Abstract:Two polynuclear Ni(II) complexes, [Ni 2 (L)(C 5 H 5 N) 2 ]⋅CH 2 Cl 2 (1) and [Ni 3 (L)( μ 1,3 OAc) 2 ] (2), were synthesized through the reaction of a double-armed salamo-type ligand H 4 L with Ni(NO 3 ) 2 ·6H 2 O (using pyridine as an auxiliary ligand) and Ni(OAc) 2 ·4H 2 O, respectively. Both complexes were characterized by X-ray crystallography, PXRD, and a range of spectroscopic techniques. Crystal structure analysis revealed that complex 1 has a dinuclear linear structure with coordinated pyridine, while complex 2 exhibits a trinuclear structure with central symmetry, where two phenoxides and two acetates bridge one of the units. The acetate bridges adopt the μ 2 -η 1 : η 1 coordination mode. Theoretical calculations, including molecular electrostatic potentials (MEPs) and density functional theory (DFT), were employed to predict reaction sites and assess the overall reactivity of the complexes. Both complexes 1 and 2 demonstrated catalytic activity, oxidizing 3,5-Ditert‐butyl catechol (3,5-DTBC) to the corresponding o-quinone (3,5-DTBQ), as monitored by UV-visible spectroscopy under ambient, aerobic conditions. Kinetic analysis using the Michaelis-Menten equation indicated turnover numbers (K cat ) of 599 h−1 and 443 h−1 for complexes 1 and 2, respectively. The mechanistic pathway of the catalytic reaction was further elucidated through ESI-mass spectrometry and cyclic voltammetry.

Abstract:Superoxide plays a key role in cell signaling, but can be cytotoxic within cells unless well regulated by enzymes known as superoxide dismutases (SOD). Nickel superoxide dismutase (NiSOD) catalyzes the disproportion of the harmful superoxide radical into hydrogen peroxide and dioxygen. NiSOD has a unique active site structure that plays an important role in tuning the potential of the nickel center to function as an effective catalyst for superoxide dismutation with diffusion controlled rates. The synthesis of structural and functional analogues of NiSOD provides a route to better understand the role of the nickel active site in superoxide dismutation. In this work, the synthesis of a series of nickel complexes supported by nitrogen rich pentadentate ligands is reported. The complexes have been characterized through absorption spectroscopy, mass spectrometry, and elemental analysis. X-ray absorption spectroscopy was employed to establish the oxidation state and the coordination geometry around the metal center. The reactivity of these complexes toward KO2 was evaluated to elucidate the role of the coordination sphere in controlling superoxide dismutation reactivity.
Influence of mixed thiolate/thioether versus dithiolate coordination on the accessibility of the uncommon +I and +III oxidation states for the nickel ion: an experimental and computational study

Abstract:Sulfur-rich nickel metalloenzymes are capable of stabilizing Ni(I) and Ni(III) oxidation states in catalytically relevant species. In an effort to better understand the structural and electronic features that allow the stabilization of such species, we have investigated the electrochemical properties of two mononuclear N(2)S(2) Ni(II) complexes that differ in their sulfur environment. Complex 1 features aliphatic dithiolate coordination ([NiL], 1), and complex 2I is characterized by mixed thiolate/thioether coordination ([NiL(Me)]I, 2I). The latter results from the methylation of a single sulfur of 1. The X-ray structure of 2I reveals a distorted square planar geometry around the Ni(II) ion, similar to what was previously reported by us for 1. The electrochemical investigation of 1 and 2(+) shows that the addition of a methyl group shifts the potentials of both redox Ni(II)/Ni(I) and Ni(III)/Ni(II) redox couples by about 0.7 and 0.6 V to more positive values. Through bulk electrolyses, only the mononuclear dithiolate [Ni(I)L](-) (1(-)) and the mixed thiolate/thioether [Ni(III)L(Me)](2+) (2(2+)) complexes were generated, and their electronic properties were investigated by UV-vis and EPR spectroscopy. For 1(-) (Ni(I), d(9) configuration) the EPR data are consistent with a d(x(2))(-)(y(2)) based singly occupied molecular orbitals (SOMOs). However, DFT calculations suggest that there is also pronounced radical character. This is consistent with the small g-anisotropy observed in the EPR experiments. The spin population (Mulliken analysis) analysis of 1(-) reveals that the main contribution to the SOMO (64%) is due to the bipyridine unit. Time dependent density functional theory (TD-DFT) calculations attribute the most prominent features observed in the electronic absorption spectrum of 1(-) to metal to ligand charge transfer (MLCT) transitions. Concerning 2(2+), the EPR spectrum displays a rhombic signal with g(x) = 2.236, g(y) = 2.180, and g(z) = 2.039 in CH(3)CN. The g(iso) value is larger than 2.0, which is consistent with metal based oxidation. The unpaired electron (Ni(III), d(7) configuration) occupies a Ni-d(z(2)) based molecular orbital, consistent with DFT calculations. Nitrogen hyperfine structure is observed as a triplet in the g(z) component of the EPR spectrum with A(N) = 51 MHz. This result indicates the coordination of a CH(3)CN molecule in the axial position. DFT calculations confirm that the presence of a fifth ligand in the coordination sphere of the Ni ion is required for the metal-based oxidation process. Finally, we have shown that 1 exhibits catalytic reductive dehalogenation activity below potentials of -2.00 V versus Fc/Fc(+) in CH(2)Cl(2).

