Abstract:This paper examines the spillover effects of promotions when products from different firms are consumed in a bundle. Using data from the HIV/AIDS category, a canonical example of combination therapy, we estimate a hierarchical Bayesian logit model across treatment regimens and show that detailing for one drug can increase demand for other drugs that are often combined with the focal drug. Such spillover effects could lead to free riding by the drugs benefitting from the spillover. We investigate the managerial and policy implications of detailing spillover effects via counterfactual policy simulations based on a dynamic oligopoly game of detailing. For managers, we show how firms can internalize the spillover effects and reduce the incentive for free riding. For policy makers, the implications of our findings relate to detailing restrictions that are often proposed on branded drugs. These restrictions aim to increase social welfare by encouraging the use of generic drugs. However, in combination therapies generic drugs may actually benefit from the detailing of complementary branded products. If detailing was curtailed, this could adversely affect generic drug prescriptions as well, which runs counter to policy makers' objectives of encouraging use of generic drugs.Data and the online appendix are available at https://doi.org/10.1287/mksc.2016.1014.
Comparative Marketing Communication: the Case of Drug Detailing

Abstract:Comparative marketing communication has emerged as an important area of managerial and scholarly inquiry. In the pharmaceutical industry, it has been practiced in detailing, the personal selling to physicians. With physician-level panel data of detailing and prescriptions, we examine the effectiveness of comparative detailing versus noncomparative detailing. In particular, we investigate whether a brand’s comparative detailing directly damages competing brands or provides them free exposure to physicians. The Bayesian hierarchical probit model with reduced-form detailing policy functions allows us to examine comparative detailing at both aggregate level and individual physician level, while controlling for possible simultaneity issues in noncomparative detailing and comparative detailing. We find that (1) comparative detailing is less or equally effective than noncomparative detailing at the aggregate level, but there is strong heterogeneity across individual physicians; (2) some brands strategically focus their comparative detailing efforts on physicians who are less responsive to noncomparative detailing but more responsive to comparative detailing than others; (3) the market leader faces a denigrating loss if underdog brands compare against it in their detailing visits, but a generic brand enjoys a free-exposure benefit when other brands in the category compare against it in their detailing visits. Our estimates provide rich managerial implications by identifying physicians who are more profitable for comparative detailing. Finally, our policy simulation shows that a ban of comparative detailing can reduce the market share of generic drugs by 1.76% in statin market.

Abstract:Drug treatment services of varying types have been scaled up in China over the past decade. Yet, barriers to treatment remain among the population of drug users in China. In this paper, we use a person-centered approach to examine external barriers to drug treatment among a sample of Chinese drug users. Specifically, we used a latent class analysis to determine a typology of external barriers to treatment among a sample of 262 drug users. The results of the analyses suggest three-classes of drug users with respect to their perceptions of external barriers to treatment--Major Barriers, Low Barriers, and Systems-level Barriers--indicating that drug users are a heterogeneous population on this matter. Age and types of drugs used were predictors of class membership. In this regard, different tactics must be utilized in order to successfully reach this wide ranging group of individuals.

Abstract:This paper develops a simple dynamic oligopoly model and a quick estimation procedure for empirical applications.I develop a simple dynamic oligopoly model for empirical work. A unique feature of the model is that any Markov-perfect equilibrium that survives some intuitive refinements can be quickly computed from low-dimensional contraction mappings in seconds. After computation, it is easy to check the uniqueness of the refined equilibrium. These results facilitate fast estimation using a full-solution approach and produce reliable counterfactual analysis. Model estimation at its minimum only requires panel data on firm presence, yet it quantifies important market primitives, such as toughness of competition and entry costs. I provide a step-by-step illustration of the estimation approach by applying it to the Dutch retail grocery industry, in which chain stores slowly replace local stores. A counterfactual simulation computing equilibrium under a large number of different primitive values shows that relaxing restrictions on chain-store entry will not only quicken the destruction of local stores, but also hurt chain stores’ profits. Abstract:Based on previous work done in this field, we build a dynamical system that describes changes in drug addiction in an isolated population when two addictive substances are available simultaneously. We then use our model to investigate whether the system captures the process of users switching drug habits. One of the motivations for this project is to mathematically check the conjecture that being addicted to a less-addictive substance will effectively lead individuals to become dependent on more addictive and potentially more dangerous drugs. We introduce additional assumptions, under which our model is reduced to a competitive Lotka-Volterra system. This dynamical system has three or four fixed points, stability of which then gives an implication about the outcomes of the competition between addictive substances and, therefore, the fate of individuals in the population. From the analysis of the reduced model, we determine that there actually exist parameter regimes that capture the following dynamics: depending on the initial distribution of the drug preference; either the use of both drugs will die out, the usage of one of the drugs will become prevalent, or addictions to both drugs will coexist.

