Abstract:Crude,bioactive water-soluble polysaccharides were isolated from the leaves of Taxus mairei using hot water extraction followed by ethanol precipitation.Three polysaccharides named PTM-1,PTM-2 and PTM-3 were fractionated by DEAE-Sepharose FF anion ion-exchange chromatograph.The column was pre-equilibrated using HCl-Tris buffer with the pH value being 8.5,and was washed with de-ionized water followed by successive elution with sodium chloride solutions of increasing concentration(0.1,0.3,0.5,0.7 and 0.9 mol/L) in a stepwise manner.After further purified by size exclusion chromatograph followed by desalted and freeze-dried,PTM-3 was analyzed with gel permeation chromatograph(GPC),ultraviolet spectroscopy(UV),infrared spectroscopy(IR) and thin layer chromatograph(TLC).The results indicated that the PTM-3 is a homogeneous pure polysaccharide which is composed of Gal、GlcVA、Rha and Ara with β-(1,3)-linkages,and that the weight-average molecular weight(Mw) is 12.3 kD.
Purification, Characterization and Bioactivity of Different Molecular-Weight Fractions of Polysaccharide Extracted from Litchi Pulp

Abstract:Litchi polysaccharides are a kind of macromolecular polymers with various biological activities and a wide range of molecular weights. In this study, two separate fractions, with average molecular weights of 378.67 kDa (67.33%) and 16.96 kDa (6.95%), which were referred to as LP1 and LP2, respectively, were separated using an ultrafiltration membrane. Their physicochemical properties, and immunomodulatory and prebiotic activity were compared. The results revealed that LP2 contained more neutral sugar, arabinose, galactose and rhamnose, but less uronic acid, protein, mannose and glucose than LP1. Compared with LP1, LP2 possessed higher solubility and lower apparent viscosity. LP2 exhibited stronger stimulation on macrophage secretion of NO, TNF-α and IL-6, as well as better proliferation of Lactobacillus plantarum, Leuconostoc mesenteroides, Lactobacillus casei and Bifidobacterium adolescentis. These results suggest that an ultrafiltration membrane might be used to prepare a highly-active polysaccharide fraction from litchi pulp that may be used for food or drug development.

Abstract:In this paper, Lepista sordida polysaccharides (LSP) were separated from Lepista sordida ( L. sordida ) mainly using the Ultrasonic-Micro Wave Synergy Extraction (UMSE) method and purified by graded alcohol precipitation. Three polysaccharide components: 40%-LSP-UMSE, 60%-LSP-UMSE, and 80%-LSP-UMSE were obtained and further analyzed the physicochemical properties, structural characteristics, and antioxidant activity. And the effects on the proliferation of Lactobacillus casei of three polysaccharide components were studied. The characteristic absorption peaks and the β-glycosidic bond of three polysaccharide components were the direct expression at UV 200 nm using UV and FT-IR spectroscopy. The three polysaccharide components were mainly composed of glucose, mannose, galactose, and ribose using high-performance liquid chromatography (HPLC) analysis. The antioxidant activity study revealed that the polysaccharides obtained by the UMSE method had better antioxidant activity compared to the traditional "Hot Water Extraction (HWE)" method. In addition, the polysaccharide components promoted the proliferation of L. casei to some extent. 40%-LSP-UMSE, 80%-LSP-UMSE as the carbon source had better acid production than the control inulin. Three LSP-UMSE used as a carbon source compared with glucose for culturing L. casei could significantly improve its tolerance to bile salts. Results are helpful to develop the bioactive polysaccharides from Lepista sordida and beneficial to develop a unique health and functional product in the future.

Abstract:Congenital Trypanosoma cruzi infection is a highly pathogenic and underreported condition. Early recognition is essential for effective treatment. Umbilical chord blood from newborns (n = 302) to infected mothers was analyzed with microhematocrit, hemoculture, and PCR methods. Each subject was then followed serologically. In calibrated suspensions of T. cruzi in blood, the sensitivity of PCR was 27-fold higher than hemoculture. However, this advantage was not reflected during routine testing of samples from maternities, partly because of the uneven distribution of few parasites in small samples. Levels of detection of congenital infection were 2.9% (8/272) for microhematocrit, 6.3% (18/287) for hemoculture, 6.4% (15/235) for PCR, and 8.9% (27/302) for cumulated results. Evaluation against the standard of delayed serology indicates that the regular application of PCR, hemoculture, and microhematocrit to blood samples allows the rapid detection of about 90% of the congenitally infected newborns, in samples that can be obtained before the mother and child leave the maternity ward.

