Abstract:Enantioselective protonation, delivery of a proton to a carbanion intermediate, is the most straightforward and fundamental method for the preparation of a chiral tertiary carbon stereocenter. Recent efforts for this objective have been realized through enamine catalysis, which has now become a prominent catalytic strategy enabling a range of fascinating chiral transformations. This review will summarize recent advances in the field of enantioselective enamine protonation for the synthesis of optically active carbonyl compounds. Dynamic kinetic resolutions of alpha-substituted carbonyl compounds through enamine intermediates will be discussed as well.

Abstract:Enamine catalysis is a prevalent strategy for the functionalization of aldehydes/ketones with electrophiles. Recently, the advent and development of oxidative enamine catalysis have allowed for the coupling of enamines with readily available nucleophiles under oxidative conditions, significantly expanding the domain of typical enamine catalysis. In this perspective, we summarize the recent advances in asymmetric oxidative enamine catalysis. On the basis of the oxidative strategy, these could be classified as (1) oxidation of nucleophile, (2) oxidation of enamine via single-electron transfer (SOMO catalysis), and (3) oxidation of enamine to α,β-unsaturated iminium ion, i.e. oxidative iminium catalysis. These strategies have enabled efficient oxidative functionalizations of aldehydes/ketones with various O-, N-, and C-centered nucleophiles in a highly stereocontrolled manner.

Abstract:A chiral primary amine catalyzed oxidative β-C–H functionalization of ketone is described. The reaction proceeds via ketone enamine oxidation by IBX and enables highly enantioselective remote C–H functionalization of both cyclic and acyclic ketones, generating chiral ketones bearing β-stereocenters.

Abstract:Catalytic enantioselective protonation（EP） is a straightforward and effective approach to construct enantioenriched carbonyl compounds containing a tertiary stereocenter at its α-position, which has been widely applied in organic synthesis and medicinal chemistry. This field has significant advances in recent decades, and relevant reviews based on substrate category,catalytic system and reaction type have been reported successively. In this paper, new developments in this field are updated and summarized since 2019, which are classied into the directly catalytic asymmetric protonation of performed enolates and the enantioselective protonation of in situ enolates, α-carbanion, or α-radical intermediates, involving a variety of substrate categories, catalytic systems, activation modes, as well as catalytic strategies and means. It is expected that these studies can inspire chemists to continuously contribute to this field and further promote the development of carbonyl chemistry,asymmetric catalysis and other related fields, and provide facile and efficient access to high-value chiral molecules, including natural products and their intermediates, drugs, drug candidates and so on.

Abstract:Enantioselective proton transfer refers to the process of enantioselective introduction of protons to carbanions through the recognition of prochiral faces. Enantioselective proton transfer can easily construct tertiary carbon stereocenters, which widely exist in natural products and bioactive compounds. However, catalytic enantioselective proton transfer has been a long-lasting challenge in asymmetric synthesis because the proton is the smallest ion and the proton transfer process of active intermediates is generally fast with complicated pathways and strong background reaction. In recent years, a series of new catalysts and catalytic strategies have been developed for stereocontrol of proton transfer processes in various organic reactions. At the same time, more in-depth mechanism studies have revealed detailed information and regulations of proton transfer in catalytic reactions. This paper reviews the recent progress on catalytic enantioselective proton transfer reactions according reaction types. At the end of this paper, the stereocontrol strategies of catalytic proton transfer are classified and the prospects of this field are also presented.

High enantioselectivities are achieved from the combination of visible light photocatalysis with chiral organocatalysts. Considering the huge progress made in both asymmetric synthetic chemistry and visible light photocatalysis chemistry, more new chiral catalysts with varieties of transition-metals will be developed and engaged in the field of asymmetric photoreactions.

Abstract:The asymmetric ene reaction is an important transformation that provides synthetically valuable allylic derivatives efficiently and enantioselectively. Hence, significant attention has been paid to developing highly efficient chiral catalytic systems to address the issues of reactivity, stereoselectivity, substrate generality, cost and operational simplicity of the process. Abstract:Metal-catalyzed enantioselective allylation, which involves the substitution of allylic metal intermediates with a diverse range of different nucleophiles or S(N)2'-type allylic substitution, leads to the formation of C-H, -C, -O, -N, -S, and other bonds with very high levels of asymmetric induction. The reaction may tolerate a broad range of functional groups and has been applied successfully to the synthesis of many natural products and new chiral compounds.

Abstract:We describe herein an unprecedented asymmetric α-amination of β-ketocarbonyls under aerobic conditions. The process is enabled by a simple chiral primary amine through the coupling of a catalytic enamine ester intermediate and a nitrosocarbonyl (generated in situ) derived from N-hydroxycarbamate. The reaction features high chemoselectivity and excellent enantioselectivity for a broad range of substrates.

