Abstract:Nitric oxide (NO) is a messenger molecule in organisms, participating in the regulation of many biological processes. The abnormal expression of NO is often observed in a variety of diseases, including cerebral ischemia, atherosclerosis, and cancer. However, a suitable tool that can directly and sensitively detect NO in vitro and in vivo is important for understanding its various biological functions. In this report, a new fluorescent probe for nitric oxide, DHP-4 , was prepared, based on dihydropyridine-coumarin. DHP-4 was able to greatly enhance the fluorescence of NO, but did not affect the fluorescence emissions of other reactive oxygen species and nitrogen species, demonstrating its highly selective and sensitive response to NO. The probe generated stable optical signals in a buffer solution at pH values ranging from 3 to 10. In addition, DHP-4 could detect NO directly, showed low cellular toxicity, and was successfully applied to determine NO in Raw 264.7 cells, indicating its great potential as a tool for investigating the biological roles of NO in vivo .

Abstract:A rationally designed small-molecule fluorogenic probe for nitric oxide (NO) detection based on a new switching mechanism has been developed. Attaching a NO-responsive dihydropyridine pendant group to a fluorophore led to a probe that displays a very high sensitivity to NO concentrations down to the low nM range and a very high specificity to NO while being insensitive to other oxidative oxygen/nitrogen species that often interfere with the sensing of NO.

Abstract:Fluorescence imaging of nitric oxide (NO) in vitro and in vivo is essential to developing our understanding of the role of nitric oxide in biology and medicine. Current probes such as diaminofluorescein depend on reactions with oxidized NO products, but not with nitric oxide directly, and this limits their applicability. Here we report the formation of an imaging probe for nitric oxide by coordinating the highly fluorescent chemical 4-methoxy-2-(1H-naphtho[2,3-d]imidazol-2-yl)phenol (MNIP) with Cu(II). The coordination compound MNIP–Cu reacts rapidly and specifically with nitric oxide to generate a product with blue fluorescence that can be used in vitro and in vivo. In the present study MNIP–Cu was used to reveal nitric oxide produced by inducible nitric oxide synthase in lipopolysaccharide (LPS)-activated macrophages (Raw 264.7 cells) and by endothelial nitric oxide synthase in endothelial cells (HUVEC). MNIP–Cu was also used to evaluate the distribution of nitric oxide synthesis in a model of acute liver injury induced by LPS and d-galactosamine in mice. The results demonstrate that MNIP–Cu can act as a novel fluorescent probe for nitric oxide and has many potential applications in biomedical research.

Abstract:In situ fluorescence imaging of nitric oxide (NO) is a powerful tool for studying the critical roles of NO in biological events. However, the selective imaging of NO is still a challenge because most currently available fluorescent probes rely on the o-phenylenediamine (OPD) recognition site, which reacts with both NO and some abundant reactive carbonyl species (RCS) (such as dehydroascorbic acid and methylglyoxal) and some reactive oxygen/nitrogen species (ROS/RNS). To address this problem, a new fluorescent probe, NCNO, based on the N-nitrosation of aromatic secondary amine was designed to bypass the RCS, ROS, and RNS interference. As was expected, the probe NCNO could recognize NO with pronounced selectivity and sensitivity among ROS, RNS, and RCS. The probe was validated by detecting NO in live cells and deep tissues owing to its two-photon excitation and red-light emission. It was, hence, applied to monitor NO in ischemia reperfusion injury (IRI) in mice kidneys by two-photon microscopy for the first time, and the results vividly revealed the profile of NO generation in situ during the renal IRI process.

Abstract:Fluorescent probes with larger Stokes shifts in the far-visible and near-infrared spectral region (600–900 nm) are more superior for cellular imaging and biological analysis due to avoiding light scattering interference, reducing autofluorescence from biological sample and encouraging deeper tissue penetration in vivo imaging. In this work, two bis-methoxyphenyl-BODIPY fluorescent probes for the detection of nitric oxide (NO) have been firstly synthesized. Under physiological conditions, these probes can react with NO to form the corresponding triazoles with 250- and 70-fold turn-on fluorescence emitting at 590 and 620 nm, respectively. Moreover, the triazole forms of these probes have large Stokes shifts of 38 nm, in contrast to 10 nm of existing BODIPY probes for NO. Excellent selectivity has been observed against other reactive oxygen/nitrogen species, ascorbic acid and biological matrix. After the evaluation of MTT assay, new fluorescent probes have been successfully applied to fluorescence imaging of NO released from RAW 264.7 macrophages by co-stimulation of lipopolysaccharide and interferon-γ. The experimental results indicate that our fluorescent probes can be powerful candidates for fluorescence imaging of NO due to the low background interference and high detection sensitivity.

