Abstract:Covering: literature up to 2018 Terpenes are a class of natural products characterized by remarkable structural diversity. Much of this diversity arises biosynthetically from a handful of linear precursors through the so-called tail-to-head terpene cyclization reaction. This reaction is one of the most complex observed in nature, and historically attempts to replicate it with non-enzymatic means have met with little success. In recent years, however, the development of manmade binding pockets that allow such reactions to take place has been reported. This Highlight provides an overview of this nascent field, and outlines the challenges that need to be overcome moving forward.

Abstract:Biomimetic total synthesis has played a pivotal role in the development of synthetic organic chemistry. In particular, efforts aimed at mimicking the head-to-tail (HT) cation–π cyclization cascades invoked in terpene biosynthesis, such as those catalyzed by type-II cyclases, have led to a multitude of new synthetic methods, chemical concepts, and total syntheses over the past century. Conversely, synthetic methodology that mimics tail-to-head (TH) cation–π cyclization cascades, mediated by Mg 2+ type-I terpene cyclases, remains elusive in organic synthesis, despite key roles in the biosynthesis of privileged therapeutic molecules such as taxol and artemesinin. Here we report that Li + /weakly-coordinating anion (WCA) salts catalyze the TH polycyclization of linaloyl fluoride, leading to high-yielding mixtures of polycyclic terpene natural products including cedrenes, cadinadiene, epizonarene, and δ-selinene. The examples reported herein are the first small molecule-catalyzed TH polycyclization reactions enabling the shortest (formal) total synthesis of ( ± )-artemisinin. Moreover we apply this strategy to the diterpene geranyllinaloyl fluoride, resulting in a two-step total synthesis of the tricyclic core of the gersemiols (named here as α -gersemiene), a recently discovered class of marine diterpenoid natural products.

Abstract:AbstractThe synthesis of terpene natural products remains a challenging task due to the enormous structural diversity in this class of compounds. Synthetic catalysts are unable to reproduce the tail‐to‐head terpene cyclization of cyclase enzymes, which create this diversity from just a few simple linear terpene substrates. Recently, supramolecular structures have emerged as promising enzyme mimetics. In the present study, the hexameric resorcinarene capsule was utilized as an artificial cyclase to catalyze the cyclization of sesquiterpenes. With the cyclization reaction as the key step, the first total synthesis of the sesquiterpene natural product δ‐selinene was achieved. This represents the first total synthesis of a sesquiterpene natural product that is based on the cyclization of a linear terpene precursor inside a supramolecular catalyst. To elucidate the reaction mechanism, detailed kinetic studies and kinetic isotope measurements were performed. Surprisingly, the obtained kinetic data indicated that a rate‐limiting encapsulation step is operational in the cyclization of sesquiterpenes.

Abstract:The tail-to-head terpene tyclization is arguably one. of the most complex reactions found in nature. The hydrogen-bond-based resorcinarene capsule represents the first "man-made enzyme-like catalyst that is capable of catalyzing this reaction. Based on noncovalent-interactions' between the 'capsule and the substrate, the product selectivity can be tuned by using different leaving groups. A detailed mechanistic investigation was performed to elucidate the reaction, mechanisril. For the cyclization of geranyi acetate, it was found that the cleavage. of the leaving group is the rate-determining step. Furthermore, the studies revealed that trace amounts of add are required as cocatalyst. A series of control experiments demonstrate that a synergistic interplay between the supramolecular capsule and the acid traces is, required for catalytic activity.

Abstract:The domino cyclization/coupling strategy is one of the most effective methods to produce cyclized and multi-functionalized compounds from olefins, which has attracted huge attention from chemists and biochemists especially for its considerable potential of enantiocontrol. Nowadays, more and more studies are developed to achieve difunctionalization of substituted olefins through an asymmetric domino intramolecular cyclization/cascade reaction, which is still an elegant choice to accomplish several synthetic ideas such as complex natural products and drugs. This review surveys the recent advances in this field through reaction type classification. It might serve as useful knowledge desktop for the community and accelerate their research.

Due to their unique structures as critical core skeletons in these molecules, a variety of new strategies for the construction of cyclobutane rings have greatly emerged during the last decade. In this review, we wish to summarize the recent progress in the cyclobutane-containing natural product synthesis with an emphasis on disconnection tactics employed to forge the four-membered rings, aiming to provide a complement to existing reviews.

