Abstract:The intramolecular hydroaminocarbonylation of alkenes is a compelling tool to rapidly access lactam, a privileged motif ubiquitous in natural products, pharmaceuticals, and agrochemicals. However, selective carbonylation to bridged polycyclic lactams with a lactam nitrogen at a bridgehead position is less explored. We herein report a modular palladium-catalyzed approach to perform a tandem hydrocarbonylative lactamization/Diels-Alder cycloaddition reaction with 2-vinyl aryl aldimines, alkenes, and CO, which offers convenient access to furnish the bridged polycyclic lactams in high yields with high selectivities.

Abstract:A simple palladium(II)-catalyzed intramolecular addition of vinylpalladium species to ketones initiated by the carbopalladation of alkynamides under mild conditions without a Pd(II)/Pd(0) redox system was developed. This cascade reaction provides a new approach for the synthesis of highly substituted α-alkylidene-β-hydroxy-γ-lactams.

Abstract:Palladium-catalyzed intramolecular cyclization of the alkynoic acids was studied which gave the five- and six-membered lactones in moderate yields. The reaction can be conducted simply by heating a toluene solution of alkynoic acids at 100 degrees C in the presence of catalytic amounts of Pd(PPh3)(4) and (o-tol)(3)P. The key for this transformation is the use of phosphines, instead of carboxylic acids, as an additive. Similar to our previously developed catalytic system the use of carboxylic acid, instead of (o-tol)(3)P, resulted into the exclusive formation of the diene.

Abstract:beta-Lactam compounds play a key role in medicinal chemistry, specifically as the most important class of antibiotics. Here, we report a novel one-step approach for the synthesis of alpha-(trifluoromethyl)-beta-lactams and related products from fluorinated olefins, anilines and CO. Utilization of an advanced palladium catalyst system with the Ruphos ligand allows for selective cycloaminocarbonylations to give diverse fluorinated beta-lactams in high yields.
