Abstract:Fifteen compounds were isolated from the rhizomes of Acorus tatarinowii by means of various chromatographic techniques such as silica gel, ODS, Sephadex LH-20 and preparative HPLC, and their structures were elucidated as tatanone A (1), calamusenone (2), acoronene (3), 2-acetyloxyacoronene (4), acorenone (5), alpha-asarone (6), beta-asarone (7), 1,2-dimethoxy-4-(1'Z-propenyl) benzene (8), methyleugenol (9), asarylaldehyde (10), acoramone (11), gamma-asarone (12), 5-hydroxymethyl-2-furaldehyde (13), galgravin (14) and eudesmin (15) on the basis of spectroscopic data analysis. Compound 1 was a new compound, and compounds 3-5 were separated from Acorus species for the first time.

Abstract:Acorus tatarinowii Schott (A. tatarinowii) is a natural medicinal plant. It plays an indispensable role in the treatment of diseases by the empirical medicine system and has achieved remarkable curative effects. A. tatarinowii is often used to treat various diseases, such as depression, epilepsy, fever, dizziness, heartache, stomachache, etc. More than 160 compounds of different structural types have been identified in A. tatarinowii, including phenylpropanoids, terpenoids, lignans, flavonoids, alkaloids, amides, and organic acids. These bioactive ingredients make A. tatarinowii remarkable for its pharmacological effects, including antidepressant, antiepileptic, anticonvulsant, antianxiety, neuroprotective, antifatigue, and antifungal effects, improving Alzheimer's disease, and so on. It is noteworthy that A. tatarinowii has been widely used in the treatment of brain diseases and nervous system diseases and has achieved satisfactory therapeutic effects. This review focused on the research publications of A. tatarinowii and aimed to summarize the advances in the botany, traditional uses, phytochemistry, and pharmacology, which will provide a reference for further studies and applications of A. tatarinowii.

Abstract:Introduction Acorus tatarinowii Schott is a traditional Chinese medicine that aromatizes dampness, opens the body and pacifies the mind, and is used medicinally as the dried rhizome of Acorus tatarinowii, family Tenaxaceae. Research has confirmed that it has a strong anti-inflammatory activity, however, its mechanism of anti-inflammatory is still unclear, and we continue to investigate its chemical composition and anti-inflammatory mechanism. Methods The separation and purification of the EtOH extract was carried out by preparative thin layer chromatography and column chromatography. The structures of the isolated compounds were identified by NMR spectroscopy and comparing their spectral data with those previously reported in literature. CCK-8 method was used to evaluate the anti-inflammatory activity of all compounds. The MDA, NO, SOD, IL-10, IL-1β, IL-6 and TNF-α levels were analyzed using ELISA kits. TNF-α and Beclin-1 expression were examined by western blot and immunofluorescence. Results Ten compounds, named cinnamic acid(1), sinapic acid(2), isoferulic acid(3), α-asarone(4), ferulic acid(5), arbutin (6), caffeic acid (7), 3-(3-hydroxyl phenyl)propanol(8), ethyl caffeate(9) and β-asarone(10) were isolated from the rhizome of Acorus tatarinowii Schott. Their structures were elucidated on the basis of extensive spectroscopic analyses, as well as by comparison with literature data. The anti-inflammatory effects of the all compounds were evaluated in lipopolysaccharide(LPS)-induced PC12 cell models in our study. The results of activity test showed that compounds 3 and 6 had good inhibitory effects on 5 μg·mL−1 LPS-induced PC12 cells. The IC50 of compounds 3 and 6 against PC12 cells were 27.41 ± 0.09 µmol·L−1 and 30.19 ± 0.10 µmol·L−1, respectively. Moreover, compounds 3 and 6 showed a significant protective effect on inflammation by increasing the IL-10 and SOD levels, and reducing the TNF-α, IL-6, IL-1β, MDA and NO levels. The most important point is that their anti-inflammatory effect is related to the autophagy factor Beclin-1. Conclusion These findings imply that compounds 3 and 6 exerts anti-inflammatory effects by regulating TNF-α/Beclin-1 pathway, which provide a new basis for the anti-inflammatory activity of Acorus tatarinowii Schott.
Compounds from Acorus Tatarinowii: Determination of Absolute Configuration by Quantum Computations and Camp Regulation Activity.

Abstract:A new cadinane-type sesquiterpenoid, tatarinowin A (1), two phenylpropanoids, tatarinoids A (2) and B (3), and a trinorlignan, tatarinoid C (4), along with 15 known compounds including two pairs of mixtures were isolated from the rhizome of Acorus tatarinowii. The absolute configurations of 1-4 were established by computation of specific rotation values. The isolated compounds were evaluated for their cAMP regulatory activity by the AlphaScreen assay.

