Ergot alkaloid control in biotechnological processes and pharmaceuticals (a mini review)

Abstract:The control of ergot alkaloids in biotechnological processes is important in the context of obtaining new strain producers and studying the mechanisms of the biosynthesis, accumulation and secretion of alkaloids and the manufacturing of alkaloids. In pharmaceuticals, it is important to analyze the purity of raw materials, especially those capable of racemization, quality control of dosage forms and bulk drugs, stability during storage, etc. This review describes the methods used for qualitative and quantitative chemical analysis of ergot alkaloids in tablets and pharmaceutic forms, liquid cultural media and mycelia from submerged cultures of ergot and other organisms producing ergoalkaloid, sclerotias of industrial Claviceps spp. parasitic strains. We reviewed analytical approaches for the determination of ergopeptines (including their dihydro- and bromine derivatives) and semisynthetic ergot-derived medicines such as cabergoline, necergoline and pergolide, including precursors for their synthesis. Over the last few decades, strategies and approaches for the analysis of ergoalkaloids for medical use have changed, but the general principles and objectives have remained the same as before. These changes are related to the development of new genetically improved strains producing ergoalkaloids and the development of technologies for the online control of biotechnological processes and pharmaceutical manufacturing ("process analytical technologies," PAT). Overall, the industry is moving toward "smart manufacturing." The development of approaches to production cost estimation and product quality management, manufacturing management, increasing profitability and reducing the negative impact on personnel and the environment are integral components of sustainable development. Analytical approaches for the analysis of ergot alkaloids in pharmaceutical raw materials should have high enough specificity for the separation of dihydro derivatives, enantiomers and R-S epimers of alkaloids, but low values of the quantitative detection limit are less frequently needed. In terms of methodology, detection methods based on mass spectrometry have become more developed and widespread, but NMR analysis remains in demand because of its high accuracy and specificity. Both rapid methods and liquid chromatography remain in demand in routine practice, with rapid analysis evolving toward higher accuracy owing to improved analytical performance and new equipment. New composite electrochemical sensors (including disposable sensors) have demonstrated potential for real-time process control.

Abstract:This work is dedicated respectfully to Prof. H. Ila, JNCASR Bangalore, India. An efficient asymmetric approach to the ergot alkaloids has been achieved via a highly regioselective Heck cyclization. Asymmetric induction of the key intermediate was achieved via a catalytic enantioselective α-aminoxylation catalyzed by d -proline (98% ee). Utilizing the aforementioned strategy, formal total synthesis of the ergot alkaloids (+)-lysergine and (+)-isolysergine has been achieved. Importantly, an asymmetric approach to the unnatural analogues (–)-lysergine and (–)-isolysergine has also been achieved via catalytic enantioselective α-aminoxylation using l -proline. Abstract:Claviceps purpurea (Fries) Tulasne is a valuable source of many bioactive metabolites (ergot alkaloids) for the pharmaceutical industry. Selection and cultivation of pharmaceutical ergot Claviceps purpurea needs quick qualitative and quantitative analyses for alkaloid identification and chemotaxonomic strain passportisation; however, express methods can be uninformative for some conditions. We used an alternative method of reversed-phase HPLC-UV (based on formic acid elution) for the identification and determination of concentrations for ergot alkaloids in sclerotias from parasitic cultures of 3 strains of pharmaceutical Claviceps purpurea . Two different types of standard solutions of ergocriptine—sum of the α+β isomers and the ergotoxine mixture (ergocornine +α+β ergocriptine with a known ratio)—allowed us to clearly identify each peak in the analysis of standard solutions and sclerotia samples of parasitic ergot. Parameters of retention time and concentrations were determined for ergotamine (25.4 ± 0.11 min, 0.57 ± 0.04 g/100 g respectively), ergometrine (7.67 ± 0.01 min, 0.02 ± 0.004 g/100 g), ergocristine (27.83 ± 0.01 min, 0.62 ± 0.05 g/100 g), ergocornine (25.61 ± 0.11 min, 0.27 ± 0.04 g/100 g), and α- and β-isomers of ergocriptine (α-ergocriptine – 27.19 ± 0.01 min, 0.17 ± 0.01 g/100 g; β-ergocriptine – 27.42 ± 0.01 min, 0.061 ± 0.05 g/100 g). Ergotamine, ergometrine, ergocristine, ergocornine, α- and β-isomers of ergocriptine were identified and quantitatively determined in sclerotias from different strains of pharmaceutical ergot Claviceps purpurea .

