Abstract:Survivin is a newly discovered inhibitor of apoptosis proteins in recent years,which has a relationship with cell proliferation and cell division,it always expressed in malignant tumors and embryonic tissues but not expressed or weakly expressed in normal adult differentiated tissues.Although Endometriosis(EMs)is a benign disease,it has some malignant biological behaviors.Research suggests that survivin may plays an important role in the occurrence and development of EMs.In this paper,we reviewed its biological characteristics and the role in EMs.

Abstract:Objective To study the implication of a novel gene survivin,an inhibitor of apoptosis.Methods The expression of survivin mRNA was quantitatively determined by RT-PCR and survivin protein was examined immunohistochemically in 46 gastric cancer tissues and corresponding normal gastric tissues.Results Suvivin mRNA was determined in 32 6% gastric cancer while in 10 87% normal tissues.Suvivin protein expressed in 78% gastric cancer while in 30% normal tissues.There were significant differences (P0 005,P0 05) between gastric cancer and normal tissues.The expression of suvivin mRNA and protein was independent of sex,diameter of tumors,the position of tumor occurrence,gross type,tumor depth,the level of differentiation,vein invasion,lymph nodes metastasis,distant metastasis and TMN staging.Conclusions Suvivin gene may play a role in inhibiting apoptosis in gastric cancer and may function importantly in tumor production but not development.

Abstract:Objective:To study the expression of apoptosis inhibitor protein of Survivin in colorectal adenocarcinoma (CAC)and its correlation with clinicalpathologic parameters and prognosis and to examine the relationship between the Survivin expression and the expression of Bcl-2 and P53 in colorectal adenocarcinoma.Methods:115 cases of colorectal adenocarcinoma, by immnohistochemical staining,were examined survivin,Bcl-2,and P53.The cases were followed up for 5 years.Results: Survivin was detected in 74 of 115 cases(64.3%).No expression of survivin was observed in normal tissues.There was no correlation between Survivin immnoactivity and clinicalpathologic parameters(P＞0.05).Survivin expression was significantly higher in Bcl-2 positive patients than in Bcl-2 negative cases(88.9% versus 42.6%,p＜0.001),but was not associated with P53(P＞0.05). The 5-year survival rate for the padents with survivin positive was siginificantly lower than that for the patients with survivin negative(p=0.001).Conclusions:Examining surviving in CAC tissue is of significance for tumor patients' prognosis and targeted therapy based on antiapoptosis mechanism.

Abstract:The factor of antiapoptosis in cancer is recently identified as survivin. Survivin is prominently overexpressed in embryonic and fetal tissues, and various human cancers. It is potentially involved in both inhibition of apoptosis and control of cell division. Recently, survivin is found to be correlated with the genesis, progress, and prognosis of urological cancer. Abstract:Survivin holds significant importance as a member of the inhibitor of apoptosis protein (IAP) family due to its predominant expression in tumours rather than normal terminally differentiated adult tissues. The high expression level of survivin in tumours is closely linked to chemotherapy resistance, heightened tumour recurrence, and increased tumour aggressiveness and serves as a negative prognostic factor for cancer patients. Consequently, survivin has emerged as a promising therapeutic target for cancer treatment. In this review, we delve into the various biological characteristics of survivin in cancers and its pivotal role in maintaining immune system homeostasis. Additionally, we explore different therapeutic strategies aimed at targeting survivin.

The expression of survivin in human gastric carcinoma and precancerous lesions.

Abstract:Objective To investigate the expressions of survivin in gastric carcinomas and precancerous lesions. Methods Survivin mRNA expression was quantitatively determined by RT-PCR, and the protein contents were quantitatively determined by ELISA in 46 specimens of gastric carcinomas and 46 specimens of normal gastric tissues. Survivin protein expressions were also examined immunohistochemically in 46 specimens of gastric carcinoma tissues and 114 specimens of gastritis and precancerous lesion tissues. Results Survivin mRNA was expressed in 32.6% of gastric carcinomas (15/46), while it was expressed in 10.8% of neighboring normal tissues (P<0.05). The survivin protein contents in gastric carcinoma were significantly higher than those in normal tissue(P<0.05). There were no significant differences in survivin protein expression among superficial gastritis, atrophic gastritis, and intestinal metaplasia, but significant differences were detected among superficial gastritis, atrophic gastritis, intestinal metaplasia, gastric dysplasia, gastric carcinoma (P<0.05). The survivin mRNA expression, protein expression and the protein contents were not associated to gender of patient, the size, depth, location, gross type, differentiation, vein invasion, lymph nodes metastasis, distant metastasis, TNM staging of tumor (all P>0.05). Conclusion These data suggest that survivin may be involved in the inhibition of apoptosis in gastric carcinomas, and may be relevant to the genesis of gastric carcinoma, but irrelevant to the development and invasion of tumors.
Effects of Survivin Mrna Expression on Apoptosis and Proliferation and MDR-1 in Hepatocellular Carcinoma