Abstract:With the cost/yield-ratio of drug development becoming increasingly unfavourable, recent work has explored machine learning to accelerate early stages of the development process. Given the current success of deep generative models across domains, we here investigated their application to the property-based proposal of new small molecules for drug development. Specifically, we trained a latent diffusion model - DrugDiff - paired with predictor guidance to generate novel compounds with a variety of desired molecular properties. The architecture was designed to be highly flexible and easily adaptable to future scenarios. Our experiments showed successful generation of unique, diverse and novel small molecules with targeted properties. The code is available at https://github.com/MarieOestreich/DrugDiff.

Abstract:Abstract The current drug development is a very expensive and prolonged process with high attrition rate, especially in the oncology drug development. New tools with dynamic and quantitative modeling and experimental support are needed to accelerate the drug discovery process and increase productivity. The process begins with building a quantitative model of biological regulatory system based on the extensive data-mining from literature and experiments. By incorporating the preclinical PK/PD data relevant to the target system, a new computational model under drug perturbation can be built. Through iterative process of hypotheses generating and experimental testing, such models can be fine-tuned. Consequences of particular perturbations, such as optimal dosage and treatment schedules, optimal drug target, and combination therapy can be simulated in time course using such models. A better understanding of the biological system will be gained and better therapeutic interventions suggestions can be provided. In this study, we use NF-κB pathway as an example study considering NF-κB pathway has become a focal point for intense drug discovery and development efforts. Dynamic drug perturbation is incorporated in the model stem from Hoffmann et al. (science 2002 Nov 8;298) through hybrid system theory to link dose – concentration relationships (PK) and concentration – effect relationships (PD). This approach facilitates the description and prediction of the time course of drug effects resulting from a certain dose regimen. Extensive simulations of the proposed model are performed in MATLAB/SIMULINK. An important observation is that there exists a “sweet spot” for certain dosage and frequency arrangements given the totaled delivered drug. The perturbation effect for different targets is simulated and compared in this model. However, the specificity of some drugs to inhibit several of these components of the NF-κB pathway is one of the concerns. For example, if we block the degradation pathway of IκB by using proteasome inhibitor (such as Bortezomib) to inhibit ubiquitination of IκB by the 26S protesome. The side effects are unacceptable when the drug is effective. Hence, we design combination therapy to induce a acceptable combined effect and using lower doses for each drugs. We observe that some combinations of the drugs with non-overlapping toxicities might provide enormous benefit. While such dynamical modeling approaches are still in their infancy, advancing techniques of the quantitative single live cell measurement of the protein processes will provide us more accurate parameters and help us to test and confirm some observations. This will allow a better understanding of the kinetics and function of mechanisms within regulatory networks. The authors believe that appropriate models can help establishing a mechanistic understanding of the disease and of drug action instead of the traditional “black-box” approach in drug discovery. Abstract:Problems with drug ADME are responsible for many clinical failures. By understanding the ADME properties of marketed drugs and modeling how chemical structure contributes to these inherent properties, we can help new projects reduce their risk profiles. Kinetic aqueous solubility, the parallel artificial membrane permeability assay (PAMPA), and rat liver microsomal stability constitute the Tier I ADME assays at the National Center for Advancing Translational Sciences (NCATS). Using recent data generated from in-house lead optimization Tier I studies, we update quantitative structure–activity relationship (QSAR) models for these three endpoints and validate in silico performance against a set of marketed drugs (balanced accuracies range between 71% and 85%). Improved models and experimental datasets are of direct relevance to drug discovery projects and, together with the prediction services that have been made publicly available at the web portal (<a class="ext-link" href="https://opendata.ncats.nih.gov/adme/">https://opendata.ncats.nih.gov/adme/</a>), provide important tools for the drug discovery community. DrugGym: A testbed for the economics of autonomous drug discovery

