Abstract:The moving bed bioreactor (MBBR) process has drawn more attention as a promising biological wastewater treatment process. Nevertheless, achieving quick start-up and microbial biofilm formation remains a significant challenge. Consequently, the present study investigated a novel chitosan-based natural sludge (CS@NGS) seeding strategy for the accelerated start-up of MBBR. Three identical bioreactors were employed; the first bioreactor was without sludge seed as the control (BR1), the second was inoculated only with sludge (BR2), and the third was inoculated with CS@NGS according to the proposed seeding method (BR3). All bioreactors were utilised to treat simulated recirculating aquaculture systems (RAS) effluent. Resultantly, the CS@NGS shortened the start-up period from over twenty to seven days due to the enhanced initial microbial adhesion and biofilm formation. Under optimal conditions, the ammonium removal in BR3 approached 100%, which was relatively higher than BR2 (96.35 +/- 1.12%) and BR1 (92.56 +/- 2.17%). Moreover, a low nitrite accumulation was exhibited in the effluents, approximately <= 0.03 mg L-1. The process performance correlated positively with core bacteria from the genera Nakamurella , Hyphomicrobium , Nitrospira , Paenarthrobacter , Rhodococcus , and Stenotrophobacter. The quantitative polymerase chain reaction (qPCR) results demonstrated that the CS@NGS enhanced the expressions of amoA, nxrB, nirK, nirS, narG, and napA nitrogen metabolism-related functional genes to varying degrees. The present study findings can assist the rapid start-up of aquaculture biofilters utilised to solve high nitrite and ammonia accumulation in recirculated water from industrial RAS.
Start-up of Anammox Bioreactor with Denitrification Biofilm

Abstract:The anaerobic ammonia oxidation (Anammox) bioreactor was successfully set up with the denitrification biofilm. After a three-month operation, the volumetric total nitrogen loading rate was 0.143?kg·m~(-3)·d~(-1) and the total nitrogen removal was 86.52% with effluent ammonia and nitrite less than 1?mg·L~(-1). The ratio of NH~+_4-N removal, NO~-_2-N removal and NO~-_3-N production as well as the biofilm color could indicate the development of Anammox bioreactor performance.
Studies on Start-Up Technique of Anammox Bioreactor

Abstract:The feasibility of start-up of anammox bioreactor, using autotropic denitrifying biofilm was studied. The anammox bioreactor could be started up successfully within 110d, adopting the method of cultivating firstly autotrophic denitrifying biofilm, then start-up of the bioreactor. The volumetric total nitrogen loading was 0.145kg/(m3.d), and the removal rate of NH4+-N and NO2--N was 98.59% and 99.08%, respectively. At the initial period of start-up, the effluent pH value of the bioreactor was lower than the influent pH value, and tended to be unanimous with the advance of the start-up process. At steady operation, the liquid pH value in the bioreactor was alkaline, the ratio of removed NH4+-N, NO2--N and produced NO3--N was 1:1.01:0.19. In the start-up process, the ratio of removed NH4+-N, NO2--N and produced NO3--N as well as the change of pH value and biofilm color could indicate the start-up course of the bioreactor.
[Isolation, identification of two aerobic denitrifiers and bioaugmentation for enhancing denitrificaition of biofilm under oligotrophic conditions]

Abstract:Two aerobic denitrifying bacteria were isolated from the sludge of municipal wastewater treatment plant by enrichment, preliminary screening with BTB culture medium and the denitrification potential test and identified through 16S rDNA sequence analysis. Then they were bioaugmented to oligotrophic biofilm system respectively, aiming to enhance the denitrification capacity. The two strains were identified as Pseudomonas aeruginosa and Pseudomonas putida respectively. They could remove 78% or 82% of the total nitrogen in the simulate wastewater when existed alone. And the total nitrogen removal efficiency of the biofilm system reached 68% and 64% after bioaugmentation with two trains, increased by 47% and 43% compared to the control, respectively. Meanwhile, the ammonia nitrogen was nearly 100% removed. It can be concluded that two aerobic denitrifiers can enhance the denitrification of biofilm system significantly under oligotrophic conditions and will not inhibit the nitrification process, therefore can help biofilm system achieve simultaneous nitrification and denitrification.

