Abstract:Exopolysaccharides (EPSs) are carbohydrate polymers that can be produced from probiotic bacteria. This study characterized the EPSs from Enterococcus faecium (EPS-LB13) and Streptococcus thermophilus (EPS-MLB10) and evaluated their biological and technological potential. The EPSs had high molecular weight and different monosaccharide compositions. The EPSs exhibited various biological activities at 250 mg/L, such as scavenging free radicals (10 % to 88.8 %), enhancing antioxidant capacity (714 to 2848 µg/mL), inhibiting pathogens (53 % to 74 %), and suppressing enzymes and cancer cells (2 % to 83 %), etc. The EPSs supported the growth of beneficial gut bacteria from Proteobacteria, Bacteroidetes, Firmicutes, and Acinetobacter in fecal fermentation with total Short-chain fatty acids production from 5548 to 6023 PPM. Moreover, the EPSs reduced the gelation time of fermented skimmed bovine milk by more than half. These results suggest that the EPSs from LB13 and MLB10 have promising applications in the dairy and pharmaceutical industries.

Abstract:Exopolysaccharides (EPSs)-producing lactic acid bacteria (LAB) and Bifidobacteria are widely used as starter cultures to make fermented food products due to their technological benefits. Moreover, LAB-EPSs and bifido-EPSs have been demonstrated to possess various physiological functions such as antioxidant, anticancer, antibacterial and immunological activities, among which the immunoregulatory activity has attracted a lot of scientific interests in recent years. Generally, these functions are closely related to the chemical compositions and configurations of EPSs. This review sheds light on structures and functions of LAB-EPSs and bifido-EPSs. Among them, in vitro and in vivo models used to evaluate the immune effects of EPSs are summarized in detail, with particular emphasis on the structure-immunity relationship of EPSs. Furthermore, the applications of EPSs in the food industry, especially the different roles EPSs played in the yogurt texture, are discussed carefully in this review. Finally, some strategies of EPSs production are also proposed to provide some information about the enhancement of EPSs yield.
Effects of bifidobacteria-produced exopolysaccharides on human gut microbiota in vitro

Abstract:Exopolysaccharides (EPSs) are carbohydrate polymers that are synthesized and present on the surface of bifidobacteria. Due to their potential applications in diverse sectors, such as food, biotechnology, cosmetics, and medicine, EPSs synthesized by bifidobacteria have recently attracted more attention. EPS production not only has benefits in food and health but also has effects on probiotics in the microbial ecosystem. In this study, we investigated the interaction between bifidobacteria EPSs and human gut microbiota in vitro using thin-layer chromatography, 16S rDNA high-throughput sequencing, and gas chromatography. The results showed that human gut microbiota has the capacity to degrade EPSs, although the degradation rate was approximately 50% after fermenting for 48 h. On the other hand, EPSs regulate the human gut microbiota. Fermented samples in the VI_Bif group clustered together according to the bacterial community compared to the VI_Starch group, in which starch was added as a carbon source. The bifidobacteria EPS promoted the growth of phylum Deinococcus_Thermus, class Deinococci, order Deinococcales, and genus Coprococcus. EPSs also increased the production of propionic acid compared to the starch group. The detection results of Dionex ICS 5000 high-purity capillary ion chromatography system showed that EPSs had absorption peaks of fucose, rhamnose, galactose/acetyl glucosamine, glucose, and ribose, and the molecular proportion of these monosaccharides was approximately 2: 2: 440: 3: 53. The monosaccharide composition of this EPS appears to be more complex than previously reported for bifidobacteria EPS. Additional studies are needed to elucidate its structure and functions.

Abstract:Probiotic bacteria are producers of secretory products such as bacteriocins and polysaccharides. Both homopolymeric and heteropolymeric exopolysaccharides (EPS) present on the surface of microorganisms have shown beneficial properties. While the fact that they play a role in bacterial homeostasis is well established, studies exploring their health promoting effects have also gained traction. Some exopolysaccharides function by inducing immune tolerance, others act by evading immune responses such as those by B and T cells. The interaction between the EPS and the immune system helps protect the bacteria against an attack by the host immune system. Several exopolysaccharides also show tolerogenic properties by reducing the amount of pro-inflammatory cytokines and increasing IL-10 production. They are also associated with anti-bacterial activity, anti-biofilm activity and anti-tumour properties. This review highlights the different types of exopolysaccharides and their health benefiting potentials. Better understanding of these mechanisms will pave the way for harnessing their potential to improve our health.

