Abstract:The techniques for improving the stereoselectivity of asymmetric reduction of 3-oxo ester to chiral 3-hydroxy ester catalyzed by yeast cells were studied. Ethyl 4-chloro-3-oxobutanoate was chosen as the model substrate, and the effects of heating and enzyme inhibitor pretreatment of yeast cells on the stereoselectivity of the reduction reaction were investigated in detail. The results showed that the heating pretreatment and preincubation with allyl alcohol could remarkably improve the stereoselectivity of the S-(D)-product, respectively. The stereoselectivity of the S-form product increased with increasing temperature and treatment time in the heating pretreatment. Using allyl alcohol as enzyme inhibitor, the stereoselectivity of the S-form product also increased with increasing the concentration of allyl alcohol. Under appropriate pretreatment conditions, the ee of (S)-ethyl 4-chloro-3-hydroxybutanoate in the asymmetric reduction could reach as high as 97%.

Abstract:Optically active aryl alcohols are a kind of important chiral building blocks for the synthesis of chiral drugs. In recent years, the preparations of enantiopure aryl alcohols have become one of the focus points in organic chemistry. Compared with conventional chemical processes, biocatalysis is more attractive due to its high enantioselectivity, mild and safe reaction conditions and less environmental hazards. Asymmetric bioreduction of aryl ketones is the most effective method for the synthesis of enantiopure aryl alcohols. This paper mainly reviews the recent progress in the preparation of chiral aryl alcohols by asymmetric bioreduction catalyzed by microbial whole-cells, enzymes, genetically engineered bacteria or yeasts, as well as immobilized cells. The effects of substrate co-solvents such as organic solvents, surfactants and ionic liquids on asymmetric bioreduction are further summarized. The prospect of the preparation of enantiopure aryl alcohols by asymmetric bioreduction is also discussed.

Abstract:Chiral alcohols are essential building blocks of numerous pharmaceuticals and fine chemicals. Aldo-keto reductases (AKRs) constitute a superfamily of oxidoreductases that catalyze the reduction of aldehydes and ketones to their corresponding alcohols using NAD(P)H as a coenzyme. Knowledge about the crucial roles of AKRs immobilization in the biocatalytic synthesis of chiral alcohols is expanding. Herein, we reviewed the characteristics of various AKRs immobilization approaches, the applications of different immobilization materials, and the prospects of continuous flow bioreactor construction by employing these immobilized biocatalysts for synthesizing chiral alcohols. Finally, the opportunities and ongoing challenges for AKR immobilization are discussed and the outlook for this emerging area is analyzed.

Abstract:Optically active 1-phenylethanol and its derivatives are versatile chiral precursors for many pharmaceuticals. The increasing market demand of enantiopure alcohols calls for exploration of more robust biocatalysts capable of delivering high conversion rates at high substrate concentrations. The carbonyl reductase YueD from Bacillus subtilis was engineered for improved reduction of halogenated acetophenones. Based on in silico docking and Alanine screening, mutant Val181Ala with 97% conversion of the model substrate 3-bromoacetophenone was obtained. Further molecular simulation analysis suggested expansion of the catalytic pocket and altered substrate orientation to provide a structural basis for the improved activity upon Vail 81 mutation. To eliminate the requirement of expensive exogenous NADPH and thus afford economical synthesis of enantiopure alcohol products, a cofactor regeneration system based on glucose dehydrogenase was introduced. Since the product of glucose oxidation seemed to inhibit the activity of YueD, xylose was used instead as the co-substrate, which efficiently regenerated NADPH in situ, delivering (S) or (R) alcohols with >99% ee at high conversion rates of 85-99% in the asymmetric reduction of halogenated acetophenones (500 mM). These data demonstrate the industrial potential of the YueD mutant Val181Ala in biosynthesis of valuable chiral alcohols. (C) 2018 Elsevier B.V. All rights reserved.

According to the rate and direction of enantioselective bioreduction, enantioselectivity, and the yield of the target products, the most promising cellular catalysts for the asymmetric bioreduction of prochiral carbonyl compounds, which enable the synthesis of target alcohols of both (S)- and (R)-configurations, were identified.

Abstract:Biocatalysis is a basic method of asymmetric catalysis for preparing chiral Active Pharmaceutical Ingredients, which owns several "green merits": chemo-, regio- and high enantioselectivity. As the development of DNA seqencing, DNA synthesis and protein engineering, suitable enzymes can be efficiently developed for basic researches and industrial applications. Biocatalysis has been keeping as a hot spot in asymmetric synthesis recently. Carbonyl reductases have been widely used for stereoselectively transforming ketones to chiral second alcohols with only one stereocenter. When combining with Dynamic Kinetic Resolution (DKR), the bioreaction with carbonyl reductases can efficiently construct chiral alcohols with two stereocenters in one step. This review highlights the method of its mechanism and nearly twenty examples from research papers and patents for one decade. We attempt to analyze and conclude the characteristics of this method based on chemical structures and enzymes. At last, a practical and developing research method is recommended in three steps: screening-racemization-balance in sequence. It is hoped to be useful for future basic researches and industrial applications.