Abstract:Bio-cathode denitrification is a new and unique technology for autotrophic nitrogen removal, which uses electricity instead of chemical matter as electron donor. Functional microorganisms are the driver of bio-cathode denitrification and inocula are the origin of functional microorganisms, so three typical inocula, denitrifying, denitratating and anammox (anaerobic ammonia oxidizing) sludge, were chosen to investigate their effect on the performance of bio-cathode denitrification in this work. The results showed that the nitrogen removal rate of denitrifying, denitratating and anammox bio-cathodes was around 101.79 mg/L.d, 47.31 mg/L.d and 100.04 mg/L.d respectively. The output power was 24.01 mW/m(2), 14.11 mW/m(2) and 45.93 mW/m(2). The denitrifying, denitratating and anammox biofilms were observed to grow on cathodes, indicating that the functional microorganisms could use electricity as the sole electron donor and so they were named as electrotrophic microorganisms. The growth rate and the cell yield of denitrifying, denitratating and anammox biofilms were 24 g/m(2).d and 1.71 x 10(-3) g/g NO3--N, 7.67 g/m(2).d and 1.94 x 10(-3) g/g NO3--N, 41 g/m(2).d and 2.32 x 10(-3) g/g NO2--N, respectively. Pseudomonas, Glycocaulis and SM1A02 were found to be enriched in the three biofilms, and Pseudomonas stutzeri was deduced to be the key functional microorganism. Denitrifying sludge was the optimal inoculum when wastewater treatment was the main purpose; however, Anammox sludge turned to be the optimal inoculum when wastewater treatment and energy recovery were combined. The functional microorganisms and their metabolic characteristics were revealed to be the critical factor that determined the enrichment of electrotrophic microorganisms from inocula and the performance of bio-cathode denitrification. These findings are significant to get an insight into the new electrotrophic microorganism and to develop the novel bio-cathode denitrification technology.
Nitrogen removal intensification of biofilm through bioaugmentation with Methylobacterium gregans DC-1 during wastewater treatment

Abstract:The increasing concern for environmental remediation has led to a search for effective methods to remove eutrophic nutrients. In this study, Methylobacterium gregans DC-1 was utilized to improve nitrogen removal in a sequencing batch biofilm reactor (SBBR) via aerobic denitrification. This bacterium has the extraordinary characteristics of strong auto-aggregation and a high ability to remove nitrogen efficiently, making it an ideal candidate for enhanced treatment of nitrogen-rich wastewater. This strain was used for the bioassessment of a test reactor (SBBRbio), which showed a shorter biofilm formation time compared to a control reactor (SBBRcon) without this strain inoculation. Moreover, the enhanced biofilm was enriched in TB-EPS and had a wider variety of protein secondary structures than SBBRcon. During the stabilization phase of SBBRbio, the EPS molecules showed the highest proportion of intermolecular hydrogen bonding. It is possible that bioaugmentation with this strain positively affects the structural stability of biofilm. At influent ammonia loadings of 100 and 150 mg. L<sup>-1</sup>, the average reduction of ammonia and nitrate-nitrogen was higher in the experimental system compared to the control system. Additionally, nitrite-N accumulation was lower and N<sub>2</sub>O production decreased compared to the control. Analysis of the microbial community structure demonstrated successful colonization in the bioreactor by a highly nitrogen-tolerant strain that efficiently removed inorganic nitrogen. These results illustrate the great potential of this type of denitrifying bacteria in the application of bioaugmentation systems.

