Abstract:Photosynthetic biohybrid systems based on purple bacteria and semiconducting nanomaterials are promising platforms for sustainable solar-powered chemical production. However, these types of biohybrid systems have not been fully developed to date, and their energy utilization and electron transfer mechanisms are not well understood. Herein, a Rhodospirillum rubrum -CdS biohybrid system was successfully constructed. The photosynthetic activity and photoelectrochemical properties of biohybrid system were analyzed. Chromatographic and spectroscopic studies confirmed the metabolic activities of R. rubrum cells were effectively augmented by surface-deposited CdS nanoparticles and validated with increased H2 evolution, polyhydroxybutyric acid (PHB) production, and solid biomass accumulation. Energy consumption and metabolic profiles of R. rubrum -CdS biohybrid system exhibited a growth phase-dependent behaviour. Photoelectrochemical study confirmed that light-excited electrons from CdS enhanced photosynthetic electron flow of R. rubrum cells. Monochromatic light modulated photoexcitation of biohybrid system was utilized to explore interfacial electron transfer between CdS and R. rubrum cells, and the results showed that CdS enhanced the utilization of blue light by R. rubrum cells. This work investigated the feasibility and prospect of utilizing R. rubrum in semi-artificial photosynthesis of valuable products, and offered insights into the energy utilization and the electron transfer mechanism between nanomaterials and purple bacteria.
In-situ construction of biomineralized cadmium sulfide-Rhodopseudomonas palustris hybrid system: Mechanism of synergistic light utilization

Abstract:Sulfide biomineralization is a microorganism-induced process for transforming the environmentally hazardous cadmium into useful resource utilization. This study successfully constructed cadmium sulfide nanoparticles-Rhodopseudomonas palustris (Bio-CdS NPs-R. palustris) hybrids. For the self-assembling hybrids, Bio-CdS NPs were treated as new artificial-antennas to enhance photosynthesis, especially under low light (LL). Bacterial physiological results of hybrids were significantly increased, particularly for cells under LL, with higher enhancement photon harvesting ability. The enhancement included the pigment contents, and the ratio of the peripheral light-harvesting complex Ⅱ (LH2) to light-harvesting Ⅰ (1.33 ± 0.01 under LL), leading to the improvements of light-harvesting, transfer, and antenna conversion efficiencies. Finally, the stimulated electron chain of hybrids improved bacterial metabolism with increased nicotinamide adenine dinucleotide (NADH, 174.5% under LL) and adenosine triphosphate (ATP, 41.1% under LL). Furthermore, the modified photosynthetic units were induced by the up-regulated expression of fixK, which was activated by reduced oxygen tension of the medium for hybrids. fixK up-regulated genes encoding pigments (crt, and bch) and complexes (puf, pucAB, and pucC), leading to improved light-harvesting and transfer, and transform ability. This study provides a comprehensive understanding of the solar energy utilization mechanism of in-situ semiconductor-phototrophic microbe hybrids, contributing to further theoretical insight into their practical application.
Reversing Electron Transfer Chain for Light-Driven Hydrogen Production in Biotic–Abiotic Hybrid Systems

Abstract:The biotic–abiotic photosynthetic system integrating inorganic light absorbers with whole-cell biocatalysts innovates the way for sustainable solar-driven chemical transformation. Fundamentally, the electron transfer at the biotic–abiotic interface, which may induce biological response to photoexcited electron stimuli, plays an essential role in solar energy conversion. Herein, we selected an electro-active bacterium Shewanella oneidensis MR-1 as a model, which constitutes a hybrid photosynthetic system with a self-assembled CdS semiconductor, to demonstrate unique biotic–abiotic interfacial behavior. The photoexcited electrons from CdS nanoparticles can reverse the extracellular electron transfer (EET) chain within S. oneidensis MR-1, realizing the activation of a bacterial catalytic network with light illumination. As compared with bare S. oneidensis MR-1, a significant upregulation of hydrogen yield (711-fold), ATP, and reducing equivalent (NADH/NAD+) was achieved in the S. oneidensis MR-1-CdS under visible light. Abstract:Engineered photosynthetic bacterium Rhodo-pseudomonas palustris is excellent at one-step CO 2 biomethanation and can use near-infrared light sources, overcoming the limitations of conventional photosynthetic systems. The current study constructed a biohybrid system that deposited CdS nanoparticles on R. palustris . This biohybrid system broadens the capture of sustainable solar energy, achieving a 155 nmol·mL −1 biological CH 4 production under full visible light irradiation, 13.4-fold of that by the pure R. palustris . The transcriptome profiles revealed that gene expression related to photosynthetic electron transfer chain, nitrogenase, nanofilaments, and redox stress defense was activated. Accordingly, we attributed the much-enhanced CO 2 biomethanation in the biohybrid system to the remarkable increase in the intracellular reducing power and the stronger rigidity of the cells assisted by photoexcited electrons from CdS nanoparticles. Our discovery offers insight and a promising strategy for improving the current CO 2 –CH 4 biomanufacturing system.

