Abstract:In recent years, near-infrared (NIR) spectroscopy has gained wide acceptance in different fields by its advantages over other analytical techniques. NIR techniques have the ability to record spectrum for solid and liquid samples with rapid speed and non-destruction. This article discusses the features and development of NIR spectroscopy in the last few years; the analysis procedure and some mathematical tools to acquire specific information; some NIR techniques and new equipment that adopted in agriculture fields; the application of NIR in quality inspection of agricultural products, such as cereal, fruits, vegetables, and animal products; the application prospect of NIR spectroscopy in modern agriculture. As a rapid-response analytical tool, NIR spectroscopy may plays an important role in quality inspection of agricultural products.

Abstract:Alcohol precipitation is a critical unit operation during the manufacture of Chinese herbal injections. To facilitate enhanced process understanding and develop control strategy, the use of near-infrared spectroscopy (NIRS) combined with multivariate statistical process control (MSPC) methodology was investigated for in-line monitoring of alcohol precipitation. The effectiveness of the proposed approach was evaluated through an experimental campaign. Six batches were run under normal operating conditions to study batch-to-batch variation or batch reproducibility and establish MSPC control limits, while artificial process variations were purposefully introduced into the four test batches to assess the capability of the model for real-time fault detection. Several MSPC tools were compared and assessed. NIRS, in conjunction with MSPC, has proven to be a feasible process analytical technology (PAT) tool for monitoring batch evolution and potentially facilitating model-based advanced process control of the alcohol precipitation during the manufacture of Chinese herbal injections.

Abstract:A rapid and simultaneous quantification of the main active compounds of the in-process extract solutions is beneficial to the process monitoring as well as ensuring quality consistency of the end products during the production of traditional Chinese medicine. Here we present a near infrared (NIR) spectroscopy-based method for rapid analysis of extract solutions in the production of compound E Jiao oral liquid. The partial least-squares regression (PLSR) models for four quality indicators (viz. total flavonoids, total saponins, total saccharides and soluble solid contents) were established and validated. The results showed that all of the four models exhibited satisfactory fitting and predictive capacity. The root mean squares error of prediction (RMSEP) was 0.0384 mg mL−1, 0.0154 mg mL−1, 3.80 mg mL−1 and 0.199% for total flavonoids, total saponins, total saccharides and soluble solid contents, respectively. This work here demonstrated that NIR spectroscopy coupled with PLSR calibration can offer a reliable and non-destructive alternative in the routine monitoring of the extraction process in the production of compound E Jiao oral liquid. The presented approach is expected to be equally applicable to the mixed decoction of other herbal medicines.
Rapid Quantification of Multi-Components in Alcohol Precipitation Liquid of Codonopsis Radix Using Near Infrared Spectroscopy (NIRS)

Abstract:A near infrared spectroscopy (NIRS) approach was established for quality control of the alcohol precipitation liquid in the manufacture of Codonopsis Radix. By applying NIRS with multivariate analysis, it was possible to build variation into the calibration sample set, and the Plackett-Burman design, Box-Behnken design, and a concentrating-diluting method were used to obtain the sample set covered with sufficient fluctuation of process parameters and extended concentration information. NIR data were calibrated to predict the four quality indicators using partial least squares regression (PLSR). In the four calibration models, the root mean squares errors of prediction (RMSEPs) were 1.22 μg/ml, 10.5 μg/ml, 1.43 μg/ml, and 0.433% for lobetyolin, total flavonoids, pigments, and total solid contents, respectively. The results indicated that multi-components quantification of the alcohol precipitation liquid of Codonopsis Radix could be achieved with an NIRS-based method, which offers a useful tool for real-time release testing (RTRT) of intermediates in the manufacture of Codonopsis Radix.
Rapid Analysis of Purification Process of Grape Seed Extracts Using Near Infrared Spectroscopy