Abstract:Novel tetranuclear Ni(II) complex [Ni 4 (L)(acac) 4 (EtOH) 2 ]⋅4CH 2 Cl 2 was synthesized by a rigid bis(salamo)-like ligand H 4 L and Ni(acac) 2 in ethanol/dichloromethane mixed solvents, and characterized by different physicochemical methods. The X-ray diffraction result showed that all Ni(II) atoms are hexa-coordinated with slightly distorted octahedral geometries, and four Ni(II) atoms are correlated with each other through inversion. Acetylacetone anions acted as auxiliary ligands for bidentate chelation, and the monodentate ethanol molecules of solvent participated in the coordination. The properties of the ligand H 4 L and its Ni(II) complex were studied, including molecular electrostatic potential surface (MEPs), Hirshfeld surface and interaction region indicator (IRI) analysis. Through the above study, the reaction active sites were visualized, and the intermolecular interactions of the Ni(II) complex were clarified, which is consistent with the crystal structure of the Ni(II) complex. In addition, fluorescence spectra of the ligand H 4 L and its Ni(II) complex were investigated.

Abstract:A new series of hexadentate thioether-containing ligands was designed. Five nickel(II) dicationic pseudo-octahedral complexes synthesised with them revealed different degrees of deviation from ideal geometry. The electronic absorption spectra confirm the formation of high-spin octahedral [NiN 4 S 2 ] 2+ chromophores with a large covalency typical for Ni(II) thioether complexes. DFT calculations help assign the chelating agent transitions, and support the Ni(II) high-spin ground state assignments, while suggesting that the SOMOs have mainly ligand character. The electrochemical behaviour of the complexes is consistent with irreversible n = 1 redox processes around −1.0 V, formally assignable to the Ni(II)/Ni(I) couple. Ligand reduction and large current augmentation due to catalytic H 2 production are also observed in this region

Abstract:Inorganic Chemistry teaches students the concept that modifications to ligand structures, especiallythe donor properties, can have a drastic impact on the reactivity and stability of the metal complexes. Experiments described here reinforce this concept through the investigation of two tetradentateligands derived from o-phenylenediamine and salicylaldehyde. The Schiff base ligand, H2salophen,reacts with Ni(OAc)2•4H2O to yield a maroon colored, square planar complex, Ni(salophen). Under thesame conditions, the amine-type ligand, H2salophan, forms a light-blue compound with a formulaNi(salophan)(HOAc). Complex Ni(salophan) free of acetate may be produced from the reaction of H2salophan with Ni(OAc)2•4H2O in the presence of NaOH, but undergoes ligand dehydrogenation toyield Ni(salophen). Students conducting these experiments have the opportunity to learn synthetictechniques and various characterization methods. Most importantly, the inquiry-guided experimentaldesign helps them develop critical thinking skills and apply acquired knowledge to solving a researchproblem in a laboratory course.
Organometallic Nickel Complexes Supported by Pyridinophane Ligands and Their Oxidative Reactivity