Abstract:Semiconductor-based biointerfaces are typically established either on the surface of the plasma membrane or within the cytoplasm. In Gram-negative bacteria, the periplasmic space, characterized by its confinement and the presence of numerous enzymes and peptidoglycans, offers additional opportunities for biomineralization, allowing for nongenetic modulation interfaces. We demonstrate semiconductor nanocluster precipitation containing single- and multiple-metal elements within the periplasm, as observed through various electron- and x-ray-based imaging techniques. The periplasmic semiconductors are metastable and display defect-dominant fluorescent properties. Unexpectedly, the defect-rich (i.e., the low-grade) semiconductor nanoclusters produced in situ can still increase adenosine triphosphate levels and malate production when coupled with photosensitization. We expand the sustainability levels of the biohybrid system to include reducing heavy metals at the primary level, building living bioreactors at the secondary level, and creating semi-artificial photosynthesis at the tertiary level. The biomineralization-enabled periplasmic biohybrids have the potential to serve as defect-tolerant platforms for diverse sustainable applications.

Abstract:Microbe–semiconductor biohybrids, which integrate microbial enzymatic synthesis with the light-harvesting capabilities of inorganic semiconductors, have emerged as promising solar-to-chemical conversion systems. Improving the electron transport at the nano–bio interface and inside cells is important for boosting conversion efficiencies, yet the underlying mechanism is challenging to study by bulk measurements owing to the heterogeneities of both constituents. Here we develop a generalizable, quantitative multimodal microscopy platform that combines multi-channel optical imaging and photocurrent mapping to probe such biohybrids down to single- to sub-cell/particle levels. We uncover and differentiate the critical roles of different hydrogenases in the lithoautotrophic bacterium Ralstonia eutropha for bioplastic formation, discover this bacterium's surprisingly large nanoampere-level electron-uptake capability, and dissect the cross-membrane electron-transport pathways. This imaging platform, and the associated analytical framework, can uncover electron-transport mechanisms in various types of biohybrid, and potentially offers a means to use and engineer R. eutropha for efficient chemical production coupled with photocatalytic materials.

Abstract:This study investigated the mediating effect of Triethanolamine on Fe@C- Rhodobacter sphaeroides hybrid photosynthetic system to achieve efficient biohydrogen production. The biocompatible Fe@C generates excited electrons upon exposure to light, releasing ferrum for nitrogenase synthesis, and regulating the pH of the fermentation environment. Triethanolamine was introduced to optimize the electron transfer chain, thereby improving system stability, prolonging electron lifespan, and facilitating ferrum corrosion. This, in turn, stimulated the lactic acid synthetic metabolic pathway of Rhodobacter sphaeroides , resulting in increased reducing power in the biohybrid system. The ternary coupling system was analyzed through the regulation of concentration, initial pH, and light intensity. The system achieved the highest total H 2 production of 5410.9 mL/L, 1.29 times higher than the control (2360.5 mL/L). This research provides a valuable strategy for constructing ferrum-carbon-based composite-cellular biohybrid systems for photo-fermentation H 2 production.

