Abstract:Purpose To synthesize the dimer of GX1 and identify whether its affinity and targeting are better than those of GX1. To prepare Ga-68-DOTA-KEK-(GX1)(2) and to apply it to PET and Cerenkov imaging of gastric cancer. Methods Ga-68-DOTA-KEK-(GX1)(2) was prepared, and the labeling yield and stability were determined. Its specificity and affinity were verified using an in vitro cell binding assay and competitive inhibition test, cell immunofluorescence, and cell uptake and efflux study. Its tumor-targeting ability was determined by nano PET/CT and Cerenkov imaging, standardized uptake value (SUV), signal-to-background ratio (SBR) quantification, and a biodistribution study in tumor-bearing nude mice. Results Ga-68-DOTA-KEK-(GX1)(2) was successfully prepared, and the labeling yield was more than 97%. It existed stably for 90 min in serum. The binding of Ga-68-DOTA-KEK-(GX1)(2) to cocultured HUVECs (Co-HUVECs) was higher than that to human umbilical vein endothelial cells (HUVECs), BGC823 cells, and GES cells. It was also higher than that of Ga-68-DOTA-GX1, indicating that the dimer did improve the specificity and affinity of GX1. The binding of KEK-(GX1)(2) to Co-HUVECs was significantly higher than that of GX1. Additionally, the uptake of Ga-68-DOTA-KEK-(GX1)(2) by Co-HUVECs was higher than that of Ga-68-DOTA-GX1 and reached a maximum at 60 min. Nano PET/CT and Cerenkov imaging showed that the tumor imaging of the nude mice injected with Ga-68-DOTA-KEK-(GX1)(2) was clear, and the SUV and SBR value of the tumor sites were significantly higher than those of the nude mice injected with Ga-68-DOTA-GX1, indicating that the probe had better targeting in vivo. Finally, the biodistribution showed quantitatively that when organs such as the kidney and liver metabolized rapidly, the radioactivity of the tumor site of the nude mice injected with Ga-68-DOTA-KEK-(GX1)(2) decreased relatively slowly. At the same time, the percentage of injected dose per gram (%ID/g) of the tumor site was higher than that of other normal organs except the liver and kidney at 60 min, which indicated that the tumor had good absorption of the probe. Conclusion GX1 was modified successfully, and the in vivo and in vitro properties of the GX1 dimer were significantly better than those of GX1. The imaging probe, Ga-68-DOTA-KEK-(GX1)(2), was successfully prepared, which provides a candidate probe for PET and Cerenkov diagnosis of gastric cancer.
Novel Dual-Function Near-Infrared II Fluorescence and PET Probe for Tumor Delineation and Image-Guided Surgery.

Abstract:Accurate tumor identification is essential in cancer management. Incomplete excision of tumor tissue, however, negatively affects the prognosis of the patient. To accomplish radical excision of tumor tissue, radiotracers can be used that target tumor tissue and can be detected using a gamma probe during surgery. Intraoperative fluorescence imaging could allow accurate real-time tumor delineation. Herein, a novel dual-modal imaging platform using base-catalyzed double addition of thiols into a propiolamide scaffold has been developed, allowing for the highly efficient and selective assembly of various thiol units in a protecting-group-free manner. The first small-molecule based αvβ3-targeted NIR-II/PET probe 68Ga-SCH2 was concisely generated via this strategy and subsequently evaluated in mice bearing the U87MG xenograft. Excellent imaging properties such as good tumor uptake, high tumor contrast and specificity, tumor delineation and image-guided surgery were achieved in the small animal models. These attractive results of 68Ga-SCH2 allow it to be a promising αvβ3-targeted NIR-II/PET probe for clinical translation.
PET imaging of a 68Ga labeled modified HER2 affibody in breast cancers: from xenografts to patients

