Abstract:The leucine zipper lipopeptide is a dimer composed of two lipopeptides with alpha-helical structures that interact through hydrophobic forces. When heated to its phase transition temperature, the helixes unwind and form a disordered conformation. Based on the thermo-sensitivity of these leucine zipper lipopeptides, we designed and synthesized a series of zipper-structured lipopeptides and then mixed them with lipids to form thermo-sensitive hybrid liposomes. Circular dichroism was used to investigate the secondary structure of the lipopeptides anchored in the liposomal bilayer. Dynamic light scattering measurements were used to assess the diameters and zeta potentials of the liposomes. Fluorescence polarization was measured to study the fluidity of the liposomal bilayer. An ultraviolet spectrophotometer was used to stimulate Doxorubicin (DOX) release in vitro at 37.0 and 45.0 degrees C. We found that Lp-Lipo (hybrid of lipopeptide and liposome) displayed higher thermo-sensitivity than pure liposome. The cholesterol content and fluidity of the liposomal bilayer affected the thermo-controlled on-off switch of the lipopeptides. Lp-Lipo shows great potential as a novel thermo-sensitive drug carrier.
Dual-Sensitive On-Off Switch in Liposome Bilayer for Controllable Drug Release.

Abstract:To improve the controllability of drug release from liposome, a series of pH- and photosensitive block copolymers C7H15-AZO- b-PDPAn- b-mPEG were designed and served as an on-off switch in the liposome bilayer. The functional properties of liposomes were studied by dynamic light scattering, fluorophotometry, UV-vis spectroscopy, and fluorescence spectrophotometry. The liposomes with or without copolymer were relatively uniform in size. Their membrane stability was improved obviously after inserting copolymer under pH 7.4, but it decreased in an acidic environment and caused a large amount of drug release. Meanwhile, UV irradiation could also result in more drug release because of the photoisomerization of the azobenzene (AZO) group. Furthermore, intermittent drug-release experiments showed that the PDPA blocks could reversibly get in and out of the liposome bilayer and ultimately realized the complete drug release. All results suggested that the designed copolymers could be inserted into liposome bilayer through self-assembly and could act as a switch for controllable drug release.
Melting Behavior of Zipper-Structured Lipopeptides in Lipid Bilayer

Abstract:A zipper-structured lipopeptide is expected to play a role of "intelligent valve" in the lipid bilayer. In this paper, a series of zipper-structured lipopeptides have been designed for preparing thermocontrollable hybrid liposomes. Their conformational transition as a function of temperature in lipid bilayer has been investigated for understanding the influences of molecular structure and bilayer property on biofunction. The melting temperatures Tm of the lipopeptides have been found to depend on their molecular structures. When the lipopeptides have been doped in bilayer, an increase of size of alkyl chain increases the stability of the α-helix resulting in a decrease in fluidity of lipid bilayer. However, an increase of amino groups at N-terminal is found to decrease the stability of the spatial structure. The thermocontrollability of the "valve" in lipid bilayer is confirmed by drug release experiments under different temperatures. Meanwhile, effects of bilayer properties on the thermosensitivity of lipopeptides have also been investigated. Results show the Tm of lipopeptide doped in bilayer decreases with the increase of membrane fluidity. Furthermore, the reversibility of the thermocontrolled "valve" is also proven by release drug under intermittent temperatures. It could be concluded that the molecular structure of the lipopeptide, as well as the property of bilayer, give great influence on the biofunction of the hybrid liposomes.
Tuning Liposome Membrane Permeability by Competitive Coiled Coil Heterodimerization and Heterodimer Exchange

