Abstract:Weaned piglets are vulnerable to nutritional, physiological, and psychological stressors, leading to abrupt taxonomic and functional shifts in the intestinal microbiome. In this study, an integrated approach combination of 16S rDNA gene sequencing and the mass spectrometry-based metabolomics techniques was used to investigate the effects of weaning stress on intestinal microbial composition and its metabolic profiles of piglets. Three litters of suckling piglets with same parity were chosen. The samples of colonic contents were collected from each selected piglets (weaned day, 3 days after weaned) for microbial and metabolomics analysis. The results showed that Lachnospiraceae, Negativicutes, Selenomonadales, Campylobacterales and other 15 species increased after weaning, while Porphyromonadaceace, Alloprevotella, Barnesiella and Oscillibacter decreased. Based on the function profiles prediction and metabolomic analysis, five key metabolic pathways including Phenylalanine metabolism, Citrate cycle (TCA cycle), Glycolysis or Gluconeogenesis, Propanoate metabolism, Nicotinate and nicotinamide metabolism might be the relevant pathways involved in weaning stress-induced gut microbiota dysbiosis. Taken together, these results indicated that weaning stress not only changed microbial composition and function but altered the microbial metabolic profiles in the intestine, which might provide a new insight in alleviating weaning stress and facilitating disease prevention during the period of weaning in piglets.
Intestinal Fatty Acid Binding Protein and Dipeptide Transporter 1 Mrna in the Small Intestinal Mucosa of Weaner Piglets: Developmental Expression and Influence of Glutamine.

Abstract:This study was conducted to investigate the developmental expression of intestinal fatty acid binding protein(I-FABP) and dipeptide transporter 1(PEPT1) mRNA in the small intestinal mucosa of weaner piglets and the influence of glutamine(Gln).Sixty nine crossed piglets(Duroc×Landrace×Large white) aged(21±3) d were used as the exprimental animals,3 piglets were slaughted at the day of weaning,and the rest 66 piglets were divided into two gruops with 3 replicates in each group and 11 piglets per relicate.Piglets in the control group and the experimental group were fed a basal diet and the basal diet+1% Gln,respectively.Three piglets were slaughtered at 3,5,7 and 14 d after weaning,respectively.Mucosal tissues were collected from the duodenum,jejunum and ileum.Real-time PCR was applied to determine the mRNA expressions of I-FABP and PEPT1.The results showed as follows: 1) no significant difference was observed in the mRNA expression levels of I-FABP and PEPT1 in the duodenum,jejunum and ileum(P0.05);2) the mRNA expression levels of I-FABP and PEPT1 in the duodenum,jejunum and ileum were decreased obviously due to the weaning,and the expression levels of piglets at the 3rd day postweaning reached the lowest and were significantly lower than those at the wearning day(P0.05),then the expression levels were increased and reached a peak at the 14th day postweaning;3) there was no significant difference in mRNA expression levels of I-FABP and PEPT1 between the control group and the experimental group(P0.05),however,the experimental group showed a trend that I-FABP mRNA expression level in the duodemum,jejunum and ileum as well as PEPT1 mRNA expression level in the duodemum recovered to normal levels early.The results indicate that the mRNA expression levels of I-FABP and PEPT1 in the duodenum,jejunum and ileum of weaner piglets change over time,and Gln plays a role in promoting the recovery of I-FABP and PEPT1 mRNA expression levels of weaner piglets.
Effects of Peptides Produced by Enzymatic Hydrolysis from Silkworm Protein on Serum Biochemical Indexes and Intestinal Mucosa in Weaned Piglets

Abstract:A total of 64 Duroc×Large Yorkshire×Landrace 21-day weaned piglets were allocated to two groups,each of which had 4 replicates,8 piglets per replicate.The control group and experimental group were fed with basal diet and basal diet + 0.3% peptides produced by enzymatic hydrolysis from silkworm protein,respectively.Results show as follows:(1) Average daily gain is higher significantly,whereas F/G and diarrhea ratio are lower dramatically in the experimental group relative to the control group.(2) The alkaline phosphatase activity and iron level in the serum of piglets are higher in the experimental group than those of control.(3) The diet supplemented with the peptides increases the villus height and decreases the crypt depth of duodenum,jejunum and ileum on day 14 postweaning.(4) The diet supplemented with the peptides increases significantly the activities of sucrase and maltase in jejunum mucosa during 1-14d of postweaning.
Effects of Soybean Antigen Protein on the Growth Performance of Weaning Piglets