Abstract:Currently, most of the polysaccharide fractions are purified using the methods usually limited to the laboratory scale or easily raising question of residual reagent, which may obstruct the future application of polysaccharide. In our study, we employed sequential ultrafiltration with different membranes combined with HPGPC to purify polysaccharide fractions from the water extract of Lentinus edodes directly. The chemical composition, morphology and structure characteristic were analyzed by chemical methods and instrumental analysis including automatic amino acid analyzer, Elemental Analyzer, SEM, HPGPC, HPAEC-PAD, FT-IR and NMR. Results indicated that three polysaccharide fractions named as LE-UF-1, LE-UF-2 and LE-UF-3 were successfully purified using ultrafiltration membranes with MWCO of 2.5, 5 and 10 kDa in sequence. Mw and polysaccharide content of LE-UF-1 (~136 kDa, 86.11%) were the highest, followed by LE-UF-2 (14–61 kDa, 49.82%) and LE-UF-3 (14–35 kDa, 22.44%). Surprisingly, LE-UF-2 had 11.73% amino acid, and LE-UF-3 had 19.64% amino acid. The polysaccharide in all the three fractions was heteropolysaccharide that mainly consisted of Glc, Gal and Man. There might be more β-sugar residues in LE-UF-1 and LE-UF-2, while more α-sugar residues in LE-UF-3. Our study provides a greener and more efficient potential procedure for large-scale polysaccharide purification, and is able to prepare the polysaccharide fractions from Lentinus edodes that may be used for food or drug development.

Abstract:In this study, a second water-soluble polysaccharide (designated as LDP-2) was isolated from the bulbs of Lanzhou Lily (Lilium davidii var. unicolor). Based on monosaccharide composition and methylation analysis, its structural features were investigated using a variety of characterizations, such as size-exclusion chromatography combined with laser light scattering (SEC-LLS), gas chromatography-mass spectroscopy (GC-MS), infrared (IR) spectroscopy, and nuclear magnetic resonance (NMR) spectroscopy. The results demonstrate that LDP-2 is a homogeneous heteropolysaccharide (molecular weight 6.2 × 104 Da), which includes four kinds of monosaccharides (Lyx, Man, Glc, and Gal in an approximate weight ratio of 6.74: 6.28: 76.50: 10.48). Furthermore, its morphology, thermal behavior, and preliminary hypoglycemic activities were also investigated.

Abstract:Polysaccharides, extracted from the herbs of Chinese natural Solanum lyratum Thunb (SLT), which is a traditional Chinese medicine with hot water and 0.04 M sodium hydroxide successively, were fractionated and purified by ion-exchange and gel-filtration chromatography. According to methylation, periodate oxidation, NMR spectroscopy, partial and graded acid and enzymic hydrolysis analysis, the results indicated the D-glucan to be linear and to contain both (1-->3)- and (1-->4)-linkages. The anomeric NMR and IR spectra measurements confirmed that the sugar residues were beta-glycosidically linked.

Abstract:Intracellular polysaccharides (iPs) were separated and purified from Coriolus versicolor LH1 mycelia and characterized for their α-glucosidase inhibitory properties. Three iP fractions (iPL-F5-2-1, iPL-F5-4-1, and iPL-F5-5-1) were extracted, separated, and purified from LH1 mycelia using microwave extraction technology, a DEAE-Sepharose CL-6B column, a Diaion HP20 macroporous adsorption column, and a Sephadex™ G-50 gel-permeation column. The principal constituents of iPL-F5-2-1, iPL-F5-4-1, and iPL-F5-5-1 were saponins and polyphenoic compound mixtures. The enzyme inhibition activity, IC(50) values, of these three fractions were 1.7, 1.8, and 0.8 mg/mL, respectively. The α-glucosidase inhibitory properties were related to the presence of α-(1,4) glycosidic linkages in the polysaccharide structure and the total relative percentage of d-glucose and d-galactose in the structure of polysaccharides, other than triterpenoids.