Abstract:The year 2017 marks the twentieth anniversary of terpenoid cyclase structural biology: a trio of terpenoid cyclase structures reported together in 1997 were the first to set the foundation for understanding the enzymes largely responsible for the exquisite chemodiversity of more than 80000 terpenoid natural products. Terpenoid cyclases catalyze the most complex chemical reactions in biology, in that more than half of the substrate carbon atoms undergo changes in bonding and hybridization during a single enzyme-catalyzed cyclization reaction. The past two decades have witnessed structural, functional, and computational studies illuminating the modes of substrate activation that initiate the cyclization cascade, the management and manipulation of high-energy carbocation intermediates that propagate the cyclization cascade, and the chemical strategies that terminate the cyclization cascade. The role of the terpenoid cyclase as a template for catalysis is paramount to its function, and protein engineering can be used to reprogram the cyclization cascade to generate alternative and commercially important products. Here, I review key advances in terpenoid cyclase structural and chemical biology, focusing mainly on terpenoid cyclases and related prenyltransferases for which X-ray crystal structures have informed and advanced our understanding of enzyme structure and function.

Abstract:An increasingly large number of bioactive cyclic peptides have been found in nature. The enhanced biological specificity, activity, and metabolic stability of cyclopeptides, by virtue of their conformationally constrained structural feature, have attracted much attention and made these compounds extensively studied. Cyclic peptidomimetic scaffolds and templates have been widely used to assemble various spatially defined functional groups for molecular recognition and drug discovery. These efforts are complemented optimally by NMR and X-ray based structure information, and assisted by molecular modeling based structure predictive methods. Peptide cyclization also becomes an effective and commonly employed strategy for peptide modifications. During the past several decades, great effort has been made to develop more efficient methods for the synthesis of cyclic peptides and peptidomimetics, as potential drug leads and/or as models for conformational analysis. This review is aimed at highlighting novel recently developed peptide cyclization approaches, and illustrating the profitable applications.

Abstract:In the wide area of host guest chemistry, hemicryptophanes, combining a cyclotribenzylene (or cyclotriveratrylene CTV) unit with another different C-3-symmetrical moiety, appears as a recent family of Molecular cages. The synthesis and recognition properties of the first hemicryptophane were reported in 1982 by Collet and Lehn, but the very little attention received by this class of host compounds in the 20 years following this first promising result can account for their apparent novelty. Indeed, in the last 10 years hemicryptophanes have aroused growing interest, and new aspects have been developed. Thanks to the rigid shaping unit of the north part (CTV) and also the variable and easily functionalized south moiety, hemicryptophanes are revealed to be inherently chiral ditopic host compounds, able to encapsulate various guests, including charged and neutral species. They also enter the field of stimuli-responsive supramolecular systems exhibiting controlled functions. Moreover, endohedral functionalization of their inner cavity leads to supramolecular catalysts. The confinement of the catalytic center affords nanoreactors with improved catalytic activities or selectivities when compared to model systems without a cavity. The current trend shows that reactions in the confined space of synthetic hosts, mimicking, enzyme behavior, will expand rapidly in the near future.