Abstract:Ergot alkaloids (EAs) are a class of indole derivatives used as prescription drugs for treating neurological diseases. Owing to the limited production of EAs by Claviceps and the enantioselective difficulties encountered in chemical synthesis, the sustainable supply of EAs remains challenging. Recently, numerous attempts have been made to produce EAs in heterologous hosts. However, these efforts have only brought the production of precursor, lysergic acid (LA), with low efficiency. Here, we reported the de novo high-efficient biosynthesis of LA and a series of LA-derived EAs in Aspergillus oryzae cell factories. Based on the genome sequencing of the EAs-producing strain C. purpurea 22.07, an EAs biosynthetic gene cluster was annotated and characterized. After introducing and optimizing the agroclavine (AG) biosynthetic pathway into A. oryzae, we constructed an efficient chassis strain to produce AG. We then confirmed the function of the annotated CloA' to catalyze the successive oxidation of AG into LA and isolysergic acid (ILA) in this AG-producing chassis and realize their de novo production with titers of 52.68 ± 1.49 and 6.32 ± 2.08 mg/L, respectively. The subsequent introduction of the downstream non-ribosomal peptide synthetase genes LpsB' and LpsC' enabled the complete biosynthesis of ergometrine and a series of its analogs, achieving a total titer of more than 160 mg/L. The unexpected biosynthesis of isolysergyl-glycine and lysergyl-glycine revealed a novel function of LpsC', which utilizes glycine as a substrate. Our work successfully realizes the complete biosynthesis of a series of EAs in an industrially feasible fungus, which will open the avenue for manufacturing EAs in a green and sustainable way.

First Synthesis of Ergotamine-13CD3 and Ergotaminine-13CD3 from Unlabeled Ergotamine

Abstract:Ergot alkaloids (EAs) formed by Claviceps fungi are one of the most common food contaminants worldwide, affecting cereals such as rye, wheat, and barley. To accurately determine the level of contamination and to monitor EAs maximum levels set by the European Union, the six most common EAs (so-called priority EAs) and their corresponding epimers are quantified using high-performance liquid chromatography coupled with tandem mass spectrometry (HPLC-MS/MS). The quantification of EAs in complex food matrices without appropriate internal standards is challenging but currently carried out in the standard method EN 17425:2021 due to their commercial unavailability. To address the need for isotopically labeled EAs, we focus on two semi-synthetic approaches for the synthesis of these reference standards. Therefore, we investigate the feasibility of the N6-demethylation of native ergotamine to yield norergotamine, which can subsequently be remethylated with an isotopically labeled methylating reagent, such as iodomethane (13CD3-I), to yield isotopically labeled ergotamine and its C8-epimer ergotaminine. Testing the isotopically labeled ergotamine/-inine against native ergotamine/-inine with HPLC coupled to high-resolution HR-MS/MS proved the structure of ergotamine-13CD3 and ergotaminine-13CD3. Thus, for the first time, we can describe their synthesis from unlabeled, native ergotamine. Furthermore, this approach is promising as a universal way to synthesize other isotopically labeled EAs.

Abstract:Ergot alkaloids are toxins which are produced biotechnologically on an industrial scale. The chemical investigation of endophytic Aspergillus fumigatus resulted in the isolation of five new ergot alkaloids named fumigaclavines D-H (2-6), along with three known analogues, fumigaclavine C (1), festuclavine (7), and fumigaclavine A (8). Their structures were unequivocally elucidated by extensive spectroscopic analyses in association with X-ray single-crystal diffraction. Fumigaclavines D-H are interesting clavine-type ergot alkaloids featuring a reverse prenyl moiety at C-2, with 1-4, 6, and 8 bearing additional substituents, e.g., an OH or OAc group at C-9. Compounds 2, 4, and 6-8 showed a broad spectrum of antimicrobial activity against a panel of anaerobic microorganisms, of which compounds 4 and 6 were the most active against Veillonella parvula with an MIC=16 µg/mL compared to that (0.12 µg/mL) of tinidazole, co-assayed as a positive reference.

