Abstract:Background Hydrogels, a type of three-dimensional (3-D) crosslinked network of polymers containing a high water concentration, have been receiving increasing attention in recent years. Self-healing hydrogels, which can return to their original structure and function after physical damage, are especially attractive. Some self-healable hydrogels have several kinds of properties such as injectability, adhesiveness, and conductivity, which enable them to be used in the manufacturing of drug/cell delivery vehicles, glues, electronic devices, and so on. Main Body This review will focus on the synthesis and applications of self-healing hydrogels. Their repair mechanisms and potential applications in pharmaceutical, biomedical, and other areas will be introduced. Conclusion Self-healing hydrogels are used in various fields because of their ability to recover. The prospect of self-healing hydrogels is promising, and they may be further developed for various applications.

Abstract:Self-healing hydrogels have emerged as the most promising alternatives to conventional brittle hydrogels used in the biomedical field due to the features of long-term stability and durability. While, the incompatibility between the fast self-healing property and enough mechanical strength of hydrogels remains a challenge. Therefore, hydrogels that possess not only mechanical toughness but also autonomous self-healing capacity are becoming sought after. This review presents a comprehensive summary of the latest self-healing mechanisms. Specifically, we review various systems based on dynamic bonds, ranging from dynamic covalent bonds to noncovalent bonds. Additionally, this review presents different characterization methods for self-healing hydrogels, along with highlighting their potential applications in the biomedical field, such as tissue engineering, drug delivery carriers, cell therapy, and wound dressing. Furthermore, this review aims to provide valuable guidance for constructing diverse self-healing hydrogels with tailored functions.

Abstract:Although considered promising materials for assisting organ regeneration, few hydrogels meet the stringent requirements of clinical translation on the preparation, application, mechanical property, bioadhesion, and biocompatibility of the hydrogels. Herein, we describe a facile supramolecular approach for preparing gelatin hydrogels with a wide array of desirable properties. Briefly, we first prepare a supramolecular gelatin macromer via the efficient host-guest complexation between the aromatic residues of gelatin and free diffusing photo-crosslinkable acrylated β-cyclodextrin (β-CD) monomers. The subsequent crosslinking of the macromers produces highly resilient supramolecular gelatin hydrogels that are solely crosslinked by the weak host-guest interactions between the gelatinous aromatic residues and β-cyclodextrin (β-CD). The obtained hydrogels are capable of sustaining excessive compressive and tensile strain, and they are capable of quick self healing after mechanical disruption. These hydrogels can be injected in the gelation state through surgical needles and re-molded to the targeted geometries while protecting the encapsulated cells. Moreover, the weak host-guest crosslinking likely facilitate the infiltration and migration of cells into the hydrogels. The excess β-CDs in the hydrogels enable the hydrogel-tissue adhesion and enhance the loading and sustained delivery of hydrophobic drugs. The cell and animal studies show that such hydrogels support cell recruitment, differentiation, and bone regeneration, making them promising carrier biomaterials of therapeutic cells and drugs via minimally invasive procedures.
A Novel Rapidly Self-healing Host-guest Supramolecular Hydrogel with High Mechanical Strength and Excellent Biocompatibility.

Abstract:It is still a challenge to achieve both excellent mechanical strength and biocompatibility in hydrogels. In this study, we exploited two interactions to form a novel biocompatible, slicing-resistant, and self-healing hydrogel. The first was molecular host-guest recognition between a host (isocyanatoethyl acrylate modified -cyclodextrin) and a guest (2-(2-(2-(2-(adamantyl-1-oxy)ethoxy)ethoxy)ethoxy)ethanol acrylate) to form three-arm host-guest supramolecules (HGSMs), and the second was covalent bonding between HGSMs (achieved by UV-initiated polymerization) to form strong cross-links in the hydrogel. The host-guest interaction enabled the hydrogel to rapidly self-heal. When it was cut, fresh surfaces were formed with dangling host and guest molecules (due to the breaking of host-guest recognition), which rapidly recognized each other again to heal the hydrogel by recombination of the cut surfaces. The smart hydrogels hold promise for use as biomaterials for soft-tissue repair.

Abstract:Self-healing hydrogels can heal themselves on the damaged sites, which opens up a fascinating way for enhancing lifetimes of materials. Polypeptide/poly(amino acid) is a class of polymers in which natural amino acid monomers or derivatives are linked by amide bonds with a stable and similar secondary structure as natural proteins (α-helix or β-fold). They have the advantages of nontoxicity, biodegradability, and low immunogenicity as well as easy modification. All these properties make polypeptides extremely suitable for the preparation of self-healing hydrogels for biomedical applications. In this review, we mainly focus on the progress in the fabrication strategies of polypeptide-based self-healing hydrogels and their biomedical applications in the recent 5 years. Various crosslinking methods for the preparation of polypeptide-based self-healing hydrogels are first introduced, including host-guest interactions, hydrogen bonding, electrostatic interactions, supramolecular self-assembly of β-sheets, and reversible covalent bonds of imine and hydrazone as well as molecular multi-interactions. Some representative biomedical applications of these self-healing hydrogels such as delivery system, tissue engineering, 3D-bioprinting, antibacterial and wound healing as well as bioadhesion and hemostasis are also summarized. Current challenges and perspectives in future for these “smart” hydrogels are proposed at the end .

