Abstract:This work presents a sequential approximate optimization method to achieve accurate and efficient optimization of bacteriocin production by Lactobacillus strain, ZJ317. Initially, a Plackett-Burman design study and a determination of the path of steepest ascent were effective in identifying the most significant factors and approaching the optimum region of the response. Response surface methodology (RSM) was then applied to evaluate the effects of different variables on bacteriocin production. The experimental results were fitted to a second-order polynomial model equation. The optimal values of each variable to achieve the theoretical maximum of bacteriocin activity (1742.77 U/ml) were predicted to be 2.0% (w/v) glucose, 1.02% (w/v) yeast extract and 2.06% (w/v) inoculum volume. The observed experimental value of bacteriocin activity under the predicted optimal conditions was 1883.61 U/ml compared to a pre-optimization value of 1037.19 U/ml, thus showing a marked improvement in the efficiency of bacteriocin production.
Optimizing the Fermentation Process for Milk-clotting Enzyme Production by Bacillus Subtilis

Abstract:In order to improve the yield of milk-clotting enzyme(MCE) ,response surface methodology(RSM) was used to optimize the fermentation process for the production of milk-clotting enzyme from Bacillus subtilis under liquid fermentation. The results showed that the optimal parameters for the fermenting process were as follows: glucose 16.2 g/L,53.3 mL medium in a 500 mL Erlenmeyer flask,inoculums size 0.130%,fermenting time 120.43 h,and the predicted MCE yield was 1097.30 SU/mL. Under the optimal conditions the MCE activity was(1129.05 ± 74.55) SU/mL,indicating that the optimizing process is effective.

Abstract:Natural cecropin B (CB), consisting of 35 amino acids, is a member of the cecropin family with the highest antibacterial activity. Here, a novel approach for the generation of recombinant CB in the methylotrophic yeast Pichia pastoris was explored. For this purpose, the CB gene was amplified by recursive PCR (rPCR) and cloned into the pPICZ alpha A vector. The SacI-linearized plasmid pPICZ alpha A-CB was transformed into P. pastoris SMD1168 by electroporation. The expression of recombinant CB was induced with 1.0% methanol at pH 5.0 for 60 h at 28 degrees C. Recombinant CB was purified using cationic exchange chromatography; 5.0 mg of pure active CB was obtained from 100 ml of culture broth supernatant. Antimicrobial assays demonstrated that CB has a broad spectrum of antimicrobial activity against both Gram-positive and Gram-negative bacteria. Our results suggest that the P. pastoris expression system can be used to produce large quantities of fully functional CB for both research and industrial purposes.
Optimization of Fermentation Medium for Production of Antibacterial Peptides by Bacillus Subtilis BSD-2

Abstract:Plackett-Burman design was used to evaluate the inportance of fermentation medium factors of Bacillus subtilis BSD-2 affecting the inhibitory zone diameter of produced antibacterial peptides. Subsequendy, 7 steepest ascent experiments involving 3 most important factors (i.e., peptone, starch and soybean meal) were conducted for approaching their optimal response region.Finally, the optimal fermentation medium formula was obtained by optimizing the most important factors using central composite design combined with response surface methodology. Results indicated that the optimal fermentation medium formula consisted of 14.29 g/L peptone, 14.07 g/L amylum, 6.49 g/L soybean power, 2.0 g/L CaCO_3 and 1.0 g/L MgSO_4. There was a 1.77-fold increase in the yield of antibacterial peptides obtained using the optimal fermentation medium, compared with that before optimization.

Abstract:Response surface methodology, which allows for rapid identification of important factors and optimization of them to enhance enzyme production, was employed here to optimize culture conditions for the production of cis-epoxysuccinic acid hydrolase from Bordetella sp. strain 1-3. In the first step, a Plackett-Burman design was used to evaluate the effects of nine variables (yeast extract, cis-epoxysuccinic acid, KH(2)PO(4), K(2)HPO(4).3H(2)O, MgSO(4).7H(2)O, trace minerals solution, culture volume, initial pH and incubation time) on the enzyme production. Yeast extract, cis-epoxysuccinic acid and KH(2)PO(4) had significant influences on cis-epoxysuccinic acid hydrolase production and their concentrations were further optimized using central composite design and response surface analysis. A combination of adjusting the concentration of yeast extract to 7.8 g/l, cis-epoxysuccinic acid to 9.8 g/l, and KH(2)PO(4) to 1.12 g/l would favor maximum cis-epoxysuccinic acid hydrolase production. An enhancement of cis-epoxysuccinic acid hydrolase production from 5.6 U/ml to 9.27 U/ml was gained after optimization.

