Abstract:We present a novel nanostructured micro cell chip with impedance spectroscopy for monitoring cell morphology change non-invasively, in real time and independent of any fluorescent or radioactive probes. The key strategy is to integrate the nanoporous alumina membrane with silicon based microfluidic devices for the impedance monitoring. In this configuration, the impedance of even single cell can be measured at the low frequency range but not be affected by the electrode polarization. The KYSE30 human oesophageal cancer cells have been successfully cultured on nanoporous alumina membrane. Initial electrochemical experiments with lipid layers have been done to testify the functionality of this device. Further experiments for cancer cells will be explored in near future.

Abstract:Clinical viscoelastic hemostatic assays, which have been used for decades, rely on measuring biomechanical responses to physical stimuli but face challenges related to high device and test cost, limited portability, and limited scalability.. Here, we report a differential pattern using self-induced adaptive bubble behavioral perception to refresh it. The adaptive behaviors of bubble deformation during coagulation precisely describe the transformation of viscoelastic hemostatic properties, being free of the precise and complex physical devices. And the integrated bubble array chip allows microassays and enables multi-bubble tests with good reproducibility. Recognition of the developed bubble behaviors empowers automated and user-friendly diagnosis. In a prospective clinical study (clinical model development [n = 273]; clinical assay [n = 44]), we show that the diagnostic accuracies were 99.1% for key viscoelastic hemostatic assay indicators (reaction time [R], kinetics time [K], alpha angle [Angle], maximum amplitude [MA], lysis at 30 min [LY30]; n = 220) and 100% (n = 44) for hypercoagulation, healthy, and hypocoagulation diagnoses. This should provide fresh insight into existing paradigms and help more clinical needs.
Automated measurement of cell mechanical properties using an integrated dielectrophoretic microfluidic device

Abstract:Cell mechanics is closely related to and interacts with cellular functions, which has the potential to be an effective biomarker to indicate disease onset and progression. Although several techniques have been developed for measuring cell mechanical properties, the issues of limited measurement data and biological significance because of complex and labor-intensive manipulation remain to be addressed, especially for the dielectrophoresis-based approach that is difficult to utilize flow measurement techniques. In this work, a dielectrophoresis-based solution is proposed to automatically obtain mass cellular mechanical data by combining a designed microfluidic device integrated the functions of cell capture, dielectrophoretic stretching, and cell release and an automatic control scheme. Experiments using human umbilical vein endothelial cells and breast cells revealed the automation capability of this device. The proposed method provides an effective way to address the low-throughput problem of dielectrophoresis-based cell mechanical property measurements, which enhance the biostatistical significance for cellular mechanism studies.

Abstract:The endothelium plays a vital role in the maintenance of vascular homeostasis and barrier dysfunction is a characteristic of many inflammatory conditions. In our study, an online and continuous technique based on real-time cell electronic sensing (RT-CES) was introduced into study of endothelial barrier function.Electrical impedance in human umbilical vein endothelial cells (HUVECs) was monitored in real-time on microelectronic cell sensor arrays using RT-CES from seeding to confluence. α-thrombin induced electrical impedance alteration in a dose-dependent manner which was paralleled by transendothelial FITC-dextran permeability across endothelial monolayer. Correspondingly, F-actin cytoskeleton redistribution of HUVECs accompanied the change of electrical impedance. The results suggest the RT-CES system could be a powerful tool to study endothelial barrier function model in vitro.

Abstract:Cells are the fundamental unit of life. Owing to cellular heterogeneity, traditional biochemical assays which analyze cells in bulk often overlook the rich information available when single cells are studied. Accurate and reliable determination of the chemical composition in individual cells would greatly improve the probability of discriminating infected cells from healthy ones and provide a solid foundation on study and development in various fields including biochemistry, medicine, and pathology clinic. Microfluidic chip system for single-cell analysis is now attracting broad interests. The ability of integrating the whole process on one microfluidic chip, including single cell injection, lysis, separation and detection of cellular constituents by chip-based CE, offers a new technical platform for single cell analysis. This article provides a review on recent advances in determining intracellular contents in individual cells using microfluidic devices. Focus areas include on-chip single cell manipulation by means of electroosmotic flow (EOF), pressure-driven flow combined with EOF and optical tweezer (OT) to perform sequentially single cell injection, lysis, separation and detection of cellular constituents by chip-based CE with highly sensitive detection. The approaches on on-chip derivatization are also addressed.