Abstract:Photosensitizing bacterial cells with semiconductor nanoparticles is an attractive way to enhance fermentative hydrogen production metabolism via harnessing solar energy as extra driving force, but the enhancement degree is typically limited by inefficient utilization of extracellular photo-electrons. Here, an intimately-coupled inorganic-bio hybrid with enhanced hydrogen-producing activity was constructed by utilizing self-assembled selenide/ sulfide semiconductor nanoparticles (CdSexS1-x) in Escherichia coli as the light harvester. Due to circumvention of transmembrane electron transfer limitation, this hybrid exhibited 2.6-fold higher hydrogen production rate than those with extracellular nanoparticles and achieved light energy conversion efficiency as high as 27.6%. The incorporation of photocatalysis did not impair the bacterial viability, attributed to an efficient scavenging of photo-excited holes by metabolites (e.g., lactate) and minimal reactive oxygen species production. Overall, the light-assisted fermentation system developed in this study offer opportunities for sustainable production of bio-hydrogen and may be extended to bio-photocatalytic production of other valuable chemicals.

Abstract:Members of the genus Shewanella are known for their versatile electron accepting routes, which allow them to couple decomposition of organic matter to reduction of various terminal electron acceptors for heterotrophic growth in diverse environments. Here, we report autotrophic growth of Shewanella oneidensis MR-1 with photoelectrons provided by illuminated biogenic CdS nanoparticles. This hybrid system enables photosynthetic oscillatory acetate production from CO2 for over five months, far exceeding other inorganic-biological hybrid system that can only sustain for hours or days. Biochemical, electrochemical and transcriptomic analyses reveal that the efficient electron uptake of S. oneidensis MR-1 from illuminated CdS nanoparticles supplies sufficient energy to stimulate the previously overlooked reductive glycine pathway for CO2 fixation. The continuous solar-to-chemical conversion is achieved by photon induced electric recycling in sulfur species. Overall, our findings demonstrate that this mineral-assisted photosynthesis, as a widely existing and unique model of light energy conversion, could support the sustained photoautotrophic growth of non-photosynthetic microorganisms in nutrient-lean environments and mediate the reversal of coupled carbon and sulfur cycling, consequently resulting in previously unknown environmental effects. In addition, the hybrid system provides a sustainable and flexible platform to develop a variety of solar products for carbon neutrality.

Abstract:There is an increasing trend in semi-artificial photosynthesis systems that combine living cells with inorganic semiconductors to activate a bacterial catalytic network. However, these systems face various challenges, including electron-hole recombination, photocorrosion, and the generation of photoexcited radicals by semiconductors, all of which impair the efficiency, stability, and sustainability of biohybrids. We first focus on a reverse strategy to improve highly efficient CO2 photoreduction on biosynthesized inorganic semiconductors using an electron conduit in the electroactive bacterium S. oneidensis MR-1. Due to the suppressed charge recombination and photocorrosion on CdS, the maximum photocatalytic production rate of formate in water was 2650 μmol g-1 h-1 (with a selectivity of ca.100%), which ranks high among all photocatalysts and is the highest for inorganic-biological hybrid systems in an all-inorganic aqueous environment. The reverse enhancement effect of electrogenic bacteria on photocatalysis on semiconductors inspires new insight to develop a new generation of bio-semiconductor catalysts for solar chemical production.
A surface-display biohybrid approach to light-driven hydrogen production in air.

Abstract:Solar-to-chemical production by artificial and bioinspired photosynthetic systems is of tremendous interest to help solve current global energy and environmental problems. We developed a bioinorganic hybrid system for photocatalytic hydrogen production under aerobic conditions by combining light-harvesting semiconductors, hydrogenase catalysis, and self-aggregation of whole bacterial cells. We induced hydrogen production via self-photosynthesis in engineered Escherichia coli cells, which were originally designed for bioremediation, with in situ biosynthesis of biocompatible cadmium sulfide nanoparticles using a surface-display system. We also introduced a biomimetic silica encapsulation strategy into the engineered E. coli cells, enabling this hybrid system to continuously produce hydrogen for 96 hours, even under natural aerobic conditions. This biohybrid catalytic approach may serve as a general strategy for solar-to-chemical production.
Solar-Driven CO2 Reduction Using a Semiconductor/Molecule Hybrid Photosystem: From Photocatalysts to a Monolithic Artificial Leaf

