Abstract:Listeria monocytogenes (LM) is a foodborne pathogen, and it can pose a risk of serious diseases to the human health. Hence, the development of an effective method for the detection of LM is very important. In this study, by selecting LM as the template and 3-thiopheneacetic acid as the functional monomer, an LM-imprinted polymer (LIP)-based sensor was proposed for the first time to detect LM by electropolymerizing TPA on the glassy carbon electrode (GCE) surface in the presence of LM. After the removal of the LM template from the electrode surface, the obtained sensor was denoted as LIP/GCE, which could effectively recognize and capture LM cells. By using [Fe(CN)6]4-/3- as the probe, its peak current at LIP/GCE could be restricted when the LM cells were captured into the imprinted cavity of LIP/GCE, and the current value decreased with an increase in the LM concentration. Serious conditions were optimized for achieving highly sensitive detection, and a low detection limit (6 CFU mL-1) coupled with a wide linear range (10 to 106 CFU mL-1) was obtained for LM. Finally, the inter-electrode reproducibility, stability, selectivity, and applicability of LIP/GCE were also investigated, and the obtained results were acceptable.

Abstract:Listeria monocytogenes (L. monocytogenes) is one of the most common food-borne pathogens. The authors describe a sensitive and reliable multicolorimetric assay for L. monocytogenes using a sensing system based on TMB2+ etching of gold nanorods. Apt-MNP was used as the capture probe, and IgY-BSA-MnO2 NPs was chosen as an oxidase-like nano-artificial enzyme to oxidize TMB to generate TMB2+. Under the optimized conditions, the longitudinal shift of localized surface plasmon resonances had a linear correlation with the L. monocytogenes concentration in the range between 10 to 10(6) cfu mL(-1). Meanwhile, the sensing system can generate vivid color responses as colorful as a rainbow, and the limit of detection is as low as 10 cfu mL(-1) at a glance. Recoveries ranging from 97.4 to 101.3% are found when analyzing spiked food samples without pre-enrichment. In our perception, it shows promise in rapid instrumental and on-site visual detection of L. monocytogenes. (C) 2018 Elsevier B.V. All rights reserved.

Abstract:In this paper, a sensitive immunoassay method was proposed for Listeria monocytogenes detection by using highly fluorescent bioconjugated nanoparticles probe. (FITC-IgG)-doped fluorescent silica nanoparticles (fsNPs) firstly were synthesized by a microemulsion method and characterized by TEM and fluorescent spectra. Then the prepared fsNPs were conjugated with polyclonal rabbit anti-L. monocytogenes antibody (pAb) and used as indicator probe. A sandwich-type immune affinity reaction between polyclonal rabbit anti-L. monocytogenes antibody coated onto microplate wells, target bacteria and the fsNPs-antibody conjugates subsequently was conducted to detect target L. monocytogenes and assemble the indicator probe onto the wells. The target L. monocytogenes was measured by the fluorescent signals of the assembled indicator probes. Under the optimal conditions, the calibration graph of fluorescent intensity is proportional to the amount of target bacteria over the range of 50-10,320 CFU/mL with a detection limit of 50 CFU/mL. The proposed method has been successfully applied to detect L. monocytogenes in food samples offering the advantages of sensitivity, simplicity, and stability.

Abstract:A newly designed AIEgen‐based fluorescent film sensor is developed and used successfully, for the first time, to detect gaseous 3‐hydroxy‐2‐butanone, a biomarker of Listeria monocytogenes. The sensor depicts fast, sensitive, selective, and fully reversible sensing performance toward the analyte, as evidenced by the reliable finding of the foodborne pathogen at the very early stage.Foodborne Listeria monocytogenes poses serious threats to public health. Fast and sensitive detection of the pathogen at the point of contamination is thus crucial to halt the spread of bacteria‐related diseases. Herein, we report for the first time a fluorescent film sensor to detect the biomarker of L. monocytogenes, 3‐hydroxy‐2‐butanone. The sensor demonstrated unprecedented sensing performance for the analyte with a detection limit lower than 0.05 mg/m3, response time less than 1 s, full reversibility, and excellent selectivity. Further study showed that the sensor can be used to monitor the growth of L. monocytogenes with much‐improved sensitivity. The superior performance of the sensor is ascribed to the specific binding, efficient charge transfer emission, and porous adlayer structure of the specially designed sensing fluorophore‐based film. Our work paves the way to develop a portable detector to meet the needs for on‐site and real‐time detection of foodborne pathogens.

