Abstract:The effects of plant growth regulators with different concentrations on the proliferation ofXingaopear in vitro was studied.The result showed that the suitable budding medium was 1/2MS + 6-BA 0.5 mg/L + IBA 0.2 mg/L + GA3 0.5 mg/L.The effective multiplication coefficient was up to 3.3.

Abstract:The effects of plant growth regulators(BA,NAA)and organic additives(potato homogenate,tomato homogenate,banana homogenate,peptone and yeast extract)on the growth,proliferation and differentiation of the rhizomes of Cymbidium goeringii in vitro were measured.The results showed that the optimum medium for formation of cluster shoots from the rhizomes was 1/2MS+0.1mg/L BA+0.5mg/L NAA+1.5g/L AC,and the optimum medium for proliferation of the rhizomes was 1/2MS+0.1mg/L BA+5.0mg/L NAA+1.5g/L AC.Except banana homogenate,all the organic additives promoted the growth and proliferation of the rhizomes,and among them peptone was the best.

Abstract:In order to study the plant growth regulators on rooting and transplanting of the strawberry vitreous,the vitreous of strawberry XingXiang is used as experimental material, and 1/2MS as base medium. On two conditions of light and 9 days dark at the beginning and light later, different IBA concentrations (0, 0.1,0.2 ,0.4,0.6 mg/L) are applied and NAA 0.05 mg/L added into medium as control. Results show that 1/2 MS+IBA 0.1～0.4 mg/L+NAA 0.05 mg/L is the optimal medium for rooting and transplanting. When the IBA concentration increases,the rate of rooting and transplanting survival decreases.

Abstract:Plant growth regulators significantly affected the pathway of embryogenesis and secondary embryos of loquat (Eriobotra japonica Lindl.). The addition of 2,4-dichlorophenoxyacetic acid (2,4-D) in medium was necessary for callus induction of cultured anthers. Anthers underwent embryogenesis under low concentrations of zeatin (ZT). Indirect embryogenesis occurred when ZT at appropriate concentration was used in combination with other plant growth regulators.

Abstract:The effects of various plant growth regulator combinations on proliferation and subsequent somatic embryo development of Larix leptolepis × L. olgensi embryogenic callus were investigated. The results are as follows: 2,4-D with different concentrations has significant influence on the proliferation of embryogenic callus. The proliferation rate declines with increasing of 2,4-D concentrations. Medium with different combinations of plant growth regulators has remarkable effect on maintenance of embryogenic callus, and 2,4-D 0.1 mg·L-1 + BA 0.05 mg·L-1 + KT 0.05 mg·L-1 or NAA 0.5 mg·L-1 + BA 0.05 mg·L-1 + KT 0.05 mg·L-1 is suitable for embryogenic callus maintenance. IBA 0.5 mg·L-1 + BA 0.05 mg·L-1 + KT 0.05 mg·L-1 was found to be best for embryogenic callus proliferation and somatic embryo development, with 4.1 times proliferation rate, and 88.6 somatic embryos from 1 g embryogenic callus. Both germination and plantlet regeneration rate were highest on medium containing IBA 0.5 mg·L-1 + BA 0.05 mg·L-1 + KT 0.05 mg·L-1, with 66.7% and 50%, respectively.

Abstract:The effects of different growth regulator formulations on elongating clusters,increasing fruit set rate and enlarging berries of summer black grape were studied on four years old plants.The results showed that application of GA3 together with 6-BA was better than that of GA3 alone.The best formulation for pre-anthesis application was 5 mg/L GA3 plus 10 mg/L 6-BA which could be used to elongate spikes when applied 7 to 10 days before flowering.For post-anthesis application,25 mg/L GA3 plus 20 mg/L 6-BA was the best for the prevention of fruit drop if applied 3 days after flowering.To promote the enlargement of berries,25 mg/L GA3 plus 2 mg/L CPPU could be used 10 to 15 days after flowering.