Abstract:A water-soluble polysaccharide was extracted from Tremella fuciformis Berk. Crude Tremella fuciformis polysaccharide was gained by boiling in hot-water, precipitating using ethanol, removing proteins with papain-Sevage method. Then the crude polysaccharide was applied to DEAE-52 cellulose colume and further purified through Sephadex s-400 column, followed by dialysis, lyopilization. The resulting sample was named as TFP1. Total sugar and uronic acid contents of TFP1 were determined by phenol-sulfuric acid method and m-hydroxydiphenyl combined with sulfuric acid method using glucose and glucuronic acid as standard respectively. UV spectrum showed that no protein or nucleic acid presented in TFP1. The results of the PMP precolumn derivatization HPLC analysis and IR spectroscopy showed that TFP1 was an acidic heteropolysaccharide containing mannose, glucuronic acid, xylose and fucose with the molar ratio of 5.78∶0.7∶1.1∶1. I2-KI assay suggested that many branched chains attached to the backbone chain which was composed of mannose residues.
Isolation, Structural Analysis and Anti‐Inflammatory Activity of a Polysaccharide from Ilex cornuta Fruits

Abstract:In the present study, a polysaccharide from Ilex cornuta fruits (LCFP-3) was obtained by hot water extraction, Diethyaminoethyl cellulose-52 (DEAE-52) chromatography column and Sephadex G-100 gel column purification. Its structural characteristics were further explored using high performance anion exchange chromatography (HPAEC), gas chromatography and mass spectrometry (GC/MS), scanning electron microscopy (SEM), Fourier transform infrared (FT-IR) spectroscopy and nuclear magnetic resonance (NMR) spectroscopy. Monosaccharide composition analysis revealed LCFP-3 contained mainly Galactose (31.92 %), Arabinose (25.87 %) and Galacturonic acid (23.35 %) while small percentage of Rhamnose, Glucose, Mannose and Xylose. Chemical composition analysis showed that the total sugar content of LCFP-3 was 90.31 % and the protein content was 0.246 %. Gel permeation chromatography (GPC) analysis showed that its average molecular weight was 41.199 kDa. Structural analysis showed that LCFP-3 may be composed of residues, T-α-Arap, T-α-Rhap, 1,3-α-Arap, 1,4-α-Arap, T-β-Galp, 1,4-α-GalpA(OMe), 1,4-β-Glcp, 1,3-β-Galp, 1,3,6-β-Manp, 1,6-β-Galp, 1,3,4-β-GalpA, 1,4,6-β-Manp, 1,3,6-β-Glcp, 1,2,3,4-α-Xylp. The anti-inflammatory activity of LCFP-3 was evaluated using lipopolysaccharide (LPS)-induced RAW246.7 macrophages. The results showed that 1-200 μg/mL LCFP-3 could dose-dependently protect against LPS-induced toxicity and 1 μg/mL LCFP-3 could significantly inhibit LPS-induced NO production. Therefore, LCFP-3 exerted an anti-inflammatory activity and has great potential as a functional ingredient.

Abstract:Objective: To study the extraction, isolation and composition of Lycium barbarum polysaccharides (LBP). Method: LBP was extracted from L. barbarum with water, isolationed and purified by DEAE ion-exchange cellulose and gel chromatography, and their structural composition was studied by means of SDS-PAGE gel electrophoresis, GC, amino acid automatic analysis, etc. Result: Pure LBP has four water solubie polysaccharides, M W was 1.524 x 10(5). LBP was composed of 6 kinds of monosaccharides (Ara, Rha, Xyl, Man, Gal and Glc), galacturonic acid and 18 kinds of amino acids. Conclusion: LBP is a kind of complex polysaccharides consisting of acidic heteropolysaccharides and polypeptide or protein, and LBP has Glycan-O-Ser glycopeptide structures.

