Abstract:OBJECTIVE A sensitive HPLC method based on post-column modification and fluorescence detection has been developed for determination of puerarin and daidzein in rat plasma.In order to investigate whether the pharmacokinetics of puerarin and daidzein was affected by the other drugs(Radix Salviae Miltiorrhizae and Rhizoma Chuanxiong)in Tongmai formulations,extensive study of the interactions between main drug and other drugs has been conducted.METHODS The pharmacokinetics behaviors of puerarin and daidzein in rat plasma were studied after orally administrated with extract of Pueraria Radix,extract of Pueraria Radix and Radix Salviae Miltiorrhizae,extract of Pueraria Radix and Rhizoma Chuanxiong,and Tongmai oral liquid,respectively.RESULTS Radix Salviae Miltiorrhizae and Rhizoma Chuanxiong had different effects on the absorption,distribution and elimination of Radix Puerariae.In Tongmai oral liquid group,the puerarin and daidzein showed the maximum plasma concentration and the lowest clearance rate of plasma.These changes of pharmacokinetics behaviors were the combined actions of Radix Salviae Miltiorrhizae and Rhizoma Chuanxiong.CONCLUSION The method is sensitive,fast,reliable and convenient,it may be helpful to certificate the rationality of the matching of the three herbs in Tongmai formulations.