Abstract:Inspired or learning from nature is always a great approach to solve problems. In terms of natural product synthesis, which is always challenging, it is wise to imitate nature’s way to overcome difficulty. Biomimetic synthesis has been recognized for its remarkable ability to simplify the synthetic processes toward complex natural targets. In this Thieme Cheminar, our speakers will discuss their recent results in this field.Biomimetic Catalyst Systems from the Self-assembly of AmphiphilesCooperative interactions between precisely positioned functional groups in an active site are one of the hallmarks of natural enzymes. Taking this queue from biological systems, chemists have devised numerous strategies for the pre-organization of functional groups to allow the formation of multiple contacts with a bound substrate. Recently, we demonstrated that such cooperative interactions can also be achieved within self-assembled vesicles. In this talk, we will discuss how self-assembly offers a dynamism that allows the incorporation of stimuli-responsiveness into these systems and even life-like properties.Current challenges in this research area include the design of structures where its formation and destruction can be reversibly coupled to an external stimulus (e.g. chemical fuel or light), systems that operate out of equilibrium, and the demonstration of communication between components within a chemical network (e.g. via feedback loops). The incorporation of responsive components within chemical networks can lead to new systems/materials that are self-generating, self-repairing and with the potential to display adaptive and evolutive properties.Chemical Emulation of the Biosynthesis of Natural ProductsBiomimetic synthesis is an effective strategy to streamline the synthesis of structurally complex bioactive natural products. However, the biosynthetic pathways of skeleton rearranged natural products are not disclosed yet in most cases. We developed a biosynthetic network analysis (BNA) method to address this general issue. By taking advantage of this method, we accomplished the divergent total syntheses of pentacyclic cytochalasans and merocytochalasans from a common precursor aspochalasin D based on biosynthetic network analysis of the cytochalasans biosynthetic pathways. The key steps of the syntheses include Prins-type and ene-type cyclizations to establish the core of pentacyclic aspochalasans. Here we report a bioinspired modular synthesis of merocytochalasans by taking advantage of a biomimetic [5+2] heterocycloaddition of an ortho-quinone intermediate. The synthetic efforts provide a platform for the synthesis of more than 500 valuable cytochalasans bearing different macrocycles and amino-acid residues. Furthermore, we realized the efficient synthesis of triterpene spirochensilide A from lanosterol through the chemical emulation of its biosynthetic pathway.Applications of Quantum Chemistry in Biomimetic Syntheses of Polycyclic Furanocembrane DerivativesThis talk summarizes the guidance provided by quantum chemical calculations to the biomimetic syntheses of polycyclic marine furanocembrane derivatives. Polycyclic furanocembrane derivatives are a group of structurally complex and biologically important marine natural products isolated from marine corals. Their syntheses are challenging due to their structural complexity. Biomimetic synthetic proposals have been made and some verified via chemical synthesis. Computational chemistry can support these biomimetic syntheses. In this talk, we would like to share the synthetic and computational attempts we have made in the biomimetic syntheses of polycyclic furanocembrane derivatives, including intricarene, bielschowskysin, providencin and plumarellide.

Abstract:With the increasing advancement of supramolecular chemistry, the focus is gradually shifting from the preparation of traditional polymeric materials to the development of multifunctional supramolecular materials. Among the building blocks of supramolecular materials, natural small molecules with intrinsic assembly properties are preferred over synthetic molecules due to their lower toxicity, better biocompatibility, biodegradability, and bioactivity. Terpenoids, as a kind of natural product with a wide range of supply sources, numerous modification sites, and abundant functions, have become a promising platform for the construction of supramolecular materials. Currently, many supramolecular materials based on different terpenoids have been successfully developed, which play an important role in tissue engineering, biomedicine, food science, and flexible electronics. This review comprehensively integrates and summarises current research findings on typical terpenoid-based supramolecular materials, including low-molecular-weight gels, nanocarriers for drug delivery, chiral self-assembled materials, and emulsion gels, focusing on their preparation methods, assembly processes, structural characteristics, and applications. The aim of this review is to draw attention to the potential of natural terpenoids, promote their exploitation and utilisation, and offer more alternatives for the creation of supramolecular materials.

Abstract:CONSPECTUS: Total synthesis of natural products has been one of the most exciting and dynamic areas in synthetic organic chemistry. Nowadays, the major challenge in this field is not whether a given target of interest can be synthesized but how to make it with commendable efficiency and practicality. To meet this grand challenge, a wise way is to learn from Mother Nature who is recognized for her superb capability of forging complicated and sometimes beyond-imagination molecules in her own delicate way. Indeed, since Sir Robert Robinson published his groundbreaking synthesis of tropinone in 1917, biomimetic synthesis of natural products, a process of imitating nature's way to make molecules, has evolved into one of the most popular research directions in organic synthesis. Our group has been engaging in biomimetic synthesis of natural products in the past decade. During this time, we have come to realize that the successful implementation of a biomimetic synthesis entails the orchestrated combination of bioinspiration and rational design. On the one hand, we prefer to utilize some elegant bioinspired transformations (e.g., Diels-Alder dimerization, 6p-electrocyclization, and [2 + 2]-photocycloaddition) as the key steps of our synthesis, which enable rapid construction of the core skeletons of the chased targets with high efficiency; on the other hand, various powerful reactions (e.g., dyotropic rearrangement of beta-lactone, tandem aldol condensation/Grob fragmentation reaction, and organocatalytic asymmetric Mukaiyama-Michael addition) are rationally designed by us, which allow for facile access to the requisite precursors for attempting biomimetic transformations. In some cases, the proposed biomimetic transformation may fail to give a satisfactory result in practice, and thus we opt to develop creative tactics (e.g., hydrogen atom transfer-triggered vinyl cyclobutane ring opening/oxygen insertion/cyclization cascade) that can meet the challenge. Guided by this synthesis concept, we have achieved the total syntheses of multiple families of natural products of great importance in both chemistry and biology, representatives of which include xanthanolides, cytochalasans, and plakortin-type polyketides. Of note, most of these targets could be accessed in a concise, efficient, and scalable manner, which paves the way for further exploration of their biological functions and medicinal potential. Moreover, owing to their biomimetic nature, our syntheses provide valuable information for deciphering the underlying biosynthetic pathways of the chased targets, which could not be attained by other synthetic modes.