Abstract:Forty-eight piglets(Duroc×Large white×Landrace) weaned at 21 days were used to study the effects of soybean antigen protein on the growth performance in a period of 15 days.The results showed that soybean antigen protein significantly decreased AFI by 25.88%(P0.01)and increased diarrhea rate by 58.77%(P0.01) compared with the control group.The serum D-xylose detection indicated that the serum D-xylose concentration was decreased by 62.39%(P0.01),68.54%(P0.01),67.27%(P0.01),67.98%(P0.01) and 69.95%(P0.01) after 1,2,3,4 h and 5 h respectively compared with the control group.The lengths of the villi within the jejunum and ileum tissues of the test group significantly decreased by 27.56% and 22.43%(P0.01) and the crypt depth significantly increased by 30.01% and 53.03%(P0.01) compared with the control group.There was no significant difference between duodenal villi length and crypt length.The activity of NF-κB in jejunum were increased 71.87%(P0.01).The results indicated that soybean antigen protein in diet exerted a negative influence on growth performance of weaning piglets.Soybean antigen protein could regulate the nuclear factor activity of intestinal mucosa tissue,and affect the morphology and structure of intestinal mucosa tissue,furthermore,affect feed intake,diarrhea rate and growth performance of weaning piglets.
Comparison between <i>Lactobacillus rhamnosus</i> GG and LuxS-deficient strain in regulating gut barrier function and inflammation in early-weaned piglets

Abstract:BackgroundEarly weaning-induced stress impairs the intestinal barrier function and adversely affects the health of piglet. Probiotics can be used to prevent and treat various intestinal diseases. Lactobacillus rhamnosus GG (LGG) has an LuxS/AI-2 quorum sensing (QS) system that senses environmental changes through chemical signaling molecules. The aim of the study was to explore whether luxS mutant affects the protective role of LGG in the gut barrier of weaned piglets by comparing the luxS mutant (Delta luxS) with its wild-type (WT). MethodsNewborn piglets were orally administered with WT and Delta luxS at dosage of 10(9) CFU, respectively. Accordingly, newborn piglets in the Con group were orally administered with PBS. Piglets were weaned on day 21 and euthanized on day 24, three days following weaning. ResultsSupplementation of Delta luxS in advance significantly boosted the relative abundances of healthy microbes (including Catenibacterium, Eubacterium, Lachnospiraceae and Bifidobacterium). WT and Delta luxS maintain intestinal barrier function mainly by promoting intestinal villus to crypt ratio (VCR), occludin protein expression and mucus secretion (P<0.05). Furthermore, LGG reduces pro-inflammatory mediators by inhibiting TLR4 and MAPK signal transduction (P<0.05). ConclusionBoth WT and Delta luxS were shown to resist weaning stress by enhancing the intestinal barrier function of piglets. It has to be said that the ability of Delta luxS to maintain intestinal tissue morphology and promote mucus secretion significantly decreased compared with that of WT.