Abstract:We report the detection of the proteins lysozyme and cytochrome c as well as the live bacterial cells of Shewanella oneidensis MR-1 in aqueous solutions with sensitivities order(s) of magnitude higher than those previously reported. Two highly sensitive surface-enhanced Raman scattering (SERS)-based biosensors using optical fibers have been employed for such label-free macromolecule detections. The first sensor is based on a tip-coated multimode fiber (TCMMF) with a double-substrate "sandwich" structure, and a detection limit of 0.2 μg/mL is achieved in protein detections. The second sensor is based on a liquid core photonic crystal fiber (LCPCF) with a better confinement of light inside the fiber core, and a detection limit of 10(6) cells/mL is achieved for the bacteria detection. Both SERS biosensors show great potential for highly sensitive and molecule-specific detection and identification of biomolecules.

Abstract:Accurate and timely diagnosis of Listeria monocytogenes (L. monocytogenes), a well-known foodborne pathogen with a serious threat to human life and health, is critical for managing bacterial infections and food quality. Here, we report a genosensor platform based on DNA biofunctionalized superparamagnetic nickel colloidal nanocrystal clusters (SNCNCs) coupling with "IS-primer" amplification reaction (ISAR) for rapid, ultrasensitive and specific L. monocytogenes detection. Specifically, the catalytic activity of SNCNCs was identified. After three-step modification, the signal unit was formed by immobilizing AuNPs and hairpin probes on the surface of SNCNCs, achieving enhanced catalytic activity, leading to the reduction of a colorimetric substrate 4-nitrophenol (4-NP). The in-situ signal amplification of target RNA on the surface of SNCNCs coupling with ISAR showed high sensitivity and a detection limit of 1.50 × 10-3 ng mL-1 under optimized conditions As SNCNCs can bind and lyse bacteria, this genosensor can detect live L. monocytogenes without RNA extraction, with a detection limit of 3.80 × 102 CFU mL-1. Furthermore, the system could also detect L. monocytogenes in artificially contaminated milk samples within 6 h. These results indicate the great potential of our method for sensitive, specific, convenient and low-cost bacterial detection in food, environment and clinics.

Abstract:A fluorescent in situ hybridization (FISH) method in conjunction with fluorescin-labeled peptide nucleic acid (PNA) probes (PNA–FISH) for detection of Listeria species was developed. In silico analysis showed that three PNA probes Lis-16S-1, Lm-16S-2 and Liv-16S-5 were suitable for specific identification of Listeria genus, Listeria monocytogenes and Listeria ivanovii, respectively. These probes were experimentally verified by their reactivity against 19 strains of six Listeria species (excluding newly described species Listeria marthii and Listeria rocourtiae) and eight other bacterial species. The PNA–FISH method was optimized as 30min of hybridization with 0.2% Triton X-100 in the solution and used to identify 85 Listeria strains from individual putative Listeria colonies on PALCAM agar plates streaked from selectively enriched cultures of 780 food or food-related samples. Of the 85 Listeria strains, thirty-seven were identified as L. monocytogenes with the probe Lm-16S-2 and two as L. ivanovii with the probe Liv-16S-5 which was in agreement with the results obtained by the API LISTERIA method. Thus, the PNA–FISH protocol has the potential for identification of pathogenic Listeria spp. from food or food-related samples.
Dual recognition and highly sensitive detection of Listeria monocytogenes in food by fluorescence enhancement effect based on Fe3O4@ZIF-8-aptamer