Abstract:Alkaloids are naturally occurring compounds with complex structures found in natural plants. To further improve the understanding of plant alkaloids, this review focuses on the classification, toxicity and mechanisms of action, providing insight into the occurrence of alkaloid-poisoning events and guiding the safe use of alkaloids in food, supplements and clinical applications. Based on their chemical structure, alkaloids can be divided into organic amines, diterpenoids, pyridines, isoquinolines, indoles, pyrrolidines, steroids, imidazoles and purines. The mechanisms of toxicity of alkaloids, including neurotoxicity, hepatoxicity, nephrotoxicity, cardiotoxicity and cytotoxicity, have also been reviewed. Some cases of alkaloid poisoning have been introduced when used as food or clinically, including accidental food poisoning, excessive consumption, and poisoning caused by the improper use of alkaloids in a clinical setting, and the importance of safety evaluation was illustrated. This review summarizes the toxicity and mechanism of action of alkaloids and provides evidence for the need for the safe use of alkaloids in food, supplements and clinical applications.

Abstract:EA have been a major benefit, and a major detriment, to humans since early in recorded history. Their medicinal properties have been used, and continue to be used, to aid in childbirth, with new uses being found in the treatment of neurological and cardiovascular disorders. The surprisingly broad range of pharmaceutical uses for EA stems from their affinities for multiple receptors for three distinct neurotransmitters (serotonin, dopamine, and adrenaline), from the great structural diversity of natural EA, and from the application of chemical techniques that further expand that structural diversity. The dangers posed by EA to humans and their livestock stem from the ubiquity of ergot fungi (Claviceps species) as parasites of cereals, and of related grass endophytes (Epichloë, Neotyphodium, and Balansia species) that may inhabit pasture grasses and produce toxic levels of EA. Further concerns stem from saprophytic EA producers in the genera Aspergillus and Penicillium, especially A. fumigatus, an opportunistic pathogen of humans. Numerous fungal species produce EA with a wide variety of structures and properties. These alkaloids are associated with plants in the families Poaceae, Cyperaceae, and Convolvulaceae, apparently because these plants can have symbiotic fungi that produce EA. Pharmacological activities of EA relate to their specific structures. Known as potent vasoconstrictors, the ergopeptines include a lysergic acid substituent with an amide linkage to a complex cyclol-lactam ring structure generated from three amino acids. Simpler lysergyl amides and clavines are more apt to have oxytonic or psychotropic activities. One of the lysergyl amides is LSD (5), the most potent hallucinogen known. The EA biosynthetic pathway in Claviceps species has been studied extensively for many decades, and recent studies have also employed epichloës and A. fumigatus. The early pathway, shared among these fungi, begins with the action of an aromatic prenyl transferase, DMATrp synthase, which links a dimethylallyl chain to L-tryptophan. When the dmaW gene encoding DMATrp synthase was cloned and sequenced, the predicted product bore no identifiable resemblance to other known prenyl transferases. The dma W genes of Claviceps species are present in clusters of genes, several of which also have demonstrated roles in EA biosynthesis. In many other fungi, dma W homologues are identifiable in otherwise very different gene clusters. The roles of DMA Trp synthase homologues in these other fungi are probably quite variable. One of them is thought to prenylate the phenolic oxygen of L-tyrosine, and another catalyzes the unusual reverse prenylation reaction in the biosynthesis of fumigaclavine C(10), an EA characteristic of A. fumigatus. The second step of the EA pathway is N-methylation of DMATrp (12) to form 13, which is then subjected to a series of oxidation/oxygenation and reduction reactions to generate, in order, chanoclavine-I (16), agroclavine (19), and elymoclavine (6). Shunt reactions generate a wide variety of other clavines. Two epimerizations occur in this pathway: one from 12 to 16, the other from 16 to 19. Further oxidation of 6, catalyzed by the cytochrome-P450 CloA, generates lysergic acid (1). An unusual NRPS complex, lysergyl peptide synthetase (LPS), is responsible for linking 1 to three hydrophobic L-amino acids to generate the ergopeptide lactams. The LPS complex includes two polypeptides, one (LPS 2) possessing a single module for activation of 1, and the other (LPS 1) possessing three modules, each specifying one of the L-amino acids. Variations in LPS 1 sequences are associated with variations in the incorporated amino acids, leading to differences between strain chemotypes, and even multiple ergopeptines within strains. For example, C. purpurea P1 produces two distinct ergopeptines (ergotamine (4) and ergocryptine (Table I)), each of which is believed to be generated by multiple LPS 1 subunits encoded by separate, but related, genes (lpsA1 and lpsA2). The main ecological roles of EA in nature are probably to protect the fungi from consumption by vertebrate and invertebrate animals. The EA produced by plant-symbiotic fungi (such as epichloë endophytes) may protect the fungus by protecting the health and productivity of the host, which may otherwise suffer excessive grazing by animals. The EA, at levels typical of plants bearing these symbionts, can negatively affect the health of large mammals as well herbivorous insects. Some clavines have substantial anti-bacterial properties, which might protect the fungus and, in some cases, their host plants from infection. However, the fact that a large number of epichloë, and even several Claviceps species, produce no detectable EA indicates that the selection for their production is not universal. An unfortunate fact for many livestock producers is that some of the most popular forage grasses tend to possess EA-producing epichloë endophytes. Such endophytes are easily eliminated, but confer such fitness enhancements to their hosts that their presence is often preferred, despite the toxic EA. The future looks promising for continued interest in EA. Research continues into their pharmacological properties, medicinal uses, and structure-function relationships. New clavines and lysergic acid derivatives are identified regularly from new sources, such as marine animals. Also, programs are well underway to modify or replace epichloë endophytes of forage grasses in order to produce new grass cultivars that lack these toxins.
Enzymatic and chemo-enzymatic approaches towards natural and non-natural alkaloids: indoles, isoquinolines, and others.

