Abstract:Voltage imaging has become an emerging technique to record membrane potential change in living cells. Yet, compared to the conventional electrophysiology, imaging approaches are still limited to relative membrane potential changes, losing important information conveyed by absolute value of membrane voltage. This challenge comes from several factors affecting the signal intensity, such as concentration, illumination intensity, and photobleaching. Spectroscopy is a quantitative method that shows potential to report the state of molecules in situ. Here, we apply electronic pre-resonance stimulated Raman scattering (SRS) imaging to detect near-infrared absorbing microbial rhodopsin voltage sensors in E. coli. The use of newly developed near-infrared microbial rhodopsins (Ganapathy et. al. 2017. JACS, 2017, 139(6):2338-44) enables electronic pre-resonance SRS imaging with single cell sensitivity. By spectral profile analysis, we identified voltage-sensitive SRS peaks. The spectral signature can be used as part of a quantitative approach to measure membrane potential and enable mapping of absolute voltage in a neural network.

Abstract:We developed membrane voltage nanosensors that are based on inorganic semiconductor nanoparticles. We provide here a feasibility study for their utilization. We use a rationally designed peptide to functionalize the nanosensors, imparting them with the ability to self-insert into a lipid membrane with a desired orientation. Once inserted, these nanosensors could sense membrane potential via the quantum confined Stark effect, with a single-particle sensitivity. With further improvements, these nanosensors could potentially be used for simultaneous recording of action potentials from multiple neurons in a large field of view over a long duration and for recording electrical signals on the nanoscale, such as across one synapse.

Abstract:Nanometer-sized glass and quartz pipettes have been widely used as a core of chemical sensors, patch clamps, and scanning probe microscope tips. Many of those applications require the control of the surface charge and chemical state of the inner pipette wall. Both objectives can be attained by coating the inner wall of a quartz pipette with a nanometer-thick layer of carbon. In this letter, we demonstrate the possibility of using open carbon nanopipettes (CNP) produced by chemical vapor deposition as resistive-pulse sensors, rectification sensors, and electrochemical nanoprobes. By applying a potential to the carbon layer, one can change the surface charge and electrical double-layer at the pipette wall, which, in turn, affect the ion current rectification and adsorption/desorption processes essential for resistive-pulse sensors. CNPs can also be used as versatile electrochemical probes such as asymmetric bipolar nanoelectrodes and dual electrodes based on simultaneous recording of the ion current through the pipette and the current produced by oxidation/reduction of molecules at the carbon nanoring.

Abstract:A self-consistent numerical model for silicon-based field effect nanowire biosensors is developed to study the impact of various surface-related physical and chemical processes, including transport of semiconductor carriers and electrolyte mobile ions, protonation and deprotonation of surface charge groups, and charges, and orientations and surface binding dynamics of immobilized biomolecules. It is shown that the sensing signal levels are affected by the gate biasing points, nonlinear screening from both electrolytes and surface charge groups, as well as the biomolecule charges and orientations. The critical role of the nanowire surface heterogeneity in determining the sensing input dynamic range is indicated based on correlations with experimental data.

Abstract:Although conventional patch-clamp methods provide high information content, they are labor-intensive and suffer from low throughput and high overall cost. Several approaches for achieving high throughput electrophysiology are under development, among which microchip-based patch-clamp systems uniquely achieve a higher degree of miniaturization, faster perfusion and mixing, and lower reagent cost without losing information content. The goal of this study was to establish a benchmark for our biochip technology with 52 chips tested sequentially. We demonstrate that our microfabrication and processing technology is sufficiently mature to produce a consistent hole size. We further demonstrate high-quality planar whole-cell patch clamping with >75% overall success rate at achieving gigaohm seals, followed by stable whole-cell access lasting at least 15 min with. access resistance (Ra) below 15 MOmega and membrane resistance (Rm) above 200 MOmega. These biochips are ideally suited for high throughput compound screening for ion channel targets.

