Abstract:Background: Fusarium graminearum causes Fusarium head blight (FHB), a major disease problem worldwide. Resistance to FHB is controlled by quantitative trait loci (QTL) of which two are located on barley chromosomes 2H bin8 and 6H bin7. The mechanisms of resistance mediated by FHB QTL are poorly defined.Results: Near-isogenic lines (NILs) carrying Chevron-derived resistant alleles for the two QTL were developed and exhibited FHB resistance in field trials. To understand the molecular responses associated with resistance, transcriptomes of the NILs and recurrent parents (M69 and Lacey) were investigated with RNA sequencing (RNA-Seq) after F. graminearum or mock inoculation. A total of 2083 FHB-responsive transcripts were detected and provide a gene expression atlas for the barley-F. graminearum interaction. Comparative analysis of the 2Hb8 resistant (R) NIL and M69 revealed that the 2Hb8 R NIL exhibited an elevated defense response in the absence of fungal infection and responded quicker than M69 upon fungal infection. The 6Hb7 R NIL displayed a more rapid induction of a set of defense genes than Lacey during the early stage of fungal infection. Overlap of differentially accumulated genes were identified between the two R NILs, suggesting that certain responses may represent basal resistance to F. graminearum and/or general biotic stress response and were expressed by both resistant genotypes. Long noncoding RNAs (lncRNAs) have emerged as potential key regulators of transcription. A total of 12,366 lncRNAs were identified, of which 604 were FHB responsive.Conclusions: The current transcriptomic analysis revealed differential responses conferred by two QTL during F. graminearum infection and identified genes and lncRNAs that were associated with FHB resistance.

Abstract:Fusarium head blight (FHB), primarily caused by Fusarium graminearum, reduces grain yield and quality in barley. Resistance to FHB is partial and quantitatively inherited. Previously, major FHB resistant QTL were detected on barley chromosome 2H Bin 8 and 2H Bin 10, and another QTL for reduced deoxynivalenol (DON) accumulation was identified on chromosome 3H Bin 6. To develop an understanding of the molecular responses controlled by these loci, we examined DON and fungal biomass levels and the transcriptome differences in near-isogenic line (NIL) pairs carrying contrasting resistant and susceptible alleles at these QTL during F. graminearum infection. No overlap was found among the differentially accumulated transcripts of the three NIL pairs, indicating that the response to infection controlled by the resistance alleles at each QTL may be distinct. Transcripts showing differential accumulation between resistant and susceptible NILs were compared to results from previous wheat/barley–F. graminearum studies and integrated into a wheat/barley–F. graminearum interaction model.

Abstract:Fusarium head blight (FHB), caused primarily by Fusarium graminearum, decreases grain yield and quality in wheat and barley. Disease severity, deoxynivalenol (DON), fungal biomass, and transcript accumulation were examined in a wheat near-isogenic line pair carrying either the resistant or susceptible allele for the chromosome 3BS FHB-resistance quantitative trait locus (Fhb1). Fhb1 restricts spread of disease symptoms but does not provide resistance to initial infection or initial DON accumulation. Wheat exhibits both induction and repression of large sets of gene transcripts during F. graminearum infection. In addition, a difference in the general timing of transcript accumulation in plants carrying either the resistant or susceptible allele at the Fhb1 locus was detected, and 14 wheat gene transcripts were detected that exhibited accumulation differences between the resistant and susceptible alleles. These results indicate that these may be host responses that differentiate the resistant from the susceptible interaction. Comparative analysis of the wheat-F. graminearum and the barley-F. graminearum interactions revealed a large set of conserved transcript accumulation patterns. However, we also detected gene transcripts that were repressed in wheat but not in barley. Based on the disease symptoms, transcript accumulation data, and comparative analysis of the barley and wheat host response to F. graminearum infection, we developed an integrated model for the interactions of wheat and barley with F. graminearum.

Abstract:Gibberella ear rot（GER） caused by Fusarium graminearum（teleomorph Gibberella zeae） is a common maize disease that not only severely reduces grain yield but also contaminates maize grain with mycotoxins.We investigated the molecular mechanism underlying the host defense responses against pathogen infection using comparative transcriptomic analysis.We injected F.graminearum spore suspensions into plants of resistant（IBM-81） and a susceptible（IBM-85） maize inbred line after pollination and performed RNA-seq 48,72,and 96 h after inoculation.Respectively 487 and 410 differentially expressed genes（DEGs）were induced in the resistant and susceptible lines across three time points,indicating that a stronger defense response was activated in the resistant than in the susceptible line.Among them,198 genes commonly induced in the two lines were subjected to pathway analysis,revealing that most of the DEGs were closely associated with defense and a wide range of metabolic activities.DEGs associated with pathogenesis-related protein 1（PR1） and regulation of salicylic acid were significantly enriched during F.graminearum infection,suggesting that these DEGs play dominant roles in maize resistance to GER.Our results provide a resource for future gene discovery and facilitate elucidation of the complex defense mechanisms involved in resistance to GER.