Abstract:Strains producing β-glucosidase were isolated by solid medium containing esculin and ferric.Then six strains having high β-glucosidase activity were selected with p-nitropheny l-β-D-glucopyranoside as substrate.Strain having high ginsenoside bioconversion was finally chosen with Rbl as substrate and converted products were determined by TLC and HPLC.β-glucosidase activity of this strain was 0.803 U/mL and ginsenoside Rd conversation rate was 97.234 2％.The selected strain was identified as Saccharomycopsisfibuligera based on the morphology and nucleotide sequence analysis of the 18S rDNA.Fermentation medium was optimized to contain lactose as carbon source and corn flour as nitrogen source,and Ca2+ as metal ion.Finally inducer was optimized to promote the ginsenoside bioconversion.Results indicated that inducer had little impact on the bioconversion but rutin,a kind of inducer,could promote the bioconversion of Rd to CK.The selected strain with high ginsenoside bioconversion had good potential application.

Abstract:Fermentation process of cephalosporin C was studied. The DO (dissolved oxygen) tension, CER (carbon dioxide evolution rate), OUR (oxygen uptake rate) and RQ (respiratory quotient) were monitored on line and the specific growth rate, specific production rate were analyzed. The highest CER and OUR are 90.2 mol/(m3·h) and 69.8 mol/(m3·h) respectively. It reveals that cephalosporin C biosynthesis is a high oxygen demanding process. The specific production rate reaches its highest value at 60-100 h. At the same time, methionine was analyzed off line. For cephalosporin C biosynthesis it is essential to keep methionine concentration at certain level in the broth. By controlling the biomass at the earlier stage of fermentation under the existing equipments and air compression capacity, the fermentation with twice precultures is found to be superior to that with thrice precultures in terms of productivity.

Abstract:Single factor test and orthogonal chart were used to optimize the fermentation medium of Bacillus pumilus TY079.The results indicated that the medium was,glucose 0.5%,cornmeal 0.5%,peptone 1.5%,soybean power 0.5%,KH2PO4 0.75%,MgSO4·7H2O 0.2%.The mean size of inhibition zone increased to 1.42 times of the original.

Abstract:[Background] Cecropin is extensively studied and which is an effective antimicrobial peptide. It will lay the foundation for the practical application of cecropin in aquaculture and agriculture to realize the commercial production of cecropin. [Objective] To obtain a genetically engineered strain for efficient production of cecropin AD. [Methods] The recombinant vector pGAPZαA-CAD was firstly constructed and then transformed to P. pastoris X33 strain by electric shock. Cecropin AD gene was successfully expressed and X33/GCAD strain was obtained. Secondly, the recombinant vector pUCGAP-CAD was constructed and transformed into X33/GCAD strain. The pGAPZαA-CAD plasmid was integrated into the GAPDH promoter region of P. pastoris X33 with Zeocin as resistance screening label, while pUCGAP-CAD plasmid was integrated into the non-translated rDNA region of P. pastoris X33 with Geneticin as resistance screening label. Finally, a recombinant X33/GUCAD strain with an efficiently expressed cecropin AD gene was obtained. [Results] The antimicrobial compound of X33/GUCAD was cecropin AD which was identified by mass spectrometry. Afterward optimizing its fermentation conditions, we found that the X33/GUCAD strain having higher potential to express cecropin AD by consuming glycerol as a sole carbon source while peptone and yeast extract for organic nitrogen source. [Conclusion] The higher copy number is more beneficial to increase the yield of cecropin AD, and the engineering strain is more stable in the later fermentation process and suitable for industrial production.
Effect of Fermentation Substrate on the Production Rule of Subticin 112 and Optimization of Fermentation Parameters

Abstract:This paper mainly focused on the optimization of fermentation parameters of subticin 112 and elucidating the effect of different media on the production of bacteriocin,to minimize the organic nitrogen content,for the procedure and cost reduction of isolation and purification and offering theoretical information for the efficient improvement of bacteriocin production.The optimum medium components were evaluated by fractional factorial designs(FFD) and central composite design(CCD) response surface methodology(RSM).Compasion among different media was deserved in the sign of biomass,antimicrobial activity,specific production rate and specific growth rate.The results showed that the content of organic nitrogen was reduced by 74.3%,while total nitrogen by 56%.Antibacterial activity was 1.7-fold higher than the un-optimized one,to be equivalent to 163.8 mg/L penicillin.Maximum specific production rate was improved by 184%.The intimate relationship of Mn+ and subticin 112 biosynthesis was identified.Subticin 112 was produced in the term of mixed metabolite kinetics.
Improved Elastase Production by Bacillus Sp. EL31410--further Optimization and Kinetics Studies of Culture Medium for Batch Fermentation.