Abstract:Along with more prevalent solid-state nanopores, glass or quartz nanopipettes have found applications in resistive-pulse and rectification sensing. Their advantages include the ease of fabrication, small physical size and needle-like geometry, rendering them useful for local measurements in small spaces and delivery of nanoparticles/biomolecules. Carbon nanopipettes fabricated by depositing a thin carbon layer on the inner wall of a quartz pipette provide additional means for detecting electroactive species and fine-tuning the current rectification properties. In this paper, we discuss the fundamentals of resistive-pulse sensing with nanopipettes and our recent studies of current rectification in carbon pipettes.

Abstract:This report studies the charge-based sensing modality of FET-embedded nanopore biosensors through FEM simulation. PNP equation is solved to analyze the mirror charge introduced by charged biomolecule while threading through the nanopore-FET sensor. Negative and positive charged molecules are analyzed respectively. Obvious local potential change induced by the presenting of charged molecules nearby is observed. In addition, the transport-induced descreening effect is observed under intensive bias, which might explain the capability of charge sensing even under high concentrations such as 1 M for FET-nanopore biosensors.

Abstract:We propose and fabricate solid-state nanopore devices that monolithically integrate solution-gated, vertical thin-film transistors (TFTs) inside the nanopores for charge-based sensing of translocating biomolecules. The TFTs consist of zinc oxide semiconductor channels and aluminum oxide gate dielectrics, which are both conformally deposited along the inner surfaces of the nanopores via atomic layer deposition. The resultant TFT channel lengths and nanopore diameters both reach the similar to 10 nm range. In translocation experiments using lambda-DNAs or bovine serum antibody (BSA) proteins, the TFT-nanopore devices demonstrate concurrent detection of the ion conductance blockade signals and modulation signals in the TFT electrical current. The TFT signals show opposite signs for the negatively charged DNAs and positively charged BSAs as well as staircase signal shapes that correspond to the folding and knotting of lambda-DNAs. Further experiments under various electrical biases and solution ionic strengths show that the ion blockade signals and the TFT signals have different dependence upon these experimental conditions. The TFT signals are analyzed to be consistent with the field effect sensing of the biomolecular charge, and the induced mirror charge is estimated from the signal amplitudes. This study could be a step forward to achieve charge-based single-biomolecular technology for basic research as well as for biosensing applications. It may also stimulate the development of TFT technologies for conformal integration of semiconductor electronics at the front end of nanostructures.

Abstract:Unraveling emergent brain activities requires simultaneous recording of action potentials from a large number of neurons. Electrical recording methods such as patch clamp and optical recording by voltage sensing dyes and proteins have been developed for years and are widely utilized. However, such techniques have insufficient spatial and/or temporal resolutions and/or suffer from poor photostability, posing a need for probes that circumvent these limitations. Improved probes, with high sensitivity and photostability, could afford the study of large neural networks (in a large filed-of-view) and/or at very high spatial resolution. Using bandgap-engineering and colloidal synthesis methods, we have synthesized seeded semiconductor (SC) nanorods (NRs) with Type-II heterojunctions that exhibit a large Quantum Confined Stark Effect (QCSE) at room temperature (1). For using these NRs as voltage sensors, however, one needs to impart them with membrane-protein like properties so that they can be stably inserted into the membrane. We report here spontaneous insertion of SC NRs into liposomes and cell membranes by functionalizing them with specially designed peptides. We provide evidences for insertion from cryo transmission electron microscopy (TEM) and polarized light microscopy. We also report on first attempts to sense membrane potential with these particles with single-particle sensitivity. With further improvements, SC NRs could potentially be used to study signals from whole neural networks in a large field-of-view. Abstract:Last decade has seen a growing trend toward smartphone-based biochemical sensing systems. Meanwhile, flexible electrochemical sensing devices like wristbands, patches, and tattoos have been widely developed for in situ detections of analyzes in accessible biofluids. For these devices, the connectivity with smartphone and the flexibility of the whole device are hard to achieve at the same time, due to the need for rigid batteries or wired connections. Here, a smartphone-based battery-free and flexible electrochemical patch is developed for real-time calcium and chloride ions detections in various biofluids. The patch is integrated with near field communication (NFC) module, on-site signal processing circuitry, and an all-printed stretchable electrode array which can maintain stable conductivity during stretching, without the need for serpentine designs. The device enables wireless power harvesting, on-site signal processing, and wireless data transmission capabilities. NFC-enabled smartphones can wirelessly power the patch and get the detection results through inductive coupling between antennas. The calcium and chloride sensors showed good sensitivity, repeatability, selectivity, and stability in quantitative detections of target ions. Ex situ measurements in serum, urine, tear, and sweat demonstrated good consistency with specialized instrument. Real-time on-body sweat analysis with the patch was also performed, which further indicated the usability and stability of the device in wearable applications. This platform provides a battery-free, wireless, and flexible solution for smartphone-based electrochemical sensing systems, which can be applied to rapid analysis of various biofluids.

