Abstract:Stimulated emission depletion (STED) microscopy is one of the breakthrough technologies that belong to far-field optical microscopy and can achieve nanoscale spatial resolution. We demonstrate a far-field optical nanoscopy based on continuous wave lasers with different wavelengths, i.e., violet and green lasers for excitation and STED, respectively. Fluorescent dyes Coumarin 102 and Atto 390 are used for validating the depletion efficiency. Fluorescent nanoparticles are selected for characterizing the spatial resolution of the STED system. Linear scanning of the laser beams of the STED system along one line of a microscope slide, which is coated with the nanoparticles, indicates that a spatial resolution of about 70 nm has so far been achieved. A two-dimensional image of the particle pattern of the STED system is constructed and compared with scanning confocal microscope. The present work has further extended the application of the STED microscopy into the blue regime.

Abstract:Stimulated emission depletion (STED) nanoscopy is one of the most well-developed nanoscopy techniques that can provide subdiffraction spatial resolution imaging. Here, we introduce dual-modulation difference STED microscopy (dmdSTED) to suppress the background noise in traditional STED imaging. By applying respective time-domain modulations to the two continuous-wave lasers, signals are distributed discretely in the frequency spectrum and thus are obtained through lock-in demodulation of the corresponding frequencies. The background signals can be selectively eliminated from the effective signal without compromise of temporal resolution. We used nanoparticle, fixed cell, and perovskite coating experiments, as well as theoretical demonstration, to confirm the effectiveness of this method. We highlight dmdSTED as an idea and approach with simple implementation for improving the imaging quality, which substantially enlarges the versatility of STED nanoscopy.

Abstract:Abstract Stimulated emission depletion microscopy (STED) is a powerful tool for studying nanoscale cell structure and activity, but the difficulties it encounters in multicolor imaging limit its application in biological research. To overcome the disadvantages of limited number of channels and high cost of multicolor STED imaging based on spectral identity, we introduced lifetime into live-cell multicolor STED imaging by separating selected dyes of the same spectrum by phasor analysis. Experimental results show that our method enables live-cell STED imaging with at least 4 colors, enabling observation of cellular activity beyond the diffraction limit.

Abstract:Stimulated emission depletion (STED) microscopy is a powerful diffraction-unlimited technique for fluorescence imaging. Despite its rapid evolution, STED fundamentally suffers from high-intensity light illumination, sophisticated probe-defined laser schemes, and limited photon budget of the probes. Here, we demonstrate a versatile strategy, stimulated-emission induced excitation depletion (STExD), to deplete the emission of multi-chromatic probes using a single pair of low-power, near-infrared (NIR), continuous-wave (CW) lasers with fixed wavelengths. With the effect of cascade amplified depletion in lanthanide upconversion systems, we achieve emission inhibition for a wide range of emitters (e.g., Nd3+, Yb3+, Er3+, Ho3+, Pr3+, Eu3+, Tm3+, Gd3+, and Tb3+) by manipulating their common sensitizer, i.e., Nd3+ ions, using a 1064-nm laser. With NaYF4:Nd nanoparticles, we demonstrate an ultrahigh depletion efficiency of 99.3 ± 0.3% for the 450 nm emission with a low saturation intensity of 23.8 ± 0.4 kW cm-2. We further demonstrate nanoscopic imaging with a series of multi-chromatic nanoprobes with a lateral resolution down to 34 nm, two-color STExD imaging, and subcellular imaging of the immunolabelled actin filaments. The strategy expounded here promotes single wavelength-pair nanoscopy for multi-chromatic probes and for multi-color imaging under low-intensity-level NIR-II CW laser depletion.

Abstract:Through the strategic application of upconversion rare-earth nanoparticles (UCNPs), this work has reduced the intensity of the traditional super-resolution by 2-3 orders of magnitude. It reveals a new mechanism of stimulated emission caused by the photon avalanche effect. With only 30mW continuous laser, resolution down to 28nm has been attained, which is only 1/36 of the excitation wavelength.