Abstract:Fluorescent probes for nitric oxide (NO), or more frequently for its oxidized surrogate dinitrogen trioxide (N2O3), have enabled scientists to study the contributions of this signaling molecule to many physiological processes. Seeking to improve upon limitations of other probes, we have developed a family of fluorescent probes based on a 2-amino-3 '-dialkylaminobiphenyl core. This core condenses with N2O3 to form benzo[c]cinnoline structures, incorporating the analyte into the newly formed fluorophore, which results in product fluorescence with virtually no background contribution from the initial probe. We varied the substituents in the core in order to optimize both the reactivity of the probes with N2O3 and their cinnoline products' fluorescence wavelengths and brightness. The top candidates were then applied to cultured cells to verify that they could respond to NO within cellular milieus, and the top performer, NO530, was compared with a "gold standard" commercial probe, DAF-FM, in a macrophage-derived cell line, RAW 264.7, stimulated to produce NO. NO530 demonstrated similar or better sensitivity and higher selectivity for NO than DAF, making it an attractive potential alternative for NO tracking in various applications.
An Oxazine-Based Near-Infrared Fluorescent Probe for Selective in Cellular Imaging of Exogenous Nitrite

Abstract:A near-infrared fluorescent probe based on benzo[a]phenoxazine containing o-phenylenediamine group as reactive site was designed and synthesized. The titration experiment showed that the probe gave colorimetric response towards sodium nitrite with the absorption maxima at 630 nm and 542 nm in pH = 5.0 disodium hydrogen phosphate-citric acid buffers containing 30% DMSO; its emission was located in 590-820 nm with the 9.1-fold fluorescent enhancement under excitation at 580 nm. Moreover, the probe can detect nitrite quantitatively in a concentration range of 0.0-26.0 mu M and the detection limit is as low as 45 nM. The salient features of this method are high sensitivity and selectivity, fast detection and low detection limit. Fluorescence confocal imaging experiments with V79 and HeLa cells indicate that the probe has the potential application in quantitative detection exogenous nitrite level for in cellular imaging. Abstract:Small-molecule fluorescent probes in combination with fluorescent microscopy can be a powerful tool to provide real-time detection and high spatiotemporal resolution of transient molecules in cells and bodies. For the design of fluorescent probes for transient molecule imaging, high detection sensitivity is crucial. In this report, two new fluorescent probes, 8-(3,4-diaminophenyl)-4,4-difluoro-4-bora-3a,4a-diaza-di(1,2-dihydro)naphtho[b,g]-s-indacene (DANPBO-H) and 8-(3,4-diaminophenyl)4,7-dimethyl-4,4-difluoro-4-bora-3a,4a-diaza-di(l,2-dihydro)naphtho[b,g]-s-indacene (DANPBO-M), have been developed for nitric oxide (NO) imaging. The detection sensitivity has been efficiently improved by use of these probes through increasing NO detection signals and decreasing background fluorescence. Fluorescence in the far-red region is enhanced by 400- and 550-fold after reaction with NO is achieved and remains stable for at least 24 h under the irradiation of xenon lamp. Excitation and emission wavelengths longer than 600 nm and excellent intracellular retention of these probes and their NO products create dark background inside and outside cells and tissues. What is more, the excellent intracellular retention of these compounds is obtained by their strong lipophilicity, which is a novel design concept diametrically opposite to the traditional approaches. The high sensitivity and dark background make DANPBO-H and DANPBO-M competitive for NO imaging in cells and tissues. The lipophilicity-based intracellular retention mechanism as a design strategy has great potential in the development of fluorescent probes for bioimaging.

Abstract:A cyanine dye (Cy7) based turn-on fluorescence probe has been successfully synthesized for the detection of nitric oxide through the attachment of a methyl amino group at the meso position of Cy7 dye. The probe exhibits weak fluorescent emission at 800 nm due to intramolecular charge transfer process, while it quickly turns into highly NIR fluorescent state upon the treatment with NO. The detection limit is as low as 11.3 nM. In addition, the selectivity of the probe toward NO over reactive oxygen species, reactive nitrogen species, and metal ions was also demonstrated. Importantly, the fluorescence response of this probe to NO occurs in the physiologically favorable NIR region, enabling its further use to image endogenous NO in an inflamed mouse model with low background interference and deep penetration depth.