Abstract:The pyridinophane ligands RN3CX (X = H, Br) are well-established scaffolds that facilitat and stabilize nickel oxidative addition complexes to the proximal C(aryl)–X bond. In this study, we report the synthesis, detailed characterization, and reactivity of a series of Ni(II) and Ni(III) complexes supported by the MeN3CX ligand. Our findings demonstrate that Ni(II) complexes can be oxidized both chemically and aerobically to yield well-defined Ni(III) species. Excitingly, the Ni-disolvento complexes exhibit catalytic trifluoroethoxylation to generate the C–O coupled product. In addition, the Ni(III)-halide complex undergoes transmetallation with a Grignard reagent and subsequent C–C reductive elimination, while the β-hydride elimination side reaction is suppressed, outperforming its Ni(II) analogue.

Abstract:The key findings and significance of this report is the synthesis and characterization of two mononuclear nickel(II) complexes, [Ni(L)(val)].0.5MeOH ( 1 ) and [Ni(HL) 2 ](ClO 4 ) 2 ( 2 ) derived from a tetradentate Schiff base ligand HL and two different nickel(II) salts. These complexes were thoroughly characterized using techniques such as elemental analysis, IR spectroscopy, single crystal X-ray diffraction, and Hirshfeld surface analysis. The results revealed distinct coordination modes of the ligands in the complexes, leading to different non-covalent interactions and packing arrangements in their crystal structures. Specifically, complex 1 exhibited tetradentate coordination in addition to the bidentate o -vanillinate ligand, while complex 2 featured tridentate coordination with the zwitterionic form of the ligand. Hirshfeld surface analysis provided insights into the relative contributions of various intermolecular interactions, highlighting the significance of hydrogen bonding, tetrel bonding, and other non-covalent interactions in stabilizing the crystal structures. In general, this study contributes to the differences in structural integrity on variation in metal salts and the understanding in depth of various non-covalent interactions using Hirshfeld surface analysis tool.

Abstract:The nickel(II) complex with a tetradentate Schiff base ligand obtained by condensation of 1,3-propanediamine with salicylaldehyde (H2salpn) was studied in a variety of solvents at room temperature. The product, that is, the N',N''-propylenebis(salicylaldiminato)nickel(II) ([Ni(salpn)]) complex, is brown in color in the solid state. The properties of the ligand and complex were characterized by elemental analysis, solubility in common solvents, molar conductivities, and ultraviolet (UV) and visible (Vis) spectroscopy. The [Ni(salpn)] complex is easily soluble in common solvents such as chloroform, methanol, ethanol, dimethyl formamide, dimethyl sulfoxide, acetonitrile, dioxane, acetone, 2-propanol, and toluene-a necessary condition for observing solvatochromism. The molar conductivity values, equal to 0.0 S mol-1 cm2 in these solvents, point to a typical non-electrolyte behavior for this complex. Spectroscopic measurements were used to confirm the square-planar geometry of the species in solution and to determine the coordination properties of the donor atoms and their bonding abilities (CFM/AOM parameters), as well as trichromaticity coordinate calculations. The results obtained show that the interactions of the metal with the donors depend on the polarity of the solvent.

Abstract:Bimetallic transition metal complexes with site-specific redox properties offer a versatile platform for understanding electron polarization, intramolecular electron transfer processes, and customizing electronic and magnetic properties that might impact reactivity and catalyst design. Inspired by the dissymmetric nickel sites in the Acetyl CoA Synthase (ACS) Active Site, three new bimetallic Ni(N2S2)-Ni(S2C2R2) complexes based on Ni(N2S2) metalloligand donor synthons, Nid, in mimicry of the nickel site distal to the redox-active iron sulfur cluster of ACS, and nickel dithiolene receiver units, designated as Nip, , the nickel proximal to the 4Fe4S cluster, were combined to explore the influence of ligand environment on electronic structure and redox properties of each unit. The combination of synthons gave a matrix of three S-bridged dinickel complexes, characterized by x-ray crystallography, and appropriate spectroscopies. Computational modeling is connected to the electronic characteristics of the nickel donor and receiver units. This study demonstrated the intricacies of identifying sites of electrochemical redox processes, within multi-metallic systems containing non-innocent ligands.