Abstract:A newly isolated and identified Diaphorobacter polyhydroxybutyrativorans strain (SL-205) was employed to enhance the denitrification performance of a laboratory-scale solid-phase denitrification (SPD) reactor using poly (3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) as a carbon source, and dynamic variations in microbial communities in the reactor were investigated. Results indicated that bioaugmentation with strain SL-205 enabled rapid reactor startup and improved denitrification performance relative to the reactor inoculated with activated sludge. Illumina sequencing revealed that bioaugmentation also significantly increased Proteobacteria abundance along with increased influent nitrate loading. Additionally, two genera of PHBV-degrading denitrifers, Diaphorobacter and Acidovorax, exhibited higher abundance, and elevated expression of denitrificationassociated genes (narG, nirK, and nirS) was observed following bioaugmentation relative to the control at influent nitrate loading ranging from 1.28 g N/(L.d) to 1.6 g N/(L.d).

Abstract:Bioaugmentation of denitrifying bacteria can serve as a promising technique to improve nutrient removal during wastewater treatment. While denitrification inhibition by bacterial quorum sensing (QS) in Pseudomonas aeruginosa has been indicated, the application of bacterial QS disruption to improve nitrate removal from wastewater has not been investigated. In this study, the effect of bioaugmentation of P. aeruginosa SD-1 on nitrate removal in sequencing batch reactors that treat nitrate rich wastewater was assessed. Additionally, the potential of a quorum sensing inhibitor (QSI) to improve denitrification following bacterial bioaugmentation was evaluated. Curcumin, a natural plant extract, was used as a QSI. The chemical oxygen demand (COD) and initial nitrate concentration of the influent were 700±20 mg/L and 200±10 mg/L respectively, and their respective concentrations in the effluent were 56.9±3.2 mg/L and 9.0±3.2 mg/L. Thus, the results revealed that bioaugmentation of P. aeruginosa SD-1 resulted in an increased nitrate removal to 82%±1%. Further, nitrate was almost completely removed following the addition of the QSI, and activities of nitrate reductase and nitrite reductase increased by 88%±2% and 74%±2% respectively. The nitrogen mass balance indicated that aerobic denitrification was employed as the main pathway for nitrogen removal in the reactors. The results imply that bioaugmentation and modulation of QS in denitrifying bacteria, through the use of a QSI, can enhance nitrate removal during wastewater treatment.
Rapid start-up of mainstream partial denitrification /anammox and enhanced nitrogen removal through inoculation of precultured biofilm for treating low-strength municipal sewage

Abstract:This study investigated the rapid start-up of mainstream partial denitrification coupled with anammox (PD/A) and nitrogen removal performance by inoculating precultured PD/A biofilm. The results showed mainstream PD/A in the anaerobic-anoxic-aerobic (A2O) process was rapidly established within 30 days. Nitrogen removal efficiency (NRE) improved by 23.8 % contrasted to the traditional A2O process. The mass balance showed that anammox contribution to total nitrogen (TN) removal were maintained at 37.9 %∼55.7 %, and reducing hydraulic retention time (HRT) strengthened simultaneously denitrification and anammox activity. The microbial community showed that the dominant bacteria such as denitrifying bacteria (DNBs) and glycogen accumulating organisms (GAOs) both in biofilm and flocculent sludge (floc), integrating with anammox bacteria (AnAOB) in biofilm might lead to enhanced nitrogen removal. Overall, this study offered a fast start-up strategy of mainstream PD/A with enhanced nitrogen removal, which are valuable for upgradation and renovation of existed municipal wastewater treatment plants (WWTPs).
Treatment of Nitrogen-Rich Wastewater by Mixed Aeration Combined with Bioaugmentation in a Sequencing Batch Biofilm Reactor: Biofilm Formation and Nitrogen-Removal Capacity Analysis