Abstract:The multi-step enzyme catalysed biosyntheses of monoterpenoid indole and isoquinoline alkaloids are described. Special emphasis is placed on those pathways leading to alkaloids of pharmacological and medicinal significance which have been fully elucidated at the enzyme level. The successful identification and cloning of cDNAs of single enzymes and their application provides great opportunities to develop novel strategies for both in vitro and in vivo alkaloid production in whole plants or tissue cultures, as well as in microbial systems such as Escherichia coli and yeast. Enzyme crystallisation, 3D analyses and site-directed mutation allowed rational engineering of enzyme substrate acceptance, which in turn can be used for reprogramming in vivo alkaloid biosynthesis and for the design of biomimetic alkaloid syntheses. These strategies broaden structural diversity and allow the creation of large libraries of unnatural alkaloid with expected optimised or novel biological activities. The chemo-enzymatic syntheses of the above-mentioned alkaloid groups and their precursors (in addition to selected examples of other alkaloid families) provides an overview of how enzyme reactions are integrated into the development of total chemical syntheses.

Abstract:Privileged ergot alkaloids (EAs) produced by the fungal genus Claviceps are used to treat a wide range of diseases. However, their use and research have been hampered by the challenging genetic engineering of Claviceps. Here we systematically refactored and rationally engineered the EA biosynthetic pathway in heterologous host Aspergillus nidulans by using a Fungal-Yeast-Shuttle-Vector protocol. The obtained strains allowed the production of diverse EAs and related intermediates, including prechanoclavine (PCC, 333.8 mg/L), chanoclavine (CC, 241.0 mg/L), agroclavine (AC, 78.7 mg/L), and festuclavine (FC, 99.2 mg/L), etc. This fungal platform also enabled the access to the methyl-oxidized EAs (MOEAs), including elymoclavine (EC), lysergic acid (LA), dihydroelysergol (DHLG), and dihydrolysergic acid (DHLA), by overexpressing a P450 enzyme CloA. Furthermore, by optimizing the P450 electron transfer (ET) pathway and using multi-copy of cloA, the titers of EC and DHLG have been improved by 17.3- and 9.4-fold, respectively. Beyond our demonstration of A. nidulans as a robust platform for EA overproduction, our study offers a proof of concept for engineering the eukaryotic P450s-contained biosynthetic pathways in a filamentous fungal host.