Abstract:A method was developed for the determination of purifying process of grape seed extracts by near infrared spectroscopy (NIRS) coupled with partial least squares (PLS) algorithm. The colourimetric assay of Folin-Ciocalteus was used as the reference method to determine the content of polyphenol. The samples in the purification process, which did not participate in modeling, were used to test the performance of the models. PLS factors, correlation coefficient of calibration set (R-2), t root mean square of cross-validation (RMSECV), root mean square of prediction (RMSEP), relative standard error of calibration (RSEC) and relative standard error of prediction (RSEP) were used to evaluate the performance of the models. The optimal calibration model was obtained with R-2=0.9995, RMSEC=0.713, RMSEP=2.67, RSEC=2.39% and RSEP=8.48% in the 5500-6200 cm(-1) spectral region by using Savitzky-Golay and second derivative spectral transform method. The results showed that the method is fast, nondestructive, environmental friendly, and can be used to determine the active components in macroporous adsorption resin purification process of traditional Chinese medicine.
Fast Determination of Multiple Quality Control Indexes for Concentrating Process of Carthamus Tinctorius L.alcohol Sedimentation Solution by NIRS

Abstract:Objective:To really realize quality control of Chinese herb concentrating process, near-infrared spectroscopy (NIRS) was used for fast monitoring multiple quality-control indexes of the process simultaneously, which could real-timely reflect its state.Methods: Taking the concentrating process of Carthamus tinctorius L.(CT)alcohol sedimentation solution as an example, samples were composed of two parts, one collected in the process and another prepared by diluting concentrated CT alcohol sedimentation solution.Models of HSYA content and density were established based on PLS algorithm, which were applied to predict the samples in the process of concentration which didn't participate in modeling to test the performance of the models.Results: The correlation coefficients of both calibration models achieved 0.990.And the predicted coefficients were up to 0.980, RSEP for them were less than 10%.Moreover, the fitting curve of the density and alcohol concentration were developed, the correlation coefficients of which reached 0.998.Therefore, the alcohol concentration could be quickly attained by the curve with known density so that the complicated operations of gas chromatography were avoided and analytical time was cut.Conclusion: It is concluded that NIRS can be used as a rapid method to determinate three key quality control indexes (the density, concentration of HSYA and ethanol) at one time in the process of concentrating process of Carthamus tinctorius L.alcohol sedimentation solution.

Abstract:The application of near infrared (NIR) spectroscopy for on-line quantitative monitoring of alcohol precipitation of the Danhong injection was investigated. For the NIR measurements, two fiber optic probes designed to transmit NIR radiation through a 2mm path length flow cell were applied to collect spectra in real-time. Particle swarm optimization- (PSO-) based least square support vector machines (LS-SVM) and partial least squares (PLS) models were developed for quantitative analysis of the critical intermediate quality attributes: the soluble solid content (SSC) and concentrations of danshensu (DSS), protocatechuic aldehyde (PA), hydroxysafflor yellow A (HSYA) and salvianolic acid B (SAB). The optimal models were then used for on-line quantitative monitoring of alcohol precipitation. The results showed that the PSO-based LS-SVM with a radial basis function (RBF) kernel was slightly better than the conventional PLS method, even though both methods exhibited satisfactory fitting results and predictive abilities. In this study, successful models were built and applied on-line; these models proffer real-time data and instant feedback about alcohol precipitation.

Abstract:As a rapid and accurate technique, near infrared (NIR) spectroscopy has been widely used in process monitoring to improve efficiency and ensure quality consistency. In the present work, the application of NIR spectroscopy for on-line quantitative monitoring of the concentration process of Wangbi tablets was investigated. Partial least squares regression (PLSR) models based on the full-range spectra or the key wavelengths selected by the competitive adaptive reweighted sampling (CARS) method were established. The accuracy of the CARS-PLSR method was greater than that of full-spectrum PLSR for three quality parameters (soluble solid contents, paeoniflorin and icariin). For the prediction set samples using the CARS-PLSR models the coefficients of determination (r(2)) and root mean square error [RMSEP (%)] were 0.97 and 1.29 for soluble solid contents, 0.93 and 0.014 for paeoniflorin and 0.85 and 0.009 for icariin, respectively. Overall the results indicated that NIR spectroscopy coupled with CARS-PLSR calibration is a reliable and non-destructive alternative method for on-line monitoring of the concentration process of Wangbi tablets on an industrial scale.