Abstract:Drug discovery is stochastic. The effectiveness of candidate compounds in satisfying design objectives is unknown ahead of time, and the tools used for prioritization---predictive models and assays---are inaccurate and noisy. In a typical discovery campaign, thousands of compounds may be synthesized and tested before design objectives are achieved, with many others ideated but deprioritized. These challenges are well-documented, but assessing potential remedies has been difficult. We introduce DrugGym, a framework for modeling the stochastic process of drug discovery. Emulating biochemical assays with realistic surrogate models, we simulate the progression from weak hits to sub-micromolar leads with viable ADME. We use this testbed to examine how different ideation, scoring, and decision-making strategies impact statistical measures of utility, such as the probability of program success within predefined budgets and the expected costs to achieve target candidate profile (TCP) goals. We also assess the influence of affinity model inaccuracy, chemical creativity, batch size, and multi-step reasoning. Our findings suggest that reducing affinity model inaccuracy from 2 to 0.5 pIC50 units improves budget-constrained success rates tenfold. DrugGym represents a realistic testbed for machine learning methods applied to the hit-to-lead phase. Source code is available at www.drug-gym.org.

Dynamics of drug trafficking: Results from a simple compartmental model

Abstract:In this work we propose a simple model for the emergence of drug dealers. For this purpose, we built a compartmental model considering four subpopulations, namely susceptibles, passive supporters, drug dealers and arrested drug dealers. The target is to study the influence of the passive supporters on the long-time prevalence of drug dealers. Passive supporters are people who are passively consenting to the drug trafficking cause. First we consider the model on a fully-connected newtork, in such a way that we can write a rate equation for each subpopulation. Our analytical and numerical results show that the emergence of drug dealers is a consequence of the rapid increase number of passive supporters. Such increase is associated with a nonequilibrium active-absorbing phase transition. After that, we consider the model on a two-dimensional square lattice, in order to compare the results in the presence of a simple social network with the previous results. The Monte Carlo simulation results suggest a similar behavior in comparison with the fully-connected network case, but the location of the critical point of the transition is distinct, due to the neighbors' correlations introduced by the presence of the lattice.

Abstract:We take on two subjects of controversy among economists-advertising and trademarks-in the context of the market for generic drugs. We outline a model in which trademarks for drug names reduce search costs but increase product differentiation. In this particular framework, trademarks may not benefit consumers. In contrast, the generic names of drugs or "International Nonproprietary Names" (INN) have unquestionable benefits in both economic theory and empirical studies. We offer a second model where advertising of a brand-name drug creates recognition for the generic name. The monopoly patent-holder advertises less than in the absence of a competitive spillover.
DecompDiff: Diffusion Models with Decomposed Priors for Structure-Based Drug Design

Abstract:Designing 3D ligands within a target binding site is a fundamental task in drug discovery. Existing structured-based drug design methods treat all ligand atoms equally, which ignores different roles of atoms in the ligand for drug design and can be less efficient for exploring the large drug-like molecule space. In this paper, inspired by the convention in pharmaceutical practice, we decompose the ligand molecule into two parts, namely arms and scaffold, and propose a new diffusion model, DecompDiff, with decomposed priors over arms and scaffold. In order to facilitate the decomposed generation and improve the properties of the generated molecules, we incorporate both bond diffusion in the model and additional validity guidance in the sampling phase. Extensive experiments on CrossDocked2020 show that our approach achieves state-of-the-art performance in generating high-affinity molecules while maintaining proper molecular properties and conformational stability, with up to -8.39 Avg. Manifold-Constrained Nucleus-Level Denoising Diffusion Model for Structure-Based Drug Design