Abstract:Engineered nanomaterials have been increasingly utilized in industry for various consumer products, environmental treatments, energy storage, and biomedical applications. Meanwhile, it has been established that certain nanomaterials can be toxic to biological cells from extensive experimental and theoretical studies. Despite that the exact molecular mechanisms of this nanomaterial toxicity are still not well understood, it is ubiquitous that their interactions with cell membranes, through either endocytosis or penetration (and thus potential lysis), act as the first step toward the inflammation or even the death of a cell. To facilitate the study of nanomaterial-membrane interactions, here we demonstrate a nanopore-based single-molecule approach that can be applied to monitor a specific nanomaterial-membrane interaction in real time. Combined with molecular dynamics and experimental approaches, we show how an ionic current can be used to detect membrane damage by a graphene nanosheet and illustrate the underlying molecular mechanism. More generally, we expect that measured transmembrane ionic currents (both DC and AC) can signify many particle-induced membrane modifications, such as hole formation, particle adsorption, and protein insertion.
Characterizing the Quantum Confined Stark Effect in Semiconductor Quantum Dots and Nanorods for Single-Molecule Electrophysiology

Abstract:We optimized the performance of quantum confined Stark effect QCSE based voltage nanosensors. A high throughput approach for single particle QCSE characterization was developed and utilized to screen a library of such nanosensors. Type II ZnSe CdS seeded nanorods were found to have the best performance among the different nanosensors evaluated in this work. The degree of correlation between intensity changes and spectral changes of the excitons emission under applied field was characterized. An upper limit for the temporal response of individual ZnSe CdS nanorods to voltage modulation was characterized by high throughput, high temporal resolution intensity measurements using a novel photon counting camera. The measured 3.5 us response time is limited by the voltage modulation electronics and represents about 30 times higher bandwidth than needed for recording an action potential in a neuron.

Abstract:We report on a silicon wafer-based device that can be used for recording macroscopic ion channel protein activities across a diverse group of cell-types. Gigaohm seals were achieved for CHO-K1 and RIN m5F cells, and both cell-attached and whole-cell mode configurations were also demonstrated. Two distinct intrinsic potassium ion channels were recorded in whole-cell mode for HIT-T15 and RAW 264.7 cells. Polydimethylsiloxane (PDMS) microfluidics were also coupled with the micromachined silicon chips in order to demonstrate that a single cell could be selectively directed to a micropore, and membrane protein currents could subsequently be recorded. These silicon chip-based devices have significant advantages over traditional micropipette approaches, and may serve as combinatorial tools for investigating membrane biophysics, pharmaceutical screening, and other bio-sensing tasks.

