Abstract:Zeolitic Imidazolate Framework-8 (ZIF-8) is a type of typical metal organic framework (MOF), which is an emerging porous coordination polymers that is bridged by zinc metal ions and organic linkages.Due to its intrinsic porous characteristics, unique pH sensitivity as well as superior drug loading efficiency, zeolitic imidazolate framework-8 has attracted extensive attention and received a wide range of applications in the field of biomedicine.In this review, we briefly discuss the effective delivery and controlled release of various drugs in ZIF-8, such as functional proteins, hydrophilic and hydrophobic drugs.Understanding the processes of various drugs loading is important for developing new drug delivery systems.In addition, we also explored the cooperation mechanism between ZIF and different materials.The combination of chemotherapy, photodynamic therapy, photothermal therapy and medical imaging have been applied in the field of nanomedicine and have made emarkable achievements.

Abstract:Due to the limitations resulting from hypoxia and the self-aggregation of photosensitizers, photodynamic therapy (PDT) has not been applied clinically to treat most types of solid tumors. Zeolitic imidazolate framework-8 (ZIF-8) is a common metal-organic framework that has ultra-high porosity, an adjustable structure, good biocompatibility, and pH-induced biodegradability. In this review, we summarize the applications of ZIF-8 and its derivatives in PDT. This review is divided into two parts. In the first part, we summarize progress in the application of ZIF-8 to enhance PDT and realize theranostics. We discuss the use of ZIF-8 to avoid the self-aggregation of photosensitizers, alleviate hypoxia, increase the PDT penetration depth, and combine PDT with multi-modal imaging. In the second part, we summarize how ZIF-8 can achieve synergistic PDT with other anti-tumor therapies, including chemotherapy, photothermal therapy, chemodynamic therapy, starvation therapy, protein therapy, gene therapy, and immunotherapy. Finally, we highlight the challenges that must be overcome for ZIF-8 to be widely applied in PDT. To the best of our knowledge, this is the first review of ZIF-8-based nanoplatforms for PDT.

Abstract:Cancer severely threatens human health and has remained the leading cause of disease–related death for decades. With the rapid advancement of nanomedicine, nanoscale metal–organic frameworks are believed to be potentially applied in the treatment and biomedical imaging for various tumors. Zeolite imidazole framework (ZIF)–8 attracts increasing attention due to its high porosity, large specific surface area, and pH–responsiveness. The designs and modifications of ZIF–8 nanoparticles, as well as the strategy of drug loading, demand a multifaceted and comprehensive understanding of nanomaterial features and tumor characteristics. We searched for studies on ZIF–8–based nanoplatforms in tumor theranostics on Web of Science from 2015 to 2022, mainly focused on the research published in the past 3 years, summarized the progress of their applications in tumor imaging and treatment, and discussed the favorable aspects of ZIF–8 nanoparticles for tumor theranostics as well as the future opportunities and potential challenges. As a kind of metal–organic framework material full of potential, ZIF–8 can be expected to be combined with more therapeutic systems in the future and continue to contribute to all aspects of tumor therapy and diagnosis.

Abstract:Photodynamic therapy (PDT) and photothermal therapy (PTT) are emerging cancer treatments that induce tumor cell death by generating cytotoxic reactive oxygen species (ROS) under light or by increasing the temperature. However, the tumor microenvironment (TME) is very complex and influenced by the degree of penetration of the excitation light, and the clinical treatment efficiency of PDT and PTT is low. The combination of PDT and PTT with other immunotherapies has emerged as a promising treatment strategy. Zeolitic imidazolate framework-8 (ZIF-8) is a novel porous organofluoride material with a large specific surface area, good thermal and chemical stabilities, high drug loading capability, and pH-responsive biodegradability compared to other nanomaterials. These ZIF-8 properties have great potential in PDT and PTT, as well as in cancer synergistic treatment. Moreover, ZIF-8 is an effective protective carrier for anticancer materials. In this paper, the synthesis strategy for ZIF-8 as a carrier and functionalized material for cancer therapy is reviewed, as well as the applicable PDT and PTT principles. Additionally, a comprehensive overview of the significant effects of ZIF-8 in cancer treatment using PDT and PTT-driven systems is provided. Lastly, the challenges facing the application of ZIF-8 in this field is presented to clarify future developments.

