Abstract:Zwitterionic materials have moieties possessing cationic and anionic groups. This molecular structure leads to unique properties that can be the solutions of various application problems. A typical example is that zwitterionic carboxybetaine (CB) and sulfobetaine (SB) materials resist nonspecific protein adsorption in complex media. Considering the vast number of cationic and anionic groups in the current chemical inventory, there are many possible structural variations of zwitterionic materials. The diversified structures provide the possibility to achieve many desired properties and urge a better understanding of zwitterionic materials to provide design principles. Molecular simulations and modeling are a versatile tool to understand the structure–property relationships of materials at the molecular level. This progress report summarizes recent simulation and modeling studies addressing two fundamental questions regarding zwitterionic materials and their applications as biomaterials. First, what are the differences between zwitterionic and nonionic materials? Second, what are the differences among zwitterionic materials? This report also demonstrates a molecular design of new protein‐resistant zwitterionic moieties beyond conventional CB and SB based on design principles developed from these simulation studies.
Intermolecular Interactions and Solubility Behavior of Multicomponent Crystal Forms of Orotic Acid: Prediction and Experiments

Abstract:Multicomponent crystal forms for drugs with poor solubility, such as orotic acid (OA), can potentially promote dissolution behavior and bioavailability. Herein an affinity prediction was carried out to predict the formation of multicomponent crystal forms between OA and 41 coformers. Amino acids were selected as coformers to solve the solid-state solubility problem of OA. Six new multicomponent crystal forms, including five salts and one salt hydrate, were characterized and confirmed by powder X-ray diffraction, differential scanning calorimetry, and thermogravimetric analysis. Spectroscopic techniques, such as Fourier transform infrared spectroscopy and X-ray photoelectron spectroscopy, were used to validate the cocrystal/salt dichotomy. In the crystal packing, all salts exhibit N+–H···O– intermolecular interactions, which is well explained by molecular electrostatic potential surfaces. Finally, a powder dissolution study was conducted. The powder dissolution of all salts presented a higher apparent solubility (1.15–1.32 times) and a faster dissolution rate in comparison to those of OA, which is ascribed to the strength of the crystal lattice and the solvation of components.

Abstract:Isostructural multicomponent crystals provide a promising way for fine-tuning physicochemical prop-erties,whereas their design remains quite challenging.The purpose of this work was to provide a new strategy for obtaining isostructural multicomponent crystals by introducing coformers with func-tional group positional isomerism.Five isostructural salts of an antitumor drug dimethylaminomicheli-olide(DMAMCL)were reported and designed with a series of dihydroxybenzoic acid regioisomers for the first time,which were identified by power and single-crystal X-ray diffractions.Similar lattice parame-ters suggested these obtained salts may have the same crystal packing mode.The quantitative similar-ity parameters via XPac,CrystalCMP and Mercury program further proved these crystal structures are 3D isostructural.Hirshfeld surface maps and 2D fingerprint plots show that the isostructural salts have similar intermolecular interactions.Compared with DMAMCL,obvious improvement was observed in the thermal stability,hygroscopicity,and solubility of these isostructural salts.Meanwhile,isostructural crys-tals may have different physicochemical properties,even though the shape and molecular size are similar and the packing of crystal structures is equally matched.

Abstract:Cocrystal engineering for bioavailability enhancement is still a serendipitous process, which requires systematic solution crystallization, in vivo predictive dissolution, and biopharmaceutical simulation. The purpose of this study is to improve bioavailability through cocrystal engineering, with a focus on understanding the mechanism of supersaturation maintenance during dissolution. BI 639667 is a poorly soluble and fast precipitating compound. The ability of its cocrystal with salicylic acid (SA) to maintain supersaturation was assessed using a modified two-step dissolution method, and its bioavailability was determined in rats. Biopharmaceutical simulation and dissolution modeling were performed to predict the in vivo performance in humans. Conductor-like screening model for realistic solvents and molecular dynamics (MD) simulation were used to study the interactions between the active pharmaceutical ingredient (API) and coformers in vacuum and aqueous media, respectively. A high melting cocrystal with SA, crystallized in dichloromethane, showed moderate solubility enhancement with prolonged supersaturation during dissolution, which was able to enhance bioavailability with reduced C max, compared to amorphous dispersion. The radial distribution function (RDF) between the API and selected coformers was calculated using MD simulation to determine the mean distance between the API and coformer molecules in aqueous media. The result suggests that SA could better compete against water for interactions with the API and could penetrate the API molecular clusters to inhibit nucleation. Thus, the RDF by MD simulation may be used to determine the disruptive effect of water on the interaction between the API and coformer and improve cocrystal engineering for bioavailability enhancement.