Abstract:Claviceps purpurea (Fries) Tulasne are used in parasitic and saprophytic cultures to obtain ergoalkaloids (EA), including ergocriptine. Ergocryptine has a variety of pharmacological effects, including reducing the secretion of prolactin and somatotropic hormone, suppresses physiological lactation. It also has an antiparkinsonian effect, and ergocriptine increases motor activity. Ergocriptine- based drugs are used in the treatment of hyperprolactinemia and adenomas that produce prolactin, infertility in men and women. The importance of C. purpurea for medicine requires the study of reproduction characteristics of the fungus cultivated in different areas and substrates. The results of studying features of the exogenous reproductive structures the ergocriptine line fungus Claviceps purpurea in saprophytic condition, as well as the composition of ergot alkaloids in sclerotia and in samples of C. purpurea mycelia, are presented. The exogenous reproductive structures play a decisive role in artificial infection in saprophytic and parasitic conditions. The aim of the work was to study the growth and reproductive in saprophytic of the line of the fungus parasite C. purpurea producing ergocriptine in saprophytic conditions under the conditions of saprophytic cultivation (by surface and submerged methods cultivation). It was found that the growth rate of conidia, including width and length, were faster under the conditions of saprophytic cultivation of C. purpurea on the surface of an agar growth medium. The features of the dynamics of conidioproduction in submerged and surface cultivation on an agar growth medium. It has been shown that the HPLC method, in comparison with TLC for the analysis of ergocriptine, is more effective, since it allows the determination of its insignificant content.

Abstract:A new approach for the divergent total synthesis of eight ergot alkaloids is reported. The approach allows the first total syntheses of pyroclavine, pibocin A, 9-deacetoxyfumigaclavine C, and fumigaclavine G and also enables the efficient synthesis of festuclavine, costaclavine, epi-costaclavine, and dihydrosetoclavine. Abstract:Background: Claviceps purpurea is a phytopathogenic fungus infecting a broad range of grasses including economically important cereal crop plants. The infection cycle ends with the formation of the typical purple-black pigmented sclerotia containing the toxic ergot alkaloids. Besides these ergot alkaloids little is known about the secondary metabolism of the fungus. Red anthraquinone derivatives and yellow xanthone dimers (ergochromes) have been isolated from sclerotia and described as ergot pigments, but the corresponding gene cluster has remained unknown. Fungal pigments gain increasing interest for example as environmentally friendly alternatives to existing dyes. Furthermore, several pigments show biological activities and may have some pharmaceutical value. Results: This study identified the gene cluster responsible for the synthesis of the ergot pigments. Overexpression of the cluster-specific transcription factor led to activation of the gene cluster and to the production of several known ergot pigments. Knock out of the cluster key enzyme, a nonreducing polyketide synthase, clearly showed that this cluster is responsible for the production of red anthraquinones as well as yellow ergochromes. Furthermore, a tentative biosynthetic pathway for the ergot pigments is proposed. By changing the culture conditions, pigment production was activated in axenic culture so that high concentration of phosphate and low concentration of sucrose induced pigment syntheses. Conclusions: This is the first functional analysis of a secondary metabolite gene cluster in the ergot fungus besides that for the classical ergot alkaloids. We demonstrated that this gene cluster is responsible for the typical purple-black color of the ergot sclerotia and showed that the red and yellow ergot pigments are products of the same biosynthetic pathway. Activation of the gene cluster in axenic culture opened up new possibilities for biotechnological applications like the dye production or the development of new pharmaceuticals.