Abstract:Two experiments were conducted to evaluate developmental gene expression of antimicrobial peptide PR-39 and effect of zinc oxide on gene regulation of PR-39 in piglets using semi-quantitative RT-PCR analysis. In experiment 1, fifteen female Tai-Hu pigs (a local breed in China) in five groups, each of three pigs at 1, 14, 28, 42 and 56 days of age were used to determine effect of age and weaning on mRNA expression of PR-39. In experiment 2, nine groups of pigs (total seventy-two female 36 days-age weaning Tai-Hu piglets) were assigned to three treatments (ZnO0, ZnO100 and ZnO3,000). The feeding experimental period lasted 15 days. After feeding experiment, nine pigs with three animals in each treatment were chosen to determine the effect of ZnO on PR-39 mRNA expression of pigs. The results showed that PR-39 mRNA levels increased steadily in postnatal day 1-28 (preweaning), and weaning significantly decreased PR-39 mRNA expression of piglets (p<0.05). ZnO3,000 (3,000 mg zinc/kg diet) significantly increased PR-39 mRNA expression (p<0.05) when piglets were feed ZnO3,000 diet for 15 days. ZnO100 (100 mg zinc/kg diet) also increased PR-39 gene expression, but the result was not statistically significant (p>0.05). The result was in accordance with the effect of ZnO3,000 and ZnO100 on weight gain of piglets and prevention of diarrhea.
Transcriptome profiling reveals transcriptional regulation of Protegrin-1 on immune defense and development in porcine granulosa cells

Abstract:Protegrin-1 (PG1) is an antimicrobial peptide (AMP) that has garnered increasing attention due to its potent immune defense activity. Our previous studies demonstrated the ability of PG1 to enhance proliferation and inhibit apoptosis of porcine granulosa cells (GCs) under oxidative stress. GCs play a crucial role in ovary follicular development. However, the specific function and underlying mechanisms of AMP in follicular development still need further elucidation. The present study aimed to comprehensively explore the biological effects of PG1 on porcine GCs using transcriptome profiling by RNA sequencing technology. Isolated GCs were incubated with or without PG1 for 24 h and transcriptome-wide analysis was exerted to identify differentially expressed genes (DEGs). The results of expression analysis revealed 1,235 DEGs, including 242 up-regulated genes and 993 down-regulated genes (|log2 (FoldChange)| > 1; adjusted P-value < 0.05). The expression levels of 7 selected DEGs were validated by quantitative reverse transcription-polymerase chain reaction (RT-qPCR) analysis, which was consistent with the RNA-sequencing data. Among the significant DEGs, several genes associated with GC function and ovarian follicle development were identified, such as estrogen receptor 2 (ESR2), growth and differentiation factor 6 (GDF6), cell division cycle 20 homolog (CDC20), Notch3, ephrin and Eph receptor system, Egl nine homolog 3 (EGLN3), and BCL2 like 14 (BCL2L14). Gene Ontology (GO) analysis revealed that the top three significant GO terms were inflammatory response, defense response, and granulocyte migration. Additionally, Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis presented that DEGs were mainly enriched in the immune system, infectious disease, signaling molecules and interaction, and immune disease. Furthermore, Ingenuity Pathway Analysis (IPA) predicted that the top activated pathway was Liver X Receptor (LXR)/ Retinoid X Receptor (RXR) Activation which is known to be associated with female reproduction. Predicted protein-protein interactions (PPIs) analysis identified complement C3 (C3) as the top node with the highest degree of network connection and revealed that DEGs in the sub-networks were involved in cytokine-cytokine receptor interaction, neuroactive ligand-receptor interaction, chemokine signaling pathway, and metabolic process. In conclusion, this study expanded the understanding of the effects of PG1 on porcine GCs at the transcriptomic level and provided a theoretical basis for further investigation into the role of PG1 in immune defense and mammalian ovarian follicular development.
Developmental Gene Expression of Lactoferrin in Duodenum and Effect of Weaning Age on Gene Expression of Lactoferrin in Piglets