Abstract:The genus Acorus, a perennial monocotyledonous-class herb and part of the Acoraceae family, is widely distributed in the temperate and subtropical zones of the Northern and Southern Hemispheres. Acorus is rich in biological activities and can be used to treat various diseases of the nervous system, cardiovascular system, and digestive system, including Alzheimer’s disease, depression, epilepsy, hyperlipidemia, and indigestion. Recently, it has been widely used to improve eutrophic water and control heavy-metal-polluted water. Thus far, only three species of Acorus have been reported in terms of chemical components and pharmacological activities. Previously published reviews have not further distinguished or comprehensively expounded the chemical components and pharmacological activities of Acorus plants. By carrying out a literature search, we collected documents closely related to Acorus published from 1956 to 2022. We then performed a comprehensive and systematic review of the genus Acorus from different perspectives, including botanical aspects, ethnic applications, phytochemistry aspects, and pharmacological aspects. Our aim was to provide a basis for further research and the development of new concepts.

Acori Tatarinowii Rhizoma: A comprehensive review of its chemical composition, pharmacology, pharmacokinetics and toxicity

Abstract:Acori Tatarinowii Rhizoma (ATR, Shi Chang Pu in Chinese), a natural product with multiple targets in various diseases. This review provides the comprehensive summary of the chemical composition, pharmacological effects, pharmacokinetics parameters and toxicity of ATR. The results indicated that ATR possesses a wide spectrum of chemical composition, including volatile oil, terpenoids, organic acids, flavonoids, amino acids, lignin, carbohydrates and so on. Accumulating evidence from various studies has shown that ATR exerts a wide range of pharmacological properties, including protecting nerve cells, alleviating learning and memory impairment, anti-ischemic, anti-myocardial ischemia, anti-arrhythmic, anti-tumor, anti-bacterial, and anti-oxidant activities. Currently, ATR is widely used in the central nervous system, cardiovascular system, gastrointestinal digestive system, respiratory system in China, and for the treatment of epilepsy, depression, amnesia, consciousness, anxiety, insomnia, aphasia, tinnitus, cancers, dementia, stroke, skin diseases, and other complex diseases. Pharmacokinetic studies indicated that β-asarone, α-asarone, cis-methylisoeugenol, and asarylaldehyde, the active components of ATR, were absorbed slowly after oral administration of ATR. Moreover, toxicity studies have suggested that ATR has no carcinogenic, teratogenic and mutagenic toxicity. Nevertheless, long term or high-dose toxicity testing in animals to explore the acute and chronic toxicity of acori Tatarinowii Rhizoma is still lacking. In view of good pharmacological activities, ATR is expected to be a potential drug candidate for the treatment of Alzheimer's disease, depression, or ulcerative colitis. However, further studies are needed to elucidate its chemical composition, pharmacological effects, molecular mechanisms and targets, improve its oral bioavailability, and clarify its potential toxicity.
Antioxidant Lignans and Neolignans from Acorus tatarinowii

Abstract:Eleven new lignans and neolignans, named acortatarinowins G–N ( 1 − 8 ), including three pairs of enantiomers ( 1a / 1b − 3a / 3b ) and five optically pure lignans and neolignans ( 4 − 8 ), along with five known analogs ( 9 − 14 ), were isolated from the rhizomes of Acorus tatarinowii Schott. Compounds 1 − 3 were successfully separated by chiral HPLC to afford 1a / 1b − 3a / 3b . The planar structures of 1 − 8 were elucidated by extensive spectroscopic analyses including HRESIMS and NMR. Their absolute configurations were determined by analyses of single-crystal X-ray diffraction and a modified Mosher’s method, assisted by experimental and calculated electronic circular dichroism (ECD) data. Compounds 1a and 1b were rare 7,8′-epoxy-8,7′-oxyneolignane. Compounds 1 − 8 were evaluated for their antioxidant activities using 2,2-diphenyl-1-picrylhydrazyl (DPPH) reducing antioxidant power assay. Compound 6, exhibiting strong DPPH radical scavenging capacity with IC 50 value of 16.4 ± 0.22 μg/mL, could interpret the herbal traditional usage.
Characterization and Authentication of Acori Tatarinowii Rhizoma and Its Adulterants by UPLC-Orbitrap-MS/MS Chromatographic Fingerprints, Elements Profiles and Chemometric Methods