Abstract:An efficient culture medium producing a bacterial elastase with high yields was developed further following preliminary studies by means of response surface method. Central composite design (CCD) and response surface methodology were applied to optimize the medium constituents. A central composite design was used to explain the combined effect of three medium constituents, viz, glucose, K2HPO4, MgSO4·7H2O. The strain produced more elastase in the completely optimized medium, as compared with the partially optimized medium. The fitted model of the second model, as per RSM, showed that glucose was 7.4 g/100 ml, casein 1.13 g/100 ml, corn steep flour 0.616 g/100 ml, K2HPO4 0.206 g/100 ml and MgSO4·7H2O 0.034 g/100 ml. The fermentation kinetics of these two culture media in the flask experiments were analyzed. It was found that the highest elastase productivity occurred at 54 hours. Higher glucose concentration had inhibitory effect on elastase production. At the same time, we observed that the glucose consumption rate was slow in the completely optimized medium, which can explain the lag period of the highest elastase production. Some metal ions and surfactant additives also affected elastase production and cell growth.

Abstract:Plackett-Burman experiment was used to evaluate the optimal level of 8 factors in Streptomyces fungicidious ERS42-101108 Enramycin initial fermentation medium production , results showed that cottonseed meal , gluten meal and KH 2 PO4 had significantly effect on enramycin production .Steepest ascent was used to approach the key factors of the maximum response region , and by central composite design and response surface , optimum fermentation medium was established .Following the above optimization process , fermentation unit Enramycin increased from 8300 u/mL to 10250 u/mL.

Abstract:By shake flask fermentation,the fermentation medium for the production of protease by Bacillus subtilis BS3 was optimized by response surface method.Based on single factor experiments,Plackett-Burman experiment(PB) was used to analyze the effects of medium components on protease production.The main impact factors were determined as corn steep liquor,ammonium chloride and corn starch.Subsequently,in order to define the optimal composition of the fermentation medium,the three factors were further optimized using steepest ascent experiment,central composite design and response surface analysis.The optimized medium was as follows: corn steep liquor 1.586g/100g,corn starch 3.170g/100g,ammonium chloride 1.998 g/100g,and other components which were added at a low level.With the optimized medium,the viable count and protease activity were 12.640×109cfu/g and 162.309u/g,respectively.

Abstract:The fermentation medium of Saccharopolyspora erythraea for erythromycin production was optimized by response surface methodology (RSM). Minimum Run Equireplicated Res IV design was used to evaluate the influence of six factors added in the initial fermentation medium. Four factors playing important roles were screened out, including MgSO 4, Betaine, CuSO 4 and CoCl 2. Then the steepest ascent path was employed to determine the optimal region of the medium composition for the further central composite design. The optimum concentration of MgSO 4, Betaine, CuSO 4 and CoCl 2 were determined to be 0.106% (w/v), 0.0185% (w/v), 0.106 mmol/L and 0.0003% (w/v) respectively by response surface analysis. Under the optimized medium the erythromycin yield was increased by 30% compared with the control.

Abstract:The fermentation conditions of recombinant Escherichia coli for cell growth and β1-3,1-4-glucanase production were investigated.The optimized medium was obtained: yeast extract 12g/L,glycerol 6g/L,NaCl 10g/L,NaNO_3 7.21g/L,KH_2PO_4 2.4g/L,K_2HPO_4 12.5g/L.The maximum of 297.71U/mL β-glucanase activity was obtained almost 14.3 times higher than that of the initial conditions.