Abstract:The mixed aerated and bioaugmented sequencing batch biofilm reactor (SBBR) was developed to improve the efficiency of nitrogen-rich wastewater treatment, and the mechanism of biofilm formation and nitrogen removal was analyzed. The results showed that the addition of aerobic denitrifiers could shorten the start-up time and significantly improve the biofilm formation ability of SBBRs. The presence of nanobubbles could make the biofilm more compact and secrete more EPS, which promoted the activity and stability of the biofilm. During the stable operation stage, the bioaugmented SBBR under mixed aeration mode showed the best nitrogen removal performance and the removal rates of NH4+-N (95.65) and TN (94.88%） were increased by 19.86% and 14.36% compared with the control group, respectively. Microbial community structure and nitrogen metabolism pathway analysis revealed that strain ZS1 colonized the reactor. Hyphomonas, Defluviicoccus and Hydrogenophaga played important roles in N removal in the mixed aerated and bioaugmented SBBR. Meanwhile, the suitable oxidative environment created by mixed aeration made autotrophic nitrifiers（mainly Nitrospira and Nitrosomonas）and aerobic denitrifiers (mainly Hydrogenophaga, Hyphomonas and Pseudofulvimonas) coexist successfully, increasing the relative abundance of the key functional genes (amoABC, hao, napAB genes) in nitrification and denitrification processes to improve the N removal performance. Furthermore, mixed aeration combined with bioaugmented stimulated the nirA, gltBD and GLUD genes, thus promoting the process of nitrogen assimilation and providing a pathway of the direct removal of NH4+-N.
Application Potential of Aerobic Denitrifiers Coupled with a Biostimulant for Nitrogen Removal from Urban River Sediment

Abstract:Aerobic denitrifiers coupled with a denitrification agent were applied in the sediment of an urban river for the bioremediation of nitrogen pollution. The results revealed that 14.7% of the total nitrogen in the sediment was removed after 115 days of treatment and the nitrate nitrogen concentration removal rate was enhanced in the overlying water. Compared with the control, the total transferable nitrogen in the sediment increased from 0.097 to 0.166 mg/g, indicating that more nitrogen is likely to be involved in the biogeochemical cycling of nitrogen. Increased urease activity indicated the possible further potential of nitrogen biodegradation, while the decreased protease pointed to the low concentration of protein remaining in the sediment. Sequencing revealed that the bacterial community diversity in the sediment increased significantly after 43 days of treatment and that the effect persisted. Compared with other microcosms, the dominant phyla in the sediment after 43 days were Firmicutes, Elusimicrobia, Spirochaetae and Fibrobacteres; whereas, after 115 of treatment, the dominant bacteria were Nitrospirae, Deferribacteres and Chloroflexi. The dominant bacteria in the sediment are mainly associated with nitrogen cycling and thus contributed considerably to nitrogen removal in the sediment. Overall, the direction of species succession was similar to natural succession; namely, there were no undesirable ecological risks involved. This study highlights the possible benefits and feasibility of using bioaugmentation technology coupled with biostimulation to remediate nitrogen-polluted sediments.
Achieving Partial Denitrification Through Control of Biofilm Structure During Biofilm Growth in Denitrifying Biofilter

Abstract:Partial denitrification was one of most effective ways to provide nitrite for annamox; whereas very limited research has been done to achieve nitrite accumulation in biofilm system. In this study, partial denitrification was studied in a lab-scale denitrifying biofilter (DNBF). The results showed biofilm structure variations caused the differences between nitrate specific reduction rate (NaSRR) and nitrite specific reduction rate (NiSRR), which led to nitrite accumulation in different degree at different biofilm formation phases. Hydrodynamic conditions also significantly influenced biofilm structure, nitrate and nitrite reduction activities. At the filtration velocity of 3.86 m h(1), not only biofilm structure, NaSRR and NiSRR kept relatively stable, but also 60% of nitrite accumulation and no nitrate in the effluent were achieved. Furthermore, Thauera genus bacteria, benefited for nitrite accumulation, became the dominant communities in high nitrite accumulation conditions. Rapid establishment of municipal sewage partial denitrification-anammox for nitrogen removal through inoculation with side-stream anammox biofilm without domestication