Abstract:The crude polysaccharide LDP was extracted from mycelia of Lactarius deliciosus Gray and then purified by DEAE-52 cellulose and Sephadex G-200 to obtain a novel polysaccharide named LDP-CP. LDP-CP was mainly composed of mannose, glucose and galactose with an average molecular weight of 2.33 × 103 kDa. The structure of LDP-CP was determined by FT-IR, methylation and NMR analysis, and the results showed that the sugar linkage units of LDP-CP were composed of (1 → 3)-linked β-D-Manp, (1 → 2,4)-linked α-D-Manp, (1→)-linked α-D-Manp, (1 → 4)-linked β-D-Glcp, (1 → 2)-linked β-D-Manp, (1 → 4,6)-linked α-D-Manp, (1 → 4)-linked α-D-Galp, (1 → 2,3)-linked α-D-Glcp and (1→)-linked α-D-Glcf. The protective effects of LDP and LDP-CP on PC12 cells against H2O2-induced oxidative injury were exhibited by enhancing cell viability and morphological protection. The improvement to the level of LDH, SOD and GSH further indicated that LDP and LDP-CP had ability to alleviate H2O2-induced oxidative damage on PC12 cells. The polysaccharides in Lactarius deliciosus Gray mycelia exhibited the great advantages in the management of oxidative toxicity, which indicated that the polysaccharides can be further developed in application of natural functional food source.

Abstract:An innovative acidic hydrolysate fingerprinting workflow was proposed for the characterization of Lyophyllum Decastes polysaccharide (LDP) by ultra performance liquid chromatography-mass spectrometry (UPLC-MS). The crude polysaccharides were firstly separated and purified by using DE-52 column and the BRT GPC purification system, respectively. The molecular weight and monosaccharide content of homogeneous polysaccharides were ascertained by utilizing HPGPC and ion chromatography separately. Secondly, the linkage of LDP was identified by methylation analysis and 1D/2D NMR spectra. The UPLC-MS/MS was used to scan and identify the acidic hydrolysate products of LDP using the PGC column. The oligosaccharides were collected by chromatography and identified by mass spectrometry. Thirdly, the expression of IL-1β, IL-6, iNOS, TNF-α and IFNAR-I was measured in order to assess the immunological activity of LDP. Besides, the targeted receptors identification of polysaccharides was performed by screening the expression of TLRs family protein. The results showed that oligosaccharide fragments with different molecular weights can be obtained by partial hydrolysis, which further verified that the structures of LDP polysaccharides was a 1-6-linked β-glucan. Moreover, the LDP polysaccharide can up-regulate the content of IL-1β, IL-6, iNOS, TNF-α and IFNAR-I and plays an important immunoregulation role through TLRs family.
Structural characteristics and intestinal flora metabolism mediated immunoregulatory effects of Lactarius deliciosus polysaccharide

Abstract:Lactarius deliciosus, a widely appreciated mushroom with delightful tastes and texture, has exhibited immunomodulatory activity in vitro, while the effects on intestinal flora metabolisms in vivo are ambiguous. In this study, a L. deliciosus polysaccharide (LDP) was extracted and purified, and the structural characteristics were evaluated, as well as the immunological enhancement on tumor-bearing mice through regulating intestinal flora metabolisms. Results showed that LDP was a heteropolysaccharide (average molecular weight of 1.44 × 107 Da) with a backbone of α-(1 → 6)-Manp and branches of α-(1 → 6)-Galp, α-(1 → 3)-Fucp, α-(1 → 6)-Glcp, α-(1 → 4)-Glcp. Animal experiments indicated that LDP could significantly protect immune organs of tumor-beraing mice and suppress solid tumors growth with inhibitory rate of 51.61 % (high-dose, 100 mg/kg), and improve the intestinal lactobacillus contents, promote adenine mediated zeatin biosynthesis, then competitively antagonize A2A receptor and enhance the activities of CD4+ T cells and CD8+ T cells, finally effectively facilitate the apoptosis and elimination of tumor cells. These results would provide powerful data supports for the further antitumor mechanisms development and practical applications of L. deliciosus polysaccharide in food and drug industries.