Abstract:Nitric oxide (NO) is an important signaling molecule involved in many physiological and pathological processes. To understand these NO-mediated processes, it is a key to develop rapid and specific detection methods for NO. In the past 2 decades, numerous excellent fluorescent probes for NO have been designed; however, it still remains limitations such as slow response, low selectivity, and short excitation wavelength (<600 nm). In this Article, a two-photon fluorescent probe, NO-QA5, has been developed with 3-dimethylaminophenyl linking at the 6-position of 5-aminoquinoline as both the active site and prefluorophore for detection of NO. The nonfluorescent NO-QA5 can fast react with NO via a diazonium intermediate to generate two azoic regioisomers, one of which exhibits intramolecular charge transfer (ICT) emission, and two-photon absorption behavior (δΦ = 57 GM), giving a turn-on fluorescence rapid response. The sensing reaction is pH-insensitive in the range of 6-11 and highly selective and well sensitive (LOD = 15 nM), possible undergoing the same intermediate diazonium with the reaction under diazotization condition (NaNO2/HCl). Also, as a nitrite fluorescent probe NO-QA5 exhibits highly sensitive (LOD = 7 nM). Therefore, NO-QA5 can serve as a dual functional fluorescent probe for NO and NO2-. Furthermore, NO-QA5 as a specific imaging agent has been demonstrated by achieving both exogenous and endogenous detections of NO in living cells under both one- and two-photon excitation and high resolution in tissue slices under two-photon excitation.
A Ratiometric Fluorescent Probe Based on A Bodipy-Coumarin Conjugate for Sensing of Nitroxyl in Living Cells

Abstract:A ratiometric fluorescent HNO probe Bod-Cou-HNO based on a Bodipy-Coumarin platform, and a sensitive HNO recognition site, triarylphosphine was constructed. The probe Bod-Cou-HNO exhibits desirable properties such as high sensitivity, high selectivity, remarkable emission shift (up to 129 nm) and low cytotoxicity. Significantly, we have demonstrated that the probe Bod-Cou-HNO is suitable for fluorescence ratiometric imaging of HNO in living cells. The further application of the ratiometric fluorescence probe Bod-Cou-HNO for the investigation of biological functions of HNO in biosystems is under progress. (C) 2016 Elsevier B.V. Abstract:In situ imaging of nitric oxide (NO) is of great importance to study the critical roles of NO in various physiological and pathological processes. In most currently available fluorescent probes for NO, the o-phenylenediamine (OPD) based derivatives are the most greeted and studied, in which the OPD serves as the reactive site and fluorescence quencher to modulate a turn-on process via photoinduced electron transfer (PET). In this work, a unique turn-off probe containing OPD moiety, N-1, was designed and synthesized, which was radically different from the typical PET-based turn-on fluorescent probes. By theoretical calculation of molecular orbital, it was verified that not the widely used PET mechanism, but the intramolecular charge transfer (ICT) mechanism should be responsible for the turn-off process. Probe N-1 exhibited a remarkable performance with 7-folds fluorescence intensity decrease (at 490 nm), a low detection limit (20 nM), a fast response to NO within 12 min and a high selectivity for NO over other relevant analytes. Furthermore, probe N-1 was also applied to imaging NO in living cells. All results demonstrated that the new probe N-1 could be a practical and ideal NO indicator with good biological significance.

Abstract:An effective bioanalytical method for rapid, sensitive, specific, and in situ sensing of nitric oxide (NO) is the key for further unveiling the biological functions of this gasotransmitter molecule in vitro and in vivo. In this contribution, a new fluorescence probe for sensing and imaging of NO in live systems was developed. The probe, FP-NO, was designed by exploring a novel sensing mechanism, i.e., the rotation of the N-N single bond of a coumarin derivative. FP-NO was prepared by incorporating a recognition unit, thiosemicarbazide moiety into a coumarin fluorophore. The weakly fluorescent FP-NO quickly and selectively reacts with NO to form a highly fluorescent product, FP-P. Such an enhancement of fluorescence emission allows NO detection with high sensitivity. The detection limit was 47.6 nM. The reaction mechanism was validated by HRMS titration analysis and the "OFF-ON" fluorescence response mechanism was rationalized by theoretical computation. FP-NO is biocompatible and live cell membrane permeable. The feasibility of FP-NO as the fluorescence probe for imaging and flow cytometry analysis of exogenous NO in MCF-7 cells and exogenous NO production in inflamed J774A.1 macrophage cells was then evaluated. Visualization of exogenous and endogenous NO production in live zebrafish was then achieved, implying the potential application of FP-NO in the studies of the NO roles in live organisms.
Near-infrared Fluorescent Probe for Imaging Nitroxyl in Living Cells and Zebrafish Model