Abstract:In this work a novel microfluidic platform for cell culture and assay is developed. On the chip a static cell culture region is coupled with dynamic fluidic nutrition supply structures. The cell culture unit has a sandwich structure with liquid channels on the top, the cell culture reservoir in the middle and gas channels on the bottom. Samples can be easily loaded into the reservoir and exchange constantly with the external liquid environment by diffusion. Since the flow direction is perpendicular to the liquid channel on the top of the reservoir, the cells in the reservoir are shielded from shear-force. By assembling the basic units into an array, a steady concentration gradient can be generated. Cell culture models both for continuous perfusion and one-off perfusion were established on the chip. Both adherent and suspended cells were successfully cultured on the chip in 2D and 3D culture modes. After culturing, the trapped cells were recovered for use in a later assay. As a competitive candidate for a standard cell culture and assay platform, this chip is also adaptable for cytotoxicity and cell growth assays.

Abstract:Dynamic curved interfaces are fundamental and ubiquitous structures in biological systems. However, replicating the structure and function associated with these interfaces for mechanobiology and drug screening is challenging. Here, we develop a dynamic curvature-enabled microfluidic organ chip of two fluid–solid dynamic curved interfaces. One interface effectively integrates adjustable biomechanics, and the other controls drug release with open microfluidics. The fluid–solid interface sensed by the cells can modulate the residual stress, stiffness, strain of the solid phase, and the flow shear stress of the fluid phase. Using the chip, we investigate the mechanotransductive responses of endothelial and epithelial cells, including Piezo1, Ca2+, and YAP, and reveal that the response of the endothelium to combined dynamic cyclic strain and flow shear stress is different from separate stimulation and also disparate from the epithelium. Furthermore, direct and high-efficiency drug release to cells is realized by constructing the other fluid–solid interface on the back side of cells, where drugs are encapsulated within cross-linked alginate hydrogel in the open microfluidic channel. Then, we replicate object-specific and location-specific biomechanical environments within carotid bifurcation and prove the effectiveness of drug delivery. Our design exemplifies dynamic curved biological interfaces with controlled mechanical environments and holds potential for patient-specific medicine.

Abstract:Vascular science is an active area of medicine and biological research. In recent years, intensive research has been focused on the reaction of endothelial cells (ECs) to relevant biological, chemical, or physical cues in vitro. The primary thing of these studies is to make a biomimic environment of ECs which is closer to the in vivo conditions. Here we developed a microfluidic system and fabricated a grooved micropattern thin film to simulate inner blood vessel wall. The micropattern structure was generated by using the elastic biocompatible material poly(dimethylsiloxane) (PDMS). Human umbilical vein endothelial cells (HUVECs) were cultured on the grooved micropattern film. After the cells reached confluence, the thin PDMS film with cells was inserted into the biological grade plastic tube. Then cell culture medium was perfused into the tube and the cellular responses under shear stress and pressure were investigated. The F-actin cytoskeleton and the nuclei of the cells were stained for examination. This microfluidic system provides a convenient and cost-effective platform for the studies of cellular response to mechanical forces. Moreover, this system could also be used for studying cellular responses to drugs under mechanical forces.

