Abstract:Lotaustralin from the root of Rhodiola rosea L was extracted using supercritical carbon dioxide with methanol as modifier. Response surface methodology using Box-Behnken experimental design was utilized to explore parameters for supercritical carbon dioxide extraction. The effects of various values of temperatures (50-70 degrees C), pressures (200-400 bar), and percentages of methanol modifier (80%-100%) on the extraction yields of lotaustralin were evaluated. Extract identification was performed using high-performance liquid chromatography. The experimental data obtained were fitted to second-order polynomial equations and assessed using analysis of variance. The highest yields predicted from the experiments were 2.05 g kg(1) lotaustralin at the optimal values, i.e. temperature 65 degrees C, pressure 316 bar, 88% methanol modifier at a flow rate of 0.4 mL min(1), and dynamic extraction time 90 min.
On-line monitoring of total saponins in extracting process of Astragali Radix

Abstract:Objective: To investigate the effective components dissolution using near-infrared (NIR) spectroscopy for the on-line monitoring of total saponins in the extracting process of Astragali Radix. Methods: The extracts and their NIR spectra were collected using a long-distance fibre-optics flow cell in the extracting process of Astragali Radix, and the contents of four compounds, astrgaloside II, isoastragaloside II, astragaloside I, and astragaloside IV, were determined as control values with HPLC-MS method. The quantitative analysis models of the above four compounds and total saponins in Astragali Radix were established using partial least square method. The established quantitative analysis model of total saponins in Astragali Radix was applied to the on-line monitoring in the extracting process of Astragali Radix to reveal the concentration of total saponins in the extracting process. Results: The correlation coefficients of the four compounds reached 0.9876, 0.9640, 0.8571, and 0.9816, respectively, and 0.9932 for the total saponins. The root mean square error of calibration (RMSEC) was 5.94, the correlation coefficient of inner cross-validation was 0.9766, and root mean square error of cross validation (RMSECV) was 11.1. Using the calibration model of the total saponins for the on-line monitoring of Astragali Radix extracting process, the dissolving information of the total saponins was obtained synchronously and exactly. Conclusion: This method is beneficial to the estimating of the end point and the optimizing of the process parameters.

Abstract:Objective To optimize the extraction process of water soluble alkaloid from Rauwolfia vomitoria Afzel.Methods Researched the influences of solvent volume,extracting time and solvent pH value on extraction techniques for Rauwolfia vomitoris Afzel.With the water soluble alkaloids in extracts as index,the optimal condition by L9(34) orthogonal test was studied.The data obtained were analyzed for GLM using SAS 8.0 software package.Results The optimum process conditions were determined as follows: adding 10 times amount of 95% industrial alcohol(pH=2) extracting 5 hours.Conclusion This extraction technology is simple,reasonable,time efficient with high yielding(12.8%).

Abstract:The enzyme-assisted extraction of rosemary acid from Perilla frutescens was optimized applying one-factor-at-a-time method (OFAT) and response surface methodology (RSM) and the antioxidant activity of the rosemary acid was investigated. Preliminary investigations were conducted into the effect of enzyme dosage, hydrolysis temperature, time and pH on the extraction yield of rosemary acid. Furthermore, the optimization of process parameters for extracting rosemary acid by cellulase hydrolysis of Perilla frutescens was done by RSM using Box Behnken design. Meanwhile, the antioxidant activity was assessed by superoxide anion and 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging assays. The optimal conditions for rosemary acid extraction were determined as follows:enzyme dosage, 3%;hydrolysis temperature, 45℃;time, 12 min;and pH, 4. The maximum extraction yield of rosemary acid of 0.617%was obtained in experiments conducted under these optimal conditions, being not significantly different from the predicted value (0.621%). The rosemary acid showed strong scavenging capacity on superoxide anion and DPPH radicals, thereby having potent antioxidant activity.

Abstract:An orthogonal design with multi-index comprehensive evaluation was adopted to optimize the alcohol precipitation process of Salviae miltiorrhizae water-extraction solution with impurity removal and retention rate of total salvianolic acids as indexes.The optimized process was to add 90% ethanol to 1.20 g/ml of Salviae miltiorrhizae water-extraction solution at a constant rate of 60.9 ml/min and stirring speed of 450 r/min until the final ethanol concentration of 80%.The impurity removal and retention rate of total salvianolic acids were 6.03 g and 67.2%.

Abstract:Objective To optimize the extract conditions for baicalein of Scutellaria baicalensis.Methods The orthogonal design with three levels of four factors including solvent,extracting time,extracting methods and extracting times were adopted,the contents of baicalein were determined by RP-HPLC.Results The optimum extracting conditions were A2B1C2D3,that was cold-maceration,water as the solvents and to extract for 90 min and totally for 4 times.Conclusion The solvents greatly influenced the extraction of baicalein.The extract technology of baicalein is feasible and reasonable.