Abstract:Two experiments were conducted to evaluate duodenal gene expression of lactoferrin and effect of weaning age on mRNA expression of lactoferrin in piglets using semi-quantitative RT-PCR analysis. In experiment 1, a total of 15 female Duroc x Landrace x Yorkshire piglets of five groups, each group pigs at 1, 14, 28, 42 and 56 days of age were used to determine developmental gene expression of lactoferrin in duodenum. In experiment 2, a total of 18 female neonatal piglets were divided into three groups, which were weaned at 21, 28 and 35 days of age respectively. In each group, three piglets' duodena were sampled at 21, 28 and 35 days of age and the other three piglets' duodena were sampled 7 days after weaning in each group. The samples were collected for detecting the effect of weaning age on lactoferrin mRNA expression of piglets. The results show that lactoferrin mRNA levels decreased steadily in postnatal day 1 - 56. But only from day 28 - 42 (14 days after weaned), the levels of lactoferrin gene expression were decreased markedly (p < 0.05), and the difference of lactoferrin mRNA levels at other stages was not significant. This result suggested that weaning had an effect on gene expression of lactoferrin. The results of experiment 2 showed that when the piglets were weaned at 21-28 and 35 days of age respectively, the expression levels of lactoferrin were decreased by 77%, 53% and 59% at the seventh day after weaning. Our results showed that weaning significantly decreased lactoferrin mRNA expression of piglets.
The Changes of Intestinal Morphology and Immune-Related Protein Gene Expressions in Piglets before and after Weaning

Abstract:This study was conducted to investigate the changes of intestinal morphology and immune-related protein gene expressions in piglets before and after weaning. A total of 24 healthy piglets from 6 litters with 1 piglet per litter were slaughtered at 7, 14, 21 and 35 days of age (weaned at 21 days of age), then duodenum and jejunum were collected. The intestinal morphology, intra-epithelial lymphocytes number and goblet cells number, jejunal secretory immunoglobulin A ( sIgA) concentration and defensin gene expressions were tested using haematoxylin and eosin staining, ELISA or real time quantitative PCR, respectively. The results showed as follows:during 7 d to 21 d after born, we found that the villous height of duodenum and jejunum was shor-ter in 14 and 21 d compared with 7 d ( P<0 . 05 ) , meanwhile the crypt depth in duodenum tended to be gradu-ally decreased following age, but the intestinal epithelial goblet cells number and intra-epithelial lymphocytes number were not significantly changed (P>0. 05). After weaning (35 d after born), in jejunum, the villous height was significantly decreased compared with 7 d ( P <0 . 05 ) and the crypt depth was significantly in-creased compared with that before weaning (during 7 d to 21 d after born) (P<0. 05). Compared with that before weaning, the intra-epithelial lymphocytes number of duodenum and jejunum was significantly increased after weaning ( P<0 . 05 ) . The sIgA concentration ( P=0 . 001 ) and defensin gene expressions in jejunum of weaned piglets were significantly higher than those of before-weaning piglets ( P <0 . 05 ) . From the results, we can find that piglets have a healthy intestinal barrier and low-expression levels of sIgA and defensins before weaning. After weaning, the weaning stress disturbances the intestinal mucosal integrity because of the atrophy of the villi and the proliferation of crypt, and intestinal barrier is destroyed, meanwhile, the number of intra-epithelial lymphocytes is increased. The infection of bacteria stimulates the expressions of immunoglobulin and defensins after weaning.
Effect of Lactoferrin on the Growth Performance, Intestinal Morphology, and Expression of PR-39 and Protegrin-1 Genes in Weaned Piglets1

Abstract:A total of 90 weaned female pigs (Duroc × Landrace × Yorkshire) were used in a 15-d growth experiment to investigate the effect of lactoferrin on growth performance, intestinal morphology, and expression of PR-39 and protegrin-1 genes. The pigs were allocated on the basis of BW and litter to 3 dietary treatments in a randomized complete block design. There were 3 replicate pens per treatment, and the pigs were grouped with 10 pigs per pen. The dietary treatments were (1) basal diet; (2) basal diet + 20 mg of flavomycin/kg + 110 mg of aureomycin/kg; (3) basal diet + 1.0 g of lactoferrin/kg. Six pigs, randomly selected from each treatment (2 piglets/pen) were slaughtered for intestinal morphology and expression of PR-39 and protegrin-1 genes at the end of the experiment. Supplementation with lactoferrin improved growth performance; it increased ADG by 41.80% (P < 0.01) and efficiency of gain (G:F) by 17.20% (P < 0.05). Intestinal villus height was increased by 15.30% (P < 0.05), and crypt depth was decreased by 9.60% (P < 0.05). Supplemental lactoferrin increased the relative abundance of mRNA for PR-39 and protegrin-1 by 143% (P < 0.01) and 217% (P < 0.01), respectively. The use of lactoferrin as an additive to improve nonspecific immunity and strengthen host defenses would be good a method of defending weaned pigs from infections and weanling stress.
Weaning Disrupts Intestinal Antioxidant Status, Impairs Intestinal Barrier and Mitochondrial Function, and Triggers Mitophagy in Piglets