Abstract:The mechanical properties of cells are of great significance to their normal physiological activities. The current methods used for the measurement of a cell's mechanical properties have the problems of complicated operation, low throughput, and limited measuring range. Based on micropipette technology, we designed a double-layer micro-valve-controlled microfluidic chip with a series of micropipette arrays. The chip has adjustment pressure ranges of 0.03-1 and 0.3-10 kPa and has a pressure stabilization design, which can achieve a robust measurement of a single cell's mechanical properties under a wide pressure range and is simple to operate. Using this chip, we measured the mechanical properties of the cells treated with different concentrations of paraformaldehyde (PFA) and observed that the viscoelasticity of the cells gradually increased as the PFA concentration increased. Then, this method was also used to characterize the changes in the mechanical properties of the differentiation pathways of stem cells from the apical papilla to osteogenesis.

Abstract:<p>Most tissue‐engineered blood vessels are endothelialized by static cultures in vitro. However, it has not been clear whether endothelial cell‐shedding and local damage may occur in an endothelial layer formed by static cultures under the effect of blood flow shear post‐implantation. In this study, we report a bionic and cost‐effective vascular chip platform, and proved that a static culture of endothelialized tissue‐engineering blood vessels had the problem of a large number of endothelial cells falling off under the condition imitating the human arterial blood flow, and we addressed this challenge by regulating the flow field in a vascular chip. Electrospun membranes made of highly oriented or randomly distributed poly(ε‐caprolactone) fibers were used as the vascular scaffolds, on which endothelial cells proliferated well and eventually formed dense intima layers. We noted that the monolayers gradually adapted to the artery‐like microenvironment through the regulation of chip flow field, which also revealed improved cellular orientations. In conclusion, we have proposed a vascular chip with adaptive flow patterns to gradually accommodate the statically cultured vascular endothelia to the shear environment of arterial flow field and enhanced the orientation of the endothelial cells. This strategy may find numerous potential applications such as screening of vascular engineering biomaterials and maturation parameters, studying of vascular biology and pathology, and construction of vessel‐on‐a‐chip models for drug analysis, among others.</p><p>This article is protected by copyright. Abstract:Hydrodynamic flow is an essential stimulus in many cellular functions, regulating many mechanical sensitive pathways and closely associating with human health status and diseases. The flow pattern of blood in vessels is the key factor in causing atherosclerosis. Hemodynamics has great effect on endothelial cells' gene expression and biological functions. There are various tools that can be used for studying flow-induced cellular responses but most of them are either bulky or lack precise controllability. We develop an integrated microfluidic device that can precisely generate different flow patterns to human endothelial cells cultured on-chip. We monitored cell morphology and used small-input RNA-seq technology to depict the transcriptome profiles of human umbilical vein endothelial cells under uni- or bidirectional flow. Such integrated and miniatured device has greatly facilitated our understanding of endothelial functions with shear stimulus, not only providing new data on the transcriptomic scale but also building the connection between cell phenotypic changes and expression alternations.
On-chip Integrated Optical Stretching and Electrorotation Enabling Single-Cell Biophysical Analysis

Abstract:Cells have different intrinsic markers such as mechanical and electrical properties, which may be used as specific characteristics. Here, we present a microfluidic chip configured with two opposing optical fibers and four 3D electrodes for multiphysical parameter measurement. The chip leverages optical fibers to capture and stretch a single cell and uses 3D electrodes to achieve rotation of the single cell. According to the stretching deformation and rotation spectrum, the mechanical and dielectric properties can be extracted. We provided proof of concept by testing five types of cells (HeLa, A549, HepaRG, MCF7 and MCF10A) and determined five biophysical parameters, namely, shear modulus, steady-state viscosity, and relaxation time from the stretching deformation and area-specific membrane capacitance and cytoplasm conductivity from the rotation spectra. We showed the potential of the chip in cancer research by observing subtle changes in the cellular properties of transforming growth factor beta 1 (TGF-β1)-induced epithelial–mesenchymal transition (EMT) A549 cells. The new chip provides a microfluidic platform capable of multiparameter characterization of single cells, which can play an important role in the field of single-cell research.