Abstract:Optical tuning of lanthanide-doped upconversion nanoparticles has attracted considerable attention over the past decade because this development allows the advance of new frontiers in energy conversion, materials science, and biological imaging. Here we present a rational approach to manipulating the spectral profile and lifetime of lanthanide emission in upconversion nanoparticles by tailoring their nonlinear optical properties. We demonstrate that the incorporation of energy distributors, such as surface defects or an extra amount of dopants, into a rare-earth-based host lattice alters the decay behavior of excited sensitizers, thus markedly improving the emitters' sensitivity to excitation power. This work provides insight into mechanistic understanding of upconversion phenomena in nanoparticles and also enables exciting new opportunities of using these nanomaterials for photonic applications.

Abstract:Abstract. We have designed and built a time-gated continuous wave stimulated emission depletion (CW-STED) nanoscopy to visualize microstructures beyond the diffraction limit. An off-line time-gating detection was performed with the help of time-correlated single-photon counting technique. Experimental results showed that before time-gating, the resolution of our system was about 75 nm with a depletion beam (592 nm) power of 200 mW. By using the off-line time-gating detection, the resolution was further improved to 38 nm. Biological samples were also used to test the performance of our time-gated CW-STED, and a resolution of 70 nm was achieved with a depletion beam (592 nm) power of 85 mW. Detailed principles of time-gated CW-STED were discussed in the text. The time-gated STED provides a better resolution with finite laser power.

Abstract:Stimulated emission depletion microscopy (STED) holds great potential in biological science applications, especially in studying nanoscale subcellular structures. However, multi-color STED imaging in live-cell remains challenging due to the limited excitation wavelengths and large amount of laser radiation. Here, we develop a multiplexed live-cell STED method to observe more structures simultaneously with limited photo-bleaching and photo-cytotoxicity. By separating live-cell fluorescent probes with similar spectral properties using phasor analysis, our method enables five-color live-cell STED imaging and reveals long-term interactions between different subcellular structures. The results here provide an avenue for understanding the complex and delicate interactome of subcellular structures in live-cell.

Abstract:Stimulated emission depletion (STED) nanoscopy plays a key role in achieving sub-50 nm high spatial resolution for subcellular live-cell imaging. To avoid re-excitation, the STED wavelength has to be tuned at the red tail of the emission spectrum of fluorescent probes, leading to high depletion laser power that might damage the cell viability and functionality. Herein, with the highly emissive silica-coated core-shell organic nanoparticles (CSONPs) enabling a giant Stokes shift of 150 nm, ultralow power STED is achieved by shifting the STED wavelength to the emission maximum at 660 nm. The stimulated emission cross section is increased by ∼20-fold compared to that at the emission red tail. The measured saturation intensity and lateral resolution of our CSONP are 0.0085 MW cm-2 and 25 nm, respectively. More importantly, long-term (>3 min) dynamic super-resolution imaging of the lysosomal fusion-fission processes in living cells is performed with a resolution of 37 nm.

Abstract:We describe the use of a layer-by-layer hierarchical nanostructure to exploit the synergy of different lanthanide ions for converting single wavelength excitation into emissions spanning the whole spectral region. By lining up a set of lanthanide ions with matched energy levels in a core–shell nanostructure, we demonstrate well-defined cascades of energy transfer that gives access to optical emissions from a large collection of lanthanide ions (Tb3+, Eu3+, Dy3+, Sm3+, Nd3+, Yb3+, and Er3+) after excitation into a common sensitizer of Ce3+ featuring a broad absorption. Through optimization of the nanoparticle structure and surface coating, high quantum yields of up to 90% are achieved. Our results highlight that the controlled energy cascades at nanometer scale provide new opportunities for applications such as fighting against counterfeiting and sensing small molecules.