Abstract:UPLC-Orbitrap-MS/MS and ICP-MS were applied to analyze and characterize the chemical composition of Acori Tatarinowii Rhizoma (ATR) and its adulterants including Acori Calami Rhizoma (ACR) and Anemones Altaicae Rhizoma (AAR). Fifty-five, fifty-five and thirty-six organic components were identified in ATR, ACR and AAR by UPLC-Orbitrap-MS/MS, respectively. Forty elements were detected by ICP-MS. AAR is different from ATR and ACR. 19 organic components existed in ATR and ACR were not detected in AAR. And, concentration of β-asarone in AAR was 1000 times lower than ATR and ACR. Chemical components in ATR and ACR are very similar. Partial least squares-discriminant analysis (PLS-DA) was used to distinguish between ATR and ACR. Variable important projection (VIP) was employed to improve the classification ability of PLS-DA. Twenty-one chemical components were identified, and were used to establish the characteristic quantitative fingerprints of ATR and ACR. Our results indicated that the combination of two analytical platforms and chemometrics will provide more information to characterize and distinguish the complex analytical objects, such as similar Chinese herbs.

Abstract:OBJECTIVE To study the chemical constituents contained in Acorus calamus. METHOD The chemical constituents were separated and purified by various chromatographic methods including silica gel, ODS, HPLC and Sephadex LH-20, and their structures were identified on the basis of analysis on spectroscopic data. RESULT Ten compounds were separated from A. calamus and identified as 1beta, 4beta, 7alpha-trihydroxyeudesmane (1), bullatantriol (2), teuclatriol (3), threo-1', 2'-dihydroxyasarone (4), erythro-1', 2'-dihydroxyasarone (5), (+)-de-4'-O-methyleudesmin (6), (+)-de-4'-0-methylmagnolin (7), (+)-eudesmin (8), (+)-magnolin (9) and beta-sitosterol (10), respectively. CONCLUSION Compounds 1-2,4-9 were separated from this plant for the first time. Specifically, compounds 1-2,6-9 were obtained from Acorus genus for the first time.

Abstract:Phytochemical investigation on the stems of Alstonia scholaris (L.) R. Br. led to the isolation and identification of 30 natural products, including sixteen alkaloids (1–16), nine triterpenoids (17–25), two phenols (26–27), and three lignans (28–30). Their structures were established by comprehensive spectroscopic analyses together with the comparison with reported data. Compound 1 was a new indole alkaloid. Compound 24 was firstly isolated from Apocynaceae family, compounds 22–23 and 26–30 were obtained from the genus Alstonia for the first time, and this is the first report of the presence of 11 and 16 in A. scholaris. The chemotaxonomic significance of these isolated compounds was also discussed.

Abstract:The chemical constituents of Atalantia buxifolia which was one of the plants in Atalantia Corr.,family Rutaceae mainly contains volatile oil,limonin,alkaloids,coumarins,flavonoids,and so on.A.buxifolia's application for medicinal purposes has a long history,modern research suggests that A.buxifolia has many pharmacological effects such as reducing phlegm,anti-cough,relieving asthma.It is used to treat acute and chronic tracheitis with desired clinical results in Guangdong,Hainan,and some other places.It has a great value and good prospect in development and utilization.In this paper,the general situation of the study on A.buxifolia in chemical constituents and pharmacology activity has been reviewed based on a lot of literature searches both at home and abroad.

Study on Constituents of Aerial Parts from Alpinia officinarum

Abstract:In order to study the chemical components from the aerial parts of Alpinia officinarum, column chromatographic techniques and high speed counter-current chromatography ( HSCCC) were used in the study to separate and analyze the components in the alcoholic extracts of the aerial parts of the plant. The result showed that galangin(1), 1,7-diphenylhepta-3,5-diol(2) and 5-hydroxyl-7-[4”-hydroxyl-3”-methoxy]-1-phenylhepta-3-one (3, a peppery substance) were obtained, and the peppery substance can be knocked out of rhizome water extracts by using HSCCC technique. Chemical compositions were very similar in both rhizome and the aerial parts by comparing their HSCCC chromatograms.