Abstract:Many copper(I) complexes with aliphatic amine ligands have a strong tendency to disproportionate to copper(II) and elemental copper in solution at higher concentrations, making it difficult to isolate them and to study their reactivity. A series of copper(I) complexes with ligands based on tridentate N,N,N',N'',N''‐pentamethldiethylenetriamine (Me5dien) were synthesized that included terminal olefinic and aromatic groups. It could be shown that the olefinic side arms stabilized some of the copper(I) complexes. Whether and how strongly the complexes were stabilized depended on the position and length of the olefinic sidearm. Additionally, the reactivity of the copper(I) complexes towards dioxygen was investigated.

Abstract:Two trinuclear Co(II) and Ni(II) complexes, [{Co3(L)2(μ-OAc)2}2]·3CH2Cl2 (1) and [Ni3(L)2(μ-OAc)2(EtOH)2]·2EtOH (2) have been successfully obtained by a new nonsymmetrical salamo type ligand H2L with Co(OAc)2·4H2O and Ni(OAc)2·6H2O, respectively. Single crystal X-ray diffraction analysis revealed that complex 1 is an unsymmetric trinuclear structure, two tetradentate N2O2-donor ligand (L)2- units are coordinated to three Co(II) atoms where two terminal Co(II) atoms are penta-coordinated lateral twisting trigonal bipyramidal geometries but central Co(II) atom is hexa-coordinated lateral twisting octahedral configuration. Complex 2 is a centrosymmetrical trinuclear structure, all of the three Ni(II) atoms are hexa-coordinated lateral twisting octahedral configurations. In addition, complexes 1 and 2 have been characterized by physicochemical and spectroscopic methods. Molecular electrostatic potential analysis revealed that the phenolic oxygen atoms in the ligand H2L have nucleophilic reactions with Co(II) and Ni(II) atoms to form chemical bonds. Hirshfeld surfaces analyses showed that H···H/H···H interactions are the predominant, indicating the presence of great amount hydrogen bonds in complexes 1 and 2. IRI analyses indicated the presence of weak hydrogen bonding interactions that made complexes 1 and 2 more stable.

Abstract:Three new ONNO-donor tetradentate unsymmetrical salen ligands were synthesized by using o -phenyl diamine with substituted salicylaldehydes followed by a two-step reaction methodology. These three ligands by reaction with Cu(OAc) 2 .4H 2 O produced three new monomeric Cu(II) complexes, [ Cu II (L 1−3 ) ] ( 1 – 3 ). Elemental analysis, IR, UV–vis, NMR, and HR-ESI-MS techniques were used to analyze and characterize all the synthesized ligands and their corresponding metal complexes. Molecular structures of 1 – 3 were confirmed by the single-crystal-XRD analysis. Furthermore, the DNA binding ability of these complexes was checked through UV–vis, fluorescence spectroscopy, and also by circular dichroism studies. All the complexes were found to show an intercalation mode of binding with the K b value in the range of 10 4 –10 5 M −1 . Finally, 1 – 3 was tested against two malignant (HeLa and A549) and non-cancerous (NIH-3 T3) cell lines to check their in vitro antiproliferative activities. Among all, 1 is the most cytotoxic of the series having IC 50 values of 5.7 ± 0.9 and 6.0 ± 0.3 μM against HeLa and A549 cell lines, respectively. This result is also consistent with the DNA binding order. Furthermore, the apoptotic mode of cell death of all the complexes was also evaluated by DAPI, AO/EB, and Annexin V-FITC/PI double staining assays.