Abstract:The application of near-infrared (NIR) spectroscopy for on-line monitoring of the extraction process of red paeony root was investigated. For NIR measurements, two fiber optic probes designed to transmit NIR radiation through a 2mm pathlength flow cell were employed to collect spectra in real-time. Partial least squares regression (PLSR) calibration models were developed for the parameters of interest: total solid, paeoniflorin, and benzoic acid. The influences of flow rate and air bubble on the NIR spectra and calibration models were also investigated. The established models were used for on-line and real-time monitoring of extraction process, and a model updating method was proposed for the long-term usage of the developed models. Furthermore, both the moving block of standard deviation (MBSD) and relative concentration changing rate (RCCR) methods were used to identify the end point of extraction process. The results of this particular application of implementing NIR spectroscopy to monitor extraction process are very encouraging. Successful models have been built and applied on-line, which proffers real-time data and instant feedback about the extraction course, and in turn, provides improved control.
In-situ Monitoring of Saccharides Removal of Alcohol Precipitation Using Near-Infrared Spectroscopy

Abstract:As unsafe components in herbal medicine (HM), saccharides can affect not only the drug appearance and stabilization, but also the drug efficacy and safety. The present study focuses on the in-line monitoring of batch alcohol precipitation processes for saccharide removal using near-infrared (NIR) spectroscopy. NIR spectra in the 4000–10,000-cm[Formula: see text] wavelength range are acquired in situ using a transflectance probe. These directly acquired spectra allow characterization of the dynamic variation tendency of saccharides during alcohol precipitation. Calibration models based on partial least squares (PLS) regression have been developed for the three saccharide impurities, namely glucose, fructose, and sucrose. Model errors are estimated as the root-mean-square errors of cross-validation (RMSECVs) of internal validation and root-mean-square errors of prediction (RMSEPs) of external validation. The RMSECV values of glucose, fructose, and sucrose were 1.150, 1.535, and 3.067[Formula: see text]mg[Formula: see text]mL[Formula: see text], and the RMSEP values were 0.711, 1.547, and 3.740[Formula: see text][Formula: see text], respectively. The correlation coefficients [Formula: see text] between the NIR predictive and the reference measurement values were all above 0.94. Furthermore, NIR predictions based on the constructed models improved our understanding of sugar removal and helped develop a control strategy for alcohol precipitation. The results demonstrate that, as an alternative process analytical technology (PAT) tool for monitoring batch alcohol precipitation processes, NIR spectroscopy is advantageous for both efficient determination of quality characteristics (fast, in situ, and requiring no toxic reagents) and process stability, and evaluating the repeatability.
Rapid monitoring approaches for concentration process of lanqin oral solution by near-infrared spectroscopy and chemometric models

Abstract:Qualitative and quantitative detection methods based on near-infrared spectroscopy (NIRs) have been proposed in the process analysis of traditional Chinese medicine in recent years. In this study, rapid monitoring methods were developed for quality control of concentration process of lanqin oral solution (LOS). Partial least squares regression (PLSR) method was adopted to construct quantitative models for epigoitrin, geniposide, baicalin, berberine hydrochloride and density. Simultaneously, the genetic algorithm joint extreme learning machine (GA-ELM) was first applied in qualitative analysis of NIRs to distinguish end point of concentration process. Results of PLSR models were satisfactory with the relative standard error of calibration valued at 3.80%, 3.75%, 3.79%, 11.5% and 1.22% for epigoitrin, geniposide, baicalin, berberine hydrochloride and density respectively, and the residual predictive deviation values were higher than 3. For qualitative analysis, the GA-ELM model obtained 100% prediction accuracy. The PLSR quantitative models and the end point discrimination model constructed by GA-ELM correspond with the requirements of practical applications. The results indicate that NIRs in combination with chemometrics has great potential in improving the efficiency in production. (c) 2020 Elsevier B.V. All rights reserved.
Rapid Determination of Geniposide in the Extraction and Concentration Processes of Lanqin Oral Solution by Near-Infrared Spectroscopy Coupled with Chemometric Algorithms