Abstract:To have a better understanding on molecular mechanism of the crystal morphology manipulation, the effect of solvents on morphology of crystals was investigated by using sulfamerazine (SMR) as model compound, through combination of experimental investigations and molecular simulations. The modified attachment energy model was employed to build the crystal morphology model in different solvents. Hirshfeld surfaces and 2-D fingerprint plots were used to investigate the solute-solute interactions, and molecular dynamics simulation was employed to investigate the interactions between solvent molecules and different crystal faces. Function of factors such as surface structure, roughness and the radial distribution function were also considered and discussed. The results suggested these factors are important to crystal habits since they could affect the adsorption competition between the solute and solvent molecules. It was found that the method developed in this work could be used to predict the morphology of SMR and could guide the selection of crystallization solvents for obtaining target morphology.

Abstract:Soluble additives are widely used to control crystallization processes, modifying the morphologies, sizes, polymorphs, and physical properties of the product crystals. Here, a simple and versatile strategy is shown to significantly enhance the potency of soluble additives, ranging from ions and amino acids to large dye molecules, enabling them to be effective even at low concentrations. Addition of small amounts of miscible organic co-solvents to an aqueous crystallization solution can yield enhanced morphological changes and an order of magnitude increase of additive incorporation within single crystalsa level that cannot be achieved in pure aqueous solutions at any additive concentration. The generality of this strategy is demonstrated by application to crystals of calcium carbonate, manganese carbonate, and strontium sulfate, with a more pronounced effect observed for co-solvents with lower dielectric constants and polarities, indicating a general underlying mechanism that alters water activity. This work increases the understanding of additive/crystal interactions and may see great application in industrial-scale crystal synthesis.

Abstract:The solubility data of sorbic acid in binary systems of (ethanol + water), (1-propanol + water) and (2-propanol + water) were measured from 283.15 to 323.15 K using the static equilibrium method under atmospheric pressure. It was found that the solubility of sorbic acid in the three binary solvent systems increased with increasing temperature as well as increasing initial mole fraction of organic solvent in these systems. The van't Hoff–Jouyban–Acree model, the modified Apelblat–Jouyban–Acree model and the CNIBS/R-K model were proposed for correlating the experimental solubility values in various solution systems. Furthermore, the dissolution thermodynamic properties of Gibbs energy change (ΔsolGo), molar enthalpy change (ΔsolHo) and molar entropy change (ΔsolSo) were calculated from the experimental solubility data, using the van't Hoff equation. The positive values of ΔsolGo, ΔsolHo and ΔsolSo indicate that these dissolution processes of sorbic acid in the solvents studied were all endothermic and entropically favorable. In addition, the change of dissolution enthalpy was the main contributor to the positive value of the molar Gibbs energy of the dissolution process. The experimental solubility and the models used in this work would be conducive to purifying sorbic acid from its crude mixtures.

Quantifying solubility enhancement due to particle size reduction and crystal habit modification: case study of acetyl salicylic acid

Abstract:The poor solubility of potential drug molecules is a significant problem in the design of pharmaceutical formulations. It is well known, however, that the solubility of crystalline materials is enhanced when the particle size is reduced to submicron levels and this factor can be expected to enhance drug product bioavailability. Direct estimation of solubility enhancement, as calculated via the Gibbs-Thompson relationship, demands reasonably accurate values for the particle/solution interfacial tension and, in particular, its anisotropy with respect to the crystal product's habit and morphology. In this article, an improved, more molecule-centered, approach is presented towards the calculation of solubility enhancement factors in which molecular modeling techniques are applied, and the effects associated with both crystal habit modification and solvent choice are examined. A case study for facetted, acetyl salicylic acid (aspirin) crystals in equilibrium with saturated aqueous ethanol solution reveals that their solubility will be enhanced in the range (7-58%) for a crystal size of 0.02 microm, with significantly higher enhancement for crystal morphologies in which the hydrophobic crystal faces are more predominant than the hydrophilic faces and for solvents in which the solubility is smaller.
Multi-component Crystal Strategy for Improving Water Solubility and Antifungal Activity of Climbazole

