Antimicrobial Resistance Analysis of Campylobacter Jejuni Isolates from Chicken Production Chain

Abstract:Campylobacter jejuni is an important foodborne pathogen,which cause gastroenteritis and invasive disease may also occur.In order to study the antimicrobial resistance,the antibiotic profile,and the relationship between antimicrobial resistance phenotype and gyrA gene mutation,we investigated 180 C.jejuni isolates from poultry yard,slaughterhouse and markets of Chengdu and Ya'an cities for susceptibility against eight antimicrobial agents by plate micro-dilution method.Moreover,ciprofloxacin-resistant and ciprofloxacin-susceptible C.jejuni isolates were examined for the point mutation of gyrA gene at DNA sequence position 257 by mismatched amplification mutation assay(MAMA) PCR.The results indicated that the resistant rates for C.jejuni isolates were 94.44%,93.89%,91.67% and 78.33% against ciprofloxacin,levofloxacin,tetracycline and clindamycin respectively,showing different antimicrobial resistance to erythromycin(20.00%),streptomycin(20.56%),gentamicin(17.78%) and florfenicol(12.22%).The 95.62%(131/137) of the ciprofloxacin-resistant C.jejuni had a substitution of the DNA codon from ACA(threonine) to ATA(isoleucine).However,10 ciprofloxacin-susceptible isolates had no mutation at the same position.The sequencing results confirmed that ciprofloxacin-resistant isolates(MJF31 and MJF53) had a substitution at amino acid position 86 of the gyrA protein due to a mutation of the DNA codon from ACA(threonine) to ATA(isoleucine).Nevertheless,the same mutation did not occur in the ciprofloxacin-susceptible isolates(JF9 and JF136).The study provided evidence for surveillance of the antimicrobial resistance of C.jejuni,and additionally provided reference for the mechanism of antimicrobial resistance.