Abstract:Three-dimensional tissue platforms are rapidly becoming the method of choice for quantification of the heterogeneity of cell populations for many diagnostic and drug therapy applications. Microfluidic sensors and the integration of sensors with microfluidic systems are often described as miniature versions of their macro-scale counterparts. This technology presents unique advantages for handling costly and difficult-to-obtain samples and reagents as a typical system requires between 100 nL to 10 mu L of working fluid. The fabrication of a fully functional cell-based biosensor utilizes both biological patterning and microfabrication techniques. A digital micro-mirror (photolithographic) system is initiated to construct the tissue platform while a cell printer is used to precisely embed the cells within the construct. Tissue construct developed with these technologies will provide an early diagnostic of a drug's potential use. A three-dimensional interconnected microfluidic environment has the potential to eliminate the limitations of the traditional mainstays of two-dimensional investigations. This paper illustrates an economical and an innovative approach of fabricating a three-dimensional cell-laden microfluidic chip for detecting drug metabolism.

Abstract:Microfluidic chips have emerged as significant tools in cell culture due to their capacity for supporting cells to adopt more physiologically relevant morphologies in 3D compared with traditional cell culture in 2D. Currently, irreversible bonding methods, where chips cannot be detached from their substrates without destroying the structure, are commonly used in fabrication, making it challenging to conduct further analysis on cells that have been cultured on-chip. Although some reversible bonding techniques have been developed, they are either restricted to certain materials such as glass, or require complex processing procedures. Here, we demonstrate a simple and reversible polydimethylsiloxane (PDMS)-polystyrene (PS) bonding technique that allows devices to withstand extended operations while pressurized, and supports long-term stable cell cultures. More importantly, it allows rapid and gentle live cell extraction for downstream manipulation and characterization after long-term on-chip culturing, and even further subculturing. Our new approach could greatly facilitate microfluidic chip-based cell and tissue cultures, overcoming current analytical limitations and opening up new avenues for downstream uses of on-chip cultures, including 3D-engineered tissue structures for biomedical applications.

Abstract:To reproduce hemodynamic stress microenvironments of endothelial cells in vitro is of vital significance, by which one could exploit the quantitative impact of hemodynamic stresses on endothelial function and seek innovative approaches to prevent circulatory system diseases. Although microfluidic technology has been regarded as an effective method to create physiological microenvironments, a microfluidic system to precisely reproduce physiological arterial hemodynamic stress microenvironments has not been reported yet. In this paper, a novel microfluidic chip consisting of a cell culture chamber with on-chip afterload components designed by the principle of input impedance to mimic the global hemodynamic behaviors is proposed. An external feedback control system is developed to accurately generate the input pressure waveform. A lumped parameter hemodynamic model (LPHM) is built to represent the input impedance to mimic the on-chip global hemodynamic behaviors. Sensitivity analysis of the model parameters is also elaborated. The performance of reproducing physiological blood pressure and wall shear stress is validated by both numerical characterization and flow experiment. Investigation of intracellular calcium ion dynamics in human umbilical vein endothelial cells is finally conducted to demonstrate the biological applicability of the proposed microfluidic system.

Abstract:Direct cell-cell communication between adjacent cells is vital for the development and regulation of functional tissues. However, current biological techniques are difficult to scale up for high-throughput screening of cell-cell communication in an array format. In order to provide an effective biophysical tool for the analysis of molecular mechanisms of gap junctions that underlie intercellular communication, we have developed a microfluidic device for selective trapping of cell-pairs and simultaneous optical characterizations. Two different cell populations can be brought into membrane contact using an array of trapping channels with a 2μm by 2μm cross section. Device operation was verified by observation of dye transfer between mouse fibroblasts (NIH3T3) placed in membrane contact. Integration with lab-on-a-chip technologies offers promising applications for cell-based analytical tools such as drug screening, clinical diagnostics, and soft-state biophysical devices for the study of gap junction protein channels in cellular communications. Understanding electrical transport mechanisms via gap junctions in soft membranes will impact quantitative biomedical sciences as well as clinical applications.