Abstract:Objective: Overexpression of human epidermal growth factor receptor-2 (HER2) in breast cancers provides promising opportunities for imaging and targeted therapy. Developing HER2 targeted positron emission tomography (PET) probes might be benefit for management of the disease. Small high-affinity scaffold proteins, affibodies, are ideal vectors for imaging HER2 overexpressed tumors. Despite of the initial success on development of 18F labeled ZHER2:342 affibody, the tedious synthesis producers, low yields and unfavorable pharmacokinetics may hinder the clinical use. 68Ga is an attractive positron emitter for PET imaging. A simple preparation of 68Ga labeled ZHER2:342 analog, 68Ga-NOTA-MAL-Cys-MZHER2:342, was reported in the study. The in vivo performances of the tracer for assessing HER2 status in breast cancers were also evaluated. Methods: NOTA-MAL conjugated Cys-MZHER2:342 was radiolabeled with 68Ga. The probe was evaluated by in vitro tests including stability and cell binding studies in breast cancer cells with different HER2 levels. In vivo evaluation was performed in mice bearing tumors using microPET imaging and biodistribution experiments. A PET/CT imaging study was initially performed in patients with breast cancers. Results: The tracer was synthesized in a straightforward chelation method with satisfactory non-decay corrected yield (81±5%) and radiochemical purity (>95%). In vivo micro-PET imaging showed that HER2 high levels expressed BT474 xenografts were more clear visualized than HER2 low levels expressed MCF-7 tumors (16.12 ± 2.69 ID%/g vs 1.32 ± 0.19 ID%/g at 1 h post-injection). The outcome was consistent with the immunohistochemical analysis. No significant radioactivity was accumulated in healthy tissues (less than 2% ID/g) except kidneys. In a preliminary clinical study, 68Ga-NOTA-MAL-Cys-MZHER2:342 PET imaging allowed more high-contrast detection of HER2 positive primary tumors (maximum standardized uptake value = 2.16±0.27) than those in HER2 negative primary focus (maximum standardized uptake value = 0.32±0.05). No detectable side-effects were found. Conclusion: In summary, this study indicates the significant efficiency of the 68Ga labeled HER2 affibody. Preclinical and clinical studies support the possibility of monitoring HER2 levels in breast cancers using 68Ga-NOTA-MAL-Cys-MZHER2:342. Advances in knowledge: The research investigated the feasibility of a 68Ga labeled HER2 affibody modified with a hydrophilic linker for breast cancer PET imaging. Favorable outcomes showed that the probe might be valuable for determining HER2 status of the disease.

Abstract:Breast cancer has become a huge burden with continued rise of incidence and death rate worldwide. Various methods for diagnosis and therapy of breast cancer have met the challenges of lack of complete information about the tumor location and limited therapy efficacy. Although aggregation-induced emission luminogens (AIEgens) have shown great promise for various cancer treatment applications, they may be incompetent for deep-seated tumor diagnosis due to the limited penetration depth. Herein, we designed and prepared a radiolabeled AIEgen-based organic photothermal agent for bimodal PET/fluorescence imaging-guided breast tumor photothermal therapy. The prepared multifunctional nanoparticles (Ga-68-TPA-TTINC NPs) with NIR-II fluorescence, gamma irradiation and photothermal conversion property could be efficiently taken up by tumor cells and induce reactive oxygen species burst in vitro, further boosting the photothermal treatment of tumor in vivo. More importantly, the nanoprobe could target and clearly visualize 4T1 tumor xenografts through PET and NIR-II fluorescence imaging with high tumor/muscle ratio up to 4.8, which provides a promising tool and solution for breast tumor theranostics.

Abstract:The overexpression of HER2 is pivotal in the initiation and progression of breast cancer. Developing HER2-targeted radiotracers is crucial for noninvasive assessment of HER2 expression, patient selection for HER2-targeted therapy, monitoring treatment response, and identifying resistance. Here, we reported a nonsite-specific coupled radiotracer, 68Ga-NOTA-SNA004-His6, and a site-specific coupled radiotracer, 68Ga-NODAGA-SNA004-GSC, based on a novel HER2 nanobody, SNA004. Both radiotracers exhibited high affinity, specific targeting, and rapid clearance in vitro and in vivo. Additionally, these tracers and trastuzumab showed noncompetitive binding to HER2. Compared to 68Ga-NOTA-SNA004-His6, 68Ga-NODAGA-SNA004-GSC demonstrated significantly reduced renal and liver uptake. PET/CT imaging with 68Ga-NODAGA-SNA004-GSC sensitively detected the responsiveness of various tumor models to trastuzumab and its antibody-drug conjugates (ADCs). Overall, the site-specific coupled radiotracer 68Ga-NODAGA-SNA004-GSC offered significant advantages in biodistribution and signal-to-noise ratio, making it a valuable tool for monitoring HER2 expression levels before, during, and after trastuzumab and ADC treatment.
A novel 68Ga-labeled cyclic peptide molecular probe based on the computer-aided design for noninvasive imaging of PD-L1 expression in tumors