Abstract:Artificial intelligence models have shown great potential in structure-based drug design, generating ligands with high binding affinities. However, existing models have often overlooked a crucial physical constraint: atoms must maintain a minimum pairwise distance to avoid separation violation, a phenomenon governed by the balance of attractive and repulsive forces. To mitigate such separation violations, we propose NucleusDiff. It models the interactions between atomic nuclei and their surrounding electron clouds by enforcing the distance constraint between the nuclei and manifolds. We quantitatively evaluate NucleusDiff using the CrossDocked2020 dataset and a COVID-19 therapeutic target, demonstrating that NucleusDiff reduces violation rate by up to 100.00% and enhances binding affinity by up to 22.16%, surpassing state-of-the-art models for structure-based drug design. We also provide qualitative analysis through manifold sampling, visually confirming the effectiveness of NucleusDiff in reducing separation violations and improving binding affinities.

Abstract:The drug development pipeline involves several stages including in vitro assays, in vivo assays, and clinical trials. For candidate selection, it is important to consider that a compound will successfully pass through these stages. Using graph neural networks, we developed three subdivisional models to individually predict the capacity of a compound to enter in vivo testing, clinical trials, and market approval stages. Furthermore, we proposed a strategy combing both active learning and ensemble learning to improve the quality of the models. The models achieved satisfactory performance in the internal test datasets and four self-collected external test datasets. We also employed the models as a general index to make an evaluation on a widely known benchmark dataset DEKOIS 2.0, and surprisingly found a powerful ability on virtual screening tasks. Our model system (termed as miDruglikeness) provides a comprehensive drug-likeness prediction tool for drug discovery and development.

Abstract:The objective of this paper is to provide two-stage game models explaining the 'Generic Competition Paradox' that demonstrates an increase of brand-name drug price in response to generic entry. Under the assumption that there are two groups of consumers who are segmented by their insurance status, high insurance coverage and low insurance coverage consumers, the models indicate that the decisive factor is the market share of the high insurance coverage consumer and the size of cross-substitute factor relative to certain characteristics of market demand. The paper analyses both the case of only true generic entry and the case of pseudo-generic and true generic entry. The models prove that a brand-name price will increase when both the market share of high insurance coverage consumer and the factor of cross-substitute are small. Also, the 'Generic Competition Paradox' more likely occurs in the market where less pseudo-generic products are produced.

Abstract:China has emerged as a major global player in the pharmaceutical industry and a significant consumer of drugs. However, the rapid growth of the raw material pharmaceutical manufacturing sector has resulted in a substantial amount of waste generation, leading to environmental pollution issues. In order to mitigate the pollution problems associated with the pharmaceutical manufacturing industry, this research proposes a tripartite game model with two dynamic parameters and develops optimization models for green supply chains under three decision-making scenarios, aiming to contribute to the efforts for achieving low-carbon development. Additionally, the study considers the government's green subsidy system and incorporates the optimization of green pharmaceutical practices and goodwill of the enterprises as dynamic parameters. By employing differential game methods, equilibrium solutions among the three parties are determined. The research findings reveal that: firstly, the green pharmaceutical levels are equivalent among the three models; secondly, in scenarios where retailers receive subsidies and make decentralized decisions, API manufacturers achieve the highest level of goodwill and the supply chain profit is maximized; thirdly, when manufacturers have sufficiently high marginal profits, adopting a decentralized decision-making approach encourages retailers and enhances the overall goodwill level of the supply chain. This study provides valuable insights for the formulation of effective strategies and policies to promote sustainable development within the pharmaceutical industry.

Abstract:Introduction: Generic substitution is encouraged to reduce pharmaceutical spending in China, and with incentive policies, the market size of the generic drug continues to rise. To find out how the generic competition affects drug price in this area, this study examines how the quantity of generic drug manufacturers can influence average drug price in the Chinese market. Methods: This study uses a rigorous selection of drugs from the 2021 China's National Reimbursement Drug List (NRDL), and uses drug-level fixed effects regressions to estimate the relationship between competition and price within each drug. Results: We note that drug prices decline with increasing competition in the Chinese market, but not in a perfectly linear manner, with marginal price declines decreasing after the fourth entrant and "rebounding" at subsequent entrants, especially the sixth. Discussion: The findings suggest the importance of maintaining effective competition between suppliers to control prices, and that the government needs to further control generic pricing, especially for late entry generics, to ensure effective competition in the Chinese market.