Abstract:In the present study, we investigated the influence of weaning on antioxidant status, intestinal integrity, mitochondrial function, and the mitophagy level in piglets (weaned at 21 d) during the 1 wk after weaning. The redox status was measured by antioxidant enzymes activities, related genes expression, and malondialdehyde (MDA) content in jejunum. The intestinal barrier function was assessed by the Ussing chamber and expression of tight junction proteins in the jejunum. The function of intestine mitochondria was measured by mitochondrial DNA (mtDNA) content and activities of mitochondria oxidative phosphorylation complexes. The levels of light chain 3-1 (LC3-I), light chain 3-II (LC3-II), PTEN-induced putative kinase 1 (PINK1), and Parkin were determined to investigate whether mitophagy is involved in the weaning process. The results showed that, as compared with the preweaning phase (d 0), weaning suppressed (P < 0.05) the activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) on d 3 and d 7 postweaning, decreased (P < 0.05) the expression of copper and zinc superoxide dismutase (Cu/Zn-SOD), manganese-containing superoxide dismutase (Mn-SOD) on d 3 postweaning, declined (P < 0.05) the level of glutathione peroxidase 1 (GPX-1) and glutathione peroxidase 4 (GPX-4) on d 3 and d 7 postweaning, and increased (P < 0.05) MDA content in jejunum on d 3 and d 7 postweaning. The jejunal transepithelial electrical resistance and levels of occludin, claudin-1, and zonula occludens-1 on d 3 and d 7 postweaning were reduced (P < 0.05), and paracellular flux of fluorescein isothiocyanatedextran (4 kDa) on d 3 and d 7 postweaning was increased (P < 0.05). Weaning induced mitochondrial dysfunction, as demonstrated by decreased (P < 0.05) content of mtDNA on d 3 and d 7 postweaning and declined (P < 0.05) activities of mitochondria complexes (I, II, III, IV) in jejunum on d 1, d 3, and d 7 postweaning. Weaning led to an increased (P < 0.05) expression level of mitophagy-related proteins, PINK1 and Parkin, in the intestinal mitochondria, as well as an enhancement (P < 0.05) of the ratio of LC3-II to LC3-I content in the jejunal mucosa on d 1, d 3, and d 7 postweaning. These results suggest that weaning disrupted intestinal oxidative balance, and this imbalance may impair intestinal barrier and mitochondrial function and trigger mitophagy in piglets.

Abstract:The small intestine plays an essential role in the health and well-being of animals. Previous studies have shown that Lactobacillus has a protective effect on intestinal morphology, intestinal epithelium integrity and appropriate maturation of gut-associated tissues. Here, gene expression in jejunum tissue of weaned piglets was investigated by RNA-seq analysis after administration of sterile saline, Lactobacillus reuteri , or an antibiotic (chlortetracycline). In total, 401 and 293 genes were significantly regulated by chlortetracycline and L. reuteri , respectively, compared with control treatment. Notably, the HP , NOX1 and GPX2 genes were significantly up-regulated in the L. reuteri group compared with control, which is related to the antioxidant ability of this strain. In addition, the expression of genes related to arachidonic acid metabolism and linoleic acid metabolism enriched after treatment with L. reuteri . The fatty acid composition in the jejunum and colon was examined by GC-MS analysis and suggested that the MUFA C18:1n9c, and PUFAs C18:2n6c and C20:4n6 were increased in the L. reuteri group, verifying the GO enrichment and KEGG pathway analyses of the RNA-seq results. The results contribute to our understanding of the probiotic activity of this strain and its application in pig production.
Antibacterial, Cytotoxicity and Mechanism of the Antimicrobial Peptide KR-32 in Weaning Piglets