Abstract:Purpose: The primary problem with climbazole (CLB), a broad-spectrum imidazole antifungal drug, is its low water solubility. In order to increase its water solubility and antifungal activity, three new multi-component crystals were synthesized in this work, and the intermolecular interactions were systematically studied. This work helps to optimize the CLB product formulation and extend its application prospects. Methods: In this work, three novel multi-component crystals, CLB-malonic acid (CLB-MA) salt, CLB-succinic acid (CLB-SA) cocrystal and CLB-adipic acid (CLB-AA) cocrystal, were successfully synthesized. And the crystal structure, thermodynamic properties, solubility, dissolution, hygroscopicity, and antifungal activity of the three multi-component crystals were fully characterized by single-crystal X-ray diffraction (SCXRD), thermogravimetric analysis (TGA), differential scanning calorimetry (DSC), dynamic water vapor adsorption (DVS) and powder dissolution tests, etc. The molecular interactions and molecular stacking in multi-component crystals were studied by Hirshfeld surface (HS), molecular surface electrostatic potential (MEP), interaction region indication (IRI) and atom and molecule (AIM) techniques. Results: The results show that the three multi-component crystals have good moisture resistance stability, and their water solubility is 6-22 times that of pure CLB. Meanwhile, the measurement of the minimum inhibitory concentration (MIC) proves that the cocrystal/salt has a stronger antifungal activity than climbazole. Quantum chemistry calculations of crystal structure visualized and quantified the interactions that exist in multi-component crystals, and explored the microscopic mechanisms underlying the different performance of multi-component crystals.

Abstract:The oil/water interfacial properties of different zwitterionic surfactant (N-dodecyl-N, N-dimethyl-3-ammonio-1-propanesulfonate, ASB; 3-(1-dodecyl-3- imidazolio)propanesulfonate, ImS3; 3-(1-dodecyl-3-benzimidazolio)propanesulfonate, BzImS3) combined with anionic surfactant (sodium dodecyl benzene sulfonate, SDBS) have been studied via experimental and simulation methods. Both experimental and simulation results indicate that the synergistic effect between surfactants is greatly affected by the aromatic cation part of the zwitterionic surfactants. For ASB/SDBS system, the SDBS and ASB molecules are adsorbed at same position of the oil/water interface, which generates competitive adsorption and resulting in a relatively loose surfactant monolayer. For ImS3/SDBS and BzImS3/SDBS systems, the additional π-π stacking interaction increases the compactness of surfactant monolayer, which contributes to the greatly reduced oil/water interfacial tension. The adsorption mechanism of mixed zwitterionic/anionic surfactant systems at oil/water interface is supplemented at a molecular level, which provides a fundamental guidance for an in-depth understanding of optimal selection of surfactant in enhancing oil recovery.

Abstract:We combine molecular dynamics simulations with experiments to estimate solubilities of an organic salt in complex growth environments. We predict the solubility by simulations of the growth and dissolution of ions at the crystal surface kink sites at different solution concentrations. Thereby, the solubility is identified as the solution's salt concentration, where the energy of the ion pair dissolved in solution equals the energy of the ion pair crystallized at the kink sites. The simulation methodology is demonstrated for the case of anhydrous sodium acetate crystallized from various solvent-antisolvent mixtures. To validate the predicted solubilities, we have measured the solubilities of sodium acetate in-house, using an experimental setup and measurement protocol that guarantees moisture-free conditions, which is key for a hygroscopic compound like sodium acetate. We observe excellent agreement between the experimental and the computationally evaluated solubilities for sodium acetate in different solvent-antisolvent mixtures. Given the agreement and the rich data the simulations produce, we can use them to complement experimental tasks, which in turn will reduce time and capital in the design of complicated industrial crystallization processes of organic salts.