Abstract:<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Ethnopharmacology relevance</h3>As a traditional folk medicine, Acorus tatarinowii Schott was used to treat digestive diseases, such as diarrhea, which may be related to Candida albicans infection; however according to literature surveys, there have been few studies of A. tatarinowii focusing on its antimicrobial activity, and almost all describe investigations using crude extracts or fractions.<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Aim of the study</h3>The aims of the current study were to isolate and identify antifungal fractions of A. tatarinowii based on their antifungal activity, explore the preliminary mechanism of 60% ethanol elution (AT60) by metabonomics, and evaluate the antifungal activity of AT60 in vivo and in vitro, to provide natural resources against fungal infections.<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Materials and methods</h3>As a pilot evaluation of activity, A. tatarinowii fractions and compounds with antifungal bioactivity were isolated by bioactive-guided column chromatography, and identified by LC-QTOF-MS/MS and NMR spectroscopy. The antifungal effects of the active ingredients against resistant C. albicans were evaluated by in vivo and in vitro colony forming unit assays. The mechanism underlying the activity of AT60 against C. albicans was explored using an LC-QTOF-based metabonomics approach and fluorescence microscopy imaging.<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Results</h3>AT60 showed better activity against C. albicans than the same dose of the first line antifungal drugs, fluconazole and itraconazole (positive control drugs). Subsequent phytochemical investigation of AT60 identified twenty-five known compounds, six of which were isolated: asaraldehyde (7), 1-(2,4,5-trimethoxyphenyl)-1,2-propanediol (12), α-asarone (14), β-asarone (15), γ-asarone (18), acotatarone C (19). Further, the compounds α-asarone (14) and acotatarone C (19) may be responsible for the antifungal activity, and exhibit synergistic effects. Metabonomics analysis indicated that AT60 can inhibit biofilm formation by regulating the C. albicans protein kinase C pathway.<h3 class="u-h4 u-margin-m-top u-margin-xs-bottom">Conclusions</h3>Our results show that A. tatarinowii has potent bioactivity against C. albicans in vitro and in vivo, and can be considered an antifungal botanic agent.

[Chemical Constituents from Ethyl Acetate-Soluble Extraction of Valeriana Jatamansi].

Abstract:Application of a combination of various chromatographic techniques including column chromatography over silica gel, Sephadex LH-20, macroporous adsorbent resin, and reversed-phase HPLC, led to the isolation of 173 compounds including irdidoids, monoterpenes, sesquiterpenes, triterpenes, lignans, flavonoids, and simple aromatic derivatives from the ethyl acetate-soluble fraction of the whole plants of Valeriana jatamansi(Valerianaceae), and their structures were elucidated by spectroscopic methods including 1D, 2D NMR UV, IR, and MS techniques. Among them, 77 compounds were new. In previous reports, we have described the isolation, structure elucidation, and bioactivities of 68 new and 25 known compounds. As a consequence, we herein reported the isolation and structure elucidation of the remaining 9 new and 71 known compounds, the structure revision of valeriotriate A(8a), as well as cytotoxicity of some compounds.
Discrimination and characterization of the volatile organic compounds of Acori tatarinowii rhizoma based on headspace-gas chromatography-ion mobility spectrometry and headspace solid phase microextraction-gas chromatography-mass spectrometry

Abstract:Acori tatarinowii rhizome (ATR) is a Traditional Chinese Medicine (TCM), which has multiple effects, such as neuroprotective activity, antidepressant and other activity. However, the widespread cultivation of ATR has led to it varying quality. Therefore, it is important to find a method to quickly identify the components of ATR. Headspace-gas chromatography-ion mobility spectrometry (HS-GC-IMS) and headspace solid phase microextraction-gas chromatography-mass spectrometry (HS-SPME-GC–MS) were applied to analyze and characterize the volatile organic compounds (VOCs) of ATR. 33 VOCs were identified by HS-GC-IMS and 95 VOCs were identified by HS-SPME-GC–MS from 15 batches of ATR. Then, quantification of estragole, methyleugenol, γ-asarone, β-asarone and asarone by gas chromatography-mass spectrometry (GC–MS). The fingerprint of HS-GC-IMS and the heatmap of HS-SPME-GC–MS were established. Which compared differential components of ATR. In addition, principal component analysis (PCA) was performed on the results of both instruments. The VOCs in the ATR were significantly correlated with β-asarone and asarone by PatternHunter analysis. It assisted HS-GC-IMS determine ATR quality. It is the first report regarding the method development of HS-GC-IMS and HS-SPME-GC–MS that targets the VOCs characterization of ATR, and the findings obtained would benefit the quality control and distinguish the complex analytical objects of ATR.