Abstract:Exopolysaccharides (EPS) are extracellular sugar metabolites/polymers of some slim microorganisms and, a wide variety of probiotics have been broadly investigated for their ability to produce EPS. EPS originated from probiotics have potential applications in food, pharmaceutical, cosmetology, wastewater treatment, and textiles industries, nevertheless slight is recognized about their function. The present review purposes to comprehensively discuss the structure, classification, biosynthesis, extraction, purification, sources, health-promoting properties, techno-functional benefits, application in the food industry, safety, toxicology, analysis, and characterization methods of EPS originated from probiotic microorganisms. Various studies have shown that probiotic EPS used as stabilizers, emulsifiers, gelling agents, viscosifiers, and prebiotics can alter the nutritional, texture, and rheological characteristics of food and beverages and play a major role in improving the quality of these products. Numerous studies have also proven the beneficial health effects of probiotic EPS, including antioxidant, antimicrobial, anti-inflammatory, immunomodulatory, anticancer, antidiabetic, antibiofilm, antiulcer, and antitoxin activities. Although the use of probiotic EPS has health effects and improves the organoleptic and textural properties of food and pharmaceutical products and there is a high tendency for their use in related industries, the production yield of these products is low and requires basic studies to support their products in large scale.

Abstract:This study was to investigate the potential prebiotic function of exopolysaccharide (EPS) from a medicinal fungus and the relationship to the molecular properties by in vitro human fecal fermentation. The EPS from Cordyceps sinensis Cs-HK1 mycelial fermentation was processed into three fractions with different monosaccharide contents, a higher molecular weight (MW) and a lower MW attained by two-step ethanol precipitation, and an intermediate MW by ultrasound-degradation of EPS. All the EPS fractions were well utilized during 24-48 h of fecal fermentation, leading to significant increases in the short chain fatty acid (SCFA) production. The consumption rate and production level of SCFAs varied slightly with the different EPS fractions. The EPS also influenced the composition and diversity of the fecal microflora, increasing the relative abundance of Firmicutes but suppressing that of Proteobacteria, which may be a beneficial effect for human health. Overall the results have shown that the Cs-HK1 EPS has significant prebiotic activity which is dependent on its molecular properties. (C) 2019 Elsevier B.V. All rights reserved.

Abstract:Exopolysaccharide (EPS) show remarkable properties in various food applications. In this review paper, EPS composition, structural characterization, biosynthesis pathways, and recent advancements in the context of application of EPS-producing Lactobacillus spp. in different food industries are discussed. Various chemical and physical properties of Lactobacillus EPS, such as the structural, rheological, and shelf-life enhancement of different food products, are mentioned. Moreover, EPSs play a characteristic role in starter culture techniques, yogurt production, immunomodulation, and potential prebiotics. It has been seen that the wastes of fermented and non-fermented products are used as biological food for EPS extraction. The main capabilities of probiotics are the use of EPS for technological properties such as texture and flavor enhancement, juiciness, and water holding capacities of specific food products. For these reasons, EPSs are used in functional and fermented food products to enhance the healthy activity of the human digestive system as well as for the benefit of the food industry to lower product damage and increase consumer demand. Additionally, some pseudocereals such as amaranth and quinoa that produce EPS also play an important role in improving the organoleptic properties of food-grade products. In conclusion, more attention should be given to sustainable extraction techniques of LAB EPS to enhance structural and functional use in the developmental process of food products to meet consumer preferences.

The Ecological Regulating Effect of Exopolysaccharides Produced by Bifidobacterium on Intestinal Microecological Imbalance

Abstract:The paper investigates the ecological effect of exopolysaccharides (EPS) produced by Bifidobacterium animalis RH on mice intestinal flora in terms of behavioral ecology. The Bifidobacterium produced two kinds of EPS, named as EPSa and EPSb. Five intestinal floras were detected to explore what role EPS plays in regulating intestinal floral and fighting against intestinal flora disturbance. After 14-days gavage, in comparison with control group, EPSa and EPSb remarkably increased the quantity of Bifidobacterium and Lactobacillus (P 0.05). That indicates that Bifidobacterium EPS is capable of regulate the intestinal flora of normal mice. On the 15th day, cyclophosphamide (CTX) is intraperitoneally injected to build mucous-injured mice models. EPSa and EPSb also significantly antagonized the decreasing numbers of Bifidobacterium and Lactobacillus (P<0.05) and increasing amounts of Clostridium perfringens, Enterococcus and Enterobacteriaceae (P<0.05) to resist the intestinal dysbacteriosis induced by large dose of CTX and maintain intestinal flora balance.