Abstract:The synthesis of organic chemicals from H2O and CO2 using solar energy is important for recycling CO2 through cyclical use of chemical ingredients produced from CO2 or molecular energy carriers based on CO2. Similar to photosynthesis in plants, the CO2 molecules are reduced by electrons and protons, which are extracted from H2O molecules, to produce O2. This reaction is uphill; therefore, the solar energy is stored as the chemical bonding energy in the organic molecules. This artificial photosynthetic technology mimicking green vegetation should be implemented as a self-standing system for on-site direct solar energy storage that supports CO2 recycling in a circular economy. Herein, we explain our interdisciplinary fusion methodology to develop hybrid photocatalysts and photoelectrodes for an artificial photosynthetic system for the CO2 reduction reaction (CO2RR) in aqueous solutions. The key factor for the system is the integration of uniquely different functions of molecular transition-metal complexes and solid semiconductors. A metal complex catalyst and a semiconductor appropriate for a CO2RR and visible-light absorption, respectively, are linked, and they function complementary way to catalyze CO2RR under visible-light irradiation as a particulate photocatalyst dispersion in solution. It has also been proven that Ru complexes with bipyridine ligands can catalyze a CO2RR as photocathodes when they are linked with various semiconductor surfaces, such as those of doped tantalum oxides, doped iron oxides, indium phosphides, copper-based sulfides, selenides, silicon, and others. These photocathodes can produce formate and carbon monoxide using electrons and protons extracted from water through potential-matched connections with photoanodes such as TiO2 or SrTiO3 for oxygen evolution reactions (OERs). Benefiting from the very low overpotential of an aqueous CO2RR at metal complexes approaching the theoretical lower limit, the semiconductor/molecule hybrid system demonstrates a single tablet-formed monolithic electrode called "artificial leaf." This single electrode device can generate formate (HCOO-) from H2O and CO2 in a water-filled single-compartment reactor without requiring a separation membrane under unassisted or bias-free conditions, either electrically or chemically. The reaction proceeds with a stoichiometric electron/hole ratio and stores solar energy with a solar-to-chemical energy conversion efficiency of 4.6%, which exceeds that of plants. In this Account, the key results that marked our milestones in technological progress of the semiconductor/molecule hybrid photosystem are concisely explained. These results include design, proof of the principle, and understanding of the phenomena by time-resolved spectroscopies, synchrotron radiation analyses, and DFT calculations. These results enable us to address challenges toward further scientific progress and the social implementation, including the use of earth-abundant elements and the scale-up of the solar-driven CO2RR system.
Solar-driven carbon dioxide fixation using photosynthetic semiconductor bio-hybrids

Abstract:Solar-driven conversion of carbon dioxide to value-added carbon products is an ambitious objective of ongoing research efforts. However, high overpotential, low selectivity and poor CO2 mass transfer plague purely inorganic electrocatalysts. In this instance, we can consider a class of biological organisms that have evolved to achieve CO2 fixation. We can harness and combine the streamlined CO2 fixation pathways of these whole organisms with the exceptional ability of semiconducting nanomaterials to harvest solar energy. A novel nanomaterial-biological interface has been pioneered in which light-capturing cadmium sulfide nanoparticles reside within individual organisms essentially powering biological CO2 fixation by solar energy. In order to further develop the photosensitized organism platform, more biocompatible photosensitizers and cytoprotective strategies are required as well as elucidation of charge transfer mechanisms. Here, we discuss the ability of gold nanoclusters to photosensitize a model acetogen effectively and biocompatibly. Additionally, we present innovative materials including two-dimensional metal organic framework sheets and alginate hydrogels to shield photosensitized cells. Finally, we delve into original work using transient absorption spectroscopy to inform on charge transfer mechanisms.
Heterostructure-Engineered Semiconductor Quantum Dots toward Photocatalyzed-Redox Cooperative Coupling Reaction