Abstract:Process monitoring is essential for the pharmaceutical industry to control the quality uniformity of final products. In this study, the prompt quantitative methods of geniposide in Lanqin oral solution (LOS) during the process of extraction and concentration were developed by near-infrared spectroscopy (NIRs). Simultaneously, the feasibility of the method for universally monitoring the two processes was investigated. Methods including interval partial least square (iPLS), synergy interval partial least square (siPLS) and competitive adaptive reweighted sampling (CARS) were performed for characteristic variables selection. The single-process models of extraction, concentration and the multi-process model based on the combination of the two processes were constructed and discussed. Results showed that the performance of siPLS models were superior to others. For the extraction, concentration and the combination of these two processes, the Rc values were 0.9486, 0.9746 and 0.9980; while the RSEP values were 10.42 %, 5.41 % and 6.17 %, respectively. Compared with the specific models, the strong accuracy and stability of the multi-process model were also demonstrated. In addition, Pearson correlation analysis and paired t-test were also applied to verify the predictive performance of models. The overall results indicate that NIRs combined with siPLS algorithm is a robust method for predicting the geniposide in the extraction and concentration of LOS. Furthermore, the multi-process model of combination is feasible for the universal supervision of geniposide, which is convenient for industrial production applications.
On-line Monitoring of Traditional Chinese Medicinal Powder Blending Process by Using Near-Infrared Spectroscopy

Abstract:OBJECTIVE To establish a rapid method based on near-infrared diffuse reflectance spectroscopy(NIR) to monitor the blending process of traditional Chinese medicinal powder on-line.METHODS NIR spectra were collected during the blending processes of mixtures consisting of Red Peony Root powder and defatted Spine Date Seed powder.Multivariate calibration models based on PLS regression were developed to correlate the collected spectra with the content of Red Peony Root powder.The optimal calibration model was applied to monitor the blending process in real time with noninterference.RESULTS After spectral pretreatment using multiplicative signal correction(MSC),first derivative and Savitzky-Golay filter,Red Peony Root powder contents were quantified using PLS algorithm in the spectral ranges of 5 900-5 950 cm-1,6 700-6 750 cm-1 and 6 950-7 100 cm-1.The correlation coefficient of the calibration model was 0.999 0.The RMSEC,RMSEP and RMSECV were 1.14,1.16 and 1.26,respectively.CONCLUSION It was demonstrated that a miniature near-infrared analyzer could be used to monitor the contents of the constituents and the blending homogeneity of traditional Chinese medicinal powder in real time,and to rapidly determine the end-point of blending process.In addition,it can be used to judge whether the physical state of powder mixture conforms to the standard.Therefore,NIR spectroscopy is considered to be an appropriate PAT tool for end-point control of blending process of traditional Chinese medicinal powder.

Abstract:This paper used near-infrared (NIR) spectroscopy for the on-line quantitative monitoring of water precipitation during Danhong injection. For these NIR measurements, two fiber optic probes designed to transmit NIR radiation through a 2 mm flow cell were used to collect spectra in real-time. Partial least squares regression (PLSR) was developed as the preferred chemometrics quantitative analysis of the critical intermediate qualities: the danshensu (DSS, (R)-3, 4-dihydroxyphenyllactic acid), protocatechuic aldehyde (PA), rosmarinic acid (RA), and salvianolic acid B (SAB) concentrations. Optimized PLSR models were successfully built and used for on-line detecting of the concentrations of DSS, PA, RA, and SAB of water precipitation during Danhong injection. Besides, the information of DSS, PA, RA, and SAB concentrations would be instantly fed back to site technical personnel for control and adjustment timely. The verification experiments determined that the predicted values agreed with the actual homologic value.
[On-line monitoring of multiple component parameters during ethanol fermentation by near-infrared spectroscopy].

Abstract:The quantity of biomass, glucose concentration and ethanol concentration are important parameters in ethanol fermentation. Traditional methods are usually based on samples for off-line measurement, which not only requires multiple instruments for test and analysis but also consumes notable time and effort, and therefore is inconvenient for real-time process control and optimization. In this study, an in-situ detection method based on the near-infrared (NIR) spectroscopy is proposed for measuring the above process parameters in real time. The in-situ measurement is carried out by using an immersion type NIR spectroscopy. A multi-output prediction model for simultaneously estimating the quantity of glucose, biomass and ethanol is established based on a multi-output least-squares support vector regression algorithm. The experimental results show that the proposed method can precisely measure the quantity of glucose, biomass and ethanol during the ethanol fermentation process. Compared to the existing partial-least-squares method for modeling and prediction of individual components, the proposed method could evidently improve the measurement accuracy and reliability.