Abstract:Membrane-active peptides that enable the triggered release of liposomal cargo are of great interest for the development of liposome-based drug delivery systems but require peptide-lipid membrane interactions that are highly defined and tunable. To this end, we have explored the possibility to use the competing interactions between membrane partitioning and heterodimerization and the folding of a set of four different de novo designed coiled coil peptides. Covalent conjugation of the cationic peptides triggered rapid destabilization of membrane integrity and the release of encapsulated species. The release was inhibited when introducing complementary peptides as a result of heterodimerization and folding into coiled coils. The degree of inhibition was shown to be dictated by the coiled coil peptide heterodimer dissociation constants, and liposomal release could be reactivated by a heterodimer exchange to render the membrane bound peptide free and thus membrane-active. The possibility to tune the permeability of lipid membranes using highly specific peptide-folding-dependent interactions delineates a new possible approach for the further development of responsive liposome-based drug delivery systems.
Physical Stimuli-Responsive Liposomes and Polymersomes As Drug Delivery Vehicles Based on Phase Transitions in the Membrane

Abstract:This paper reviews liposomes with crystalline phase and polymersomes exhibiting crystalline and thermotropic liquid crystalline phases in the membrane. Intriguing morphologies of vesicles are described, including spherical, ellipsoidal and faceted vesicles, produced by a large variety of amphiphilic molecules and polymers with nematic phase, smectic phase or crystalline phase. It is highlighted how the phase transitions and the phase grain boundaries could be used ingeniously to destabilize the vesicular structure and to achieve cargo-release under the action of external stimulation. These liposomes and polymersomes are responsive to physical stimuli, such as temperature variation, shear stress, light illumination, and magnetic and electric fields. These stimuli-responsive properties make them promising candidates as new smart drug delivery systems.
Sequence and length optimization of membrane active coiled coils for triggered liposome release

Abstract:Defined and tunable peptide-lipid membrane interactions that trigger the release of liposome encapsulated drugs may offer a route to improving the efficiency and specificity of liposome-based drug delivery systems, but this require means to tailor the performance of the membrane active peptides. In this paper, the membrane activity of a de novo designed coiled coil peptide has been optimized with respect to sequence and size to improve release efficiency of liposome encapsulated cargo. The peptides were only membrane active when covalently conjugated to the liposomes. Two amino acid substitutions were made to enhance the amphipathic characteristics of the peptide, which increased the release by a factor of five at 1 μM. Moreover, the effect of peptide length was investigated by varying the number of heptad repeats from 2 to 5, yielding the peptides KVC2-KVC5. The shortest peptide (KVC2) showed the least interaction with the membrane and proved less efficient than the longer peptides in releasing the liposomal cargo. The peptide with three heptads (KVC3) caused liposome aggregation whereas KVC4 proved to effectively release the liposomal cargo without causing aggregation. The longest peptide (KVC5) demonstrated the most defined α-helical secondary structure and the highest liposome surface concentration but showed slower release kinetics than KVC4. The four heptad peptide KVC4 consequently displayed optimal properties for triggering the release and is an interesting candidate for further development of bioresponsive and tunable liposomal drug delivery systems.

Abstract:One switchable nanodelivery system was constructed. Liposomes were functionalized by a novel dual-recognition peptide STP, which is pH-responsive as well as the affinity ligand of tumor marker VEGFR2 (the angiogenesis marker vascular endothelial growth factor receptor 2). Efficient drug delivery and in-vivo therapy could be "turned on" and accelerated only in the conditions of VEGFR2 overexpression and a mild acidic environment. We envisioned that the successful demonstration of this switchable nanocarrier system would open a new avenue on rapid cytoplasmic delivery for specific cancer diagnostics and therapeutics.
Modulation of Phase Transition of Thermosensitive Liposomes with Leucine Zipper-Structured Lipopeptides

Abstract:Targeted therapy for cancer requires thermosensitive components in drug carriers for controlled drug release against viral cells. The conformational transition characteristic of leucine zipper-structured lipopeptides is utilized in our lab to modulate the phase transition temperature of liposomes, thus achieving temperature-responsive control. In this study, we computationally examined the conformational transition behaviors of leucine zipper-structured lipopeptides that were modified at the N-terminus by distinct functional groups. The conformational transition temperatures of these lipopeptides were determined by structural analysis of the implicit-solvent replica exchange molecular dynamics simulation trajectories using the dihedral angle principal component analysis and the dictionary of protein secondary structure method. Our calculations revealed that the computed transition temperatures of the lipopeptides are in good agreement with the experimental measurements. The effect of hydrogen bonds on the conformational stability of the lipopeptide dimers was examined in conventional explicit-solvent molecular dynamics simulations. A quantitative correlation of the degree of structural dissociation of the dimers and their binding strength is well described by an exponential fit of the binding free energies to the conformation transition temperatures of the lipopeptides.