Abstract:Gibberella stalk rot caused by Fusarium graminearum is one of the most destructive soil-borne diseases of maize (Zea mays L.). Chemical means of controlling Gibberella stalk rot are not very effective; development of highly resistant hybrids is the best choice for disease control. Hence, understanding of the molecular basis underlying maize resistance against Gibberella stalk rot would undoubtedly facilitate the resistance breeding for stalk rot. Two quantitative trait loci (QTL), qRfg1 and qRfg2, conferring resistance to Gibberella stalk rot were detected in our previous study. Three near-isogenic lines (NILs) of maize with either qRfg1 (NIL1) or qRfg2 (NIL2), or neither (NIL3) were generated and subjected to RNA sequencing to study the transcriptional changes after F. graminearum inoculation at 0 (control), 6, and 18 h post-inoculation (hpi). In total, 536,184,652 clean reads were generated, and gene expression levels were calculated using FPKM (fragments per kilobase of exon model per million mapped reads). A total of 7252 differentially expressed genes (DEGs) were found in the three NILs after F. graminearum inoculation. As many as 2499 DEGs were detected between NIL1 and NIL3 at 0 hpi, of which 884 DEGs were more abundant in NIL1 and enriched in defense responses. After F. graminearum inoculation, 1070 and 751 genes were exclusively up- and downregulated, respectively, in NIL1 as compared to NIL3. The 1070 upregulated DEGs were enriched in growth/development, photosynthesis/biogenesis, and defense-related responses. Genes encoding putative auxin-induced proteins and GH3 family proteins in auxin signaling pathway were highly induced and lasted longer in NIL3. Genes involved in polar auxin transport (PAT) were more abundant in NIL3 as compared with NIL2. The qRfg1 confers its resistance to Gibberella stalk rot through both constitutive and induced high expression of defense-related genes; while qRfg2 enhances maize resistance to the disease via relatively lower induction of auxin signaling and repression of PAT. The defense-related transcriptional changes underlying each QTL will undoubtedly facilitate our understanding of the resistance mechanism and resistance breeding for maize stalk rot.

Abstract:Fusarium head blight (FHB) is one of the most prevalent diseases of wheat and other small grain cereals that is predominantly caused by the fungal pathogen Fusarium graminearum. Extraction of total proteins were from tissues of A061-3 and A061-4 plants. Three biological replicates were carried out for each line at four time points. Samples were performed using iTRAQ (Isobaric tags for relative and absolute quantification). This data is being made available to increase the understanding of FHB resistance proteomics. The data from this study are related to the research article "Isobaric tags for relative and absolute quantification-based proteomic analysis of defense responses triggered by the fungal pathogen Fusarium graminearum in wheat" [1].

Abstract:SummaryFusarium head blight, caused by Fusarium graminearum, is a devastating disease of wheat. We developed near‐isogenic lines (NILs) differing in the two strongest known F. graminearum resistance quantitative trait loci (QTLs), Qfhs.ndsu‐3BS (also known as resistance gene Fhb1) and Qfhs.ifa‐5A, which are located on the short arm of chromosome 3B and on chromosome 5A, respectively. These NILs showing different levels of resistance were used to identify transcripts that are changed significantly in a QTL‐specific manner in response to the pathogen and between mock‐inoculated samples. After inoculation with F. graminearum spores, 16 transcripts showed a significantly different response for Fhb1 and 352 for Qfhs.ifa‐5A. Notably, we identified a lipid transfer protein which is constitutively at least 50‐fold more abundant in plants carrying the resistant allele of Qfhs.ifa‐5A. In addition to this candidate gene associated with Qfhs.ifa‐5A, we identified a uridine diphosphate (UDP)‐glycosyltransferase gene, designated TaUGT12887, exhibiting a positive difference in response to the pathogen in lines harbouring both QTLs relative to lines carrying only the Qfhs.ifa‐5A resistance allele, suggesting Fhb1 dependence of this transcript. Yet, this dependence was observed only in the NIL with already higher basal resistance. The complete cDNA of TaUGT12887 was reconstituted from available wheat genomic sequences, and a synthetic recoded gene was expressed in a toxin‐sensitive strain of Saccharomyces cerevisiae. This gene conferred deoxynivalenol resistance, albeit much weaker than that observed with the previously characterized barley HvUGT13248.

Abstract:Molecular mapping led to the identification of two major Fusarium head blight (FHB) resistance QTL, Fhb1 and Qfhs.ifa-5A. The actual function of the resistance genes is still unknown. The resistant line CM82036, the susceptible line Remus and six closely related lines were analyzed for differential gene expression after Fusarium attack. The related lines show contrasting levels of FHB resistance due to the possession of Fhb1 and Qfhs.ifa-5A. At anthesis plants were challenged by Fusarium graminearum or water and at six time points after inoculation gene expression of specific wheat floral tissue was analyzed by cDNA-AFLPs and the Affymetrix Wheat GeneChip. 0.44% of the analyzed gene tags by cDNA-AFLPs displayed differential expressions after Fusarium attack depending on the genotype. Five of the gene tags were associated with the FHB resistance level of the genotypes and the possession of resistance alleles at Fhb1 and Qfhs.ifa-5A. These gene tags show homologies to a UDP-glucosyltransferase, wheat phenylalanine ammonia-lyase, DnaJ-like protein, pathogenesis-related family protein and a rice cDNA clone with unknown function. A thorough comparative analysis with the data gained by the Wheat GeneChip experiments is in progress.