Abstract:Mainstream partial denitrification anammox was achieved through inoculation of side-stream mature partial nitritation anammox biofilm without domestication. The contribution of anammox to nitrogen removal was 29.4 %. Moreover, prolonging anoxic hydraulic retention time and introducing side-stream nitrite under different carbon/nitrogen ratios enriched anammox bacteria. The abundance of anammox bacteria increased by ∼ 10 times ((2.19 ± 0.17) × 10 12 copies gene / g dry sludge) with a total relative abundance of 18.51 %. During 258 days of operation, the contribution of anammox to nitrogen removal gradually increased to 68.8 %. The total nitrogen in the effluent decreased to 8.84 mg/L with a total nitrogen removal efficiency of 76.4 % under a carbon/nitrogen ratio of 3. This paper proposes a novel way to rapidly achieve mainstream partial denitrification anammox via inoculation with side-stream mature partial nitritation anammox biofilm. This method achieves advanced nitrogen removal from municipal wastewater, even under low carbon/nitrogen ratios.

STUDY OF SIMULTANEOUS DENITRIFICATION AND DESULFURIZATION BY BIOLOGICAL PACKED TOWER

Abstract:This study investigated the artificial simulated wastewater containing sulfide and nitrate nitrogen,in which sulfide is regarded as an electron donor and nitrate is as an electron acceptor. Anaerobic biological packed tower is used to achieve simultaneous denitrification and desulfurization to study the start-up performance of the tower and the biofilm growth characteristics of the flora. The results showed that: under the condition of S/N( molar ratio) of 5:3,the initial pH of 8. 3 to 8. 5,the temperature of 28 ℃,and the influent sulphide load of 500g/( m3·d),the sulfur and nitrogen removal rate reached 96. 7% and 87. 5%,respectively. The dominant bacteria in the anaerobic biological packed tower is denitrification thiobacilli,whose suitable growth environment is a temperature of 28 ℃ to 30 ℃,pH range of 6 to 7. And its tolerance of the initial sulfide concentration after domestication is increased to 625 to 1250g/m3.
Improving denitrification performance of biofilm technology with salt-tolerant denitrifying bacteria agent for treating high-strength nitrate and sulfate wastewater from lab-scale to pilot-scale

Abstract:This study focused on the application of salt-tolerant denitrifying bacteria (DBA) in an optimized biofilm process to treat high sulfate-nitrate wastewater from lab-scale to pilot-scale. Lab-scale results demonstrated the salinity, DBA inoculum, supplementary carbon and phosphorus source significantly varied the startup periods at the range of 36-74 d, and the optimum initial start-up conditions were as follows: >0.6 g/L of DBA, 2-4 of C/N ratio, 0.3-0.6 mg/L of phosphorus and a salinity-gradient domestication method. A pilot scale of biofilm technology with DBA was further developed for treating real wastewater from the desulfuration and denitration with both high nitrate (≈200 mg/L) and sulfate (2.7%). The denitrification efficiency reached above 90% after one-month gradient-salinity of 0.5%-2.7%. Mature biofilm had dominant genera Hyphomicrobium (31.80%-61.35%), Methylotenera (0.85%-20.21%) and Thauera (1.42%-8.40%), etc. Notably, the largest genera Hyphomicrobium covered the complete denitrification genes.

Abstract:The denitrifier community and associated nitrate and nitrite reduction in the bioaugmented and general sequencing batch reactors (SBRs) during the treatment of coking wastewater containing pyridine and quinoline were investigated. The efficiency and stability of nitrate and nitrite reduction in SBR was considerably improved after inoculation with four pyridine- or quinoline-degrading bacterial strains (including three denitrifying strains). Terminal restriction fragment length polymorphism (T-RFLP) based on the nosZ gene revealed that the structures of the denitrifier communities in bioaugmented and non-bioaugmented reactors were distinct and varied during the course of the experiment. Bioaugmentation protected indigenous denitrifiers from disruptions caused by pyridine and quinoline. Deep Treatment of Tailwater by Aerobic Denitrifying Bacteria Enhanced Biofilter