Abstract:The asymmetric cross-coupling reaction is developed as a straightforward strategy toward 1,1-diaryl alkanes, which are a key skeleton in a series of natural products and bioactive molecules in recent years. Here we report an enantioselective benzylic C(sp(3))-H bond arylation via photoredox/nickel dual catalysis. Sterically hindered chiral biimidazoline ligands are designed for this asymmetric cross-coupling reaction. Readily available alkyl benzenes and aryl bromides with various functional groups tolerance can be easily and directly transferred to useful chiral 1,1-diaryl alkanes including pharmaceutical intermediates and bioactive molecules. This reaction proceeds smoothly under mild conditions without the use of external redox reagents.

Abstract:Herein we report a highly enantioselective kinetic resolution of tertiary benzyl alcohols via palladium/chiral norbornene cooperative catalysis. With simple aryl iodides as the resolution reagent, a wide range of readily available racemic tertiary benzyl alcohols are applicable to this method. Both chiral tertiary benzyl alcohols and benzo[c]chromene products are obtained in good to excellent enantioselectivities (selectivity factor up to 544). The appealing synthetic utility of the obtained enantioenriched tertiary alcohols is demonstrated by the facile preparation of several valuable chiral heterocycles. Preliminary mechanism studies include DFT calculations to explain the origin of enantiodiscrimination and control experiments to uncover the formation of a transient axial chirality during the kinetic resolution step.
Rational Design of Dehydrogenase/Reductases Based on Comparative Structural Analysis of Prereaction-State and Free-State Simulations for Efficient Asymmetric Reduction of Bulky Aryl Ketones

Abstract:Inspired by the conformational change of the enzyme-substrate complex in molecular dynamics (MD) simulation with distance restriction, we propose a strategy for identifying the engineering targets based on the comparative analysis of enzyme-/substrate- binding modes in MD simulations with and without distance restriction (prereaction-state simulation and free-state simulation). Taking the short-chain dehydrogenase/reductase (SDR) mutant EbSDR8-G94A/S153L (Mu0) with poor activity toward bulky aryl ketones as an example, H145 and Y188 were identified as the engineering targets due to the distinct conformation difference in the two simulation modes. To break the "beam" structure formed by these residues at the entry of cavity C2 in free-state simulation, the substrate-binding pocket was reconstructed, and meanwhile the relative size of cavities C1 and C2 was modulated to improve the enantioselectivity. In this way, mutants for efficient asymmetric reduction of o-halogenated acetophenones, propiophenones, aromatic ketoesters, and diaryl ketones were designed, delivering chiral alcohols with >99% conversion and >98% ee. The effectiveness of this design strategy was also validated by the successful redesign of PpYSDR, generating a variant for efficient reduction of (4-chlorophenyl) 2-pyridyl ketone into the S-product with >99% conversion and 96% ee. MD simulations suggested a suitable binding pocket with proper energy contribution as the ubiquitous mechanism for the improvement of activity and enantioselectivity toward substrates with varied structures. The success in this study provides hints for the rational design of alcohol dehydrogenases/reductases with both a broad substrate spectrum and high enantioselectivity.
Engineering carbonyl reductase for one-pot chemobiocatalytic enantioselective synthesis of value-added N-containing chiral alcohol from N-acetyl-D-glucosamine

Abstract:Direct chemobiocatalytic conversion of biomass into value-added chemicals is promising yet challenging, because of the combined advantages of the two technologies and their incompatible issues. In this work, structure-guided engineering of carbonyl reductase from Streptomyces coelicolor (ScCR) was performed to improve its catalytic activity as well as stability, thus allowing for direct conversion of chitin-derived N-acetyl-D-glucosamine (NAG) into chiral 3-acetamido-5-(1-hydroxyethyl)furan (3A5HEF) by integrating its variants with chemical catalysts, with no isolation of any intermediate. Upon three rounds of site-directed mutagenesis, two robust variants M3 (S167F/P168S) and M4 (S167Y/P168S) were identified, with 6-fold higher catalytic efficiency (kcat/Km) toward 3-acetamido-5-acetylfuran (3A5AF) than the parent enzyme and improved stability. Mechanistic insights into the improved activities of variant M3 were provided on the basis of the molecular docking study as well as molecular dynamics simulations. Chemobiocatalytic conversion of NAG into (R)-3A5HEF was performed via sequential chemical dehydration by tyrosine hydrochloride/CaCl2 and biocatalytic asymmetric reduction by variant M4, with 53% yield and >99% ee. In the scale-up two-step synthesis, (R)-3A5HEF was obtained with 42% isolated yield. In addition to 3A5AF, greatly improved catalytic activities of the two variants toward other carbonyl compounds, particularly acetophenones, were observed (up to 41-fold higher activity than the parent enzyme). Various carbonyl compounds were reduced to the target chiral alcohols by the variants, with 46-99% yields and >99% ee. This work demonstrates that protein engineering is a powerful strategy to address the incompatibility between chemo- and biocatalysts in chemobiocatalysis.