Abstract:Lactic acid bacteria (LAB) are capable of synthesising metabolites known as exopolysaccharides (EPS) during fermentation. Traditionally, EPS plays an important role in fermented dairy products through their gelling and thickening properties, but they can also be beneficial to human health. This bioactivity has gained attention in applications for functional foods, which leads them to have prebiotic, immunomodulatory, antioxidant, anti-tumour, cholesterol-lowering and anti-obesity activity. Understanding the parameters and conditions is crucial to optimising the EPS yields from LAB for applications in the food industry. This review provides an overview of the functional food market together with the biosynthesis of EPS. Factors influencing the production of EPS as well as methods for isolation, characterisation and quantification are reviewed. Finally, the health benefits associated with EPS are discussed.

Abstract:The production of exopolysaccharides (EPS) by lactic acid bacteria (LAB) has attracted particular interest in the food industry. EPS can be considered as natural biothickeners as they are produced in situ by LAB and improve the rheological properties of fermented foods. Moreover, much research has been conducted on the beneficial effects of EPS produced by LAB on modulating the gut microbiome and promoting health. The EPS, which varies widely in composition and structure, may have diverse health effects, such as glycemic control, calcium and magnesium absorption, cholesterol-lowering, anticarcinogenic, immunomodulatory, and antioxidant effects. In this article, the latest advances on structure, biosynthesis, and physicochemical properties of LAB-derived EPS are described in detail. This is followed by a summary of up-to-date methods used to detect, characterize and elucidate the structure of EPS produced by LAB. In addition, current strategies on the use of LAB-produced EPS in food products have been discussed, focusing on beneficial applications in dairy products, gluten-free bakery products, and low-fat meat products, as they positively influence the consistency, stability, and quality of the final product. Highlighting is also placed on reports of health-promoting effects, with particular emphasis on prebiotic, immunomodulatory, antioxidant, cholesterol-lowering, anti-biofilm, antimicrobial, anticancer, and drug-delivery activities.

Abstract:Microbial exopolysaccharides (EPS) are long-chain polysaccharides that are synthesized and secreted by microorganisms into the external matrix. In bacteria, EPS can either be associated with the cell surface in the form of capsules or be secreted into the environment. The yield of EPS is influenced by various factors, including the growth conditions and composition of the growth media. EPSs are high molecular-weight carbohydrates that consist of a backbone of repeated subunits of monosaccharides in varying ratios. Recently, there has been an increasing interest in EPS-producing microbes with Generally Recognized as Safe (GRAS) status. These food-grade organisms have the potential to produce polymers that can control the rheological and functional properties of food systems. EPSs have also been reported to have prebiotic and immune-modulating functions like anticancer, antidiabetic, antiviral, etc. The increasing recognition of the association between EPS and health benefits suggests the potential of EPS-producing starters with functional characteristics in the production of value-added functional products. Such products align with consumer demand for natural and healthy alternatives with fewer additives. The exploration of functional means of EPS in Pharmacology will provide an opportunity to identify novel and robust microbial resources producing unique EPSs.
Effects of exopolysaccharides form Lactobacillus plantarum KX041 on high fat diet-induced gut microbiota and inflammatory obesity

Abstract:Lactobacillus plantarum KX041 is a probiotics obtained from early screening, which can relieve inflammation and enrich the anti-obesity intestinal flora, and produce high yield of exopolysaccharides (EPS). The extraction, structure and physicochemical properties of EPS have been completed earlier. However, whether the functional activity of L. plantarum KX041 is related to the EPS is still unclear. In this study, combined in vivo and in vitro tests explored the effects of EPS on inflammatory obesity. In vitro tests, EPS relieased inflammation by inhibiting TLR4/p-NFκB, and inhibited adipocyte differentiation and reduced the transcriptional level of inflammation and lipid-synthesis genes (especially CD36 and peroxisome proliferators-activated receptors-γ PPARγ). In vivo tests demonstrated that EPS could alleviate weight gain, white adipocyte expansion, organ damage, insulin resistance, hyperlipidemia, and inflammatory response caused by high fat diet. Combined with in vitro tests, the transcription level of inflammation and lipid-synthesis genes in adipose tissue identified that IL-1β and PPARγ were the key genes for the effect of EPS on inflammatory obesity. Moreover, EPS increased the abundance of Akkermansia and Lachnospiraceae_NK4A136_group, and intestinal levels of isobutyrate, which is one of the key factors in alleviating inflammatory obesity. These insights may inform the development of functionalized dietary interventions in obesity.