Abstract:Terpenoids comprise the largest family of natural products and include various structurally different genus which play important roles in living organisms. Biosynthetically, diterpenoids are derived from (E,E,E)-geranylgeranyl diphosphate (GGPP). From GGPP, diterpene cyclase catalyzes a sequence of carbocation-mediated cyclizations, rearrangements, and further oxidations, leading to a class of structurally unique ent-kaurenes, such as cafestol, gibberellin A3 and oridonin. According to the biosynthesis pathway of ent-kaurene, we designed a chiral acetal-enabled and SnCl4-promoted biomimetic polyene cationic cyclization. With a following Birch reduction/alkylation cascade, a core skeleton of representative ent-kaurenes diterpenoids was completed.

Abstract:Oligomeric sesquiterpenoids, biogenetically assembled from two or three monomeric sesquiterpenoid units via diverse pathways, represent a unique class of natural products. The synthetic studies inspired by their biogenesis have offered significant impetus for the efficient construction of these architecturally complex frameworks. Here we provide an overview for the biomimetic syntheses of these dimeric and trimeric molecules based on the different strategies of bond formation, including Diels-Alder reaction, hetero-Diels-Alder reaction, [2 + 2] cycloaddition, and C-C bond coupling.

Abstract:Abstract Terpenoids, formed by cyclization and/or permutation of isoprenes, are the most diverse and abundant class of natural products with a broad range of significant functions. One family of the critical enzymes involved in terpenoid biosynthesis is terpene cyclases (TCs), also known as terpene synthases (TSs), which are responsible for forming the ring structure as a backbone of functionally diverse terpenoids. With the recent advances in biotechnology, the researches on terpene cyclases have gradually shifted from the genomic mining of novel enzyme resources to the analysis of their structures and mechanisms. In this review, we summarize both the new methods for genomic mining and the structural mechanisms of some typical terpene cyclases, which are helpful for the discovery, engineering and application of more and new TCs.

Abstract:Tetracyclic terpenoid-derived natural products are a broad class of medically relevant agents that include well-known steroid hormones and related structures, as well as more synthetically challenging congeners such as limonoids, cardenolides, lanostanes, and cucurbitanes, among others. These structurally related compound classes present synthetically disparate challenges based, in part, on the position and stereochemistry of the numerous quaternary carbon centers that are common to their tetracyclic skeletons. While de novo syntheses of such targets have been a topic of great interest for over 50 years, semisynthesis is often how synthetic variants of these natural products are explored as biologically relevant materials and how such agents are further matured as therapeutics. Here, focus was directed at establishing an efficient, stereoselective, and molecularly flexible de novo synthetic approach that could offer what semisynthetic approaches do not. In short, a unified strategy to access common molecular features of these natural product families is described that proceeds in four stages: (1) conversion of epichlorohydrin to stereodefined enynes, (2) metallacycle-mediated annulative cross-coupling to generate highly substituted hydrindanes, (3) tetracycle formation by stereoselective forging of the C9-C10 bond, and (4) group-selective oxidative rearrangement that repositions a quaternary center from C9 to C10. These studies have defined the structural features required for highly stereoselective C9-C10 bond formation and document the generality of this four-stage synthetic strategy to access a range of unique stereodefined systems, many of which bear stereochemistry/substitution/functionality not readily accessible from semisynthesis.