Abstract:Nitroxyl (HNO) is one of the important derivatives of nitric oxide (NO). It was intertwined with various biological and pharmacological events as a well-defined active molecule. Developing fluorescent probes for high specific and in situ trapping of HNO in living samples is still challenging. In this project, we constructed a near-infrared (NIR) metal-free fluorescent probe (DCM-P) for monitoring HNO in vitro and in vivo. The novel probe, DCM-P, contains a dicyanomethylene-4H-pyran (DCM) fluorophore as the reporter and a triarylphosphine as the recognition moiety. Upon exposure to HNO, the probe emits fluorescence at the wavelength of 688 nm, which belongs to near-infrared region and endows great beneficial for imaging in vivo. Moreover, DCM-P shows high chemoselective detection and fast response to HNO in the presence of various biological relevant reductants, and is successfully employed to trap nitroxyl in different living cells and zebrafish models. These results suggest that DCM-P is a suitable near-infrared metal-free fluorescent probe for motoring nitroxyl in living samples.

Abstract:Nitric oxide (NO) plays a crucial role in a variety of pathophysiological processes. Although a large number of NO fluorescent probes have been developed, it remains barely unexplored how the electrical effect of the recognition site affects responsiveness. In this work, we demonstrate an electronic-tuning strategy to enhance the sensitivity and responsiveness of NO probes based on BODIPY by decorating various groups with different electronic capabilities in the recognition unit. Experimental results show that the electronic effect at the identification section has a significant impact on the signal response. BODIPY 5 , containing the OH group at the para -position of aniline, exhibits the greatest fluorescence responsiveness with NO. The signal does not vary for the strong electron-drawing and electron-donating groups, in which electronic or charge transfer is always in the On or OFF state, as predicted by TD-DFT calculations. Additionally, the cellular assay demonstrated that the probe possesses the ability to visualize exogenous NO in lipid droplets of HeLa cells and the endogenous NO produced in Raw 264.7 cells. This study offers a useful NO imaging tool as well as a guideline for designing high-performance NO probes for practical application.

Abstract:Nitric oxide (NO) is an important mediator involved in various physiological phenomena. To obtain direct evidence for NO functions in vivo, a novel fluorescent probe for NO, 1,3,5,7-tetramethyl-8-(3′,4′-diaminophenyl)-difluoroboradiaza-s-indacence (TMDABODIPY) was designed and synthesized, which features high photostability and no pH dependency over a wide pH range. The fluorescence of 1,3,5,7-tetramethyl-8-(3′,4′-diaminophenyl)-difluoroboradiaza-s-indacence is very strong, however, when the reagent traps NO in the presence of dioxygen, the weakly fluorescent triazole form is obtained, which offers the advantages of specificity and sensitivity for direct detection of NO. Calibration for NO showed the response has good linearity (0.01–0.4μmoll−1) and its detection limit is 2.5nmoll−1 (s/n=3). The proposed method has been used to monitor the release of NO from S-nitrosocysteine, a NO-releasing agent.
A Highly Specific Rhodamine B Based Turn-on Fluorescent Probe for Nitric Oxide and Application in Living Cells

Abstract:The design and synthesis of two rhodamine derivatives with the same luminescent group, which have an interesting "turn-on" fluorescent response toward NO and can be readily prepared through one-step reaction in acetonitrile. Of the two compounds, rh-NO-1 showed high selectivity for NO among various reactive oxygen/nitrogen species, accompanied by a 24-fold fluorescent enhancement. With an increase of the amount of NO from 0 to 100 mu M, the linear correlation between the fluorescent intensity of rh-NO-1 at 600 nm and the concentration of NO was found. The probe rh-NO-1 had good sensitivity and the detection limit was as low as 36.8 nM. Upon addition of NO, rh-NO-1 had a clear response within 2 min. This was a colour change from colorless to pink, which was visible to the naked eye. In addition, both rh-NO-1 and its derivative can be used to detect NO in living cells. (C) 2020 Elsevier Ltd. A Rhodamine-Based Fast and Selective Fluorescent Probe for Monitoring Exogenous and Endogenous Nitric Oxide in Live Cells

Abstract:Nitric oxide, an important cellular messenger molecule, plays a wide range of physiological roles in neurotransmission, vascular relaxation, platelet activation, and immune response. In this work, we synthesized a selective and sensitive cationic fluorescent probe (ROPD) for monitoring exogenous and endogenous nitric oxide in live cells by grafting a NO-trapper o-phenylenediamine (OPD), via one of the amino groups of OPD, onto a rhodamine fluorophore at the C-3 position. The probe exhibited high selectivity and sensitivity towards NO with fast response in the water phase over a wide pH range. At the same time, the probe ROPD is cell-membrane permeable and can be used for the real-time visualization of exogenous NO in both L929 and HeLa cells, and endogenous NO in stressed RAW 264.7 cells.