Abstract:A highly sensitive and selective foodborne pathogen detection system is essential to ensure food quality and public health. Therefore, in this study, a dual recognition and highly sensitive detection of Listeria monocytogenes by fluorescence enhancement strategy based on Fe3O4 @ZIF-8 have been developed. Aptamer absorbed on Fe3O4 @ZIF-8 were used for the specific capture of Listeria monocytogenes, and the conjugates of anti-Listeria monocytogenes antibody-biotin and streptavidin-FITC were used as signal amplification and fluorescence recognition probes binding with target bacteria. Simultaneously, the quantitative detection of Listeria monocytogenes was realized by monitoring the increase of FITC fluorescence signal due to hydrophobic environment. The linear range of the detection of pure culture was from 1.4 × 101 to 1.4 × 107 CFU/mL, with the detection limits as low as 0.88 CFU/mL and the coefficient of variation of each concentration no more than 2%. A good specificity of the detection system was also showed in the results, and the detection took about 70 min. Furthermore, the spiked samples of pork and milk were also tested by the detection system, and the recoveries ranged from 88.0% to 103.8%, with the coefficient of variation less than 12.8%. Taking advantages of sensitivity, rapidly and good selectivity, the proposed strategy in this study has great application potential in detection of foodborne pathogens in food.

Abstract:The rapid detection of low concentrations of Salmonella Typhimurium ( S. Typhimurium) is an essential preventive measure for food safety and prevention of foodborne illness. The study presented in this paper addresses this critical issue by proposing a single mode-tapered seven core-single mode (STSS) fiber ring laser (FRL) biosensor for S. Typhimurium detection. The experimental results show that the specific detection time of S. Typhimurium is less than 20 min and the wavelength shift can achieve −0.906 nm for an S. Typhimurium solution (10 cells/mL). Furthermore, at a lower concentration of 1 cell/mL applied to the biosensor, a result of −0.183 nm is observed in 9% of samples (1/11), which indicates that the proposed FRL biosensor has the ability to detect 1 cell/mL of S. Typhimurium. In addition, the detection results in chicken and pickled pork samples present an average deviation of −27% and −23%, respectively, from the measured results in phosphate buffered saline. Taken together, these results show the proposed FRL biosensor may have potential applications in the fields of food safety monitoring, medical diagnostics, etc.

Abstract:In this paper, a novel high-sensitivity optical sensor based on cascaded two Fabry-Perot interferometers (FPIs) has been investigated theoretically and experimentally. In order to improve the sensitivity, the two FP interferometers were designed with slightly different free spectrum ranges (FSRs) to produce Vernier effect. The experimental results showed that the sensitivity of the sensor is 23,794.6 nm/RIU for the wavelength interrogation and 4602.3 dB/RIU for the intensity interrogation, much higher than that of the conventional sensors for the bulk refractive index (RI) sensing. Finally, the reaction between Streptomycin monoclonal antibody and Streptomycin-BSA was measured, showing a great potential for low-cost and portable systems in biochemical sensing.
Homogenous Detection of Listeria Monocytohenes Dna with Molecule Beacon and Highly Fluorescent Bioconjugated Nanoparticles Probe

Abstract:Based on molecular beacon recognition and nanoparticles labeling technique, we constructed a sensitive detection method for target DNA of Listeria monocytogenes in homogeneous system. Using FITC-IgG complex as nuclear materials, FITC-IgG@SiO2 core/shell fluorescence nanoparticles were successfully synthesized. It effectively prevented the disclosure of FITC in traditional preparation process. Then the prepared fluorescence nanoparticles and Au nanoparticles were used to label 5' and 3' end of L. monocytogenes sequence-specific molecular beacon probe, respectively. The molecular beacon-fluorescent nanoparticles probe was successfully constructed. Under the optimized conditions, it was found that the fluorescent intensity is proportional to the concentration of target DNA of L. monocytogenes over the range of 1 similar to 200 pmol/L with a detection limit of 0.3 pmol/L (3S/N), and the relative standard deviation is 2.6% (50 pmol/L, n=11). The proposed method was applied to detect L. monocytogenes in food samples, the results obtained are identical to that of by an official standard method.