Abstract:Streptococcus thermophilus is the most important thermophilic dairy starter, and is widely used in the dairy industry. Streptococcus thermophilus exopolysaccharides received wide attention over recent decades, because they can improve the properties of the dairy product and confer beneficial health effects. The understanding of the regulatory and biosynthetic mechanisms of EPS will improve the EPS biosynthesis, increase the productivity of EPSs, and develop EPSs with desirable properties. The structure of EPSs is the focus of this study. Revealing the structure-function relationship can lead to increase the knowledge base and from there to increased research of EPS. The EPS yield is a key limiting factor in the research and utilization of EPS. In the present review, biosynthetic pathways and genetics of S. thermophilus EPSs were described and reviewed. At the same time, functional properties and applications of EPS, and strategies for enhancement of EPS production are discussed.
Exopolysaccharides from Lactobacillus plantarum YW11 improve immune response and ameliorate inflammatory bowel disease symptoms

Abstract:Exopolysaccharides (EPSs) possess many bioactivities such as immune regulation, antioxidant, anti-tumor and modulation of intestinal microbial balance but their direct effect on inflammatory bowel disease (IBD) response has not been studied. The purpose of this study was to evaluate the anti-inflammatory effect of EPS produced by L. plantarum YW11 administered at different dosages in IBD mouse model induced with 5% dextran sulphate sodium (DSS). The DSS-induced colitis, accompanied by body weight loss, reduction of colon coefficient and histological colon injury was considerably ameliorated in mice fed the EPS (10 mg/kg). The middle dose of the EPS (25 mg/kg) could effectively recover the intestinal microbial diversity and increase the abundance of Roseburia, Ruminococcus and Blautia with increased content of butyric acid. Moreover, EPS also reduced the production of pro-inflammatory cytokines (TNF-α, IL-1β, IL-6, IFN-γ, IL-12 and IL-18) and enhanced the anti-inflammatory cytokine IL-10. This study showed that EPS might help in modulation of gut microbiota and improve the immunity of the host to reduce the risk of IBD symptoms.

Abstract:A wide range of lactic acid bacteria (LAB) is able to produce capsular or extracellular polysaccharides, with various chemical compositions and properties. Polysaccharides produced by LAB alter the rheological properties of the matrix in which they are dispersed, leading to typically viscous and “ropy” products. Polysaccharides are involved in several mechanisms such as prebiosis and probiosis, tolerance to stress associated to food process, and technological properties of food. In this paper, we summarize the beneficial properties of exopolysaccharides (EPS) produced by LAB with particular attention to prebiotic properties and to the effect of exopolysaccharides on the LAB-host interaction mechanisms, such as bacterial tolerance to gastrointestinal tract conditions, ability of ESP-producing probiotics to adhere to intestinal epithelium, their immune-modulatory activity, and their role in biofilm formation. The pro-technological aspect of exopolysaccharides is discussed, focusing on advantageous applications of EPS in the food industry, i.e., yogurt and gluten-free bakery products, since it was found that these microbial biopolymers positively affect the texture of foods. Finally, the involvement of EPS in tolerance to stress conditions that are commonly encountered in fermented beverages such as wine is discussed.