Abstract:The membrane potential of a cell is produced by the difference between intracellular and extracellular ion concentrations, which can be categorized into either resting potential or action potential. Voltage clamp and patch clamp are the main techniques to detect and record the membrane potential. And the electrode in the voltage clamp and glass tube in the patch clamp can stimulate the cell membrane and can't monitor or record the potential online. Cell-field effect transistor is a kind of sensor for membrane potential detection which controls the leakage source current of the device at the grid, so as to sense the membrane potential of cell. It is a new technology of membrane potential detection and record, and its advantage is that detecting without stimulation to the cell membrane, and monitoring at real time for a long time length. And it can be widely applied to the fields such as scientific research, drug development, medical device, inspection and quarantine, environmental protection, public security, and so on. This paper introduced the structure, transfer characteristic, equivalent circuit, preparation technology of the cell-FET sensor, and application in action potential recording, real-time monitoring of cell growth, monitoring of exocytosis, and detecting of tumor biomarker.

Abstract:Cell-based biosensors constitute a fundamental tool in biotechnology, and their relevance has greatly increased in recent years as a result of a surging demand for reduced animal testing and for high-throughput and cost-effective in vitro screening platforms dedicated to environmental and biomedical diagnostics, drug development and toxicology. In this context, electrochemical/electronic cell-based biosensors represent a promising class of devices that enable long-term and real-time monitoring of cell physiology in a non-invasive and label-free fashion, with a remarkable potential for process automation and parallelization. Common limitations of this class of devices at large include the need for substrate surface modification strategies to ensure cell adhesion and immobilization, limited compatibility with complementary optical cell-probing techniques, and need for frequency-dependent measurements, which rely on elaborated equivalent electrical circuit models for data analysis and interpretation. We hereby demonstrate the monitoring of cell adhesion and detachment through the time-dependent variations in the quasi-static characteristic current curves of a highly stable electrolyte-gated transistor, based on an optically transparent network of printable polymer-wrapped semiconducting carbon-nanotubes.

Abstract:During the dielectric breakdown process of thin solid-state nanopores, the application of high voltages may cause the formation of multi-nanopores on one chip, which number and sizes are important for their applications. Here, simulations were conducted to mimic the investigation of in situ nanopore detection with scanning ion conductance microscopy (SICM). Results show that SICM can provide accurate nanopore location and relative pore size. Detection resolution is influenced by the dimensions of the applied probe and separation between the probe and membranes, which can be enhanced under large voltages or a concentration gradient.

Abstract:A Schottky barrier can be formed at the interface between a metal electrode and a semiconductor. The current passing through the metal‐semiconductor contact is mainly controlled by the barrier height and barrier width. In conventional nanodevices, Schottky contacts are usually avoided in order to enhance the contribution made by the nanowires or nanotubes to the detected signal. We present a key idea of using the Schottky contact to achieve supersensitive and fast response nanowire‐based nanosensors. We have illustrated this idea on several platforms: UV sensors, biosensors, and gas sensors. The gigantic enhancement in sensitivity of up to 5 orders of magnitude shows that an effective usage of the Schottky contact can be very beneficial to the sensitivity of nanosensors.

Abstract:Nanopores utilize their confined space for achieving single-molecule detection. Reading the ionic fluctuation induced by a single molecule inside nanopores reveals characteristics of individual molecules such as structures, sizes, electrical properties and interactions. Most of the current commercialized instrumentations for nanopores contains only one recording channel, which acquire the ionic blockages only from a single nanopore in the single experiment. Therefore, the low throughput limits the application of nanopore detection in the real application. In this study, we designed and discussed the appropriate circuit configuration for multi-channel nanopore measurements. Two kinds of trans-impedance amplifiers were compared aiming to improve the throughput of the home-made single-channel electrochemical sensor instrument Cube-D2. Moreover, the bandwidth, noise and bias voltage supply of these circuits are explored and optimized by using circuit simulation softwares, which verifies the feasibility of our design in multi-channel nanopore instrumentations.