Abstract:Programmed death-ligand 1 (PD-L1) serves as a crucial biomarker for guiding the screening of cancer patients and the stratification of immunotherapy. However, due to the high heterogeneity of tumors, the current gold standard for detecting PD-L1 expression (immunohistochemistry) fails to comprehensively evaluate the overall PD-L1 expression levels in the body. Fortunately, the use of PD-L1 targeted radiotracers enables quantitative, real-time, and noninvasive assessment of PD-L1 expression levels and dynamics in tumors. Notably, analyzing the binding mode between the precursor and the target protein to find linker binding sites that do not affect the activity of the target molecule can greatly enhance the successful development of molecular probes. This study introduced a groundbreaking cyclic peptide molecular probe called 68Ga-DOTA-PG1. It was derived from the BMS-71 cyclic peptide and was specifically designed to evaluate the expression of PD-L1 in tumors. The radiolabeling yield of 68Ga-DOTA-PG1 surpassed 97% while maintaining a radiochemical purity of over 99%. In vitro experiments demonstrated the effective targeting of PD-L1 in tumor cells by 68Ga-DOTA-PG1, with significantly higher cellular uptake observed in A375-hPD-L1 cells (PD-L1 + ) compared to A375 cells (PD-L1-). Biodistribution and PET imaging studies consistently showed specific accumulation of 68Ga-DOTA-PG1 in A375-hPD-L1 tumors, with a maximum uptake of 11.06 ± 1.70% ID/g at 2 h, significantly higher than the tumor uptake in A375 cells (1.70 ± 0.17% ID/g). These results strongly indicated that 68Ga-DOTA-PG1 held great promise as a PET radiotracer for imaging PD-L1-positive tumors.

Preclinical evaluation of a dual-receptor targeted tracer [68Ga]Ga-HX01 in 10 different subcutaneous and orthotopic tumor models

Abstract:Purpose: The integrin αvβ3 and aminopeptidase N (APN/CD13) play vital roles in the tumor angiogenesis process. They are highly expressed in a variety of tumor cells and proliferating endothelial cells during angiogenesis, which have been considered as highly promising targets for tumor imaging. Arginine-glycine-aspartic (RGD) and asparagine-glycine-arginine (NGR) are two peptides specifically binding to the integrin αvβ3 and CD13, respectively. In this study, we optimized our previously developed probe and preclinically evaluated the new integrin αvβ3 and CD13 dual-targeted probe, NOTA-RGD-NGR (denoted as HX01) radiolabeled with 68Ga, in 10 different subcutaneous and orthotopic tumor models. Methods: The specific activity and radiochemical purity of [68Ga]Ga-HX01 were identified. The dual-receptor targeting ability was confirmed by a series of blocking studies and partly muted tracers using BxPC-3 xenograft model. The dynamic imaging study and dose escalation study were explored to determine the optimal imaging time point and dosage in the BxPC-3 xenograft model. Next, we established a variety of subcutaneous and orthotopic tumor models including pancreas (BxPC-3), breast (MCF-7), gallbladder (NOZ), lung (HCC827), ovary (SK-OV-3), colorectal (HCT-8), liver (HuH-7), stomach (NUGC-4), and glioma (U87) cancers. All models underwent [68Ga]Ga-HX01 PET/CT imaging about 2 weeks post-inoculation, with a subset of them undergoing [18F]FDG PET/CT scan performed concurrently, and their results were compared. In addition, ex vivo biodistribution studies were also performed for verifying the semi-quantitative results of the non-invasive PET images. Results: [68Ga]Ga-HX01 significantly outperformed single target probes in the BxPC-3 xenograft model. All blocking and single target groups exhibited significantly descending tumor uptake. The high tumor uptakes were found in BxPC-3, MCF-7, and NOZ subcutaneous tumors (%ID/g > 1.1), while middle uptakes were observed in HCC827, SK-OV-3, HCT-8, and HuH-7 subcutaneous tumor (%ID/g 0.7-1.0). Due to the low background, the tumor-to-muscle and tumor-to-blood ratios of [68Ga]Ga-HX01 were higher than that of [18F]FDG. Conclusions: [68Ga]Ga-HX01, as a dual target imaging agent, exhibited superior in vivo performance in different subcutaneous and orthotopic mice models of human tumors over [18F]FDG and its respectively mono-receptor targeted agents, which warrants the future clinical translation for tumor imaging.
Targeting HER2-positive Gastric Cancer with a Novel 18F-Labeled ZHER2:342 Probe