Abstract:Cancer severely threatens human health, which makes it particularly urgent to develop effective strategies for cancer diagnosis and therapy. Gene therapy and nucleic acid-based cancer diagnosis play important roles in cancer theranostic, but their applicability is challenged by the low cellular uptake and enzymatic degradation. In response, safe and efficient carrier metal–organic frameworks (MOFs) have been proposed. Zeolite imidazole frameworks (ZIFs), a promising MOF type, can easily encapsulate negatively charged nucleic acid while offering a high loading efficiency, adjustable structure, and conditional responsiveness (pH, adenosine triphosphate (ATP), or glutathione (GSH)). In this review, we studied recent articles on nucleic acid-loading ZIFs-based nanoplatforms in tumor theranostics on the Pubmed database, with a focus on the synthesis and applications in tumor diagnosis and treatment. The relevant favorable aspects, potential challenges, and future opportunities are also discussed in this review.

Abstract:Zeolitic Imidazole Frameworks (ZIFs) are widely applied in nanomedicine for their high drug loading, suitable pore size, pH-responsive drug release, and so on. However, fast drug release during circulation, unexpected toxicity to mice major organs, undesirable long-term accumulation in the lung and even death currently hinder their in vivo biomedical applications. Herein, we report an amorphous ZIF-8 (aZIF-8) with high loading of 5-Fu through pressure-induced amorphization. This nano-system avoids early drug release during circulation and provides tumor microenvironment-responsive drug release with improved in vitro cell viability, and survival rate in in vivo evaluations as compared to ZIF-8. Furthermore, aZIF-8 shows longer blood circulation and lower lung accumulation than ZIF-8 at same injected doses. Less drug release during circulation, longer blood circulation, and better biocompatibility of aZIF-8/5-Fu significantly improves its therapeutic efficacy in ECA-109 tumor-bearing mouse, and result in 100% survival rate over 50 days after treatment. Therefore, aZIF-8 with favorable biocompatibility and long blood circulation is expected to be a promising nano-system for efficacious cancer therapy in vivo.

Abstract:With the rapid advancement of nanotechnology, stimuli-responsive nanomaterials have emerged as a feasible choice for the designing of controlled drug delivery systems. Zeolitic imidazolates frameworks are a subclass of Metal-organic frameworks (MOFs) that are recognized by their excellent porosity, structural tunability and chemical modifications make them promising materials for loading targeted molecules and therapeutics agents. The biomedical industry uses these porous materials extensively as nano-carriers in drug delivery systems. These MOFs not only possess excellent targeted imaging ability but also cause the death of tumor cells drawing considerable attention in the current framework of anticancer drug delivery systems. In this review, the outline of stability, porosity, mechanism of encapsulation and release of anticancer drug have been reported extensively. In the end, we also discuss a brief outline of current challenges and future perspectives of ZIFs in the biomedical world.

Abstract:Zeolitic imidazolate framework (ZIF-8) is specifically promising for drug carrier due to its excellent intrinsic properties. However, the high toxicity of ZIF-8 nanoparticles severely limits its further research and clinical application. In the work, the biocompatibility of ZIF-8 nanoparticles are greatly improved by coating ZrO2 onto the surface. The survival rate of cells and mice in the ZIF-8@ZrO2 nanocomposites group is significantly increased compared with the undecorated ZIF-8 nanoparticles group. Doxorubicin (DOX) as a chemotherapeutic drug is deposited during the ZIF-8 growth by a facile one-pot method. Ionic liquid (IL) is loaded into the pore of the ZIF-8/DOX@ZrO2 nanocomposites for enhanced microwave thermal therapy. The tumor inhibition rate of ZIF-8/DOX@ZrO2@IL nanocomposites with synergistic microwave thermal therapy and chemotherapy is obviously higher than other groups. In addition, the ZIF-8/DOX@ZrO2@IL nanocomposites are used for real-time monitoring of the therapeutic outcomes due to the excellent CT contrast agent of ZrO2. Therefore, such ZrO2 coating strategy shows great promise for overcoming high toxicity of ZIF-8 nanoparticles, which offers a new platform for tumor synergistic microwave thermal therapy and chemotherapy using ZIF-8/DOX@ZrO2@IL nanocomposite as a theranostic nanocarrier.

