Human-Derived Cytokine Functionalized Sericin/Agarose Composite Gel Material with Cell Proliferation-Promoting Activity Fabricated Using Genetically Engineered Silk for Medical Application

Abstract:With the development of silkworm genetic manipulation techniques, it is simple to create new functional silk material containing artificial functional target protein molecules, a promising material for the manufacture of functional medical biomaterials for damaged tissue repair and regeneration in the future. However, these biomaterials often have weak structural stability and mechanical properties. In this study, a human-derived cytokine functionalized sericin/agarose composite gel material was successfully fabricated using previously developed genetically engineered silk. This composite gel material not only exhibits superior mechanical properties and morphological stability, but it also efficiently and gradually releases active FGF-1 and FGF-2 protein for up to three weeks, thereby promoting significant NIH/3T3 cell proliferation by activating the extracellular signal-regulated kinase (ERK) pathway. Most importantly, this sericin/agarose composite gel causes no obvious cytotoxicity or severe inflammation responses in vitro and in vivo. These findings suggest that this sericin/agarose composite gel material has medical application potential as functional biomaterial for damaged tissue repair and regeneration, and that the combined use strategy of agarose and silk protein in biomaterials can improve the performance and promote the application of genetically engineered silk as biomaterial in the future.