Abstract:To realize the diagnosis of HER2-positive gastric cancer via PET imaging, herein, a new kind of 18F-labeled HER2 affibody probe was created; the bifunctional maleimide derivative 1,4,7-triazacyclononane-1,4,7-triacetic acid (NOTA-MAL) was first coupled to a polypeptide, and the resulting compound was subsequently labeled with the 18FAl complex. The binding characteristics of the probe were assessed using both in vitro studies and in vivo microPET imaging and biodistribution experiments. Immunohistochemical staining was performed to confirm the expression level of HER2 in the studied cell lines and tumors. The probe was successfully produced with the radiochemical purity of more than 95%. The NCI N87 cell-associated radioactivity was 19.31 ± 1.01% AD, and it decreased to 0.83 ± 0.04% AD per 106 cells after blocking HER2 as early as 15 minutes post-incubation (p < 0.05). A competition binding assay between radiolabeled and non-radioactive affibody molecules with NCI N87 indicated that the IC50 was 8.10 nM. The microPET imaging and biodistribution of human gastric cancer xenografts demonstrated that the probe could specifically accumulate in tumors at early time points. Protein detection confirmed a strong HER2 expression in NCIN87 and a weak HER2 expression in SGC7901. In conclusion, 18FAl-NOTA-MAL-Cys-GGGRDN(M0)-ZHER2:342 was successfully prepared via a one-step method. The favorable preclinical data showed specific and effective tumor targeting capacity of the proposed probe; this revealed that the probe proposed herein might have potential application in gastric cancer imaging.

Abstract:Tc-99m- and I-131-labeled arginine-arginine-leucine (RRL) peptides have shown the feasibility of tumor imaging in our previous studies. However, there have been no reports using RRL peptide for positron emission tomography (PET) imaging. Noninvasive Detection of HER2 Expression in Gastric Cancer by 64Cu-Nota-trastuzumab in PDX Mouse Model and in Patients

Abstract:The purpose of this study was to establish the quality control and quantify the novel 64Cu-NOTA-Trastuzumab in gastric cancer patient-derived xenografts (PDX) mice models and patients by applying the molecular imaging technique. Trastuzumab was labeled with 64Cu using NCS-Bz-NOTA as bifunctional chelator, and hIgG1 was labeled with the same procedures as a negative control agent. HER2-positive (case 176, n = 12) and HER2-negative (case 168, n = 3) PDX models were established and validated by Western blot, DNA amplification, and immunohistochemistry (IHC). Both models were conducted for micro-PET imaging by tail injection of 18.5 MBq of 64Cu-NOTA-Trastuzumab or 64Cu-NOTA-hIgG1. Radioprobe uptake in tumor and main organs was quantified by region of interested (ROI) analysis of the micro-PET images and autoradiography. Finally, gastric cancer patients were enrolled in preliminary 64Cu-NOTA-Trastuzumab PET/CT scans. NOTA-Trastuzumab was efficiently radiolabeled with 64Cu over a 99% radiochemical purity and 17.5 GBq/μmol specific activity. The immune activity was preserved as the nonmodified antibody, and the radiopharmaceutical proved to be stable for up to 5 half-decay lives of 64Cu both in vitro and in vivo. Two serials of PDX gastric cancer models were successfully established: case 176 for HER2 positive and case 168 for HER2 negative. In micro-PET imaging studies, 64Cu-NOTA-Trastuzumab exhibits a significant higher tumor uptake (11.45 ± 0.42 ID%/g) compared with 64Cu-NOTA-IgG1 (3.25 ± 0.28 ID%/g, n = 5, p = 0.0004) at 36 h after intravenous injection. Lower level uptake of 64Cu-NOTA-Trastuzumab (6.35 ± 0.48 ID%/g) in HER2-negative PDX tumor models further confirmed specific binding of the radioprobe. Interestingly, the coinjection of 2.0 mg of Trastuzumab (15.52 ± 1.97 ID%/g) or 2.0 mg of hIgG1 (15.64 ± 3.54 ID%/g) increased the 64Cu-NOTA-Trastuzumab tumor uptake in PDX tumor (HER2+) models compared with 64Cu-NOTA-Trastuzumab alone ( p < 0.05) at 36 h postinjection. There were good correlations between micro-PET images and IHC ( n = 4) and autoradiography in PDX (HER2+) tumor tissues. Therefore, 64Cu-NOTA-Trastuzumab successfully translated to clinical PET imaging, and 64Cu-NOTA-Trastuzumab PET/CT scan in gastric cancer patients showed good detection ability. In conclusion, we reported quality control and application of novel 64Cu-NOTA-Trastuzumab for HER2 expression in PDX gastric cancer mice models and gastric cancer patients. Moreover, 64Cu-NOTA-Trastuzumab holds great potential for noninvasive PET detection, staging, and follow-up of HER2 expression in gastric cancer.
SPECT/CT Imaging of the Novel HER2-Targeted Peptide Probe 99Mtc-Hynic-h6f in Breast Cancer Mouse Models.