Abstract:We report an enantioselective photoredox dehalogenative protonation as a new type of asymmetric protonation. As a paradigm, with a cooperative catalytic system consisting of a chiral H-bonding catalyst and a dicyanopyrazine-derived chromophore (DPZ) photosensitizer that is irradiated with visible light, a range of cyclic and acyclic ketones with labile chiral secondary C-F, C-Cl and C-Br bonds at the α-position were obtained in high yields with good to excellent enantioselectivities (up to >99% ee) by using a secondary amine as the terminal reductant. Given the ready accessibility of halides, the success of this work should provide inspiration for constructing diverse chiral α-tertiary carbonyls and their variants.

Abstract:Asymmetric conjugate addition of bromonitromethane to alpha,beta-unsaturated ketones was studied using a series of chiral primary amines as the catalysts. The cascade intramolecular cyclopropanation provided nitrocyclopropanes efficiently. Excellent enantioselctivities and good yields were obtained for several cyclic enones catalyzed by a bifunctional thiourea-primary amine. Both acid and base additives were found to be necessary for the transformation.

Abstract:Asymmetric catalysis with benzyne remains elusive because of the highly fleeting and nonpolar nature of benzyne intermediates. Reported herein is an electrochemical approach for the oxidative generation of benzynes (cyclohexyne) and its successful merging with chiral primary aminocatalysis, formulating the first catalytic asymmetric enamine-benzyne (cyclohexyne) coupling reaction. Cobalt acetate was identified to stabilize the in situ generated arynes and facilitate its coupling with an enamine. This catalytic enamine-benzyne protocol provides a concise method for the construction of diverse α-aryl (α-cyclohexenyl) quaternary carbon stereogenic centers with good stereoselectivities.

Abstract:A chiral amine catalyst can generate polyconjugated trienamine intermediates with polyunsaturated carbonyl compounds. As such, the electron-donating property of the amine catalyst can be transmitted along the C=C bonds, raising the HOMO energy of the polyconjugated enamine systems. As a result, highly asymmetric Diels-Alder cycloaddition reactions with a variety of electron-deficient alkenes could occur at the remote beta, epsilon- or delta, epsilon-positions of trienamines, which can efficiently produce a wide range of chiral cyclic substances with highly structural diversity and complexity. This catalytic strategy not only successfully realizes the direct functionalizations of carbonyl compounds at the remote E-position, but also excellent diastereo- and enantioselectivity could be obtained even though the reactive E-site is seven bounds away from the chiral center of amine catalyst. In this respect, this catalytic protocol is of great usefulness and importance in organic synthesis because the related compounds are hard to be prepared using the conventional methods. In the review, we summarize the discovery and the development of asymmetric trienamine catalysis over the past few years, with emphasis on the asymmetric reactions of 2, 4-dienals, various types of dienones, and aromatic compounds specifically functionalized with a carbonyl group, via trienamine, cross-conjugated trienamine or formal trienamine catalysis. In addition, the future development of chiral amines is also discussed.

Abstract:The catalytic, enantioselective N-oxidation of substituted pyridines is described. The approach is predicated on a biomolecule-inspired catalytic cycle wherein high levels of asymmetric induction are provided by aspartic-acid-containing peptides as the aspartyl side chain shuttles between free acid and peracid forms. Desymmetrizations of bis(pyridine) substrates bearing a remote pro-stereogenic center substituted with a group capable of hydrogen bonding to the catalyst are demonstrated. Our approach presents a new entry into chiral pyridine frameworks in a heterocycle-rich molecular environment. Representative functionalizations of the enantioenriched pyridine N-oxides further document the utility of this approach. Demonstration of the asymmetric N-oxidation in two venerable drug-like scaffolds, Loratadine and Varenicline, show the likely generality of the method for highly variable and distinct chiral environments, while also revealing that the approach is applicable to both pyridines and 1,4-pyrazines.

Abstract:Optically active alpha-arylation carbonyl units are widely present in a wide variety of drugs, bioactive natural products, and valuable pharmacologically active molecules. Catalytic enantioselective alpha-arylation of carbonyl enolates or the related precursors with various arylating agents constitutes a powerful tactic for constructing such privileged scaffolds, which is of great interest and has gained considerable progress. This review summarizes the advances based on two major strategies with diverse arylating agents, discusses in detail the reaction mechanism, limitations, and advantages of each method and their applications, and expounds the synthetic opportunities still open for further exploration.

Abstract:Asymmetric catalytic hydrogenations of imines and enamines with chiral transition-metal complexes bearing chiral ligands are among the most green and powerful approaches for the elaboration of chiral amine structures in organic synthesis. This review focuses on recent applications of asymmetric hydrogenations of imine and enamine substrates in the total syntheses of natural products. These applications include diverse processes involving asymmetric transfer hydrogenation (ATH) and asymmetric hydrogenation (AH) to form key chiral amine motifs in natural products with good efficiency and high-level enantiocontrol.