Abstract:Semiconductor quantum dots have been emerging as one of the most ideal materials for artificial photosynthesis. Here, we report the assembled ZnS-CdS hybrid heterostructure for efficient coupling cooperative redox catalysis toward the oxidation of 1-phenylethanol to acetophenone/2,3-diphenyl-2,3-butanediol (pinacol) integrated with the reduction of protons to H2. The strong interaction and typical type-I band-position alignment between CdS quantum dots and ZnS quantum dots result in efficient separation and transfer of electron-hole pairs, thus distinctly enhancing the coupled photocatalyzed-redox activity and stability. The optimal ZnS-CdS hybrid also delivers a superior performance for various aromatic alcohol coupling photoredox reaction, and the ratio of electrons and holes consumed in such redox reaction is close to 1.0, indicating a high atom economy of cooperative coupling catalysis. In addition, by recycling the scattered light in the near field of a SiO2 sphere, the SiO2-supported ZnS-CdS (denoted as ZnS-CdS/SiO2) catalyst can further achieve a 3.5-fold higher yield than ZnS-CdS hybrid. Mechanistic research clarifies that the oxidation of 1-phenylethanol proceeds through the pivotal radical intermediates of •C(CH3)(OH)Ph. This work is expected to promote the rational design of semiconductor quantum dots-based heterostructured catalysts for coupling photoredox catalysis in organic synthesis and clean fuels production.
Enhanced photo-fermentative hydrogen production by constructing Rhodobacter capsulatus-ZnO/ZnS hybrid system

Abstract:This study incorporated ZnO/ZnS nanoparticles with Rhodobacter capsulatus SB1003, forming a hybrid system to promote photo-fermentative hydrogen production. The results indicate that the material's photocatalytic activity and concentration significantly affected hydrogen yield. The addition of ZnO/ZnS exhibited a more significant auxiliary effect than ZnO and achieved an approximately 30% increase in hydrogen production compared to the control group. ZnO/ZnS enhanced the production of extracellular polymers, thereby strengthening the synergy between the nanomaterials and the bacteria. The photogenerated electrons from ZnO/ZnS were utilized by the photosynthetic bacteria. Furthermore, the activity of nitrogenase was enhanced, resulting in improved hydrogen production performance. This study provides insights into hydrogen production by photosynthetic bacteria with the assistance of inorganic semiconductor nanomaterials.
Anaerobic self-assembly of regenerable bacteria-quantum dots hybrid for solar hydrogen production

Abstract:Inorganic-biological hybrid systems (bio-hybrid), comprising fermentative bacteria and inorganic semiconductor photosensitizers for synergistic utilization of solar energy and organic wastes, offer opportunities for sustainable fuel biosynthesis, but the low quantum efficiency, photosensitizer biotoxicity and inability for self-regeneration are remaining hurdles to overcome. Here, we unveiled a previously-neglected role of oxygen in suppressing the biosynthesis of cadmium selenide quantum dots (CdSe QDs) and the metabolic activities of Escherichia coli, and accordingly adopted a simple oxygen-regulation strategy to enable self-assembly of bacterial-QDs hybrid for efficient solar hydrogen production. Shifting from aerobic to anaerobic biosynthesis significantly lowered the intracellular reactive oxygen species level and raise the NADPH and thiol-protein production, enabling two-order-of-magnitude higher bio-QDs synthesis rate and resulting in CdSe-rich products. The bacteria with abundant biocompatible intracellular bio-QDs naturally formed high-activity, self-regenerable bio-hybrid and achieved quantum efficiency of 28.7% for visible-light-driven photocatalytic hydrogen production, outperforming all the existing existing bio-hybrids. It also exhibited high stability during cyclic operation and robust performance for treating real wastewater under simulated sunlight. Our work provides valuable new insights into the metallic nanomaterial biosynthesis process to guide the design of self-assembled bio-hybrid towards sustainable energy and environmental applications.