Abstract:Diarrhea is a serious issue among livestock, and pathogens and viral infections are the main causes of diarrhea, especially in weaned piglets. Additionally, overuse of antibiotics can lead to severe environment pollution. Therefore, our objective was to study a 32-residue engineered antimicrobial peptide, KR-32, that displays high antimicrobial activity with minimal hemolytic activity and cytotoxicity. Minimum inhibitory concentration and scanning electron microscopy results indicated that antimicrobial peptide KR-32 exerted its antimicrobial activity by destroying the cell membrane integrity. Piglets with clinical diarrhea were divided into three groups and treated with saline (control), KR-32 for 3 days. The antimicrobial peptide KR-32 alleviated the diarrhea of the weaned piglets by reducing the diarrheal rate and index, and it significantly decreased the IL-6 and TNF-α levels compared with the control group levels. To further study the inflammation of the piglets, we tested the effects of KR-32 on IPEC-J2 in vitro. We found that KR-32 attenuated lipopolysaccharides (LPS)-induced inflammation by activating the STAT-1 signaling pathway. In addition, KR-32 significantly reduced pro-inflammation factors IL-6, IL-8 and TNF-α compared with the LPS-treated group. LPS-induced inflammation also affected intestinal barrier functions, but KR-32 enhanced the intestinal barrier by increasing expression of TJ proteins (claudin-1 and occludin) in IPEC-J2. In conclusion, our research reveals AMP KR-32 may have potential application for the control of infectious disease in livestock, and furthermore, AMP KR-32 is a potent antibacterial agent.
Exposure to plant-oriented microbiome altered jejunal and colonic innate immune response and barrier function more strongly in suckling than in weaned piglets

Abstract:Weaning often leaves the piglet vulnerable to gut dysfunction. Little is known about the acute response of a gut mucosa primed by a milk-oriented microbiome before weaning to a plant-oriented microbiome (POM) after weaning. We evaluated the epithelial structure, secretory response and permeability in the small and large intestines of piglets receiving a milk-based (i.e., preweaning) or plant-based diet (i.e., postweaning) to POM inocula using intestinal loop perfusion assays (ILPA). The POM were prepared from jejunal and colonic digesta of four 7 week-old weaned (day 28 of life) piglets, having gut-site specific microbial and metabolite composition. Two consecutive ILPA were performed in 16 piglets pre- (days 24 to 27) and 16 piglets postweaning (days 38 to 41) in two replicate batches. Two jejunal and colonic loops per piglet were perfused with Krebs-Henseleit buffer (control) or the respective POM. The outflow fluid was analyzed for antimicrobial secretions. Jejunal and colonic loop tissue were collected after each ILPA for histomorphology and electrophysiology using Ussing chambers. ANOVA was performed using the MIXED procedure in SAS. The POM stimulated the secretory response by increasing mucin in the jejunal and colonic outflow by 99.7% and 54.1%, respectively, and jejunal IgA by 19.2%, whereas colonic lysozyme decreased 25.6% compared to the control (P < 0.05). Fittingly, the POM raised the number of goblet cells by 96.7% in jejunal and 56.9% in colonic loops compared to control loops (P < 0.05). The POM further flattened jejunal villi by 18.3% and reduced crypt depth in jejunal and colonic loops by 53.8% and 9.0% compared to the control (P < 0.05); observations typically made postweaning and indicative for mucosal recognition of 'foreign' compounds. The POM altered the jejunal and colonic net ion flux as indicated by 22.7% and 59.2% greater short-circuit current compared to control loops, respectively; the effect being stronger postweaning (P < 0.05). Colonic barrier function improved with age (P < 0.05), whereas POM perfusion compromised the mucosal barrier as suggested by 17.7% and 54.1% greater GT and mucosal-to-serosal flux of fluorescein-isothiocyanate dextran, respectively, compared to the control (P < 0.05). In conclusion, results demonstrated that the preweaning gut epithelium acutely responds to novel compounds in postweaning digesta by upregulating the first line of defense (i.e., mucin and lysozyme secretion) and impairment of the structural integrity.