Abstract:Hybrid breeding is the most important means of selecting pear (Pyrus) varieties, but a long juvenile period severely restricts the selection of new varieties. In this study, we used 'Yuluxiang' × 'Akituki' 4-year-old seedling trees to study the effects of plant growth regulators, ring stripping, and ring cutting on the promotion of phase change and flowering to assist in shortening the breeding cycle. A single application of 100 mg/kg 6-BA + 1000 mg/kg PP333 was most effective in promoting phase change and flowering. This treatment effectively inhibited the growth and thickening of annual shoots, significantly increased soluble sugar and protein contents in buds, increased the ABA content by 45.41%, decreased the IAA content by 7.35%, increased the expression of the flower-promoting genes FT and LFY by 2273.41% and 1153.71%, respectively, and decreased the expression of the flower-suppressing gene TFL1 by 74.92%. The flowering plant rate increased by 23.34% compared to the control. Both ring stripping and ring cutting were effective in promoting phase change and flowering, significantly increasing the flowering rate, inflorescence number, and the number of flowering plants. For improving the flowering rate, the ring-stripping treatment had the strongest effect and effectively inhibited the growth and thickening of annual shoots, while also significantly increasing the soluble sugar and protein contents in buds, reducing the contents of IAA and GA3 by 8.73% and 50.12%, respectively, increasing the expression of FT and LFY by 80.01% and 821.14%, respectively, and reducing the expression of the flower-suppressing gene TFL1 by 59.22%. In conclusion, ring stripping, ring cutting, and spraying of 100 mg/kg 6-BA + 1000 mg/kg PP333 were effective in promoting phase change and early flowering in seedling trees.

Abstract:The effect of the combination of 6-BA,NAA with CC and GA_3,sucrose with agar on the proliferation and development of Gerbera adventitious bud were studied.The result showed that combinations of 6-BA 3 mg/L, NAA 0.3 mg/L with CC 3 mg/L and GA_3 0.5 mg/L,sucrose 40 g/L with agar 6 g/L had the best effects on the proliferation and development of adventitious bud.The proliferation rate was the largest and the adventitious buds were strong on these two media,which were the most suitable for the proliferation and development of adventitious bud.

Abstract:The effects of boric acid(H3BO4) and several plant growth regulators,such as gibberellic acid(GA3),indoleacetic acid(IAA),2,4-dichlorophenoxyacetic acid(2,4-D) and naphthalene acetic acid(NAA) on the rate of pollen germination and pollen tube growth of Zizyphus jujuba Mill."zhongqiusucuizao"were studied.The results of H3BO4 alone showed that an optimal concentration of 15 mg/L increased(P0.05)the pollengermination rate to 72.07%,which was 47.62% higher than that of the control(no H3BO4).The optimal concentrationfor GA3 alone was 10.0 mg/L with a pollen germination rate of 70.41%(P0.05).The best germinationrate was found with 5.0 mg/L IAA,10.0 mg/L 2,4-D and 5.0 mg/L NAA respectively.However,the best results(pollen germination of 90.23%) was from the combination of 5.0 mg/L IAA +10.0 mg/LGA3+15.0 mg/L H3BO4.

Abstract:The different basal medium,levers of IBA,NAA,sucrose and the dark culture periods were used to compared their effects on rooting of the regenerated plant from leafs of pear cultvars Dangshansu by using the orthogonal experiment design.The results showed that the effective order of five factors on rooting of the regenerated plant was basal medium,IBA,sucrose,NAA and the dark culture periods.The different basal medium,IBA,and sucrose had significant effect on rooting of the regenerated plant.The best combination for Dangshansu pear was 1/4MS + IBA0.5 mg·L-1 + sucrose 20 g·L-1,and dark cultured for 5 d.

Abstract:The fruit and seed development of two strains of Ziziphus jujuba Mill. cv.Jinsixiaozao,Xiaozao 15 and Xiaozao 24,sprayed with growth regulators and amino acid fertilizers during flowering season,was investigated.The results showed that low concentration of GA3(5-10 mg/L) could enhance the percentage of fruits to bearing shoots(PFTBS) and decrease the percentage of fruit with seeds(PFWS);However,the PFTBS decreased when GA3 concentration reached 35 mg/L;at 150 mg/L 6-BA and 0.02 g/L amino acid fertilizers could decrease both PFTBS and PFWS after natural and self-pollination.