Abstract:Stimulated emission depletion (STED) nanoscopy is a powerful sub-diffraction imaging tool to probe subcellular structures and organelles. Conventional organic dyes require high STED power (PSTED) to obtain sub-diffraction resolution, leading to serious photo-bleaching. Herein, this study demonstrates highly emissive silica-coated core-shell organic nanoparticles (CSONPs) as a new type of photostable probe with ultrahigh stimulated emission depletion efficiency for low-power super-resolution STED nanoscopy. The CSONPs offer (i) efficient red emission with high solid-state fluorescence quantum yields around 0.6, (ii) large Stokes shift of 150 nm and (iii) high photostability owing to silica shell protection. The stimulated emission depletion efficiency (η) of CSONPs was extremely high up to η = 99% (the highest value reported so far) with a saturation intensity as low as Isat = 0.18 MW cm-2. Moreover, this research demonstrates the super-resolution imaging of living HeLa cells stained using CSONPs with a lateral spatial resolution of 63 nm at an extremely low depletion power of ISTED = 0.89 MW cm-2 and a long-term stability >600 s at η = 80% without obvious fatigue. The excellent and comprehensive performances of the CSONPs are promising for super-resolution imaging in biological applications.

Abstract:Stimulated-emission depletion (STED) microscopy has profoundly extended our horizons to the subcellular level<a href="#ref-CR1">1</a>,<a href="#ref-CR2">2</a>,<a href="/articles/s41565-021-00927-y#ref-CR3">3</a>. However, it remains challenging to perform hours-long, autofluorescence-free super-resolution imaging in near-infrared (NIR) optical windows under facile continuous-wave laser depletion at low power<a href="/articles/s41565-021-00927-y#ref-CR4">4</a>,<a href="/articles/s41565-021-00927-y#ref-CR5">5</a>. Here we report downshifting lanthanide nanoparticles that enable background-suppressed STED imaging in all-NIR spectral bands (λexcitation = 808 nm, λdepletion = 1,064 nm and λemission = 850–900 nm), with a lateral resolution of below 20 nm and zero photobleaching. With a quasi-four-level configuration and long-lived (τ > 100 μs) metastable states, these nanoparticles support near-unity (98.8%) luminescence suppression under 19 kW cm−2 saturation intensity. The all-NIR regime enables high-contrast deep-tissue (~50 μm) imaging with approximately 70 nm spatial resolution. These lanthanide nanoprobes promise to expand the application realm of STED microscopy and pave the way towards high-resolution time-lapse investigations of cellular processes at superior spatial and temporal dimensions.

Abstract:Lanthanide-doped nanoparticles exhibit unique luminescent properties, including large Stokes shift, sharp emission bandwidth, high resistance to optical blinking, and photobleaching, as well as the unique ability to convert long-wavelength stimulation into short-wavelength emission. These attributes are particularly needed for developing luminescent labels as alternatives to organic fluorophores and quantum dots. In recent years, the well-recognized advantages of upconversion nanocrystals as biomarkers have been manifested in many important applications, such as highly sensitive molecular detection and autofluorescence-free cell imaging. However, their potential in multiplexed detection and multicolor imaging is rarely exploited, largely owing to the research lagging on multicolor tuning of these particles. Lanthanide doping typically involves an insulating host matrix and a trace amount of lanthanide dopants embedded in the host lattice. The luminescence observed from these doped crystalline materials primarily originates from electronic transitions within the [Xe]4f(n) configuration of the lanthanide dopants. Thus a straightforward approach to tuning the emission is to dope different lanthanide activators in the host lattice. Meanwhile, the host lattice can exert a crystal field around the lanthanide dopants and sometimes may even exchange energy with the dopants. Therefore, the emission can also be modulated by varying the host materials. Recently, the advance in synthetic methods toward high quality core-shell nanocrystals has led to the emergence of new strategies for emission modulation. These strategies rely on precise control over either energy exchange interactions between the dopants or energy transfer involving other optical entities. To provide a set of criteria for future work in this field, we attempt to review general and emerging strategies for tuning emission spectra through lanthanide doping. With significant progress made over the past several years, we now are able to design and fabricate nanoparticles displaying tailorable optical properties. In particular, we show that, by rational control of different combinations of dopants and dopant concentration, a wealth of color output can be generated under single-wavelength excitation. Strikingly, unprecedented single-band emissions can be obtained by careful selection of host matrices. By incorporating a set of lanthanide ions at defined concentrations into different layers of a core-shell structure, the emission spectra of the particles are largely expanded to cover almost the entire visible region, which is hardly accessible by conventional bulk phosphors. Importantly, we demonstrate that an inert-shell coating provides the particles with stable emission against perturbation in surrounding environments, paving the way for their applications in the context of biological networks.