Effects of Early Weaning on Intestinal Permeability and Tight Junction Protein Occludin Mrna Expression Levels of Piglets

Abstract:The aim of this paper is to study the changes of intestinal permeability and mechanisms in early weaning piglets.A total of 32 piglets weaned on the 21st day were sellected to measure plasma D-lactate level,DAO activity and urine lactulose mannitol ratio on the 21st,28th and 35th day to evaluate the intestinal permeability.Level of Occludin mRNA expression was researched at the same time.The results indicated that plasma D-lactate level,DAO activity and urine lactulose mannitol ratio increased significantly on the 28th day and markedly maintained high level to on the 35th day compared with the figures before weaning(P0.05);Occludin mRNA expression level decreased obviously after weaning(P0.05),and recovered to some extent on the 35th day.It is suggested that damage of intestinal barrier function and the increase of intestinal permeability may be due to the declined expression of tight junction protein Occludin mRNA.

Abstract:This experiment was conducted to study the effect of bovine lactoferrn(bLF)on antimicrobial peptide PMAP-37 gene expression.In vivo,seventy-two weaning piglets(Duroc× Landrace× TaiHu)of 36 days were divided into three treatment groups randomly.The control(treatment 1)received the basal dietary without bLF.Treatments 2 and 3 received dietary contained 0.125% and 0.25% bLF,respectively.In vitro,bone marrow cells were separated and cultured with bLF with 10,100,and 1 000 μg/mL respectively for 3 and 6 h.The control wells were incubated with culture medium.Using semi-quantitative RT-PCR,18S rRNA used as inner control,effect of different concentration bLF on antimicrobial peptide PMAP-37 gene expression in diet and in bone marrow cells of pigs was detected.The results indicated that,compared with control group,0.125% and 0.25% bLF in diet increased the gene expression of antimicrobial peptide PMAP-37 by 109.62%(P <0.01)and 69.23%(P <0.05),respectively.In bone marrow cells,10,100,and 1 000 μg/mL bLF also increased gene expression of PMAP-37 respectively for 3 and 6 h.These observations suggest that antimicrobial peptide PMAP-37 gene expression can be regulated by bLF.

Abstract:Seventy-two weanling pigs (♂Duro×♀Landrace×Taihu) with age of 36 days were arranged with four experimental treatments by randomly and each treatment were consisted of three replicates and of eight weanling pigs.there were two stages in this experiment,we took the sample at the age of 58 days and 80days respectively. The control group received the basal diet (DE 13.25 MJ/kg, CP 18.5%), the experimental treatment 2, 3 and 4 received the basal diet supplemented with 100 mg/kg and 3000 mg/kg Zn as ZnSO_4 and ZnO respectively. The primers for PR-39 and β-actin were designed respectively according to the reported porcine antibacterial peptide PR-39 gene and housekeeping gene β-actin sequences. Then the suitable MgCl_2 concentration, cycles, and the competition between the two pairs of primers in PCR system were verified. Thereafter an optimized semi-quantitative RT-PCR method was construct; The expressed house-keeping geneβ-actin used as internal control, to detect the effects of different zinc source on PR-39 gene expression in weanling pigs. The results showed that supplementation of 3 000 mg/kg Zn as ZnO marked improved the gene expression of porcine antibacterial peptide PR-39 by 378.26% (P0.01);As to the weanling pig at the age of 58 days, when compared with treatment 1, the percentage of PR-39 gene expression were increased by 38.4% in the experimental treatments supplementation of 100 mg/kg Zn as ZnSO_4, but the results of statistical analysis are not significant. As to the weanling pig at the age of 80 days, when compared with treatment 1,the experiment treatments supplementation of 3000 mg/kg Zn as ZnO improved the gene expression of porcine antibacterial peptide PR-39 by 4.52%; the experiment treatments supplementation of 100 mg/kg Zn as ZnSO_4 significant improved the gene expression of porcine antibacterial peptide pr-39 by 51.97%.