Abstract:In order to increase the prodution of coenzyme Q10,coenzyme Q10-producing strain R-SL15 was used to obtain from the optimum fermentation medium.Plackett-Burman design and Box-Behnken response surface methodology(RSM) was used to optimize medium for coenzyme Q10 biosynthesis.According to the RSM analysis on the data of Box-Behnken design,optimal fermentation conditions for the highest coenzyme Q10 content in fermentation medium were set at glucose content of 18.90g/L,yeast extract content of 5.54g/L,(NH4)2SO4 content of 0.98g/L、KH2PO4 content of 0.95g/L,beef extract content of 6g/L,MgSO4·7H2O content of 0.75g/L,FeSO4·7H2O content of 100mg/L,NaCl content of 10g/L and distilled water of 1L.Under the optimal conditions,coenzyme Q10 content produced by R-SL15 could reach to 51.31mg/L,which was 99.34% higher than that of non-optimized medium which was 25.74mg/L.The regression model was highly significant(R2=0.9423) and showed good fitness to the experimental data,and it could be used to predict.
Production and optimization studies of cephalosporin C by solid state fermentation

Abstract:Production of cephalosporin C employing a strain, Cephalosporium sp. NCIM 1039 under solid state fermentation was optimized. Different substrates like wheat bran, wheat grains, rice grains, barley and rice bran were studied to optimize the best substrate. Wheat bran showed the highest antibiotic yield. The physical and chemical parameters were optimized. The maximum productivity of cephalosporin C (750 U/g) was achieved by employing wheat bran and with optimized nutritional and process parameters such as potato starch as additive 1% w/w, urea as additive 1% w/w, incubation period of 7 days, incubation temperature at 30 degrees C, inoculum level 10% w/v, moisture content of solid substrate 80% and pH 7.0.

Abstract:ABSTRACT: Probiotic bacteria, such as some strains of Clostridium butyricum, have been frequently used as the active ingredient in functional foods. However, slow growth of the probiotic bacteria has been one of the big concerns for their potential commercial application. In the present study, a fractional factorial design was applied to investigate the main factors, namely, the concentrations of the glucose, tryptone, yeast extract, beef extract, cys‐teine, (NH4) 2SO4, NaCl, K2HPO4, and the medium initial pH that affected the growth of 1 probiotic strain, C. butyricum ZJUCB, currently preserved in our laboratory. Central composite experimental design and response surface methodology were adopted to derive a statistical model for optimizing the composition of the medium. The experimental results showed that the optimum medium for incubating the C. butyricum ZJUCB was composed of 2.44% (w/v) glucose, 2.08% yeast extract (w/v), 1% tryptone (w/v), 0.1% (NH4)2SO4 (w/v), 0.1% NaHCO3 (w/v), 0.02% MnSO4. H2O (w/v), 0.02% MgSO4. 7H2O (w/v), 0.002% CaCl2 (w/v), and 2% agar (w/v) (if necessary) at pH 8.55. After incubation for 24 h in the optimum medium, the populations of the viable organisms could reach 109 colony‐forming units (CFU)/mL, which was 100 times higher than that incubated in the initial medium.

Abstract:ABSTRACT: Nisin is an effective food biopreservative widely used in food industry. However, 1 problem of concern is limited production rate and final nisin concentration. A nisin‐producing strain, L. lactis Lac2, a mutant strain with high yield of nisin, was obtained in our laboratory recently. In the present study, a fractional factorial design was applied to investigate the main factors that affect the yield of L. lactis Lac2. Central composite experimental design and response surface methodology were adopted to derive a statistical model for optimizing the composition of the medium. The results showed that the optimum medium for nisin production of L. lactis Lac2 was composed of 2.68% sucrose (w/v), 0.5% tryptone (w/v), 1% yeast extract (w/v), 0.3% Tween‐80 (w/v), 0.02% MgSO4·7H2O (w/v), 0.81% NaCl (w/v), 1.91% K2HPO4 (w/v), 0.05% ascorbic acid (w/v), and 2% agar (w/v) (if necessary) at pH 6.5. When cultured in the optimum medium, the nisin yield is an average of 3381.81 IU/mL, which nearly doubled the yield when incubated in the initial medium. Also, the concentration of tryptone was decreased while that of the sucrose was increased when compared with CM broth, which means a reduction of the fermentation cost.

Abstract:The optimization of medium components for the production of clavulanic acid by Streptomyces clavuligerus was performed using both factional factorial design and response surface methodology. The factional factorial design experiment was initially employed for this purpose and it was found that soy meal powder, FeSO4·7H2O, and ornithine were the most important medium components (confidence levels above 85%). Response surface methodology for optimization was further investigated. These three composing elements showed significant effects on producing clavulanic acid. Their optimal concentrations were: soy meal powder 38.102g/l, FeSO4·7H2O 0.395g/l, and ornithine 1.177g/l. The yield of clavulanic acid was increased by 50% by culturing strain S. clavuligerus in the nutritionally optimized fermentation medium.