Abstract:Objective To study the mRNA expression of survivin, a novel apoptosis inhibitor, and cell apoptosis and proliferation and MDR 1 in hepatocellular carcinoma (HCC) and investigate the effects of survivin mRNA expression on apoptosis and proliferation and MDR 1 in HCC. Methods RT PCR, TUNEL and SABC IHC were used to detect the expression of survivin mRNA and MDR 1 mRNA, cell apoptosis and the ki 67 expression, respectively, in samples from 31 cases of HCC. Meanwhile, the effects of survivin mRNA expression on cell apoptosis and proliferation and the relationship between survivin mRNA expression and MDR 1 mRNA expression were analyzed. Results The positive survivin mRNA expression was observed in 22 out of the 31 cases (71%). All the neighboring non tumor and cirrhotic tissues were completely negative for survivin mRNA expression. The apoptotic index (AI) was 0 4%～15% and the overall average was 4 24±3 92%. The average of AI was 4 64±4 30% in survivin mRNA positive cases and 3 36±2 98% in negative cases. There was no significant difference between the 2 groups of patients (P0 05). The ki 67 proliferation index (LI) was 2%～80% and the overall average was 26 21±14 9%. The average LI was 30 32±15 43% in the survivin mRNA positive cases and 17 56±9 44% in negative ones. The LI showed a significant positive correlation with the survivin mRNA expression (P0 05). The positive MDR 1 mRNA expression was 18/31 (58 05%) in HCC tissue. Amongst the 22 survivin mRNA positive cases, 15 were MDR 1 mRNA positive and 7 negative. As for the 9 survivin mRNA negative cases, 3 were MDR 1 mRNA positive and 6 negative. Conclusions Survivin may play an important role in the progression of HCC. There is no significant difference between survivin mRNA expression and AI. However, LI shows a significant positive correlation with the survivin mRNA expression. Though survivin is an apoptosis inhibiting agent, it does not significantly influence cell apoptosis in HCC, and it plays an important role in the progression and development of HCC by promoting cell proliferation. Survivin will be a potential new target of gene therapy in HCC. There is no significant correlation between survivin and MDR 1 expression. The formation of MDR in HCC is attributed to various kinds of pathway but not survivin.

Abstract:Objective:To explore the expression of survivin in endometrial cancer and its roles in regulating apoptosis of endometrial cancer cells.Methods:Immunohistochemistry was used to examine the expression of survivin in endometrial cancer specimens.RNA interference(RNAi) was applied to knock down survivin expression.The effect of RNAi was examined by RT-PCR and Western blotting.Cell apoptosis was detected by the FCM before and after RNAi.Western blotting was used to detect the expressions of caspase-3,caspase-8 and Bcl-2,which were closely associated with apoptosis.Results:The positive rate of survivin protein in endometrial cancer was obviously higher than those in atypical endometrium and normal endometrium(P0.05 and P0.01,respectively).RNAi effectively inhibited the expressions of survivin mRNA and survivin protein(P0.01).RNAi also induced cell apoptosis.Cell apoptosis rates in the two groups of RNAi were obviously higher than those in control groups(P0.01).Survivin RNAi also up-regulated the cleavage of caspase-3 and caspase-8.Conclusion:The aberrant expression of survivin may be closely related to the carcinogenesis of endometrial cancer,and inhibition of its expression may induce apoptosis of endometrial cancer cells.

Abstract:Pancreatic cancer is estimated to be the fourth most common cancer in men and fifth in women in the world and has poor prognosis. In recent years, more and more effort has been put on the relationship between pancreatic cancer and apoptosis. As a newly discovered inhibitor of apoptosis, survivin has drawn more attention. Strong evidence has shown that survivin is expressed in pancreatic cancer cells on frozen sections. Survivin increases in the development of pancreatic ductal adenocarcinoma and its expression can be a marker in evaluating the prognosis of pancreatic cancer patients. Abstract:Survivin was initially identified as a member of the inhibitor apoptosis (IAP) protein family and has been shown to play a critical role in the regulation of apoptosis. More recent studies showed that survivin is a component of the chromosome passenger complex and acts as an essential mediator of mitotic progression. Other potential functions of survivin, such as mitochondrial function and autophagy, have also been proposed. Survivin has emerged as an attractive target for cancer therapy because its overexpression has been found in most human cancers and is frequently associated with chemotherapy resistance, recurrence, and poor survival rates in cancer patients. In this review, we discuss our current understanding of how survivin mediates various aspects of malignant transformation and drug resistance, as well as the efforts that have been made to develop therapeutics targeting survivin for the treatment of cancer.