Abstract:Five polysaccharide fractions (LST1, LST2, LJT1, LJT2 and LJT3) from Lentinus edodes were isolated and purified. Their molecular weights (Mw) were estimated as 65.5~697kDa by using high-performance gel permeation chromatography (HPGPC). The results from monosaccharide analysis indicated that all of them contained glucose as main sugar component. The chemical and spectroscopic analyses implied that these fractions were composed of a main chain consisting of several (2~6) β-(1→3)-d-glucose residues with branching at C6 position. The predicted structural units of the purified polysaccharides were established with different degrees of branching (DB). Congo-red test, X-ray diffraction (XRD) and atomic force microscope (AFM) were used to verify that the natural lentinan contained triple helical spatial conformation. Furthermore, the results from anti-tumor tests in vitro and in vivo revealed that these polysaccharide fractions could have dual anti-tumor effects, including killing tumor cells directly and immunoregulation with low toxicity to the host. Interestingly, the high Mw polysaccharides had smaller DB values and displayed greater anti-tumor effects.

Abstract:The polysaccharides derived from Lentinula edodes exhibit highly desirable bioactivities with significant commercial potential. However, obtaining them has presented challenges due to the complex matrix of natural products. This study aims to address this issue by purifying polysaccharides from expired shiitake mushrooms using a fixed-bed column with activated carbon. Temperature and mobile phase were investigated. Results showed that 40% of the total sugars present in the initial sample can be recovered. Moreover, all assays showed antioxidant activity. In total, five polysaccharides were identified. The monosaccharide composition of all exopolysaccharides (EPS) was composed only of glucose and their respective molecular weights were 4.2 × 10 4 kDa; 2.5 × 10 4 kDa; 9.3 × 10 3 kDa; 2.6 × 10 3 kDa; and 1.1 × 10 3 kDa determined by high-performance liquid chromatography (HPLC) analysis. These results demonstrated the potential of shiitake for further exploration and optimization, indicating the possibility of even greater advancements in the extraction process and the commercial feasibility of these valuable compounds.

Abstract:The aim of the paper is to investigate the technics of isolating the immune-active polysaccharides from Herba Spirodelae seu Lemnae(PHS) by ultrafiltration.PHS were extracted and isolated through decoction,ultrafiltration,(removal) of protein and ethanol precipitation et al.The total contents of saccharides in the extraction and sample were(determined) by the methods of sulphuric acid phenol and sulphuric acid anthrone.The protein contents were determined by colorimetric method of coomassie brilliant blue G 250.Some hazel powder was gained.The purity,the contents of protein,and the recovery are 53.5%,1.2%,and 6.43%,respectively.Ultrafiltraion is a kind of method,which can isolate PHS simply,quickly and effectively.

Abstract:In this study, Phellinus linteus polysaccharides (PLPS) and proteins were simultaneously separated from P. linteus mycelia by using an aqueous two-phase system (ATPS) based on choline chloride ([Chol]Cl)/K2HPO4, and the physicochemical and antioxidant properties of PLPS after ATPS extraction were evaluated. Results demonstrated that the maximal extraction efficiencies of 68.53% ± 0.29% PLPS and 82.37% ± 0.41% proteins were obtained when the cholinium-based ATPS contained 68.9% K2HPO4, 20% [Chol]Cl, 10.0 mg mL−1 crude water extract (1.0 mL), and distilled water (4.0 mL) at shaking time and temperature of 30 min and 21.2 °C, respectively. Compared with C-PLPS obtained using traditional ethanol precipitation and isolation protocols, PLPS had higher carbohydrate content (63.58% ± 1.12%), lower molecular weight (15.2 kDa, 80%), different monosaccharide compositions, and showed similar preliminary structural characterizations. Moreover, PLPS exhibited more evident scavenging effects on free radicals and in vitro antioxidant activities than C-PLPS. Therefore, the method of [Chol]Cl/K2HPO4 ATPS can be developed as an effective strategy for the separation/purification of highly bioactive polysaccharides.