Abstract:Overexpression of human epidermal growth factor receptor 2 (HER2) plays important roles in tumorigenesis and tumor progression in breast cancer. Nuclear imaging of HER2 expression in tumors might detect all HER2-positive tumors throughout the body and guide HER2-targeted therapies for patients. We therefore aimed to develop a HER2-targeted peptide probe for breast cancer imaging. A novel SPECT imaging probe, Tc-99m-HYNIC-H6F, was prepared and then evaluated in breast cancer animal models. Methods: The HER2-targeted peptide H6F (YLFFVFER) was conjugated with the bifunctional chelator hydrazinonicotinamide (HYNIC). Tc-99m-HYNIC-H6F was prepared, and the in vivo characteristics of Tc-99m-HYNIC-H6F were investigated in MDA-MB-453 (HER2-positive) and MDA-MB-231 (HER2-negative) models using small-animal SPECT/CT. Moreover, to investigate the specificity of the H6F peptide toward HER2 and the potential applications in monitoring therapies involving trastuzumab, unlabeled H6F and trastuzumab were used as blocking agents in cell competition studies and SPECT imaging. Results: A standard tricine/trisodium triphenylphosphine-3,3',3 ''-trisulfonate labeling procedure demonstrated that the radiochemical purity was greater than 95%. Tc-99m-HYNIC-H6F displayed excellent HER2-binding specificity both in vitro and in vivo. SPECT/CT imaging revealed that the MDA-MB-453 tumors were clearly visualized (percentage injected dose per gram, 3.58 +/- 0.01 at 30 min after injection), whereas the signals in HER2-negative MDA-MB-231 tumors were much lower (0.73 +/- 0.22 at 30 min after injection). Tumor uptake of MDA-MB-453 was blocked by the coinjection of excess H6F but not by excess trastuzumab. Conclusion: The Tc-99m-HYNIC-H6F peptide probe specifically accumulates in HER2-positive tumors and is therefore promising for the diagnosis of HER2-positive cancers. Because (99m)-HYNIC-H6F and trastuzumab target different regions of the HER2 receptor, this radiotracer also has great potential for monitoring the therapeutic efficacy of trastuzumab by rechecking the expression level of HER2 without blocking effect during therapy.
Preliminary Study of Radionuclide-Labeled MerTK-Targeting PET Imaging Agents for the Diagnosis of Melanoma

Abstract:MerTK PET imaging holds potential as a promising approach for assessing tumor aggressiveness and monitoring treatment response. Synthesis and Evaluation of (68)Ga-Labeled DOTA-2-deoxy-D-glucosamine As a Potential Radiotracer in Μpet Imaging.