Impurity-induced Acceleration of Polymorphic Conversion via Crystalline Solid Solutions and the T-X Phase Diagrams of Salicylic acid and 3-Hydroxybenzoic acid

Abstract:Two T-X binary phase diagrams have been constructed between salicylic acid (SA) and two monotropic polymorphs of the isomer 3-hydroxybenzoic acid (3HBA). Crystalline solid solutions (CSS) were formed at all extremes of the phase diagrams. The solid-state miscibilities ranged from 0.5% up to 6% of the second component. The thermodynamically stable Form I of 3HBA exhibited a higher solid state miscibility than Form II of 3HBA across all investigated temperatures. The solubility changes induced by the different CSS were measured experimentally in 40 w% methanol in water at 25 °C and are presented in two ternary phase diagrams. The SA-rich CSS phase exhibited the highest solubility increase corresponding to 160% up to the solvus at 0.7% 3HBA in SA. The changes in solubility of the CSS phases belonging to the two 3HBA polymorphs were found to diverge with increasing incorporation of SA in the respective crystal lattices. This thermodynamic divergence in combination with the monotropic stability relationship caused the driving force for polymorphic conversion to increase with increasing SA content. This unusual scenario was demonstrated experimentally through the use of solution-mediated phase transformation (SMPT) experiments analyzed in-situ by Raman. It was found that the incorporation of 0.5% SA in the crystal lattice of 3HBA Form II caused the polymorphic conversion rate to Form I to double, in comparison to when 3HBA is chemically pure. The current example thus demonstrates the thermodynamic context for how solid-state miscible impurities can expedite polymorphic conversions. This and other contributions showcase how the rate of crystallizations can be enhanced or reduced solely based on formation of CSS with an impurity or additive, without accounting for any surface adsorption effects.

Abstract:The effect of cetyltrimethylammonium bromide (CTAB) on the crystal morphology and size of calcium sulfate whiskers in aqueous HCl solutions was investigated by combined experimental and molecular dynamics (MD) simulation studies. When CTAB was used, calcium sulfate whiskers with small diameters and high aspect ratios were obtained. The MD simulation results indicate that the experimental results can be attributed to the combined effect of the surface adsorption and inhibition of the solute diffusion of CTAB.
Modulation of the dissolution with ASP from a supersaturated solution on a bionic platform for gout pathology crystals

Abstract:The core to the treatment of gout is the elimination of pathologic crystal, monosodium urate monohydrate (MSUM). The primary treatment available is to gradually dissolve the "culprit crystals" by lowering the blood uric acid concentration with medications, which often takes a long time and in severe cases must still be treated surgically. Herein, we developed a dynamic bionic platform based on a hydrogel composite membrane (HCM) to screen the direct facilitated solubilization of MSUM crystals by small organic molecules in bionic saturated, or even supersaturated, solutions. The customized and biologically safe (NAGA/PEGDA/NIPAM) HCM, which is consistent with the main amino acid composition of articular cartilage, well mimics the entire process of organic molecules leading to the dissolution of MSUM crystals in the joint system. With the verifications of this platform, it is shown that l-aspartic acid (ASP) significantly promotes the dissolution of MSUM crystals not only in saturated but also in supersaturated solutions. Furthermore, a novel mechanism called "crane effect" was used to explain this "dissolution effect" of ASP on MSUM, which stems from the ability of ASP to lock onto the surface of MSUM crystals through hydrogen bonding by virtue of its two carboxyl groups, and simultaneously its amino group lifts the uric acid molecules from the surface of MSUM crystals by virtue of interactions of hydrogen bonding. The results of bulk crystallization, scanning electron microscopy (SEM), powder X-diffraction (PXRD), and density-functional theory (DFT) studies are quantitatively consistent with this hypothetical "crane effect" mechanism. Hence, this HCM-based functional platform could provide entirely novel ideas and methods for drug design and screening for the treatment of pathological crystal diseases of gout.