Abstract:The high concentration of total nitrogen in the tailwater discharged from some wastewater treatment plants is relatively high, which is easy to cause eutrophication in the receiving water bodies. In this study, the aerobic denitrifying bacteria were combined with denitrifying biofilter through bioreinforcement technology. The aerobic denitrifying bacteria strains W30 and Z66 isolated from activated sludge were used to construct a complex bacterial agent to strengthen the denitrifying biofilter, and the enhanced nitrogen removal effect of the complex bacterial agent was investigated, and the microbial community structure in the system after bioreinforcement was analyzed by high-throughput sequencing technology. The results showed that strain W30 was identified as Pannonibacter and strain Z66 as Pseudomonas by 16S rDNA sequencing analysis. The bioreinforced denitrification biofilter had a fast start-up rate of membrane hanging, and the removal rate of total nitrogen (TN) increased by 9. 36% under natural oxygenation compared with that under aeration; the single-factor test of carbon/nitrogen ratio (C/N) and hydraulic retention time (HRT) determined C/N=4. 27 and HRT=1. 0 h as the optimal working conditions of the filter cell; the highthroughput sequencing results further showed that the percentages of Pannonibacter and Pseudomonas in the denitrifying biofilter decreased from 0. 49% and 2. 56% at the beginning of operation to 0. 073% and 0. 18% after 130 days of operation. A stable denitrification system was formed with Thauera, Dechlorobacter, Exiguobacterium and other genera.

Abstract:Purifying tank as a compact biofilm reactor has been widely used to remove organic matter in rural sewage, but its potential for nitrogen removal is rare to be discussed. This study developed a lab-scale compact biofilm reactor to realize an efficient nitrogen removal performance by step-feed, intermittent aeration, and immobilization technique. The results show that an efficient simultaneous nitrification and denitrification (SND) process took place by feeding with synthetic wastewater under high C/N ratio of 2 and with real sewage as well, mainly due to the step-feed. The average removal efficiency of total inorganic nitrogen arrived at 72.7 and 63.3% for synthetic wastewater and real sewage, respectively. Besides the step-feed operation, the intermittent aeration was adopted to enhance SND, which allowed hydraulic behavior of compact biofilm reactor following the model of completely stirred tank reactor. The high-throughput sequencing analysis indicates that Sphaerotilus became the dominant genera with relative abundance of 30.29% under high C/N ratio, and the nitrifiers were not greatly inhibited. Moreover, the immobilization technique helped restore microbial activity under low temperature, promoting the satisfactory nitrogen removal performance of recovered microorganism to be rebuilt by feeding nutrient solution. Overall, the long-term SND process and maintaining effective biofilm activity can be established in the compact biofilm reactor through several improving alternatives.

Abstract:Mixed strains Delftia sp.YH01 and Acidovoiux sp.YH02, with capability of heterotrophic nitrification-aerobic denitrification, were introduced into a two-stage aerobic sequencing batch reactor to enhance NO3--N removal. With optimal C/N of 8, efficient NO3--N removal was achieved at initial NO3--N concentration of 2000 mg.L-1. Meanwhile, the massive accumulation of NO2--N was avoided during the long operation. Compared to the one-stage aerobic sequencing batch reactor, the removal efficiency of NO3--N and TN in the two-stage aerobic sequencing batch reactor was increased by 36.5% and 42.7%, which respectively was 93.8% and 88.4%. Microbial community study showed that the mixed strains have the stronger viability and can synergistically denitrify with the indigenous microorganisms in system, such as Azoarcus, Uncultured Saprospiraceae, Thauera, Paracocccus, which could be major contributors for aerobic denitrification. The proposed technology was shown to achieve high-efficiency treatment of high NO3--N wastewater through aerobic denitrification.