Constituents from the Leaves of Aquilaria Sinensis

Abstract:OBJECTIVE:To study the chemical constituents of the leaves of Aquilaria sinensis.METHOD:The compounds were isolated and purified by the methods of solvent extraction and chromatographic technique, and their structures were identified on the basis of the analyses of spectral data.RESULT:Thirty-three compounds were obtained. Among them, twelve compounds were identified as 5-hydroxyl-7,4'-dimethoxyflavone (1), acacetin (2), luteolin (3), genkwanin (4), yuankanin (genkwanin-5-O-beta-D-primeveroside, 5), adenosine (6), genkwanin-5-O-beta-D-glucopyranoside (7), hypolaetin-7-O-beta-D-glucopyranoside (8), hypoxanthine (9), uracil (10), 8-C-beta-D-galactopyranosylisovitexin (11), and 4-(1,2,3-trihydroxypropyl) -2,6-dimethoxyphenyl-1-O-beta-D-glucopyranoside (12), respectively.CONCLUSION:All compounds except for 1, 3 and 4 were isolated from the leaves of A. sinensis for the first time.

Abstract:Urtica atrichocaulis, an endemic plant to China, is commonly used to treat rheumatoid arthritis even though its pharmaceutical activities and chemical constituents were not studied. Herein, we reported our investigations on the chemical compositions of the phenolic compounds-rich fraction from U. atrichocaulis (TFUA) and their antirheumatoid arthritis activities. We found that the TFUA significantly inhibited the adjuvant-induced rats arthritis, carrageenin-induced rats paw edema, cotton pellet-induced mice granuloma, and the acetic acid-induced mice writhing response. Our phytochemical investigations on the TFUA resulted in the first-time isolation and identification of 17 phenolic constituents and a bis (5-formylfurfuryl) ether. The extensive HPLC analysis also revealed the chemical compositions of TFUA. Our further biological evaluation of the main phenolic components, individually and collectively, indicated that the antirheumatoid arthritis activities of TFUA were the combined effect of multiple phenolic constituents.

Abstract:Objective To study the chemical constituents of Taraxacum mongolicum. Method Repeated column chromatography over silica gel, C-18 and Sephadex LH-20 were used to isolate chemical constitutes and their structures were elucidated by spectroscopic methods MS, 1H-NMR, 13C-NMR including 2D-NMR techniques and by direct comparing spectral data with those reported in literatures. Result Eight compounds were identified as artemetin (1), quercetin (2), luteolin (3), luteolin-7-O-beta-D-glucopyranoside (4), caffeic acid (5), esculetin (6), stigmasterol (7), taraxasteryl acetate (8), respectively. Conclusion Compound 1 was obtained from this genus and compound 6, 8 from this species for the first time. Furthermore, the 13 carbon data of compound 6 from the literature were corrected with 2D-NMR techniques in this paper.

Abstract:OBJECTIVE:To study the chemical constituents of the leaves of Aquilaria sinensis.METHOD:The compounds were isolated and purified by the methods of solvent extraction and chromatographic technique, and their structures were identified on the basis of the analyses of spectral data.RESULT:Eleven compounds were obtained and identified as beta-sitosterol (1), hexacosanic acid (2), cryptotanshinone (3), 2alpha-hydroxyursane (4), dihydrotanshinone I (5), tanshinone I (6), tanshinone II(A) (7), 2alpha-hydroxyursolic acid (8), p-hydroxybenzoic acid (9), hydroquinone (10), and daucosterol (11), respectively.CONCLUSION:All compounds except for compound 9 were isolated from the leaves of A. sinensis for the first time.

Abstract:Objective Tamarix ramosissima is the host plant of Cistanche tubulosa. The chemical cons-tituents of T. ramosissima were studied in order to improve the development of culture base of C. tubulosa and look for the base for exploitation of T. ramossisma. Methods Various chromatographic techniques were employed for the isolation and purification of the constituents including silica gel, Sephadex LH-20, and preparative HPLC. The structures of compounds were elucidated by chemical and spectral analyses (IR, UV, NMR, and MS). Results Twelve compounds have been isolated from the tender wattle of T. ramosissima, their structures were elucidated as aleuritolic acid (D-friedoolean-14-en-3-ol-28-oic acid, Ⅰ), aleuritolonic acid (D-friedoolean-14-en-3-one-28-oic acid, Ⅱ), rhamnocitrin (Ⅲ), ellagic acid-3, 3′-dimethyl ether (Ⅳ), rhamnetin (Ⅴ), kaemferol (Ⅵ), isoferulic acid (Ⅶ), aromadendrin (Ⅷ), (2α,3β)-dihydrorhamnetin (Ⅸ), quercetin (Ⅹ), 7, 4′-dimethoxykaemferol (Ⅺ), 3-methoxykaemferol (ⅩⅡ). Conclusion Among these compounds, Ⅰ, Ⅱ, Ⅳ, Ⅴ, Ⅷ, Ⅸ, and ⅩⅡ are isolated from the plants of Tamarix L. for the first time.