Abstract:BACKGROUND The exopolysaccharides (EPS) produced by Lactobacillus and other probiotics are associated with many benefits, such as immune regulation, antioxidant properties, antitumor effect, and regulation of intestinal microbe homeostasis. In the present work, the modulatory effect of EPS produced by Lactobacillus rhamnosus ZFM231 on the intestinal flora of mice with inflammatory bowel disease (IBD) induced by dextran sulfate solution (DSS) was investigated. RESULTS Results indicated that weight loss, colonic length, the disease activity index (DAI) score and colonic tissue damage in mice were significantly improved by EPS treatment. Compared with the model group, in the EPS-treated group, the diversity of and composition of gut microbiota at both phylum and genus levels were found to recover to the levels of normal group, indicating the effective modulation on gut microbiota by EPS; short-chain fatty acids (SCFAs), including acetic acid, propionic acid and butyric acid produced by intestinal microbial metabolism, increased significantly; the level of anti-inflammatory factor TGF-β significantly increased and the level of pro-inflammatory factor TNF-α significantly decreased in the colonic cells of EPS-treated mice. CONCLUSION It is clear that EPS produced by L. rhamnosus ZFM231 could find application in functional foods with the property of anti-ulcerative colitis. Abstract:The immunoregulation effect to polysaccharides from Bifidobacterium spp. was investigated on the base of functional assessment standards of health food. Effects of the EPS on immunity were investigated by promoted the proliferation of spleen lymph cells, delayed type hypersensitivity reaction, the HC50 value and macrophage function assay in mice. Data showed that the EPS could obviously increase the ratio of swallowed chicken red blood cell by macrophage and the HC50 value in mice. However, no significant effect was found on the delayed type hypersensitive induced by sheep red blood cell, for only the low dose of 100 mg/(kg·d) EPS promoted the proliferation of spleen lymph cells. Bifidobacterium spp. EPS can certain immunomodulating function.

Abstract:Lactic acid bacteria (LAB) used in the food industry, mainly for the production of dairy products, are able to synthetize exopolysaccharides (EPS). EPS play a central role in the assessment of rheological and sensory characteristics of dairy products since they positively influence texture and organoleptic properties. Besides these, EPS have gained relevant interest for pharmacological and nutraceutical applications due to their biocompatibility, non-toxicity and biodegradability. These bioactive compounds may act as antioxidant, cholesterol-lowering, antimicrobial and prebiotic agents. This review provides an overview of exopolysaccharide-producing LAB, with an insight on the factors affecting EPS production, their dairy industrial applications and health benefits.

Abstract:Exopolysaccharides (EPSs) synthesized by lactic acid bacteria (LAB) have implications for host health and act as food ingredients. Due to the variability of LAB-EPS (lactic acid bacteria-derived exopolysaccharide) gene clusters, especially the glycosyltransferase genes that determine monosaccharide composition, the structure of EPS is very rich. EPSs are synthesized by LAB through the extracellular synthesis pathway and the Wzx/Wzy-dependent pathway. LAB-EPS has a strong immunomodulatory ability. The EPSs produced by different genera of LAB, especially Lactobacillus, Leuconostoc, and Streptococcus, have different immunomodulatory abilities because of their specific structures. LAB-EPS possesses other health effects, including antitumor, antioxidant, intestinal barrier repair, antimicrobial, antiviral, and cholesterol-lowering activities. The bioactivities of LAB-EPS are tightly related to their structures such us monosaccharide composition, glycosidic bonds, and molecular weight (MW). For the excellent physicochemical property, LAB-EPS acts as product improvers in dairy, bakery food, and meat in terms of stability, emulsification, thickening, and gelling. We systematically summarize the detailed process of EPS from synthesis to application, with emphasis on physiological mechanisms of EPS, and specific structure-function relationship, which provides theoretical support for the potential commercial value in the pharmaceutical, chemical, food, and cosmetic industries.

Abstract:Bifidobacteria are a prominent type of bacteria that have garnered significant research attention for their exceptional probiotic properties and capacity to produce exopolysaccharides (EPSs). These compounds exhibit diverse physical, chemical, and biological characteristics, prompting numerous investigations into their potential applications. Researchers have noted their beneficial effects as immune modulators within the host's body across various industries. Extensive research has been conducted on the immunomodulatory effects of bifidobacteria-derived EPSs, with emerging engineering strategies aimed at enhancing their immune-modulating capabilities. Understanding the structure, physicochemical properties, and biological activities of these compounds is crucial for their effective utilization across different industries. Our review encompassed numerous studies exploring Bifidobacterium and its metabolites, including EPSs, across various sectors, drawing from diverse databases. The distinctive properties of EPSs have spurred investigations into their applications, revealing their potential to bolster the immune system, combat inflammation, and treat various ailments. Additionally, these compounds possess antioxidant and antimicrobial properties, making them suitable for incorporation into a range of products spanning food, health, and medicine.