Abstract:Objective:To optimize extraction technology of Baiqing gel.Method:With the content of paeonol as index,distillation extraction technology of Cynanchum paniculatum was investigated by single factor test;With the content of matrine,berberine hydrochloride and extract yield as comprehensive index,soaking time,the amount of water reflux times and time were chosen as factors,extraction technology of Baiqing gel was optimized by orthogonal test.Result:Distillate was collected with 10 times the amount of C.paniculatum,dregs(after distillation),Phellodendron chinense,Sophora flavescens and other herbal pieces were extracted 3 times with 10 times the amount of water,1.5 h each time.Conclusion:This optimized technology was rational and stable with high extraction rate.

Abstract:To optimize the rose extraction process and evaluate the antioxidant properties of the extracts. The contents of gallic acid and rutin in rose extracts were used as indicators. According to the results of single-factor evaluation of the crushing degree of rose during extraction, extraction method, solid-liquid ratio of extraction and ethanol concentration. The three-factor and three-level L9 (33) orthogonal design optimized extraction scheme was established. The factor levels were as follows: solid-liquid ratio (1:8, 1:10, 1:12); Ethanol concentration of extractant (50%, 80%, 100%); Extraction method (ultrasonic treatment with ice for 30min, 3 times; Ultrasonic treatment with ice for 1h and 3 times; Reflux in water bath for 1h, 3 times); The DPPH in vitro antioxidant test was applied to compare the antioxidant capacity of the optimized extract and rose water extract. The best extraction scheme of rose was as follows: the ratio of solid to liquid was 1:10, the concentration of ethanol was 80%, and the extraction method was ultrasonic for 1 hour and repeated three times. In the antioxidant experiment, it can be found that the antioxidant capacity of the extract is significantly higher than that of the rose water extract. The optimized rose extract has a significantly higher antioxidant capacity than the rose water extract. The establishment of the extraction method significantly improves the utilization rate of antioxidant components in roses. It not only helps to improve the utilization rate of roses and avoids waste, but also helps the development and utilization of rose-related health products.

Abstract:In this study, we optimized the baicalin water extraction process from Scutellaria baicalensis Georgi, Lamiaceae (a traditional Chinese medicine). Orthogonal test design L9(3)4 was used to analyze the optimization of water extraction process of baicalin from S. baicalensis. The effect of solid–liquid ratio, extraction time and soaking time on the yield of baicalin were investigated and optimized by orthogonal test. High-performance liquid chromatography was employed for the determination of extraction yield of baicalin. Analysis of variance was carried out to study the effects of the above three factors. The results showed that solid–liquid ratio plays a significant role in attaining maximum extraction yields of baicalin. However, the other two factors had some effect (not statistically significant) on the extraction yield of baicalin. Conclusively, the optimum experimental conditions such as the solid–liquid ratio (1:12), extraction time (30min) and soaking time (1h) for the water extraction of baicalin were proposed which can provide the maximum extraction yield of baicalin. In addition, the score based on the content of baicalin and total solid residues yield were used as evaluation indexes for baicalin uterus suppositories evaluation.

Abstract:Ethanol precipitation is an important separation and purification process in the traditional Chinese medicines (TCMs) industry. In the present study, a membrane dispersion micromixer was applied to achieve good mixing for the ethanol precipitation process of Astragali radix concentrate. New experimental apparatus was set up to rapidly lower the temperature of ethanol solution before mixing with the concentrate. Ethanol precipitation process was optimized according to Quality by design concept. To identify critical material attributes (CMAs), ten batches of Astragali radix were used to prepare concentrates. Calycosin-7-O-β-D-glucoside content, the sucrose content, and the electrical conductivity were found to be CMAs after the correlation analysis and stepwise regression modelling. Definitive screening design was used to investigate the relationships among critical process parameters, CMAs, and process critical quality attributes (CQAs). Quadratic models were developed and design space was calculated according to the probability of attaining process CQA standards. A material quality control strategy was proposed. High quality and low quality Astragali radix concentrates can be discriminated by the inequalities. Low quality Astragali radix concentrates should not be released for ethanol precipitation process directly. Verification experiment results indicated accurate models and reliable design space. The temperature control method and control strategy are promising for ethanol precipitation process of other TCMs or foods.

Abstract:An activity-based approach to optimize the ultrasonic-assisted extraction of antioxidants from Pericarpium Citri Reticulatae (Chenpi in Chinese) was developed. Response surface optimization based on a quantitative composition-activity relationship model showed the relationships among product chemical composition, antioxidant activity of extract, and parameters of extraction process. Three parameters of ultrasonic-assisted extraction, including the ethanol/water ratio, Chenpi amount, and alkaline amount, were investigated to give optimum extraction conditions for antioxidants of Chenpi: ethanol/water 70:30 v/v, Chenpi amount of 10 g, and alkaline amount of 28 mg. The experimental antioxidant yield under the optimum conditions was found to be 196.5 mg/g Chenpi, and the antioxidant activity was 2023.8 μmol Trolox equivalents/g of the Chenpi powder. The results agreed well with the second-order polynomial regression model. This presented approach promised great application potentials in both food and pharmaceutical industries.