Abstract:In this study, we explore the potential of 1D ZnO–Au nanocomposites as innovative label-free photoluminescence (PL) immunosensors for rapidly detecting Listeria monocytogenes, a significant concern in food safety. We synthesized ZnO nanorods (ZnO_NR) and nanowires (ZnO_NW), followed by Au deposition to create ZnO_NR/Au and ZnO_NW/Au nanocomposites. Our analyses, including SEM, TEM, Raman spectroscopy, and photoluminescence (PL), revealed distinct structural and optical properties of these nanocomposites, especially noting the superior crystallinity and stability of ZnO_NR/Au. The biosensor performance was evaluated through PL sensitivity to Anti-Listeria antibodies, demonstrating that ZnO_NR with higher concentration of Au nanoparticles exhibited higher sensitivity and a lower limit of detection (LOD), attributed to a greater density of Listeria binding sites. The developed biosensor demonstrated a remarkable limit of detection (LOD) of 8.3 × 10 2 CFU/mL, rivaling or surpassing conventional culture-based methods and some molecular techniques. This research underscores the critical role of Au deposition duration in optimizing biosensor performance and presents a promising advancement in rapid and sensitive Listeria detection, with significant implications for enhancing food safety protocols.

Abstract:Pathogenic bacteria contamination poses a major threat to human health. The detection of low-abundance bacteria in complex samples has always been a knotty problem, and high-sensitivity bacterial detection remains challenging. In this work, a novel magnetic platform with high enrichment efficiency for L. monocytogenes was developed. The magnetic platform was designed by branched polyglutamic acid-mediated indirect coupling of cefepime on magnetic nanoparticles (Cefe-PGA-MNPs), and the specific enrichment of low-abundance L. monocytogenes in real samples was achieved by an external magnet, with a capture efficiency over 90%. A controllable and highly active platinum-palladium nanozyme was synthesized and further introduced in the magnetic nanoplatform for the construction of enzymatic colorimetric biosensor. The total detection time for L. monocytogenes was within 100 min. The colorimetric signals generated by labelled nanozyme were corresponding to different concentrations of L. monocytogenes , with a limit of detection (LOD) of 3.1 × 10 1 CFU/mL, and high reliability and accuracy (with a recovery rate ranging from 96.5% to 116.4%) in the test of real samples. The concept of the developed method is applicable to various fields of biosensing that rely on magnetic separation platforms.

Abstract:In this paper, simple imine-based organic fluorophore 4-amino-3-(anthracene-9 yl methyleneamino) phenyl (phenyl) methanone (APM) has been synthesized via a greener approach and the same was used to construct a fluorescent immunoassay for the detection of Listeria monocytogenes (LM). A monoclonal antibody of LM was tagged with APM via the conjugation of the amine group in APM and the acid group of anti-LM through EDC/NHS coupling. The designed immunoassay was optimized for the specific detection of LM in the presence of other interfering pathogens based on the aggregation-induced emission mechanism and the formation of aggregates and their morphology was confirmed with the help of scanning electron microscopy. Density functional theory studies were done to further support the sensing mechanism-based changes in the energy level distribution. All photophysical parameters were measured by using fluorescence spectroscopy techniques. Specific and competitive recognition of LM was done in the presence of other relevant pathogens. The immunoassay shows a linear appreciable range from 1.6 × 106-2.7024 × 108 cfu/mL using the standard plate count method. The LOD has been calculated from the linear equation and the value is found as 3.2 cfu/mL, and this is the lowest LOD value reported for the detection of LM so far. The practical applications of the immunoassay were demonstrated in various food samples, and their accuracy obtained was highly comparable with the standard existing ELISA method.