Abstract:The purposes of this study were to develop an efficient method of labeling D-glucosamine hydrochloride with gallium 68 ((68)Ga) and investigate the imaging properties of the resulting radiotracer in a human tumor xenograft model using micro-positron emission tomography (μPET). The precursor compound 1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid (DOTA)-2-deoxy-D-glucosamine (DOTA-DG) was synthesized from D-glucosamine hydrochloride and 2-(4-isothiocyanatobenzyl)-DOTA. Radiolabeling of DOTA-DG with (68)Ga was achieved in 10 minutes using microwave heating. The labeling efficiency a nd radiochemical purity after purification of (68)Ga-DOTA-DG were ~85% and greater than 98%, respectively. In A431 cells, the percentages of (68)Ga-DOTA-DG and (18)F-FDG uptakes after 60 min incubation were 15.7% and 16.2%, respectively. In vivo, the mean ± standard deviation of (68)Ga-DOTADG uptake values in A431 tumors were 2.38±0.30, 0.75±0.13, and 0.39±0.04 percent of the injected dose per gram of tissue at 10, 30, and 60 minutes after intravenous injection, respectively. μPET imaging of A431-bearing mice clearly delineated tumors at 60 minutes after injection of (68)Ga-DOTA-DG at a dose of 3.7 MBq. (68)Ga-DOTA-DG displayed significantly higher tumor-to-heart, tumor-to-brain, and tumor-to-muscle ratios than (18)F-FDG did. Further studies are needed to identify the mechanism of tumor uptake of this new glucosamine-based PET imaging tracer.

Abstract:Objective To investigate the effect of 18 F labeled anti-HER2 affibody probe on the targeting HER2 overexpressed human gastric cancer with micro PET imaging . Methods Anti-HER2 specific affibody ZHER2:342 was obtained from chemical synthesis routes .The bifunctional maleimide derivative of 1,4,7-Triazacyclononane-1,4,7-triacetic acid (NOTA) was coupled to thiol-group of cysteine of ZHER2:342 to form the chelator-peptide conjugation .Then the newly produced 18 F was labeled to the conjugation through 18FAl one step labeling.The resulting radiolabeled product was analyzed by high performance liquid chromatography (HPLC).The characteristics of 18FAl-NOTA-MAL-Cys59 ZHER2:342 binding to NCI N87 were assessed by using the in vitro cell uptaking and blocking assays , competition binding assay.NOD SCID mice models bearing HER 2 positive human gastric cancer tumor was established . Micro PET imaging was conducted to evaluate the targeting ability of the probe with xenograft models . Results The radiochemical purity of 18 F labeled ZHER2:342 was more than 95%.The NCI N87 cell associated radioactivity was close to the platform , approximately(7.48 ±0.49)%ID, as early as 15 minute after incubation together .Blocking analysis demonstrated that the binding of 18 F-ZHER2:342 to living HER2-overexpressed cells was receptor mediated .Competition binding assay with NCI N 87 indicated IC50 was 9.4nM.The microPET imaging of human gastric cancer xenograft demonstrated that the radioactive probe could specifically accumulate in the tumor tissue of (7.22 ±0.24)%ID/g 30 min after injection.The quantitive date didn ’ t decrease significantly with time extending .The uptaking level decreased apparently after blocking HER2. Conclusions The novel 18 F labeled ZHER2:342 could be acquired easily and effectively targeting gastric cancer with HER 2 overexpression .

Abstract:In order to develop novel 68Ga‐labeled PET tracers for folate receptor imaging, two DOTA‐conjugated Pteroyl‐Lys derivatives, Pteroyl‐Lys‐DOTA and Pteroyl‐Lys‐DAV‐DOTA, were designed, synthesized and radiolabeled with 68Ga. Biological evaluations of the two radiotracers were performed with FR‐positive KB cell line and athymic nude mice bearing KB tumors. Both 68Ga‐DOTA‐Lys‐Pteroyl and 68Ga‐DOTA‐DAV‐Lys‐Pteroyl exhibited receptor specific binding in KB cells in vitro. The tumor uptake values of 68Ga‐DOTA‐Lys‐Pteroyl and 68Ga‐DOTA‐DAV‐Lys‐Pteroy were 10.06 ± 0.59%ID/g and 11.05 ± 0.60%ID/g at 2 h post‐injection, respectively. Flank KB tumor was clearly visualized with 68Ga‐DOTA‐DAV‐Lys‐Pteroyl by Micro‐PET imaging at 2 h post‐injection, suggesting the feasibility of using 68Ga‐labeled Pteroyl‐Lys conjugates as a novel class of FR targeted probes.
Preparation, biological characterization and preliminary human imaging studies of 68Ga-DOTA-IBA

