Microbial Communities in Petroleum Polluted Soils by PCR-DGGE

Abstract:Microbial communities play an important role in the efficient biodegradation of petrochemicals in polluted soils with competition among degrading bacteria populations in different microeco-systems. Microbial communities in the contaminated soils in Chinese oilfields were evaluated using PCR-DGGE. The studies analyzed the relationship between the microbial eco-system and the degradation abilities of the microbes in the eco-system. The results show that the DNA yields were 3.45~11.7 μg/g calculated by soil mass, with a DNA purity of 1.5~1.87. The total DNA yields were affected by the numbers and activities of the microbes which were functions of the soil petroleum content. The DGGE analyses shows that the microbial diversity in east China soil with the highest water content was 2~5 times higher than in contaminated soil samples in Northwest China. After two months of enhanced bioremediation, the microbial diversity decreased to about 75% of that in Northeast China.