Abstract:Ginsenoside Ro (Ro), a natural anionic biosurfactant derived from ginseng, has been found to markedly increase the solubility of saikosaponin a (SSa), which is the active ingredient of Radix Bupleuri. SSa is minimally soluble in water. To determine the mechanism by which Ro solubilizes SSa, the self-assembly behavior of Ro and the phase behavior of blended Ro and SSa systems were studied by mesoscopic dynamics (MesoDyn) and dissipative particle dynamics (DPD) simulations. The simulation results show that Ro can form vesicles via the closure of oblate membranes. At low concentrations, SSa molecules are solubilized in the palisade layer of the Ro vesicles. At high concentrations, they interact with Ro molecules to form mixed vesicles with Ro adsorbing on the surfaces of the vesicles. The evaluation of the SSa solubilization process reveals that, at low concentrations, Ro aggregates preferentially to form vesicles, which then absorb SSa into themselves. However, at high concentrations, SSa first self-aggregates and then dissolves. This is because the solubilization behavior of Ro shifts the precipitation-dissolution equilibrium in the direction of dissolution. These results of the simulations are consistent with those of transmission electron microscopy (TEM) and dynamic light scattering (DLS).

Abstract:Solution crystallization is an important separation unit operation in active pharmaceutical ingredient production. Solvent is one of the important factors affecting crystal morphology. How to select/design suitable solvents is still one of the most urgent problems in the crystallization field. In this paper, a framework for crystallization solvent design based on the developed quantitative prediction model of crystal morphology is proposed. First, molecular dynamics is used to predict the crystal morphology in solvents. Next, solvent descriptors are selected by stepwise regression method. Then, the quantitative relationship between crystal aspect ratio and solvent descriptors is developed. Subsequently, Computer-Aided Molecular Design method is integrated with the developed quantitative prediction model. The crystallization solvent design problem is expressed as a Mixed-Integer Non-Linear Programming model with maximum yield as the objective function, which is solved by decomposition algorithm. Abstract:Controlling polymorphism is one of the most difficult challenges for the pharmaceutical industry. Polymorphs of the same compound can have very different properties that might affect the quality of the final drug. Many studies have focused on trying to determine robust methods to produce specific polymorphs that exhibit the desired characteristics of the drug (solubility and rate of dissolution in particular). The choice of solvent, pH, kinetic conditions and the presence of impurities can all have very strong effects on the polymorphic outcome of cooling crystallization. In this work, the crystallization of ortho-aminobenzoic acid (OABA) in the presence of a structurally related additive (benzoic acid) will be studied using both in situ PAT tools and offline techniques. The effect on morphology, polymorphic outcome, polymorphic transformation kinetics and tendency towards agglomeration in different solvents will be studied using a large variety of instrumentation. The incorporation of benzoic acid in solid solution in the three different polymorphs of OABA will also be quantified and discussed. The results of the crystallization experiments and offline analysis will be compared and interpreted using the information on the crystal structures of the three OABA polymorphs from the Cambridge Crystallographic database.

Abstract:Crystal engineering has proved to be an effective and reliable approach to tailor the physicochemical properties of active pharmaceutical ingredients. In this work, three novel sophocarpine salts were prepared and reported, named sophocarpine-hesperetin (SC-HES), sophocarpine-L-(-) malic acid monohydrate (SC-LMA MH), and sophocarpine-p-hydroxybenzoic acid monohydrate (SC-PHBA MH). The crystal structures of the salts were confirmed by infrared spectroscopy and single crystal X-ray diffraction. SC-HES exhibited a laminated molecular stacking structure, SC-LMA MH formed a wavy layered structure, while sophocarpine molecules filled in the voids of the reticular structure to form the 3D structure of the SC-PHBA MH. Thermal analysis shows that after salification, the melting point of the compound increased from 78°C to over 120°C, overcoming its low melting point. Lattice energy and Hirshfeld surface analysis indicated that lower lattice energy provides better thermal stability of the crystal and intermolecular hydrogen bonds (O–H···O and N→H···O) play a key role in the construction of the crystal structures. In addition, the water solubility and hygroscopicity of the three salts were characterized and discussed, with SC-HES exhibiting satisfying physical stabilities and hygroscopicity, and SC-LMA MH achieving 6.7 times solubility than the free base, which provides viable options for solid dosage form development of the drug.