Abstract:The progressive development of zeolitic imidazolate frameworks (ZIFs), as a subfamily of metal-organic frameworks (MOFs), and their unique features, including tunable pore size, large surface area, high thermal stability, and biodegradability/biocompatibility, have made them attractive in the field of biomedicine, especially for drug delivery and biomineralization applications. The high porosity of ZIFs gives them the opportunity for encapsulating a high amount of therapeutic drugs, proteins, imaging cargos, or a combination of them to construct advanced multifunctional drug delivery systems (DDSs) with combined therapeutic and imaging capabilities. This review summarizes recent strategies on the design and fabrication of ZIF-based nansystems and their exploration in the biomedical field. First, recent developments for the adjustment of particle size, functionality, and morphology of ZIFs are discussed, which are important for achieving optimized therapeutic/theranostic nanosystems. Second, recent trends on the application of ZIF nanocarriers for the loading of diverse cargos, including anticancer medicines, antibiotic drugs, enzymes, proteins, photosensitizers, as well as imaging and photothermal agents, are investigated in order to understand how multifunctional DDSs can be designed based on the ZIF nanoparticles to treat different diseases, such as cancer and infection. Finally, prospects on the future research direction and applications of ZIF-based nanomedicines are discussed.

Abstract:Metal-organic frameworks (MOFs) are crystalline porous materials with periodic network structures formed by self-assembly of metal ions and organic ligands. Attributed to their tunable composition and pore size, ultrahigh surface area (1000-7000 m2/g) and pore volume (1.04-4.40 cm3/g), easy surface modification, appropriate physiological stability, etc., MOFs have been widely used in biomedical applications in the last two decades, especially for the delivery of bioactive agents. In the initial stage, MOFs were widely used to load small molecule drugs with ultra-high doses. Whereafter, more recent work has focused on the load of biomacromolecules, such as nucleic acids and proteins. Over the past years, we have devoted extensive effort to investigate the function of MOF materials for bioactive agent delivery. MOFs can be used not only as an intelligent nanocarrier to deliver or protect bioactive agents but also as an activator for their release or activation in response to the different microenvironments. Altogether, this review details the current progress of MOF materials for bioactive agent delivery and looks into their future development.

Abstract:In recent years, zeolitic imidazolate framework-8 (ZIF-8) has become an attractive metal organic framework (MOF) material in drug delivery for cancer chemotherapy. However, as a drug delivery system, ZIF-8 still shows some disadvantages, such as short blood circulation time and poor tumor targeting, leading to reduced drug delivery efficiency and unsatisfactory treatment. Herein, we developed a phosphorylcholine-based zwitterionic copolymer coated ZIF-8 nanodrug (DOX@ZIF-8@P(MPC-co-C7A)), and the obtained nanodrug was preparedviaa charge-conversional zwitterionic copolymer coating on DOX@ZIF-8 composites. In this system, DOX was encapsulated in the framework of ZIF-8, which could reduce the drug leakage in the bloodstream. The phosphorylcholine-based zwitterionic copolymer effectively extended the blood circulation time, resulting in enhanced tumor accumulation of the nanodrug. Once the nanodrug reached the tumor site, the surface charge of the system could rapidly convert to positive, resulting in an enhanced tumor cellular uptake. Finally, in the acidic environment inside intracellular organelles, DOX will be released rapidly for chemotherapy owing to the fast disintegration of ZIF-8 frameworks. Therefore, the obtained nanodrug could effectively inhibit the growth of A549-bearing tumors (93.2% tumor inhibition rate) with negligible side effects. Overall, this work significantly improved the drug delivery efficiency of ZIF-8, which may pave the way for the biomedical applications of ZIF-8 crystals in anti-tumor drug delivery.