Abstract:Purpose: In this study, DOTA-IBA was radiolabeled with 68 Ga and we determined the optimum labelling conditions and assessed the biological properties of 68 Ga-DOTA-IBA. We investigated the biodistribution of 68 Ga-DOTA-IBA in normal animals and undertook PET/CT imaging in humans. Finally, we explored the feasibility 68 Ga-DOTA-IBA as a bone imaging agent and demonstrated its potential for the therapeutic release of 177 Lu/ 225 Ac-DOTA-IBA. Methods: The controlled variables method was used to assess the impact of variables on the radiochemical purity of 68 Ga-DOTA-IBA. The biological properties of 68 Ga-DOTA-IBA were investigated. 68 Ga-DOTA-IBA micro-PET/CT imaging was performed on animals. Volunteers were recruited for 68 Ga-DOTA-IBA imaging and data were compared to 99m Tc-MDP imaging studies to calculate the target to non-target ratio (T/NT) of the lesions. Results: The prepared 68 Ga-DOTA-IBA had a radiochemical purity of >97% and demonstrated good biological properties with a good safety profile in normal mice. PET/CT imaging of the animals showed rapid blood clearance with high contrast between the bone and stroma. Human imaging showed that 68 Ga-DOTA-IBA could detect more lesions compared to 99m Tc-MDP and had a higher targeted to untargeted ratio. Conclusions: 68 Ga-DOTA-IBA is an osteophilic radiopharmaceutical that can be synthesized using a simple labelling method. 68 Ga-DOTA-IBA has high radiochemical purity and is stable in vitro stability. It is rapidly cleared from the blood, has low toxicity and has strong targeting to the bone with long retention times. We also found that it is rapidly cleared in non-target tissues and has high contrast on whole-body bone imaging. 68 Ga-DOTA-IBA PET/CT has potential as a novel bone imaging bone modality in patients with metastatic disease.

Abstract:Background and purpose: The immunogenic drugs represented by programmed cell death 1 (PD-1) and programmed death ligand-1 (PD-L1) have achieved great success in clinic. However, with the development of clinical application, the overall effective rate varies greatly. At present, detection of the expression of PD-1/PD-L1 before the start of tumor treatment is an important basis for clinical medication. However, the current detection method of PD-L1 expression requires the acquisition of patient specimens, which is traumatic. Therefore, there is an urgent need to develop a noninvasive method to detect PD-L1 expression in vivo. This study innovatively designed and synthesized a new positron emission tomography (PET) imaging agent targeting PD-L1 polypeptide 68Ga-DOTA-PDL1P. In this study, the labeling rate, radiochemical purity and stability of 68Ga-DOTA-PDL1P were evaluated for quality control. And the application was evaluated in the mouse model of melanoma in order to obtain a promising 68Ga-DOTA-PDL1P immune PET probe. Methods: Gallium-68 (68Ga) was used to label DOTA-PDL1P, and its radiochemical purity and stability were evaluated by quality control. To evaluate its specific targeting performance, the cell uptake and blocking experiments were carried out in murine melanoma B16-F10 cells. The biodistribution experiment was performed in B16-F10 tumor-bearing mice to analyze the metabolism of the PET imaging agent in vivo. The Tumor uptake of 68Ga-DOTA-PDL1P in B16-F10 tumor-bearing mice was observed by PET/CT imaging, and the tumor/muscle (T/M) ratio was semi-quantitatively analyzed. In addition, radioautography was performed to further analyze the tumor uptake of 68Ga-DOTA-PDL1P probe. Tumor cell proportion score (TPS) and combined positive score (CPS) were used to analyze the immunohistochemical staining results to evaluate the expression of PD-L1 in tumor tissues. Results: The labeling efficiency and radiochemical purity of 68Ga-DOTA-PDL1P were more than 99%, and the radiochemical purity of 68Ga-DOTA-PDL1P was more than 90% after 3 h of incubation in PBS and fetal bovine serum. Cellular uptake results showed that 68Ga-DOTA-PDL1P could be specifically taken up by B16-F10 cells, and the specific uptake could be blocked by PD-L1P peptide competitive binding. Biodistribution assay showed that 68Ga-DOTA-PDL1P was mainly excreted through the urinary system. The results of microPET/CT imaging of tumor-bearing mice showed a high uptake of 68Ga-DOTA-PDL1P in tumor tissues, and the T/M ratio was 6.7. The autoradiography results confirmed that 68Ga-DOTA-PDL1P high uptake in the tumor was consistent with the PD-L1 positive area of immunohistochemical staining, which proved that 68Ga-DOTA-PDL1P could specifically bind to PD-L1 positive malignant tumor cells. Conclusion: The novel 68Ga-DOTA-PDL1P PET imaging agent synthesized in this study has good stability and can specifically target PD-L1 positive tumor cells, which is a promising new immune PET probe targeting PD-L1 protein.