Abstract:A hydrogel is a three-dimensional structure that holds plenty of water, but brittleness largely limits its application. Self-healing hydrogels, a new type of hydrogel that can be repaired by itself after external damage, have exhibited better fatigue resistance, reusability, hydrophilicity, and responsiveness to environmental stimuli. The past decade has seen rapid progress in self-healing hydrogels. Self-healing hydrogels can automatically self-repair after external damage. Different strategies have been proposed, including dynamic covalent bonds and reversible noncovalent interactions. Compared to traditional hydrogels, self-healing gels have better durability, responsiveness, and plasticity. These features allow the hydrogel to survive in harsh environments or even to be injected as a drug carrier. Here, we summarize the common strategies for designing self-healing hydrogels and their potential applications in clinical practice.

Abstract:Biocompatible self-healing hydrogels are new-generation smart soft materials that hold great promise in biomedical fields. Chitosan-based self-healing hydrogels, mainly prepared via dynamic imine bonds, have attracted broad attention due to their mild preparation conditions, excellent biocompatibility, and self-recovery ability under a physiological environment. In this review, we present a comprehensive overview of the design and fabrication of chitosan-based self-healing hydrogels, and summarize their biomedical applications in tissue regeneration, customized drug delivery, smart biosensors, and three/four dimensional (3D/4D) printing. Finally, we will discuss the challenges and future perspectives for the development of chitosan-based self-healing hydrogels in the biomedical field.

Abstract:Self-healing hydrogels possess an ability to recover their functionality after experiencing damage by regenerating cross-links. The main challenge in making self-healing hydrogels based on host-guest (HG) interactions is their limited mechanical strength, which can be solved using beta-cyclodextrin dimers (β-CDsD). Here, β-CDsD as a host cross-linker was used to increase the mechanical property of the HG interactions. Alginate with acceptable biocompatibility was modified by dopamine (ALG-DOP) and employed as a guest polymer. Self-healing hydrogel was developed between them, and Ag nanoparticles were added to create an antibacterial activity. Dopamine with appropriate size and suitable adhesiveness established HG interactions with β-CDsD, and cells were able to grow well on hydrogel. This hydrogel showed an impressive self-healing capability <5 min. These hydrogels revealed a respectable porosity from 15 to 55 μm essential for exchanging the substances required for cell growth and cell waste elimination. Biocompatibility was investigated against NIH 3 T3 fibroblasts cells, and the results showed that the cells grew well. The in vitro release of curcumin from the hydrogel was examined in PBS at pH of 7.4. The hydrogel can be a perfect candidate for controlled drug release, and wound-dressing due to self-healing property, antibacterial activity, adhesion, and biocompatibility.

Abstract:Hydrogels as semi-solid materials with highly tunable mechanical properties are promising materials in numerous fields. The emergence of self-healing hydrogels, with the ability to recover their original structure post-fracture, has further expanded their applications in flexible devices, wound healing, and implant materials. Biopolymers have the advantages of biocompatibility, biodegradability, and environmental sustainability, making the self-healing hydrogels from biopolymers more promising materials in biomedical applications. In this paper, we started from the dynamic bonds that used to realize the self-healing properties of biopolymer-based hydrogels, and discussed their advantages and disadvantages. The construction mechanisms, self-healing properties of the hydrogels from different biopolymers were then introduced separately. We also reviewed the their potential applications, and discussed their challenges and prospects.

Abstract:The cucurbit[n]uril supramolecular hydrogels are driven by weak intermolecular interactions, of which exhibit good stimuli responsiveness and excellent self-healing properties. According to the composition of the gelling factor, supramolecular hydrogels comprise Q[n]-cross-linked small molecules and Q[n]-cross-linked polymers. According to different driving forces, hydrogels are driven by the outer-surface interaction, the host-guest inclusion interaction, and the host-guest exclusion interaction. Host-guest interactions are widely used in the construction of self-healing hydrogels, which can spontaneously recover after being damaged, thereby prolonging their service life. The smart Q[n]s-based supramolecular hydrogel composed is a kind of adjustable and low-toxicity soft material. By designing the structure of the hydrogel or modifying the fluorescent properties, etc., it can be widely used in biomedicine. In this review, we mainly focus on the preparation of Q[n]-based hydrogels and their biomedical applications including cell encapsulation for biocatalysis, biosensors for high sensitivity, 3D printing for potential tissue engineering, drug release for sustained delivery, and interfacial adhesion for self-healing materials. In addition, we also presented the current challenges and prospects in this field.

