been shown to offer potential advantages in enhancing pharmaceutical properties of TCM. This review outlines the definition, classification, preparation methods, characterization techniques, and pharmaceutical properties (such as solubility, dissolution, stability, mechanical properties, permeability, and in vivo performance) of cocrystals and coamorphous systems. More insights into the formation mechanisms of cocrystals and coamorphous systems, the identification of single-phase homogeneous coamorphous systems, and the characterization of their intermolecular interactions between components are covered. Latest research studies on the delivery of TCM via cocrystallization and coamorphization technologies are also summarized. The mechanisms of enhancement in druggability-related properties through cocrystals and coamorphous systems are further discussed. In particular, several common kinetic solubility behaviors of cocrystal and coamorphous systems, including "spring and parachute" and "spring and hover" models, are deeply understood, which will contribute to the maintenance of high supersaturation and in vivo bioavailability.

Abstract:Pharmaceutical cocrystals represent an emergent class of successful materials designed based on crystal engineering principles, in which the limitations of the extant drugs are addressed. Though the number of newly reported cocrystals is increasing exponentially, the scale-up of the cocrystallization process has not received enough attention. Solution cocrystallization techniques offer scalable processes that can utilize conventional crystallization machinery and process analytical technologies. The presence of multiple components and competing kinetics of the starting materials as well as the cocrystal make the cocrystallization process harder to study and control. This perspective emphasizes the importance of multicomponent phase diagrams for identifying operating regions in solution cocrystallization and discusses the effects of various operating parameters on the process kinetics. It also covers the intermolecular interactions involved in cocrystal development, cocrystallization methods, and quality control in pharmaceuticals. Furthermore, the role of the Design of Experiment and Quality by Design in scaling up the cocrystallization process efficiently is highlighted. A systematic decision tree has been proposed for scaling up the production of promising cocrystals using solution-based techniques, which can effectively address the current challenges in large-scale cocrystal production and help bring these potential materials to the market in the near future.

Abstract:Pharmaceutical cocrystals formed from active pharmaceutical ingredients(APIs) and coformers through noncovalent molecular interaction such as hydrogen bonding.It can improve the physicochemical properties and bioavailability of APIs so the pharmaceutical cocrystals have attracted much attention in pharmaceutical field in recent years.The preparation of cocrystals of amides,carboxylic acids,alcohol-phenols and heterocycles as APIs and their effect on physicochemical properties and bioavailability of APIs were classifiedly reviewed so as to provide a reference for the application of pharmaceutical cocrystals in pharmaceutical field.