Abstract:Infections from invasive Listeria monocytogenes (L. monocytogenes) frequently occur in food and can cause high morbidity and death. Thus, the sensitive, specific, and rapid detection of L. monocytogenes is critical for ensuring food safety and public health. Herein, a fluorescence immunoassay for trace L. monocytogenes detection was designed based on guinea pig antibody-functionalized magnetic nanoparticles (Fe3O4 NPs/pAb1) and rabbit antibody-anchored CdZnTe quantum dots (CdZnTe QDs/pAb2). Because of the antibody-directed magnetic separation and long-wave fluorescent emission for CdZnTe QD indication, the constructed immunoassay strategy presented excellent anti-interference performance toward a biological matrix. The immunosensor exhibited a wide detection range of 1 to 109 CFU mL-1 for L. monocytogenes and a low limit of detection (LOD) of 1 CFU mL-1, achieving an exceptionally sensitive detection of trace L. monocytogenes. Meanwhile, the immunosensor showed good specificity and had a short time-consumption of 60 min to realize the accurate determination of trace Listeria monocytogenes in spiked tap water and pasteurized milk samples.

Abstract:Background: Rapid and sensitive detection of foodborne pathogens in food plays a crucial role in controlling outbreaks of foodborne diseases, of which Listeria monocytogenes and Salmonella typhimurium are representative and notable pathogens. Thus, it's of great importance to achieve the effective detection of these pathogens. However, the most common detection methods (culture-based technique, Polymerase Chain Reaction and immunological methods) have disadvantages that cannot be ignored, such as time-consuming, laborious, complex sample preparation process, and the possibility of cross-reaction. Hence, it is essential to develop a facile detection method for the pathogens with high sensitivity and specificity to avoid the above-mentioned disadvantages. Results: We report a label-free visual platform for the simultaneous capture and detection of Listeria monocytogenes and Salmonella typhimurium. For the first time, we have prepared polydimethylsiloxane-Chromotrope 2R membrane which serves as the substrate for bacterial capture and enrichment through the formation of specific recognition sites. The positively charged Pt-covalent organic framework combines with the pathogens through surface charge interaction, thereby the label-free sandwich platform is formed. Remarkable peroxidase activity of Pt-covalent organic framework converts the conversion of bacterial quantity into amplified color signal by catalyzing 3,3',5,5'-Tetramethylbenzidine to oxidized 3,3',5,5'-Tetramethylbenzidine. The platform demonstrates the capability to identify two representative food-borne pathogens within a time frame of 100 min, exhibiting high sensitivity and excellent specificity without the interference from non-target bacteria. The limit of detection of the visual platform toward Listeria monocytogenes and Salmonella typhimurium was 1.61 CFU mL-1 and 1.31 CFU mL-1, respectively. And the limit of quantification toward Listeria monocytogenes and Salmonella typhimurium was 4.94 CFU mL-1 and 2.47 CFU mL-1, respectively. The relative standard derivations of the visual platform for both bacteria were lower than 4.9 %. Furthermore, our proposed platform has obtained reliable and satisfactory results on analyzing diverse food samples. Significance: This research expands the application of a label-free platform combined with unlabeled nanocomponents in the rapid isolation and detection of diverse of food-borne pathogens. The platform possesses the advantages of simple operation and real-time monitoring, without complicated sample pretreatment process. The whole detection process can realize the simultaneous monitoring of Listeria monocytogenes and Salmonella typhimurium within 100 min. Furthermore, it is also of reference significance for the detection of other common pathogens.

Abstract:A novel strip test system combining immunomagnetic separation with lateral flow immunoassay (LFIA) was established for the accurate detection of Listeria monocytogenes. In this system, a pair of matched monoclonal antibodies was used to construct a sandwich immunoassay, in which superparamagnetic particles were coupled with one of the antibodies as a labeled antibody to capture the target bacteria, while the other antibody was immobilized on the detection zone. After a 20-min reaction, the strips were analyzed by a novel instrument which could detect the magnetic signal of the immunocomplex in a magnetic field. Sensitivity evaluation showed that the limit of detection (LOD) of the superparamagnetic LFIA system for L. monocytogenes was 10(4) CFU/mL, which was at least one log lower than conventional LFIA. No cross-reaction was observed when Salmonella, Escherichia coli O157:H7, or three types of harmless Listeria strains were tested. Further evaluation with actual food samples indicated that the superparamagnetic LFIA system showed 100 % concordance with real-time PCR. Therefore, this novel superparamagnetic LFIA system could be used as a rapid, sensitive, and specific method for the detection of L. monocytogenes.