Abstract:Self-healing polymers, which can spontaneously recover themselves after being ruptured, result in enhanced lifetimes for materials and open up a fascinating direction in material science. Macrocycle-based host-guest interactions, one of the most crucial non-covalent interactions, play a key role in self-healing material fabrication. This review aims to highlight the very recent and important progress made in the area of self-healing polymer materials by focusing on cyclodextrins (CDs), crown ethers, cucurbit [n] urils (CBs), calix[n] arenes and pillar[n] arenes with special guest groups and tailored structures. In addition, we also propose future research directions and hope that this review can in a way reflect the current situation and future trends in this developing area.

Abstract:The recognition-directed host-guest interaction is recognized as a valuable tool for creating supramolecular polymers. Functional hydrogels constructed through the dynamic and reversible host-guest complexation are endowed with a great many appealing features, such as superior self-healing, injectability, flexibility, stimuli-responsiveness and biocompatibility, which are crucial for biological and medicinal applications. With numerous topological structures and host-guest combinations established previously, recent breakthroughs in this area mostly focus on further improvement and fine-tuning of various properties for practical utilizations. The current contribution provides a comprehensive overview of the latest developments in host-guest supramolecular hydrogels, with a particular emphasis on the innovative molecular-level design strategies and hydrogel formation methodologies targeting at a wide range of active biomedical domains, including drug delivery, 3D printing, wound healing, tissue engineering, artificial actuators, biosensors, etc. Furthermore, a brief conclusion and discussion on the steps forward to bring these smart hydrogels to clinical practice is also presented.

Abstract:Conductive hydrogels have generated great interest in biomedical and electrical fields. However, conventional conductive hydrogels usually lack self-healing properties, which might be unfavorable for their application. Conductive self-healing hydrogels with excellent performance for applications in the biomedical and electrical fields are growing in number. In this review paper, the progress related to conductive self-healing hydrogels is summarized. The self-healing mechanism is classified to demonstrate the design and synthesis of conductive self-healing hydrogels and their applications in tissue engineering, wound healing, electronic skin, sensors and self-repaired circuits are presented and discussed. The future development of conductive self-healing hydrogels and problems that need to be solved are also described.

Abstract:Self-healing hydrogel is a new generation smart material that has drawn great attention among researchers from multiple cutting edge areas. Self-healing hydrogels are continuously enriching our knowledge in developing new materials and changing our lives. In the recent decades, thanks to numerous dedicated researchers, series of self-healing hydrogels have been explored through dynamic chemistry and supramolecular interactions, which have been applied in interdisciplinary areas for controlled release of drugs, 3D cell culture, and scaffolds of tissue engineering, etc. Self-healing hydrogels can be injected as solid gels instead of liquid precursor, then self-heal in situ. Thus, self-healing hydrogels have been utilized as a new type of injectable carrier to deliver drugs. This method effectively avoids unwanted cargo loss throughout the cycling and dramatically improves the delivery efficiency of drugs. Thus, self-healing hydrogels have been studied in recent research as great therapeutic carrier for tumour therapy and wound-healing. This review aims to summarize some recent research progresses in the preparation of self-healing hydrogels based on different dynamic covalent bonds. The bio-medical applications of self-healing hydrogels including tumour therapy and wound-healing have also been introduced. The future development and challenges of dynamic chemistry based on self-healing hydrogels have also been discussed.

Abstract:Polymer hydrogel is a soft material with a three-dimensional network structure that can absorb and retain a large amount of water. Polymer hydrogels have good biocompatibility, mechanical properties and important application value in biomedical and bioengineering fields. Self-healing hydrogels are smart hydrogels that respond to external stimuli and repair their own damage. Compared with the traditional hydrogel, the self-healing hydrogel has the property of repairing damage, and received extensive attention in the scientific field in recent years. Dynamic chemistry-based self-healing hydrogels are novel self-healing hydrogels that can reshape three-dimensional network structures by dynamic covalent or non-covalent bonding to repair damage. The new selfhealing hydrogel can quickly repair its own damage and has good environmental adaptability, laying the foundation for the development of self-healing hydrogels as multifunctional new materials. The research progress of recent self-healing hydrogels based on dynamic chemistry was reviewed in this paper, especially focusing on the updated development on self-healing hydrogels based on hydrogen bonds, metal coordination interactions, host-guest interactions, ionic bonds, hydrophobic interactions, imine bonds/acylhydrazone bonds, borate bonds and disulphide bonds, and meanwhile, the problem of self-healing hydrogels was put forward, and the future direction of development was finally predicted.