Abstract:Intestinal commensal fungi are vital to human health, and their secondary metabolites play a key role in the reciprocal relationship. In the present study, the first example of 2,3-seco ergot alkaloids belonging to clavine-type were isolated from the fermentation of human intestinal fungus Aspergillus fumigatus CY018, including two pairs of diastereoisomers, secofumigaclavines A and B (3 and 4) and secofumigaclavines C and D (5 and 6), one analogue features a highly unsaturated skeleton, secofumigaclavine E (7), along with two known ones, fumigaclavines C (1) and D (2). Their structures were identified based on extensive spectroscopic data in a combination of quantum chemical calculations. Moreover, a single-step operation of semi-synthetic reaction based on riboflavin (RF)-dependent photocatalysis was performed to obtain the novel 2,3-seco ergot alkaloids 3 and 5 from their biosynthetic precursors 1 and 2. All the isolated compounds were evaluated for their anti-inflammatory activity. Among them, secofumigaclavine B (4) could bind to MD2 with a low micromole level of the equilibrium dissociation constant measured by surface plasmon resonance (SPR), and suppress TLR4-mediated NF-κB signaling pathway in RAW264.7 cells, resulting in its anti-inflammatory effect. Molecular dynamics revealed that amino acid residue Tyr131 played a key role in the interaction of secofumigaclavine B (4) with MD2. These findings suggested that secofumigaclavine B (4) could be considered as a potential candidate for the development of MD2 inhibitors.

Abstract:Psychoactive natural products play an integral role in the modern world. The tremendous structural complexity displayed by such molecules confers diverse biological activities of significant medicinal value and sociocultural impact. Accordingly, in the last two centuries, immense effort has been devoted towards establishing how plants, animals, and fungi synthesize complex natural products from simple metabolic precursors. The recent explosion of genomics data and molecular biology tools has enabled the identification of genes encoding proteins that catalyze individual biosynthetic steps. Once fully elucidated, the "biosynthetic pathways" are often comparable to organic syntheses in elegance and yield. Additionally, the discovery of biosynthetic enzymes provides powerful catalysts which may be repurposed for synthetic biology applications, or implemented with chemoenzymatic synthetic approaches. In this review, we discuss the progress that has been made toward biosynthetic pathway elucidation amongst four classes of psychoactive natural products: hallucinogens, stimulants, cannabinoids, and opioids. Compounds of diverse biosynthetic origin - terpene, amino acid, polyketide - are identified, and notable mechanisms of key scaffold transforming steps are highlighted. We also provide a description of subsequent applications of the biosynthetic machinery, with an emphasis placed on the synthetic biology and metabolic engineering strategies enabling heterologous production.

Abstract:The fungal family Clavicipitaceae includes plant symbionts and parasites that produce several psychoactive and bioprotective alkaloids. The family includes grass symbionts in the epichloae clade (Epichloë and Neotyphodium species), which are extraordinarily diverse both in their host interactions and in their alkaloid profiles. Epichloae produce alkaloids of four distinct classes, all of which deter insects, and some-including the infamous ergot alkaloids-have potent effects on mammals. The exceptional chemotypic diversity of the epichloae may relate to their broad range of host interactions, whereby some are pathogenic and contagious, others are mutualistic and vertically transmitted (seed-borne), and still others vary in pathogenic or mutualistic behavior. We profiled the alkaloids and sequenced the genomes of 10 epichloae, three ergot fungi (Claviceps species), a morning-glory symbiont (Periglandula ipomoeae), and a bamboo pathogen (Aciculosporium take), and compared the gene clusters for four classes of alkaloids. Results indicated a strong tendency for alkaloid loci to have conserved cores that specify the skeleton structures and peripheral genes that determine chemical variations that are known to affect their pharmacological specificities. Generally, gene locations in cluster peripheries positioned them near to transposon-derived, AT-rich repeat blocks, which were probably involved in gene losses, duplications, and neofunctionalizations. The alkaloid loci in the epichloae had unusual structures riddled with large, complex, and dynamic repeat blocks. This feature was not reflective of overall differences in repeat contents in the genomes, nor was it characteristic of most other specialized metabolism loci. The organization and dynamics of alkaloid loci and abundant repeat blocks in the epichloae suggested that these fungi are under selection for alkaloid diversification. We suggest that such selection is related to the variable life histories of the epichloae, their protective roles as symbionts, and their associations with the highly speciose and ecologically diverse cool-season grasses.