Abstract:The effects of various plant growth regulators with different concentrations and ratios on seedling growth and endogenous hormones of Cunninghamia lanceolata tissue culture were studied based on L4 (23 ) orthogonal ex-perimental design. The results showed that different plant growth regulators could promote seedling height growth and stem growth of Cunninghamia lanceolata tissue culture seedling, the more suitable plant growth regulator con-centration for proliferation was IBA 0. 3 mg/L and 6-BA 0. 8 mg/L, the KT 1 mg/L was favorable for the seedlings growth, the KT 2 mg/L was favorable for the stem segment growth. The contents of chlorophyll a, chlorophyll b, total chlorophyll content, and the content of IAA, GA, ZT, ABA, and IAA/ZR ratio were all the maximum at the combination of IBA 0. 2 mg/L, 6-BA 0. 8 mg/L, KT 1 mg/L. The effect of IBA on the seedling height growth, chlorophyll content and the content of ZR was the highest, and the effect of 6-BA on the stem segment growth and the content of ABA was greater, and the effect of KT on the content of IAA and GA was higher. An ex-tremely significant negative correlation were found between seedling height and ABA/GA, the negative correlation between stem segment multiplication coefficient and IAA/ZR was not so significant, an extremely significant nega-tive correlation were found between total chlorophyll content and ABA/ZR. The decrease of the ratio of ABA/GA was favorable for the seedling height growth, and the decrease of the ratio of IAA/ZR was favorable for the induc-tion of axillary buds, and the decrease of the ratio of ABA/ZR was favorable for the accumulation of chlorophyll.

Abstract:The effects of different plant growth regulator treatments on induction of clumpy buds and rooting in Abelmoschus esculentus(L.) Moench [Hibiscus esculentus L.] plants were studied,the stem segments with one leaf and one axillary bud of seedlings derived from sterile germination were used as explants.The results showed that when the explants were inoculated on 1/2MS basic medium,in a certain concentration range,the increase of IAA concentrations significantly promoted the ratio of differentiation and proliferation,and the presence of 6-BA could increase the role of IAA treatments,but not obviously,so the optimal combination was IAA 1.0 mg/L+6-BA 2.0 mg/L.After the buds grew to more than 1 cm,the single buds were cut out to inoculate on 1/4MS basic medium adding different NAA concentrations,and the optimal rooting rate and growth performance was found in the treatment of NAA 0.1 mg/L.

Abstract:Using the leaves of test-tube seedlings of Actinidia deliciosa as expiants, the effect of different growth regulating substance on the Actinidia deliciosa leaves excised regeneration system was researched, including the differentiation of adventitious buds, the regeneration and rooting of regenerated shoots. The results indicated that the maximum adventitious buds regeneration frequency achieved 76.6% on the medium of Murashige and Skoog medium(MS) containing 5.0 mg·L−1 6-benzylaminopurine(6-BA) and 0.2 mg·L−1 1-naphthaleneacetic acid (NAA); the most suitable medium for shoot multiplication was MS with 2.0 mg·L−1 6-BA, 0.2 mg·L−1 NAA and 0.1 mg·L−1 gibberellin acid(GA3) and the multiplication coefficient was up to 4.76; 1/2MS with 0.7 mg·L−1 1h-indolo-3-butanoic acid(IBA) was the most suitable for the rooting of regenerated shoots, and the rooting rate was 100%.
Study on Tissue Culture in Vitro of Pear Dwarfing Interstocks S2, S5 and PDR54

Abstract:The influencing factors of multiplication coefficient, regeneration quality and rooting for adventitious shoots of pear dwarfing interstocks S_ 2 , S_ 5 and PDR_ 54 were studied. The results showed that the regeneration ability of adventitious shoots was influenced by genotypes, media and plant growth regulators. MS medium was more suitable for the proliferation of S_ 2 ,S_ 5 and PDR_ 54 than QL, WPM and AS media.The middle concentrations of 6-BA (2-3 mg·L~ -1 )in combination with lower concentrations of IBA (0.1-0.2 mg·L~ -1 ) and GA_ 3 (1-2 mg·L~ -1 ) could improve the proliferation of all the three interstocks, with proliferous index of shoots from 3.71 to 5.83. The best medium for S_ 2 was MS+3.0 mg·L~ -1 6-BA+0.2mg·L~ -1 IBA+2.0 mg·L~ -1 GA_ 3 ,while MS+2.0 mg·L~ -1 6-BA+0.1 mg·L~ -1 IBA+1.0 mg·L~ -1 GA_ 3 was the optimal for S_ 5 and PDR_ 54 proliferation in comparison of the comprehensive index of shoot quality,numbers and lengths. The regenerated shoots were rooted with a rooting percentage of 67%, 50% and 86% respectively for S_ 2 , S_ 5 and PDR_ 54 .
Study on inducing and proliferating conditions of adventitious buds from bulblets of'Yixing lily'

