Pathological characteristics and susceptibility analysis of Hep-2 and Vero cells infected with respiratory syncytial virus

Abstract:Objective To compare the pathological characteristics and susceptibility of HEP-2 cells and Vero cells infected with respiratory syncytial virus(RSV), and the infectivity of the progeny virus produced by RSV on the two kinds of cells.Methods The experiment was divided into Hep-2 blank control group, Hep-2 experimental group, Vero blank control group and Vero experimental group. The blank control group was not infected with virus, and the experimental group was infected with RSV at MOI=1. The cytopathic effect(CPE) of cells was observed by microscope, the expression of RSV N protein was detected by immunofluorescence, and the virus titer was determined by plaque assay. A549 cells were further infected with the progeny viruses produced by Hep-2 and Vero. The infectivity of progeny viruses was compared by plaque assay. Results At 48 h after infection in Hep-2experimental group and 84 h in Vero experimental group,obvious CPE were formed in both groups. The characteristics of CPE formed by RSV on Hep-2 and Vero cells were obviously different. The expression of RSV N protein detected by immunofluorescence was consistent with the characteristics of CPE. Both experimental groups were able to produce high levels of virus titers. However,when A549 cells were infected with the progeny virus of produced by Hep-2 and Vero cells, it was found that there was no significant difference in the titer between the two groups at36 and 48 h after infection(P >0.05), and the titer of of Hep-2 experimental group was higher than that of Vero experimental group at 60 h after infection, with statistically significant difference(P<0.05). Conclusion Both Hep-2 and Vero cells infected with RSV could form obvious CPE, and reach a high virus titer. However, the infectivity of the progeny virus produced by RSV on Hep-2 cells was significantly stronger than that produced on Vero cells.