Abstract:The control over secondary structure has been widely studied to regulate the properties of polypeptide materials, which is used to change their functions in situ for various biomedical applications. Herein, we designed and constructed enzyme-responsive polypeptides as gating materials for mesoporous silica nanoparticles (MSNs), which underwent a distorted structure-to-helix transition to promote the release of encapsulated drugs. The polypeptide conjugated on the MSN surface adopted a negatively charged, distorted, flexible conformation, covering the pores of MSN to prevent drug leakage. Upon triggering by alkaline phosphatase (ALP) overproduced by tumor cells, the polypeptide transformed into positively charged, α-helical, rigid conformation with potent membrane-penetrating capabilities, which protruded from the MSN surface to uncover the pores. Such a transition thus enabled cancer-selective drug release and cellular internalization to efficiently kill tumor cells. This study highlights the important role of chain flexibility in modulating the biological function of polypeptides and provides a new application paradigm for synthetic polypeptides with secondary-structure transition.

Abstract:Physically coating liposomes with peptides of desirable functions is an economic, versatile, and less time-consuming approach to prepare drug delivery vehicles. In this work, we designed three peptides-Ac-WWKKKGGNNN-NH2 (W2K3), Ac-WWRRRGGNNN-NH2(W2R3), Ac-WWGGGGGNNN-NH2(W2G3)-and studied their coating ability on negatively charged liposomes. It was found that the coating was mainly driven by the electrostatic interaction between the peptides' cationic side groups and the acidic lipids, which also mediated the "anchoring " of Trp residuals in the interfacial region of lipid bilayers. At the same conditions, the amount of the coated W2R3 was more than that of W2K3, but the stability of the liposome coated with W2R3 was deteriorated. This was caused by the delocalized charge of the guanidinium group of arginine. The coating of the peptide rendered the liposome pH-responsive behavior but did not prominently change the phase transition temperature. The liposome coated with peptides displayed appropriate pH/temperature dual responsive characteristics and was able to release the content in a controlled manner.

Abstract:Stimuli-triggered disassembly of block copolymer vesicles or polymersomes has been conventionally achieved via solubility switching of the bilayer-forming block, requiring cooperative changes of most of the repeating units. Herein we report an alternative approach by incorporating hydrophobic blocks exhibiting stimuli-triggered head-to-tail cascade depolymerization features. Amphiphilic block copolymers bearing this motif self-assemble into self-immolative polymersomes (SIPsomes). By modular design of terminal capping moieties, visible light, UV light, and reductive milieu can be utilized to actuate SIPsomes disintegration into water-soluble small molecules and hydrophilic blocks. The design of SIPsomes allows for triggered drug co-release and controllable access toward protons, oxygen, and enzymatic substrates. We also demonstrate programmed (OR-, AND-, and XOR-type logic) enzymatic reactions by integrating SIPsome encapsulation and trigger/capping moiety-selective cascade depolymerization events.
Regulating Vesicle Bilayer Permeability and Selectivity Via Stimuli-Triggered Polymersome-to-picsome Transition

Abstract:Compared to liposomes, polymersomes of block copolymers (BCPs) possess enhanced stability, along with compromised bilayer permeability. Though polyion complex vesicles (PICsomes) from oppositely charged block polyelectrolytes possess semipermeable bilayers, they are unstable towards physiologically relevant ionic strength and temperature; moreover, permselectivity tuning of PICsomes has remained a challenge. Starting from a single component diblock or triblock precursor, we solve this dilemma by stimuli-triggered chemical reactions within pre-organized BCP vesicles, actuating in situ polymersome-to-PICsome transition and achieving molecular size-selective cargo release at tunable rates. UV light and reductive milieu were utilized to trigger carboxyl decaging and generate ion pairs within hydrophobic polymersome bilayers containing tertiary amines. Contrary to conventional PICsomes, in situ generated ones are highly stable towards extreme pH range (pH 2-12), ionic strength (~3 M NaCl), and elevated temperature (70 °C) due to multivalent ion-pair interactions at high local concentration and cooperative hydrogen bonding interactions of pre-organized carbamate linkages.
Modulation of the Physicochemical State of Interior Agents to Prepare Controlled Release Liposomes