Abstract:Listeria monocytogenes is one of the major foodborne pathogens, which may cause serious food safety problems and illnesses in humans and animals. Consequently, sensitive, fast and reliable detection methods, as well as effective sample preparation methods are of great demanding. This study established a magnetic separation method based on a aptamer functionalized positively charged magnetic beads (Fe3O4@aptamer) and constructed a fluorescent test strip sensor for rapid, sensitive and specific detection of Listeria monocytogenes. Benefit from the dual recognition and signal amplification process of Fe3O4@aptamer enrichment and PCR of hly gene, the fluorescent strip sensor for detection of Listeria monocytogenes was reliable and sensitive, a linear curve was obtained in the range of 1.0×102-1.9×108 CFU mL-1, and the detection limit was 1.0×102 CFU mL−1. The detection can be done in 3 h without culture enrichment. Furthermore, the developed method was successfully applied for the detection of Listeria monocytogenes in pork tenderloin, the recoveries ranged from 91.1% to 97.1%, with the coefficient of variation less than 23.4%, revealing the feasible and reliable application of this method in practical sample. The proposed fluorescent strip sensor is rapid, sensitive and specific, which has great application prospects in the field of pathogenic bacterium detection.

Abstract:In recent years, nanozymes have been widely used in the field of biosensing and food safety testing due to their advantages of low cost, high stability, easy modification and adjustable catalytic activity. However, how to reduce the signal interference generated by reducing substances, macromolecules and colored substances in the food matrix in nanozymes-based colorimetric sensing is still a major challenge. In this paper, using Listeria monocytogenes as a model analyte, sodium sulfonyl methacrylate (SBMA) polymers were modified onto cotton swabs by photothermal polymerization and combined with Listeria monocytogenes-specific aptamer (Apt1) to prepare swabs that can specifically capture and isolate Listeria monocytogenes from complex matrices (SBMA/Apt1 cotton swab). In addition, in combination with the inhibitory effect of the aptamer (Apt2) on the oxidase activity of Mn3O4 NPs, a colorimetric biosensor based on nanozymes that can quantitatively, sensitively, and specifically identify Listeria monocytogenes in food products was constructed. The results showed that the colorimetric signal of the method was linear with the concentration of Listeria monocytogenes in the range of 2.83-2.83 × 105 CFU/mL, and the limit of detection was 2.64 CFU/mL, which can be used for the detection of Listeria monocytogenes in complex environments and food samples.

Abstract:Listeria monocytogenes ( L. monocytogenes ) is a ubiquitous foodborne pathogens responsible for listeriosis, which is associated with high morbidity and mortality. Development of simple and fast methods for L. monocytogenes detection is of great needed. In this study, we present a simple method for L. monocytogenes detection combined aptamers decorated magnetic beads for selective capture and gold nanoparticles (AuNPs) mediated signal amplication. A NH 2 labeled aptamer specific to L. monocytogenes was decorated on magnetic beads for selective capture and isolation of L. monocytogenes from food sample. AuNPs was functionalized with sulfhydryl modified primers specific to hly gene of L. monocytogenes in acidic environments. The AuNP@primers could catalyze TMB-H 2 O 2 to TMB 2+ and generate a significant blue color, this reaction was found to be enhanced by DNA. AuNP@primers was used as a signal indicator for the presence of L. monocytogenes DNA by enhancing the catalytic reaction of TMB-H 2 O 2 . The colour change was observed by naked eye and the optical density of TMB 2 + was analyzed to quantify the result. The developed assay was highly specific and the limit of detection was 10 1 CFU/mL of L. monocytogenes . For the detection of L. monocytogenes in spiked food samples, the recoveries were ranging from 73.63 to 84.21% with the coefficient of variation less than 2.41% in meat, and ranged from 94.13 to 100.71% with the coefficient of variation less than 2.82% in milk, respectively. Therefore, the developed colorimetric assay is suitable for food sample testing, the detection takes less than 1.5 h, which enables on-site screening. This study offers a promising approach for rapid, sensitive and accurate detection of L. monocytogenes in food.