Abstract:This experiment was conducted to investigate the effects of immobilized antimicrobial peptides on growth performance, serum biochemical index, inflammatory factors, intestinal morphology, and microbial community of weaning piglets. A total of 21 weaning piglets [Duroc × (Landrace × Yorkshire)] with initial body weight (7.64 ± 0.65 kg) were randomly allocated to one of three treatments with seven replicates (one pig per replicate) per treatment according to sex and weight in randomized complete block design. Pigs in the three treatments were fed corn–soybean meal-based diet (CON), corn–soybean meal based diet + flavomycin (25 mg/kg) + quinone (50 mg/kg) (AB), and corn–soybean meal based diet + 1,000 mg/kg immobilized antimicrobial peptides (IAMPs), respectively. The experiment lasted for 28 days, including early stage (0–14 days) and late stage (15–28 days). The results showed the following: (1) compared with the CON group, the average daily gain in the whole experimental time ( p < 0.05) was significantly increased, and the diarrhea rate of weaning piglets was decreased ( p < 0.01) in the IAMPs group; (2) compared with the CON group, the concentrations of serum IgM and superoxide dismutase (SOD) in the IAMPs group were significantly higher than the CON and AB groups ( p < 0.01); (3) compared with CON group, the concentrations of serum interleukin (IL)-10 and transforming growth factor (TGF-β) were significantly increased ( p < 0.05), and the concentration of IL-12 was significantly decreased ( p < 0.05) in the IAMPs group; (4) compared with CON group, the concentrations of serum endotoxin and D-lactate of piglets were significantly reduced ( p < 0.05), and the relative expression of ZO-1 and occludin in the jejunum of piglets were significantly increased ( p < 0.05) in the IAMPs group; (5) compared with the CON group, the villus height of the duodenum and jejunum of weaning piglets in IAMPs and AB groups was significantly increased ( p < 0.05); and (6) compared with CON group, the relative abundance of Escherichia–Shigella in the colon and cecal digesta was decreased. In summary, the addition of 1,000 mg/kg immobilized antimicrobial peptides in the diet effectively relieved weaning stress by showing improved growth performance, antioxidant and immune capacity, intestinal morphology, and microorganisms.

Abstract:Modulation of the synthesis of endogenous host defense peptides (HDPs) by probiotics represents a novel antimicrobial approach for disease control and prevention, particularly against antibiotic-resistant infections in human and animals. However, the extent of HDP modulation by probiotics is species dependent and strain specific. In the present study, The porcine small intestinal epithelial cell line (IPEC-J2) cells and neonatal piglets were used as in-vitro and in-vivo models to test whether Lactobacillus reuteri I5007 could modulate intestinal HDP expression. Gene expressions of HDPs, toll-like receptors, and fatty acid receptors were determined, as well as colonic short chain fatty acid concentrations and microbiota. Exposure to 10⁸ colony forming units (CFU)/mL of L. reuteri I5007 for 6 h significantly increased the expression of porcine β-Defensin2 (PBD2), pBD3, pBD114, pBD129, and protegrins (PG) 1-5 in IPEC-J2 cells. Similarly, L. reuteri I5007 administration significantly increased the expression of jejunal pBD2 as well as colonic pBD2, pBD3, pBD114, and pBD129 in neonatal piglets (p < 0.05). This was probably associated with the increase in colonic butyric acid concentration and up-regulating expression of Peroxisome Proliferator-Activated Receptor-γ (PPAR-γ) and G Protein-Coupled Receptor 41 (GPR41) (p < 0.05), but not with stimulation of Pattern-Recognition Receptors. Additionally, supplementation with L. reuteri I5007 in the piglets did not affect the colonic microbiota structure. Our findings suggested that L. reuteri I5007 could modulate intestinal HDP expression and improve the gut health of neonatal piglets, probably through the increase in colonic butyric acid concentration and the up-regulation of the downstream molecules of butyric acid, PPAR-γ and GPR41, but not through modifying gut microbiota structure.