Abstract:Stimulated emission depletion microscopy provides a powerful sub-diffraction imaging modality for life science studies. Conventionally, stimulated emission depletion requires a relatively high light intensity to obtain an adequate depletion efficiency through only light–matter interaction. Here we show efficient emission depletion for a class of lanthanide-doped upconversion nanoparticles with the assistance of interionic cross relaxation, which significantly lowers the laser intensity requirements of optical depletion. We demonstrate two-color super-resolution imaging using upconversion nanoparticles (resolution ~ 66 nm) with a single pair of excitation/depletion beams. In addition, we show super-resolution imaging of immunostained cytoskeleton structures of fixed cells (resolution ~ 82 nm) using upconversion nanoparticles. These achievements provide a new perspective for the development of photoswitchable luminescent probes and will broaden the applications of lanthanide-doped nanoparticles for sub-diffraction microscopic imaging.

Abstract:Rare-earth-doped upconversion nanoparticles are attracting considerable attention because of their stable, coherent, narrowband and multi-colour luminescence, which features less dephasing effects, crosstalk, photo-blinking and photo-bleaching compared with quantum dots and organic dyes. However, due to the 4 f –4 f forbidden transitions of rare-earth ions, upconversion nanoparticles exhibit long luminescence decay times, ranging from microseconds to milliseconds, restricting their application in time-dependent nanophotonic devices. Here we fabricate a tilted plasmonic nanocavity to shorten the luminescence decay time of a rare-earth-doped nanoparticle to sub-50 ns while maintaining high quantum efficiency enhancement, tunable polarization-dependent and far-field directional emissions and selective polychromatic chirality. We expect this new type of ultrafast, directional and polarized luminescence of a rare-earth-doped nanoparticle to vigorously promote the development of coherent single-photon sources, quantum communications and nanolasers.
Spaser Nanoparticles for Ultranarrow Bandwidth STED Super-Resolution Imaging

Abstract:Super-resolution microscopy, as a powerful tool of seeing abundant spatial details, typically can only distinguish a few distinct targets at a time due to the spectral crosstalk between fluorophores. Spaser (i.e., surface plasmon laser) nanoprobes, which confine lasing emission into nanoscale, offer an opportunity to eliminate such obstacle. Here, realized is narrow band stimulated emission depletion (STED) nanoscopy on spaser nanoparticles by collecting the coherent spasing signals. Demonstrated are the physics concept and feasibility of erasing spaser emission by using a depletion beam to suppress the population inversion, which lays the foundation of spaser-based STED super-resolution. Thanks to the small size (47 nm) and narrow spectral linewidth (3.8 nm) of the spaser nanoparticles, a 74 nm spatial resolution in STED imaging within an acquisition bandwidth of 10 nm is finally obtained. These spaser nanoparticles, if multiplexing with different wavelengths, in principle, allow for spectral-multiplexed imaging, sensing, cytometry, and light operation of a large number of targets all at once.

Abstract:Upconversion nanoparticles (UCNPs) exhibit unique optical properties such as photo-emission stability, large anti-Stokes shift, and long excited-state lifetimes, allowing significant advances in a broad range of applications from biomedical sensing to super-resolution microscopy. In recent years, progress on nanoparticle synthesis led to the development of many strategies for enhancing their upconversion luminescence, focused in particular on heavy doping of lanthanide ions and core-shell structures. In this article, we investigate the non-linear emission properties of fully Yb-based core-shell UCNPs and their impact on the super-resolution performance of stimulated excitation-depletion (STED) microscopy and super-linear excitation-emission (uSEE) microscopy. Controlling the power-dependent emission curve enables us to relax constraints on the doping concentrations and to reduce the excitation power required for accessing sub-diffraction regimes. We take advantage of this feature to implement multiplexed super-resolution imaging of a two-sample mixture.

Abstract:To date, real-time adjusting the emitting color with high brightness remains a formidable challenge. In this work, compact core-shell upconversion nanocrystals emitting real-time tunable colors were presented. On the basis of rational design to suppress the energy dissipation and color crosstalk, highly intense green and red lights are respectively generated at the 980 and 1530 nm excitations.
