Construction of a prognostic model of neuroblastoma in children

Abstract:Objective:This retrospective study aimed to analyze and screen independent prognostic factors for childhood neuroblastoma, and construct a Nomogram model to predict the prognosis.Methods:The data of 160 neuroblastoma cases were retrospectively analyzed from 2014 to 2019, independent prognostic factors were screened through univariate and multivariate Cox regression analysis, and a Nomogram model was constructed to predict the 3-year and 5-year event-free survival rates of the patients. By calculating the C-index and the area under the curve, and drawing a calibration curve for internal verification, the discrimination and consistency of the prediction model were evaluated, and the prognostic value of the model factors in the high-risk group was further studied.Results:MYCN amplification, neuron-specific enolase (NSE) at diagnosis, and imaging-defined risk factors were independent prognostic factors for childhood neuroblastoma. Based on the above factors, the Nomogram prediction model was constructed. Internal verification showed that the model C-index was 0.786, and the model discrimination and consistency were good. MYCN amplification and imaging-defined risk factors were not associated with the prognosis of the high-risk group ( P>0.05), while NSE at diagnosis was a significant prognostic factor of the high-risk group ( P=0.001). Conclusions:MYCN amplification, NSE, and image-defined risk factors at diagnosis are independent prognostic factors for neuroblastoma. Incorporating these factors and constructing a Nomogram model may accurately predict the prognosis.