Abstract:A new design of a macro-to-micro interface that can be used for simple and reliable control of comprehensive microfluidic cell culture processes is introduced making microfluidic procedures easily accessible to biological laboratories. The novel macro-to-micro interface is evaluated by adapting a workflow for single-cell, cell pair, and cell cluster encapsulation into hydrogel beads acting as 3D microenvironment with subsequent long-term cultivation. For the first time, the coupling of single-cell time-lapse microscopy data with phenotypic (immunofluorescence) and genotypic (single-molecule RNA fluorescence in situ hybridization) endpoint measurements as well as downstream compatibility in a single chip is shown. The presented platform will be a valuable tool for performing studies of dynamic biological processes coupled to a multiparametric endpoint characterization at the single-cell level as well as for gaining more detailed mechanistic insights into gene function relationships and the behavior of biological systems.

Abstract:Mechanical stimulation, especially fluid shear stress (FSS), is essential for a cell to regulate regular behaviors. A high-throughput platform to provide varying FSS for cell research is desperately required for better mimicking of the complex fluidic microenvironment. This work reports an integrated microfluidic chip that could afford five different FSS gradients consistently to investigate drug toxicity on cells with the stimulation of FSS. Compared with traditional methods to provide FSS, this device would be easier to operate, have higher throughput, and could eliminate interference factors from the culture environment for cells (apart from the unique variable FSS). On such a multi-FSS platform, effects of drugs toxicity on cells were exhibited, which would be more intense than that under static conditions. The results indicated that FSS enhanced the drug toxicity. The designed biochip provides an easy and high-throughput platform to evaluate the toxicity of drugs in the more authentic microenvironment and could be promisingly applied in future drug screening tests.

Abstract:Microfluidics is a highly useful platform for culturing, monitoring, and testing biological cells. The integration of electrodes into microfluidic channels extends the functionality, sensing, and testing capabilities of microfluidic systems. By employing an electrochemical impedance spectroscopy (EIS) technique, the non-invasive, label-free detection of the activities of cells in real-time can be achieved. To address the movement toward spatially resolving cells in cell culture, we developed a sensory system capable of electro-addressing cell location within a microfluidic channel. This simple system allows for real-time cell location, integrity monitoring (of barrier producing cells), and confluency sensing without the need for frequent optical evaluation-saving time. EIS results demonstrate that cells within microfluidic channels can be located between various pairs of electrodes at different positions along the length of the device. Impedance spectra clearly differentiates between empty, sparse, and confluent microfluidic channels. The system also senses the level of cell confluence between electrode pairs-allowing for the relative quantification of cells in different areas of the microfluidic channel. The system's electrode layout can easily be incorporated into other devices. Namely, organ-on-a-chip devices, that require the monitoring of precise cell location and confluency levels for understanding tissue function, modeling diseases, and for testing therapeutics.
Quantitative Study of the Dynamic Tumor-Endothelial Cell Interactions Through an Integrated Microfluidic Coculture System.

Abstract:The interaction between tumor and endothelial cells is crucial to cancer metastasis and angiogenesis. We developed a novel microfluidic device to assess the cell-cell interaction quantitatively at the single cell resolution. This integrated chip offers 16 coculture experiments in parallel with controllable microenvironments to study interactions between cells dynamically. We applied this approach to model the tumor invasion using Hela cells and human umbilical vein endothelial cells (HUVECs) and monitored the migration of both. We observed the retreatment of HUVECs upon the approach of Hela cells during coculture, indicating that the interaction between two cells was mediated by soluble factors. This interaction was further analyzed through quantitatively processing the phase-contrast microscopic time-lapse images of each individual coculture chamber. We also confirmed this paracrine effect by varying the frequency of medium change. This microfluidic technique is highly controllable, contamination free, fully automatic, and inexpensive. This approach not only offers a unique way to quantitatively study the interaction between cells but also provides accurate spatial-temporal tunability of microenvironments for cell coculture. We believe this method, intrinsically high-throughput and quantitative, will greatly facilitate the study of cell-cell interactions and communications.