Abstract:Bulblets of'Yixing lily'were used as explants to study effects of different combinations and concentrations of plant growth regulators and cutting ways on inducing adventitious buds from the bulblets,and effects of subculture ways on the proliferation of adventitious buds were also discussed.The results showed that the combination of 0.2 mg/L NAA and 1.5 mg/L 6-BA had a better effect on inducing adventitious buds from bulblets,and the inductivity and differentiation coefficient were 98.3% and 5.32,respectively.When the bottom part of single bulblet leaf was used as an explant,the inductivity and differentiation coefficient were 87.5% and 4.11,respectively.Subculturing the fasciculate buds without cutting could get a higher proliferation coefficient and a large number of test-tube bulbs.

Abstract:Some plant growth regulators were tested in series of experiments to determine their effects on inducing adventitious shoot from leaf discs of Dendranthema morifolium cv.Xiaojinhuang.The results showed that 1.0 mg·L-1 6-BA was more favorable to shoot formation than TDZ or KT,and regeneration rate of 1.0 mg·L-1 6-BA treatment was 27.1%.Whereas the combination of 6-BA 1.0 mg·L-1 and NAA 0.6 mg·L-1 on regeneration frequency was much more efficient than others with a regeneration rate of 96.9%.The optimal rooting medium was MS + NAA 0.4 mg·L-1.

Abstract:The color and berry size of ‘Crimson Seedless’ play vital role in their marketability. In the present study, eight-year-old ‘Crimson Seedless’ was used as the experimental material to investigate the effect of plant growth regulators on the fruit weight, quality and color. Several dilutions of “Pro-Gibb” (20% gibberellic acid (A3)) and “Promalin” (1.8% 6-BA & 1.8% gibberellic acid (A4 + A7)) were applied to the ‘Crimson Seedless’ plants during the swelling period, and the results showed that the cluster weight and berry weight were significantly increased by 46.77% and 20.64% respectively when treated with 6000 times diluted solution of “Promalin”. Except for application of 15,000 times diluted solution of “Pro-Gibb”, 4000 times and 5000 times diluted solution of “Promalin”, there were no significant differences between control and other treatments in soluble solid content (SSC). The titratable acid content (TA) of the berries was also significantly reduced under various treatments of plant growth regulators. The Vc content of the berries under T2 treatment increased up to 6.40 mg/100 g compared with the control. The color of berries with 20,000 times diluted solution of “Pro-Gibb” treatment was pink, and the color of berry with other treatments was red. In conclusion, application of 6000 times diluted solution of “Promalin” improved fruit quality, texture and berry weight more effectively compared with other applied treatments. Overall, the results of the present study showed that optimized doses of the plant growth regulators could significantly improve the ‘Crimson Seedless’ fruit quality.

Abstract:The effect of plant growth regulator concentration on in vitro multiplication and rooting were studied using adventitious buds obtained in tissue culture of Populus yunnanensis Dode as raw material.The results showed that MS+0.1 mg/L 6-BA +0.02 mg/L NAA was the optimum medium for adventitious buds multiplication;and 1/2MS+0.02 mg/L NAA was good for rooting.

Abstract:The best time for sterilization,the effects of different combinations of 6-BA and NAA on the inducement and proliferation of axillary bud,the effects of different concentrations of NAA on rooting,and the survival of transplanting to different substrates for explants ofDianthus caryophyllus was studied by tissue culture technique with the stem segments of the "Masters",a cultivar of Dianthus caryophyllus as materials,respectively.The results of experiments were:Aseptic seedlings would be obtained by the treatments of 0.1％ HgCl2 for 1 min or 10％ NaClO for 4 min,as it significantly reduced the contamination and improved the survival rate and rooting rate of explants.Combinatorial treatments with 3 mg/L of 6-BA and 0.1 mg/L of NAA,resulted in the best inducement and proliferation for the axillary bud growth.The germination rate (value of 83％) as well as coefficient of proliferation (value of 8.3) was better than those of treatment with growth regulator 6-BA only.The rooting rate ofexplants rose along with the increase of NAA concentration in the medium and it was up to 90％ when the 1 mg/L of NAA,while there was significantly difference between value for rooting number ratio and the root length (p＜0.05).The vermiculite or perlite mixed with vermiculite was the suitable medium for transplanting the tissue cultures of Dianthus caryophyllus.