Abstract:OBJECTIVE:To optimize the extraction technology of Hongteng liquid suppository.METHODS:The contents of Paeoniflorin and Berberine hydrochloride were taken as the indexes to optimize the extraction technology.4 influential factors including concentration of alcohol,frequency of extraction,the dosage of solvent and time of extraction were investigated with orthogonal test L9(34).RESULTS:The optimal extraction technique was to extract herbal medicines which consisted of Sargentgloryvine Stem,Paeoniae Radix,Cortex Phellodendri,Caulis Lonicerae and Dahurian Patrinia Herb etc with 80% alcohol at 6 times the weight of herbal material for 3 times,1.5h every time.CONCLUSION:This technology is simple,stable and suitable for industrialized production.

Abstract:Radix Astragali is a famous Chinese traditional and folk medicine with a wide range of medicinal values in clinic. In this study, an analytical efficient strategy based on UHPLC-QQQ-MS/MS and UHPLC-LTQ-Orbitrap-MS/MS was established to explore and reveal the chemical transformations for Radix Astragali under different alkaline wash conditions for analytical sample preparation. Firstly, a rapid and sensitive UHPLC-QQQ-MS/MS method for the quantification of 14 main constituents in Radix Astragali has been developed and validated. Secondly, according to the standard substance comparison, accurate mass measurements, mass fragmentation behaviors and related literatures, a total of 102 components have been screened and identified using UHPLC-LTQ-Orbitrap method. Among them, 47 compounds are saponins, and the other 55 are flavonoids. Consequently, there were two chemical transformations including hydrolysis and degradation observed when Radix Astragali was treated with alkali. Besides, hydrolysis of glycosides and acetyl played a considerably important role in the process of sample preparation. It has been proved that 10 % ammonia could relatively guarantee the high content of astragaloside IV and avoid the over-degradation of most chemical ingredients in Radix Astragali. In conclusion, this work would provide a scientific and practical method for quality control of Radix Astragali as well as its compound preparations.

Abstract:ObjectiveTo investigate the influences of different parameters on the process of alcohol precipitation and to assure the optimal conditions in this study. MethodsThe influences of the concentration of alcohol,the relative density of herb liquor,the speed of adding alcohol and setting temperature on the retenthor rate of active components in Cinobufacini alcohol precipitation process were investigated by single factor experiment.Then an orthogonal test was conducted by the four factors with greater influence.The optimization of technology was performed by using orthogonal design and multi-index compositive score method. ResultsThe optimal process parameters were the relative density of herb liquor 1.10,the final concentration of alcohol 82%,the concentration of alcohol 75% and the settling temperature 10℃. ConclusionThe method is stable and feasible,and has widely industrial application prospect.

Abstract:The purpose of the research was to determine the optimum technology parameters of extraction α-linolenic acid from the Portulaca oleracea L. by using the orthogonal experiment on the basis of signle factor experiment. The extraction ratio of α-linolenic acid from the Portulaca oleracea L. was determined by RP-HPLC.The alternate action of extraction time,solid-liquid ratio,ultrasonic power and its optimum level range were also investigated. Results showed that the order influenced extraction of α-linolenic acid was:extraction time>ultrasonic power>solid-liquid ratio. The optimal extraction conditions of α-linolenic acid from the Portulaca oleracea L.were obtained as:330W of ultrasonic power,1∶20 of solid-liquid ratio,4h of extraction time,hexane as extractant solvent,60℃ of extraction temperature,200 mesh of material size.Under above optimal conditions,the extraction ratio of α-linolenic acid from the Portulaca oleracea L. was 24.302mg/100g.

Abstract:A single-factor experiment for determining the extraction temperature, liquid-to-solid ratio, sonication time, and ethanol concentration was designed to investigate the influence of these parameters on the extraction of adenosine from bamboo shavings. Afour-factor orthogonal test was then carried out to optimize the extraction performance. Results showed that all these parameters significantly influenced the extraction of adenosine. The range and variance analyses based on the orthogonal test revealed that ethanol concentration produced most significant direct effect on the extraction of adenosine, followed by liquid-to-solid ratio and sonication time. The highest yield of adenosine was achieved by extraction at temperature 50°C, liquid-to-solid ratio 15:1 (v/w), sonication time 45 min, and ethanol concentration 0% (v/v). This optimized extraction procedure resulted in 1.42 mg/g adenosine yield and a 96.7% adenosine extraction from bamboo shavings.
Optimization of the extraction process of polysaccharide in Polygonatum and Fructus Lycii with orthogonal test

Abstract:OBJECTIVE To find out the optiminal extraction process of polysaccharide from Polygonatum and Fructus Lycii. METHODS With total polysaccharides and dry extract rate as the parameters for water-extraction,times of extraction,time and the ratio of material to water were used for the optimization of water-extraction technology.RESULTS The optimum extraction process:water doses was 10,8,8×,reflux extraction for 3 times and 1.5 h,1 h,1 h each time respectively.The extraction rate of soluble polysaccharides was 10.98% in the optimum conditions.CONCLUSION The optimum extraction process is stable and feasible.