Clinical value of survivin and its underlying mechanism in ovarian cancer: A bioinformatics study based on GEO and TCGA data mining

Abstract:Objective: An increasing number of studies have confirmed that survivin (BIRC5) plays essential roles in ovarian cancer. Nevertheless, inconsistent or controversial results exist in some studies. In the present study, we sought to determine the clinical significance of survivin and its potential molecular pathways. Methods: The correlation between survivin (BIRC5) expression and diagnostic value, prognostic value and clinicopathological features was assessed by meta-analysis with more than 4000 patients from literature, GEO and TCGA. In addition, the potential molecular mechanism of survivin in ovarian cancer was also determined. Results: The pooled sensitivity and specificity were 0.71 (95%CI: 0.68-0.74) and 0.97 (95%CI: 0.94-0.98), respectively. The AUC of sROC was 0.8765. The results showed that there was also a significant relationship between survivin expression and poor overall survival (HR: 1.24, 95%CI: 1.14-1.35, p < 0.001), disease-free survival (HR: 1.53, 95%CI: 0.57-4.09, p < 0.001), as well as higher recurrence rate (HR: 1.11, 95%CI: 0.97-1.27). Moreover, survivin expression was also associated with tumor progression (cancerous vs. benign, OR: 11.29, 95%CI: 8.96-14.24, p < 0.001), TNM stage (III + IV vs. I + II, OR: 5.38, 95%CI: 4.16-6.97, p < 0.001), histological grades (G3 vs. G1 ∼ G2, OR: 4.36, 95%CI: 3.29-5.77, p < 0.001), and lymphatic metastasis (metastasis vs. non-metastasis, 3.35, 95%CI 2.36-4.75, p < 0.001). Bioinformatics analysis revealed the 50 most frequently altered neighboring genes of survivin in OC, and then Gene Oncology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis were conducted. GO analysis showed that these genes were related to signal conduction, cell cycle, apoptosis, and metabolism. KEGG pathways analysis indicated that these genes were primarily enriched in mitotic prometaphase, PLK1 signaling events and the regulation of glucokinase by the glucokinase regulatory protein. Conclusion: Survivin (BIRC5) expression might become a specific but low-sensitivity biomarker in ovarian cancer patients, and its presence indicated poor prognosis and worse TNM stages. This protein might function as an oncoprotein by influencing specific pathways involving the 50 genes identified herein. Additional studies are needed to confirm these results.

Abstract:Inhibitors of programmed cell death (apoptosis) aberrantly prolonging cell viability may contribute to cancer1 by facilitating the insurgence of mutations and by promoting resistance to therapy2. Despite the identification of several new apoptosis inhibitors related to bcl-22,3 or to the baculovirus IAP gene4–9, it is not clear whether apoptosis inhibition plays a general role in neoplasia. Here, we describe a new human gene encoding a structurally unique IAP apoptosis inhibitor, designated survivin. Survivin contains a single baculovirus IAP repeat and lacks a carboxyl-terminal RING finger. Present during fetal development, survivin is undetectable in terminally differentiated adult tissues. However, survivin becomes prominently expressed in transformed cell lines and in all the most common human cancers of lung, colon, pancreas, prostate and breast, in vivo. Survivin is also found in approximately 50% of high-grade non-Hodgkin's lymphomas (centroblastic, immunoblastic), but not in low-grade lymphomas (lymphocytic). Recombinant expression of survivin counteracts apoptosis of B lymphocyte precursors deprived of interleukin 3 (IL-3). These findings suggest that apoptosis inhibition may be a general feature of neoplasia and identify survivin as a potential new target for apoptosis-based therapy in cancer and lymphoma.

Abstract:Inhibitors of Apoptosis proteins (IAPs) were discovered through experiments aimed at rescuing apoptosis in insects. Classically associated with the inhibition of apoptosis, the IAP member Survivin also regulates cell cycle progression and is an essential component of the Chromosomal Passenger Complex (CPC), responsible for chromosomal segregation. Although undetectable in most adult tissues, Survivin is expressed in Adult Stem Cells (ASCs) and plays a crucial role in their maintenance. Survivin is overexpressed in most cancers, contributing to their clonal expansion. As a result, it has been proposed as a possible anticancer target for nearly two decades. In this discussion, we will explore the rationale behind Survivin as a therapeutic target, focusing on common cancer types such as carcinomas, sarcomas, and leukemias. We will delve into the modulation of Survivin by cancer pro-survival cell signaling, the association between SNPs and tumorigenesis, and its regulation by miRNAs. Finally, we will compare cell growth, clonogenic capacity, and apoptosis, along with different strategies for Survivin inhibition, including gene expression and protein activity modulation.