Abstract:Two new tri- and tetra-nuclear Ni(II)-based complexes, [{NiL(EtOH)(μ-OAc)}2Ni]•4EtOH (1) and [Ni4L2(μ3-OMe)4(MeOH)4] (2), supported by a more flexible salamo-type ligand H2L containing coumarin functional groups have been synthesized. Complex 1 is tri-nuclear, including three Ni(II) atoms with hexa-coordinated octahedral geometries. X-ray single-crystal structure analysis revealed that complex 2 is a novel tetra-nuclear structure previously unreported, and the geometry of four Ni(II) atoms with hexa-coordinate can be regarded as octahedron. Interestingly, due to the improvement of the flexibility of salamo-type ligand H2L, complex 2 is obtained by the form of intramolecular self-assembly. Moreover, the fluorescence properties of H2L and its Ni(II)-based complexes were studied. Subsequently, the supramolecular structure, Hirshfeld surfaces analyses and DFT calculations of the two Ni(II)-based complexes have been performed.

All the compoundswere synthesized and characterized by various spectroscopic analytical methods.Thein‐vitro antimalarial investigations revealed that the complex CuL exhibited equipotency with quinine drug having IC50 value 0.25 μg/mL.The compound L shows significant inhibition of bacterial spp.viz.E. Coli, P. Aeruginosa, and S. Aureus (MIC=12.5‐50μg/mL), while the compound CoL (MIC=12.5μg/mL) exhibited potency against gram‐positive bacteria. In the in‐vitro anti‐inflammatory study, the compound CuL has moderate activity than other tested compounds. The compound CuL showedthe highest anti‐malarial docking score with enzyme pLDH at ‐8.12 Kcal/mol. The DFT study also gives authentication of higher antimalarial activity of CuL due to high dipole moment. None of the potent compound was found cytotoxic towards verocell lines.

A Series of Ni Complexes Based on a Versatile ATCUN-Like Tripeptide Scaffold to Decipher Key Parameters for Superoxide Dismutase Activity

Abstract:The cellular level of reactive oxygen species (ROS) has to be controlled to avoid some pathologies, especially those linked to oxidative stress. One strategy for designing antioxidants consists of modeling natural enzymes involved in ROS degradation. Among them, nickel superoxide dismutase (NiSOD) catalyzes the dismutation of the superoxide radical anion, O2•-, into O2 and H2O2. We report here Ni complexes with tripeptides derived from the amino-terminal CuII- and NiII-binding (ATCUN) motif that mimics some structural features found in the active site of the NiSOD. A series of six mononuclear NiII complexes were investigated in water at physiological pH with different first coordination spheres, from compounds with a N3S to N2S2 set, and also complexes that are in equilibrium between the N-coordination (N3S) and S-coordination (N2S2). They were fully characterized by a combination of spectroscopic techniques, including 1H NMR, UV-vis, circular dichroism, and X-ray absorption spectroscopy, together with theoretical calculations and their redox properties studied by cyclic voltammetry. They all display SOD-like activity, with a kcat ranging between 0.5 and 2.0 × 106 M-1 s-1. The complexes in which the two coordination modes are in equilibrium are the most efficient, suggesting a beneficial effect of a nearby proton relay.

Abstract:What is the shape of a more flexible salamo-type ligand? Unexpectedly single crystals of the ligand and a uncommon tetranuclear chloride-bridged complex with Ni(II) [Ni 4 (L) 2 ( μ 2 Cl) 2 ( μ 3 OMe) 2 (MeOH) 2 ] were acquired. Single-crystal X-ray analyzes revealed the true structure of the ligand to be a twisted rectilinear shape, with a unique structure providing a diverse coordination environment for the metal ions. The involvement of halogens in the bridging is uncommon, and Single-crystal X-ray analysis of the complex showed that each deprotonated methoxy group bridges three Ni(II) metals, and then three Ni(II) metals are bridged by two chloride ions, each chloride ion bridges two Ni(II) metals. At this point the geometries of all four Ni(II) atoms adopt 6-coordinated octahedral configurations. It was demonstrated in UV–visible titration test that the metal-to-ligand ratios in solution and the structure of the complex obtained by single-crystal X-ray analysis are consistent. The properties were predicted analytically by DFT theory calculations and the UV–vis absorption spectra obtained by TD-DFT theory calculations in general coincide with the test results. The weak interactions in the complex were analyzed by cell freedom, Hirshfed surface analysis, ESP and IRI calculations. Finally, the fluorescence properties were explored in detail.