Abstract:To prepare controlled release liposomes, freeze-drying of double emulsions (FDE) was employed to entrap a model drug, topotecan, which has been reported to tend to leak rapidly from vesicles. In addition, hydrogenated soy phosphatidylcholine (HSPC), N-(carbonyl-methoxypolyethyleneglycol2000)-1,2-distearoyl-sn-glycero-3-phosphoethanolamine (PEG-PE), and cholesterol (3:1:1, mass ratio) were used as the vesicle lipid. Different inner aqueous phases (W1) containing topotecan together with a variety of chemicals, such as citrate, sulfate, and divalent copper ions, were used to prepare W1/O/W2 double emulsions, which were then freeze-dried to obtain dry products. Upon rehydration, the dry products formed topotecan-entrapping unilamelar liposomes with an encapsulation efficiency of 80% and a mean diameter of less than 200nm. The in vitro release experiments demonstrated that the drug release of topotecan-entrapping FDE liposomes could be successfully controlled through altering the state of the incorporated drug by means of protonation, precipitation, or forming a transition metal-complex. Specifically, the formulation of 300mM CuSO(4) had a drug release half-life of 36h. This novel method is a promising way to prepare controlled release liposomes that are more suitable for therapeutic application. In addition, the liposome formation mechanism was discussed based on micrographs and the size of the double emulsions and vesicles, as well as the small angle X-ray scattering pattern and transmission electron microscopy images.
Successively-responsive Drug-Carrier Vesicles Assembled by 'supramolecular Amphiphiles'.

Abstract:Novel vesicles assembled by 'supramolecular amphiphiles' based on the inclusion complexes between 1 and β-CDs (β-CD and HP-β-CD) were found to carry drugs and be successively-responsive to external stimuli for the first time. These vesicles were observed by TEM and SEM and confirmed by DLS. The formation patterns of 'supramolecular amphiphiles' characterized by UV and NMR can be controlled by choosing different hosts. Unlike traditional drug delivery and releasing systems, the 'useless' inclusion complexes 1·β-CDs in thinking formula can assemble into vesicles to carry both cytotoxic 1 and drugs (piroxicam and ampicillin) at one time, confirmed by TEM, UV, and NMR data. Also the vesicles assembled by 1·β-CDs can be successively-responsive to acidifying and oxidizing, and release drugs and cytotoxic compounds in order.
Liposome Crosslinked Polyacrylamide/DNA Hydrogel: a Smart Controlled-Release System for Small Molecular Payloads.

Abstract:A novel stimuli-responsive hydrogel system with liposomes serving as both noncovalent crosslinkers and functional small molecules carriers for controlled-release is developed. Liposomes can crosslink polyacrylamide copolymers functionalized with cholesterol-modified DNA motifs to yield a DNA hydrogel system, due to the hydrophobic interaction between cholesteryl groups and the lipid bilayer of liposomes. Functional information encoded DNA motifs on the polymer backbones endow the hydrogel with programmable smart responsive properties. In a model system, the hydrogel exhibits stimuli-responsive gel-to-sol transformation triggered by the opening of DNA motifs upon the presence of a restriction endonuclease enzyme, EcoR I, or temperature change, realizing the controlled-release of liposomes which are highly efficient carriers of active small molecules payloads. Two active molecules, 1,1-dioctadecyl-3,3,3,3-tetramethylindodicarbocyanine perchlorate (DiIC18(5)) and calcein, are chosen as the hydrophobic and hydrophilic model payloads, respectively, to address the feasibility of the releasing strategy. Moreover, the hydrogel exhibits injectable property as well as self-recovery behaviors.
Brain-targeted Drug Delivery by in Vivo Functionalized Liposome with Stable D-peptide Ligand.