Abstract:Artificial photosynthesis by microbe-semiconductor biohybrid systems has been demonstrated as a valuable strategy in providing sustainable energy and in carbon fixation. However, most of the developed biohybrid systems for light harvesting employ heavy metal materials, especially cadmium sulfide (CdS), which normally cause environmental pollution and restrict the widespread of the systems. Herein, we constructed an environmentally friendly biohybirid system based on a typical acetogenic bacteria, Moorella thermoacetica, coupling with a carbon-based semiconductor, graphitic carbon nitride (g-C3N4), to realize light-driven carbon fixation. The proposed biohybrid system displayed outstanding acetate productivity with a quantum yield of 2.66 ± 0.43 %. Non-targeted proteomic analysis indicated that the physiological activity of the bacteria was improved, coupling with the non-toxic material. We further proposed the mechanisms of energy generation, electron transfer and CO2 fixation of the irradiated biohybrid system by proteomic and metabolomic characterization. With the photoelectron generated in g-C3N4 under illumination, CO2 is finally converted to acetate via the Wood-Ljungdahl pathway (WLP). Other associated pathways were also proved to be activated, providing extra energy or substrates for acetate production. The study reveals that the future focus of the development of biohybrid systems for light harvesting can be on the metal-free biocompatible material, which can activate the expression of the key enzymes involved in the electron transfer and carbon metabolism under light irradiation.

Abstract:Highly efficient inorganic light absorbers combined with highly specific biocatalysts, i.e., chemolithoautotrophic microbes, have recently been proposed for solar-powered biomanufacturing to pave the way in artificial photosynthesis. In this work, we studied the global protein and metabolite changes of a Moorella thermoacetica-CdS (cadmium sulfide quantum dots) hybrid system during light harvesting. The Wood-Ljungdahl pathway (WLP) that has been considered as the main approach for CO2 fixation was found to be highly activated by CdS. Targeted metabolite quantification revealed that energy-metabolism-associated glycolysis and the tricarboxylic acid (TCA) cycle were also activated. Therefore, we propose an energy-conservation scheme to the M. thermoacetica-CdS hybrid system, i.e., glycolysis and the TCA cycle along with the oxidation of acetyl-coenzyme A (CoA) and NADH, in addition to the well-documented chemiosmotic proton gradient-driven ATP production by ATPase. We expect that the study can be helpful to guide the design of effective and biocompatible photo-biocatalytic systems in the future.

Abstract:Despite the potential of the S‐scheme artificial photosynthetic system in driving photocatalytic hydrogen generation, conventional approaches tend to focus too heavily on a single reduction site. These systems still suffer poor redox ability and charge separation impairing the photocatalytic hydrogen evolution reaction. To overcome this limitation, we propose a novel strategy of designing a double S‐scheme system that leverages dual reduction sites, thereby preserving energetic photo‐electrons and holes to enhance apparent quantum efficiency. In this work, we put forth a double S‐scheme junction consisting of CdS nanospheres decorated with anatase TiO2 nanoparticles coupled with graphitic carbon nitride. The as‐prepared catalyst exhibits a remarkable hydrogen evolution rate of 26.84 mmol g−1 h−1 and an apparent quantum efficiency as high as 40.2% at the wavelength of 365 nm. This enhancement of photocatalytic hydrogen evolution performance is ascribed to the efficient separation and transportation induced by the double S‐scheme. Supporting this, both theoretical calculations and comprehensive spectroscopy tests (both in‐situ and ex‐situ) affirm the efficient charge transportation across the catalyst interface. Moreover, substituting the reduction‐type catalyst CdS with other similar sulfides like ZnIn2S4, ZnS, MoS2 and In2S3 further confirms the feasibility of the proposed double S‐scheme configuration. Our findings provide a pathway to designing more effective double S‐scheme artificial photosynthetic systems, opening up fresh perspectives in enhancing photocatalytic hydrogen evolution performance. Abstract:The rise of inorganic–biohybrid organisms for solar-to-chemical production has spurred mechanistic investigations into the dynamics of the biotic–abiotic interface to drive the development of next-generation hybrid systems. The model system, cyanobacteria–gold nanoparticle hybrids, combines a light harvester with a photosynthetic bacterium to drive the reduction of CO2 to glycerol with improved efficiency and increased glycerol production by 14.6%, in comparison to cyanobacteria only. In this work, we report insights into this unique photochemical behavior and propose a charge-transfer pathway from Au nanoparticle to cyanobacteria. Transient absorption (TA) spectroscopy revealed that photoexcited electron transfer rates are on the order of a few ps to the potential electron acceptor in photosystem II. This work represents a promising platform to utilize a conventional spectroscopic methodology to extract insights from more complex biotic–abiotic hybrid systems.