Abstract:In this paper, the principle of NIRS (near infrared spectroscopy)-based process trajectory technology was introduced.The main steps of the technique include:① in-line collection of the processes spectra of different technics; ② unfolding of the 3-D process spectra;③ determination of the process trajectories and their normal limits;④ monitoring of the new batches with the established MSPC (multivariate statistical process control) models.Applications of the technology in the chemical and biological medicines were reviewed briefly. By a comprehensive introduction of our feasibility research on the monitoring of traditional Chinese medicine technical process using NIRS-based multivariate process trajectories, several important problems of the practical applications which need urgent solutions are proposed, and also the application prospect of the NIRS-based process trajectory technology is fully discussed and put forward in the end.
Rapid determination of purifying process of Fructus Aurantii extracts by NIR transmission spectroscopy

Abstract:OBJECTIVE To develop a rapid and nondestructive method with near infrared (NIR) transmission spectroscopy for the fast detection of the purifying process of Fructus Aurantii extracts. METHODS Partial least square regression (PLSR) was applied to build the calibration models, and the contents of constituents were determined by HPLC as reference values. RESULTS Root mean square errors of Cross-Validation (RMSECV) obtained by PLS model for naringin and neohesperidin were 0.677 and 0.554 respectively, and root mean square errors of prediction (RMSEP) were 0.363 and 0.264 respectively. CONCLUSION The results show that the proposed method can be applied for fast detection in the herbal medicine purifying process.
A Feasibility Study on the Non-Invasive Analysis of Bottled Compound E Jiao Oral Liquid Using Near Infrared Spectroscopy

Abstract:The aim of this study was to explore the feasibility of near infrared (NIR) spectroscopy for the quantitative analysis of Compound E Jiao oral liquid, a popular oral TCM preparation, through intact glass vials. Transmission spectra were acquired with a sample holder tailored to the instrument's sample compartment. Total flavonoids, total saponins and soluble solid contents were considered as quality indicators of this oral preparation and calibration models were built using partial least-squares regression (PLSR) algorithm. The sample set was comprised of production samples as well as laboratory samples prepared by a concentrating-diluting method so as to extend the concentration range beyond the production variation. The developed models gave root mean square error of prediction (RMSEP) of 0.084 mg mL(-1) for total flavonoids, 0.014 mg mL(-1) for total saponins and 0.21% for soluble solid contents. To better evaluate the prediction performance, an independent set of ten production samples was introduced for the external validation. The models are compared with those built with the transflectance spectra collected in the destructive way. The results of paired t-test and F-test at a significance level of 0.05 revealed that the two sets of models exhibited comparable model performance. Application of Near Infrared Spectroscopy Combined with Particle Swarm Optimization Based Least Square Support Vactor Machine to Rapid Quantitative Analysis of Corni Fructus

Abstract:A novel method was developed for the rapid determination of multi-indicators in corni fructus by means of near infrared (NIR) spectroscopy. Particle swarm optimization (PSO) based least squares support vector machine was investigated to increase the levels of quality control. The calibration models of moisture, extractum, morroniside and loganin were established using the PSO-LS-SVM algorithm. The performance of PSO-LS-SVM models was compared with partial least squares regression (PLSR) and back propagation artificial neural network (BP-ANN). The calibration and validation results of PSO-LS-SVM were superior to both PLS and BP-ANN. For PSO-LS-SVM models, the correlation coefficients (r) of calibrations were all above 0.942. The optimal prediction results were also achieved by PSO-LS-SVM models with the RMSEP (root mean square error of prediction) and RSEP (relative standard errors of prediction) less than 1.176 and 15.5% respectively. The results suggest that PSO-LS-SVM algorithm has a good model performance and high prediction accuracy. NIR has a potential value for rapid determination of multi-indicators in Corni Fructus.