Abstract:Brain-targeted drug delivery poses a great challenge due to the blood-brain barrier (BBB). In a previous study, we have developed a peptide-modified stealth liposome (SP-sLip) to enhance BBB penetration via the adsorption of apolipoproteins in plasma. SP is an 11-amino acid peptide derived from 25 to 35 of the Amyloid beta peptide (A beta(1-42)), which is a nature ligand of apolipoproteins. Although freshly prepared SP-sLip exhibited efficient brain targeting performance, it occured self-aggregation and instability in storage. In this study, we developed a D-peptide ligand according to the reverse sequence of SP with D-amino acids, known as (SP)-S-D, to improve the stability in storage. Notably, (SP)-S-D exhibited a reduced tendency for self-aggregation and improved stability in comparison to the SP peptide. Furthermore, compared to SP-sLip, (SP)-S-D-modified sLip ((SP)-S-D-sLip) demonstrated enhanced stability (>2 weeks), prolonged blood circulation (AUC increased 44.4%), reduced liver and spleen accumulation (reduced by 2.23 times and 1.86 times) with comparable brain-targeting efficiency. Similar to SP-sLip, (SP)-S-D-sLip selectively adsorbed apolipoprotein A1, E, and J in the blood to form functionalized protein corona, thus crossing BBB via apolipoprotein receptor-mediated transcytosis. These findings underscored the importance of ligand stability in the in vitro and in vivo performance of brain-targeted liposomes, therefore paving the way for the design and optimization of efficient and stable nanocarriers.

Abstract:Developing a targeted intelligent drug delivery system with excellent controllability of drug release remains an ongoing challenge in chemotherapy. A multistage responsive liposome was constructed by inserting a folate (FA) modified lipid and a pH/reduction dual-sensitive copolymer into the liposome bilayer. The properties of liposome were studied by fluorescence polarization, dynamic light scattering and in vitro drug release. Since the PDPA segment of the copolymer could embed in or escape from the liposomal bilayer depending on pH value, the membrane stability of the liposome was improved at pH 7.4 and decreased at pH 5.0. As a result, the drug leakage of the liposome was significantly suppressed at pH 7.4 but promoted at an acidic pH. Moreover, the liposome Lipo5-FA, which contained negatively charged FA groups protected by the longer PEG, was found to undergo membrane fusion with the positively charged Lipo(+) to verify the longer PEG was cut by GSH. All results indicated that the smart copolymer acted as a switch on liposomal bilayer for drug release and also a protective “umbrella” for FA.
Accurate Targeting and Controllable Release of Hybrid Liposome Containing a Stretchable Copolymer

Abstract:Developing an intelligent drug delivery/release system, which can transport drugs to the target tissues precisely and release drugs timely, is an important challenge in chemotherapy. A multistage sensitive drug delivery system is designed by inserting a folate (FA) modified lipid and a pH/temperature dual-sensitive amphiphilic copolymer into a liposome bilayer. The stretchable copolymer plays a role in protection on FA ligand for more accurate targeting. Then, the stretch ability of the copolymer in the liposome bilayer is verified by using the Langmuir-Blodgett film technique. The interaction between the 1,2-dipalmitoyl-sn-glycerol-3-phosphocholine (DPPC) monolayer and hybrid liposomes is found to increase, indicating the FA ligand is exposed due to the copolymer shrinking with increasing temperature. Fluorescence polarization measurements demonstrate that the insertion of the copolymer improves the stability of the liposome and offers pH-controllability for drug release. As a result, the drug leakage of the hybrid liposome is restrained significantly at pH 7.4, while at an acidic pH, the drug release is accelerated. The designed pH/temperature dual-sensitive copolymer is expected to provide more precise targeting and environmentally controlled drug release to drug delivery systems based on liposomes.