Abstract:Due to the temporal and spatial heterogeneity of human epidermal growth factor receptor 2 (HER2) expression in breast tumors, immunohistochemistry (IHC) cannot accurately reflect the HER2 status in real time, which may cause misguided treatment decisions. HER2-specific imaging can noninvasively determine HER2 status in primary and metastatic tumors. In this study , HER2 expression in breast cancer patients was determined in vivo by SPECT/CT of 99m Tc-HP-Ark2, comparing with PET/CT of 18 F-FDG lesion by lesion.

Abstract:Angiotensin-converting enzyme 2 (ACE2) is closely related to tumor formation. We developed the radiolabeled peptide pair 68Ga/177Lu-labeled 1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid (DOTA)-conjugated DX600 (68Ga/177Lu-HZ20), for the targeting and mapping of ACE2-overexpressing tumors. 68Ga/177Lu-HZ20 was prepared with a routine labeling method. HepG2ACE2+/HepG2WT cell lines were used to evaluate the specificity of 68Ga/177Lu-HZ20. Pharmacokinetics, biodistribution, and micro-PET/CT and -SPECT/CT imaging were performed, and radiation dosimetry was estimated. Immunohistochemistry (IHC) staining was performed to assess the expression of ACE2 in tumors. The radiolabeling yields of 68Ga/177Lu-HZ20 were 88.49 ± 8.57% (n > 10) and 84.71 ± 9.75% (n > 10), with specific activities of (18.74 ± 3.72) × 106 and (17.85 ± 1.62) × 106 GBq/mol, respectively. 68Ga/177Lu-HZ20 showed significant differences in the cellular uptake of HepG2ACE2+/HepG2WT cells and fast clearance in KM mice. Moreover, HepG2ACE2+ tumors were clearly visualized in 68Ga/177Lu-HZ20 micro-PET/SPECT images. Based on micro-PET/CT, the standard uptake value (SUVmax) of HepG2ACE2+ tumors was 0.66 ± 0.02 at 30 min postinjection, IHC confirmed the high expression of ACE2 in HepG2ACE2+ tumors. In PET/CT images, the SUVmean of volunteer 1 was higher than the 18F-FDG value in the same lesion. 68Ga/177Lu-HZ20 was successfully obtained and showed high and specific uptake in tumors overexpressing ACE2. They may serve as paired probes for ACE2-targeting theranostics.

Abstract:Arg-Gly-Asp(RGD) and bombesin(BBN) can specifically target integrin αvβ3 and gastrin releasing peptide receptor(GRPR),respectively.We designed and synthesized a RGD-BBN heterodimeric peptide with both the RGD and BBN motifs in one molecule.RGD-BBN was conjugated with a bifunctional chelate NOTA and then labeled with 68Ga.The in vitro cell uptake and in vivo microPET imaging of 68Ga-RGD-BBN were tested in both MDA-MB-435(GRPR-/integrin αvβ3+) and T47D(GRPR+/integrin αvβ3 low expression) breast cancer cells and nude mouse models,respectively.68Ga-NOTA-RGD and 68Ga-NOTA-BBN counterparts were also synthesized and compared.It is shown that 68Ga-RGD-BBN could sensitively detect the breast cancers with either or both high expression of integrin αvβ3 and GRPR,and the dual-targeted probe possessed higher tumor uptake than the corresponding monomeric counterparts.68Ga-NOTA-RGD-BBN is a promising agent for PET imaging of both GRPR+/(integrin αvβ3 low expression) and GRPR-/integrin αvβ3+ cancers.