Abstract:<p>With the rapid development of genomic DNA sequencing, recombinant DNA expression, and protein engineering, biocatalysis has been increasingly and widely adopted in the synthesis of pharmaceuticals, bioactive molecules, fine chemicals, and agrochemicals. In this review, we have summarized the most recent advances achieved (2018–2020) in the research area of ketoreductase (KRED)‐catalyzed asymmetric synthesis of chiral secondary alcohol intermediates to pharmaceuticals and bioactive molecules. In the first part, synthesis of chiral alcohols with one stereocenter through the bioreduction of four different ketone classes, namely acyclic aliphatic ketones, benzyl or phenylethyl ketones, cyclic aliphatic ketones, and aryl ketones, is discussed. In the second part, KRED‐catalyzed dynamic reductive kinetic resolution and reductive desymmetrization are presented for the synthesis of chiral alcohols with two contiguous stereocenters.</p>

Abstract:Chiral diols are an integral part of chiral building blocks and synthetic intermediates. Transition metal-catalyzed asymmetric hydrogenation and transfer hydrogenation of diketones are recognized as one of the most straightforward and efficient methods for the preparation of enantiomerically enriched diols. Meanwhile, the mono-reduction product of diketones, chiral hydroxyketones, has wide application in chiral fragment construction as well, among which, desymmetrization of cyclic diketones possesses the capability of establishing multiple chiral centers in one step. In this paper, research progress of asymmetric hydrogenation and transfer hydrogenation of diketone compounds in recent decades is reviewed from the perspective of different substrate types (1,2-/1,3-/1,4-diketones), with emphasis on the influence of coordination mode between substrate and catalyst species on the stereoselectivity. In addition, future challenges and development tendencies in this field are summarized and prospected.
Enantiocomplementary Bioreduction of 1-(Arylsulfanyl)propan-2-ones

Abstract:This study explored the enantiocomplementary bioreduction of substituted 1-(arylsulfanyl)propan-2-ones in batch mode using four wild-type yeast strains and two different recombinant alcohol dehydrogenases from Lactobacillus kefir and Rhodococcus aetherivorans. The selected yeast strains and recombinant alcohol dehydrogenases as whole-cell biocatalysts resulted in the corresponding 1-(arylsulfanyl)propan-2-ols with moderate to excellent conversions (60-99%) and high selectivities (ee > 95%). The best bioreductions-in terms of conversion (>90%) and enantiomeric excess (>99% ee)-at preparative scale resulted in the expected chiral alcohols with similar conversion and selectivity to the screening reactions.

Abstract:Fluorinated motifs are frequently encountered in drugs and agrochemicals. Incorporating fluorine-containing motifs in drug candidates for lead optimization in pharmaceutical research and development has emerged as a powerful tool. The construction of molecules that feature a trifluoromethyl (CF3−) group on a stereogenic carbon has accumulated broad research efforts. Unlike its well-explored, biologically active methyl counterpart, asymmetric construction of β-trifluoromethylated alcohols bearing adjacent stereocenters still remains elusive. Through retrosynthetic analysis, we posited that followed by sequential reduction of carbonyl, the initial construction of chiral α-trifluoromethylated ketones could render the desired product in a facile, one-pot fashion. Herein, we developed the first example of nickel-catalyzed asymmtric reductive cross-coupling trifluoroalkylation of acyl chlorides for enantioselective synthesis of diverse α-trifluoromethylated ketones. The one-pot reduction of these α-trifluoromethylated ketones furnished corresponding alcohols bearing β-CF3-substituted stereogenic carbons with excellent diastereoselectivity and complete enantioselective retention. High yields/enantioselectivity, mild conditions, and good functional group compatibility are shown in the system. Abstract:Biocatalysis has emerged as a strong tool for the synthesis of active pharmaceutical ingredients (APIs). In the early twentieth century, whole cell biocatalysis was used to develop the first industrial biocatalytic processes, and the precise work of enzymes was unknown. Biocatalysis has evolved over the years into an essential tool for modern, cost-effective, and sustainable pharmaceutical manufacturing. Meanwhile, advances in directed evolution enable the rapid production of process-stable enzymes with broad substrate scope and high selectivity. Large-scale synthetic pathways incorporating biocatalytic critical steps towards >130 APIs of authorized pharmaceuticals and drug prospects are compared in terms of steps, reaction conditions, and scale with the corresponding chemical procedures. This review is designed on the functional group developed during the reaction forming alcohol functional groups. Some important biocatalyst sources, techniques, and challenges are described. A few APIs and their utilization in pharmaceutical drugs are explained here in this review. Biocatalysis has provided shorter, more efficient, and more sustainable alternative pathways toward existing small molecule APIs. Furthermore, non-pharmaceutical applications of biocatalysts are also mentioned and discussed. Finally, this review includes the future outlook and challenges of biocatalysis. In conclusion, Further research and development of promising enzymes are required before they can be used in industry.