Abstract:The novel self-healing hydrogel is a typical intelligent and multi-functional biomedical material. It is of great significance to resolve the challenge of repairing damages in flexible biomaterials, which could facilitate intelligent activity and efficiency in biomaterials. The present paper reviews the design strategies and performance of recently developed self-healing hydrogels and their applications in tissue engineering, including transplantation of neural stem cells, repair of the central nervous system, bone regeneration, regeneration of cartilage-bone tissue complex, cardiac cell therapy, as well as hemostasis. Such advances and applications promote the innovative design and fabrication of intelligent multifunctional hydrogels and offer new opportunities to increase the diversity of flexible self-healing materials. Such a synthesis of research and applications lays a foundation for further development of intelligent hydrogels for tissue engineering and their application in the biomedical field.

Abstract:Shear‐thinning and self‐healing hydrogels are being investigated in various biomedical applications including drug delivery, tissue engineering, and 3D bioprinting. Such hydrogels are formed through dynamic and reversible interactions between polymers or polypeptides that allow these shear‐thinning and self‐healing properties, including physical associations (e.g., hydrogen bonds, guest–host interactions, biorecognition motifs, hydrophobicity, electrostatics, and metal–ligand coordination) and dynamic covalent chemistry (e.g., Schiff base, oxime chemistry, disulfide bonds, and reversible Diels–Alder). Their shear‐thinning properties allow for injectability, as the hydrogel exhibits viscous flow under shear, and their self‐healing nature allows for stabilization when shear is removed. Hydrogels can be formulated as uniform polymer and polypeptide assemblies, as hydrogel nanocomposites, or in granular hydrogel form. Abstract:In modern times, biological materials have received a lot of attention due to their potentials in the treatment of body defects. Among biomaterials, hydrogels have a high advantage due to their multifunctional structures and behavior. However, the mechanical failure of conventional hydrogels under normal conditions remains a significant problem. To overcome this lesion, self-healing hydrogel materials have been created that have the property of self-healing and are minimally invasive for repairing biological defects in the whole body. In this review study, by combining the developments of recent articles in synthesis and discussion of clinical applications for repairing various types of body defects, the development of self-healing hydrogels is presented. By reviewing the characteristics and behaviors of healable hydrogels, the repair of different types of defects as well as their mechanism is presented. Some properties of polymers such as molecular mass, hydrophilicity, functional groups, intramolecular and extramolecular interactions can affect the properties of self-healing hydrogels. The review examines the development of smart self-healing hydrogel processes over the past 10 years, focusing primarily on their recent ability to repair tissue defects. This paper also presents various methods of synthesis and types of polymers and discusses clinical applications that facilitate the improvement of lesions and enhance organs function. The issues presented here may be of interest to researchers interested in improving self-healing hydrogels.

Abstract:In recent years, implantable biomaterials have attracted significant interest owing to their potentials for use in the therapy of physical defects and traumas. Among the implantable biomaterials, hydrogels have received increasing attention for their tunable structures and good rheological behavior. However, the mechanical failures of traditional gel materials during normal operation remain a serious issue. To overcome this problem, hydrogel materials with self-healing and injectable abilities have been developed, with their potential for autonomous self-recovery and minimally invasive implantation. In this paper, the progress of injectable self-healing hydrogels is presented by combining developments in the fundamental knowledge of polymer designs and discussions on the practical biomedical applications of the materials. The mechanisms of different types of self-healing hydrogels are introduced first and their performances are then discussed, followed by a review of the self-healing hydrogels with injectability. The applications of the injectable self-healing hydrogels are discussed in the final section. This paper provides an overview of the progress of a smart material, injectable self-healing hydrogel, during the past ten years and mainly focuses on its recent development. This paper presents developments in the fundamental knowledge in polymer designs and discussions on the practical biomedical application of the materials, which sheds more light on the advancement of injectable self-healing hydrogels. This paper should be of interest to the readers who are curious about the advances of injectable self-healing hydrogels.

Abstract:A supramolecular hydrogel was prepared through the host-guest interaction between beta-cyclodextrin (beta-CD) and ferrocene (Fc) with two polymers as pendant groups. Reversible gel-sol transition was observed with the alternative stimuli of a positive potential (or an oxidant) and a negative potential (or a reductant). The hydrogel not only had good self-healing ability due to the dynamic host-guest interaction, but also was prepared under mild conditions and could respond to moderate electrochemical stimuli. Good biocompatibility endowed the hydrogel with potential practical and real-life applications, especially in tissue engineering and drug release field.