Abstract:Survivin, as a member of the inhibitor of apoptosis proteins (IAPs) family, acts as a suppressor of apoptosis and plays a central role in cell division. Survivin has been considered as an important cancer drug target because it is highly expressed in many types of human cancers, while it is effectively absent from terminally differentiated normal tissues. Moreover, survivin is involved in tumor cell resistance to chemotherapy and radiation. Preclinically, downregulation of survivin expression or function reduced tumor growth induced apoptosis and sensitized tumor cells to radiation and chemotherapy in different human tumor models. This review highlights the role of survivin in promoting cellular proliferation and inhibiting apoptosis and summarizes the recent advances in and challenges of developing small-molecule survivin inhibitors.

Abstract:Apoptosis is an active process of cellular self-destruction that is involved in normal development. Two kinds of anti-apoptosis genes p35 and iap have been identified in baculovirus genome. Both of them function at different point in the death pathway. Extensive studies have been made about the structures and function mechanisms of the genes and their products. This review highlights the recent advances in the researches of the genes and their function mechanisms and explores the possible utilizations in the future.

Abstract:Survivin, the smallest member of IAP (inhibitor of apoptosis) family, is a dual functional protein acting as a critical apoptosis inhibitor and key cell cycle regulator. Survivin is usually expressed in embryonic tissues during development and undetectable in most terminally differentiated tissues. Numerous studies demonstrate that survivin is selectively upregulated in almost all types of human malignancies and its overexpression positively correlates with poor prognosis, tumor recurrence, and therapeutic resistance. This differential expression of survivin in tumors and normal tissues draws a great interest to develop survivin-targeted therapy for cancer treatment. Nonetheless, the molecular mechanisms controlling survivin expression in malignant tumor cells have not been fully understood. While aberrant activation of receptor tyrosine kinases (RTKs) and the downstream signaling, such as PI-3K/Akt, MEK/MAPK, mTOR, and STAT pathways, have frequently been shown to upregulate survivin, recent data suggest that a class of noncoding RNAs, microRNAs (miRNAs) also play an important role in survivin dysregulation in human cancers. Here, we focus on survivin expression-regulated by specific miRNAs binding to the 3'-UTR of survivin mRNA, and summarize the latest advances on survivin-targeted therapy in clinical trials and the therapeutic potential of survivin-targeting miRNAs in cancer.

Abstract:Abstract Introduction: Survivin, a member of the inhibitor of apoptosis family, is upregulated in multiple cancers including ovarian cancer, but is rarely detectable in normal tissues. Therefore, it is a potential drug target. However, targeting survivin is still challenge and small molecule inhibitors failed in clinical trials. It's very important to understand how survivin contributes to tumor development. In this study, we investigated the role of survivin in ovarian cancer using ovarian cancer cell lines in vitro and mouse models in vivo and examined how survivin contributes to ovarian primary ovarian tumor growth and metastasis by examining epithelial to mesenchymal transition (EMT). Methods: To define the role of survivin in EMT in ovarian cancer cells, we established survivin knockout cell lines using lentiviral CRISPR/Cas9 nickase and pharmacologically inhibited survivin with a novel inhibitor MX106. We examined EMT markers, migration and invasion assays in survivin disrupted ovarian cancer cells using Transwell plates. We also assessed primary ovarian tumor growth and metastasis in an orthotopic ovarian cancer mouse models through intrabursal injecting survivin KO cell line and also treatment using MX106. Findings: We found that disruption of survivin expression inhibited EMT by attenuating the TGFβ pathway in ovarian cancer cells. Cell proliferation, migration, and invasion were significantly inhibited in survivin knockout or MX106 treatment in SKOV3 and OVCAR3 cell lines compared to control cells. Inhibition of survivin sensitized cell responses to paclitaxel treatment and overcame chemoresistance in vitro. Lentiviral CRISPR/Cas9 mediated survivin knockout or MX106 treatment efficiently inhibited primary ovarian tumor growth and metastasis in orthotopic ovarian cancer mouse models. Conclusions: Our data demonstrate that inhibition of survivin using either genetic knockout or a novel inhibitor MX106 suppresses primary ovarian tumor growth and metastasis by attenuating TGFβ pathway. MX106, as a novel survivin inhibitor effectively overcame chemoresistance compared to commercial YM155, which provides a novel approach for ovarian cancer therapy. Citation Format: Junming Yue.