Abstract:Zeolitic imidazolate framework-8(ZIF-8) is a class of porous crystalline materials formed by the selfassembly of zinc ions and 2-methylimidazole. It shows potential advantages in encapsulation and transportation of functional materials owing to its high porosity, large specific surface,convenient synthesis and controllable size. More importantly, ZIF-8 is an ideal carrier for drug delivery and release due to its excellent biocompatibility, stability under physiological conditions and responsiveness to the weak acidic environment associated with malignant tumors and other diseases. In fact, the small molecular drugs (doxorubicin, 5-fluorouracil) and biological macromolecules(antibody, nuclein) have all been loaded into ZIF-8 for chemotherapy, photothermal therapy, photodynamic therapy and biosensing. The particle size of ZIF-8 is very important for biomedical applications, and accurate functional regulation of ZIF-8 is agreat challenge for its biomedical application. Herein, we review the synthesis methods, size-control and biomedicalapplications of ZIF-8.

Abstract:Breast cancer, one of the three most life-threatening cancers in modern times, must be explored for treatments with low side effects and practical efficacy. Metal organic framework materials (MOFs) is made by metal ions as the center for point and organic ligands as a bridge connecting a new type of porous nano-materials, among them, the zinc base zeolite imidazole skeleton material series (ZIFs) because of its excellent biocompatibility and pH slow controlled release ability, is widely used in the tumor microenvironment in basic research and achieved remarkable curative effect. Inspired by this, in this review, we focus on the recent research progress on the application of ZIFs in the treatment of breast cancer, mainly studying the structure of ZIFs such as ZIF-8, ZIF-90 and ZIF-67 and their application in novel therapies for breast cancer treatment, such as targeted drug delivery, photothermal therapy, immunotherapy and gene therapy.We will more fully demonstrate the potential of zif in breast cancer treatment, hoping to provide an avenue for exploring breast cancer treatment.

Abstract:Zeolitic imidazolate frameworks (ZIFs) have attracted great interest as pH-sensitive drug carrier because of high drug loading and intrinsic biodegradability. In this work, a biocompatible NIR and pH-responsive drug delivery nanoplatform based on ZIFs (PDA-PCM@ZIF-8/DOX) is synthesized for in vivo cancer therapy. The biocompatibility of ZIFs is greatly improved by polydopamine (PDA) modifying and proved by cytotoxicity and in vivo acute toxicity evaluation. The degradability is also regulated in an appropriate rate. Due to mild reaction condition of ZIFs, the synthesis and drug loading is achieved in one pot with high loading (37.86%) and encapsulation rate (78.76%). Meanwhile, PDA acts as a photothermal transfer agent to trigger thermal response switch of phase change materials for NIR controlled drug release. Under the dual stimulus of NIR and acid environment, the drug release is as high as 78%, while only 21% is released without stimulus, showing a remarkable effect of control release. In vivo anti-tumor experiments demonstrate the high tumor inhibition rate of photothermal-chemotherapy group with a significant synergistic effect. The biocompatible and biodegradable drug delivery platform based on ZIFs has shown great promise for future clinic cancer therapy.

Abstract:The rapid development of nanotechnology has generated numerous ideas for cancer treatment, and a wide variety of relevant nanoparticle platforms have been reported. Metal-organic frameworks (MOFs) have been widely investigated as an anti-cancer drug delivery vehicle owing to their unique porous hybrid structure, biocompatibility, structural tunability, and multi-functionality. MOF materials with catalytic activity, known as nanozymes, have applications in photodynamic and chemodynamic therapy. Nucleic acids have also attracted increasing research attention owing to their programmability, ease of synthesis, and versatility. A variety of functional DNAs and RNAs have been applied both therapeutically (gene-targeting drugs for cancer treatment) and nontherapeutically (used as modified materials to enhance the therapeutic effects of other nanomedicines). The combined use of MOFs and functional nucleic acids have been extensively investigated and has been associated with excellent tumor-suppressor activity in various treatment methods. In this review, we summarize the progress in the research and development of tumor therapy based on MOFs and nucleic acid delivery over recent years, focusing on the combinational use of different delivery and design strategies for MOF/therapeutic nucleic acid platforms. We further summarize the strategies for combining MOFs (universal carrier, functional carrier) and nucleic acids (therapeutic nucleic acids, nontherapeutic nucleic acids) and discuss the corresponding therapeutic effects in cancer treatment. Abstract:Controlled drug release is a promising approach for cancer therapy due to its merits of reduced systemic toxicity and enhanced antitumor efficacy. Here, multifunctional Fe₃O₄@carbon@zeolitic imidazolate framework-8 (FCZ) hybrid nanoparticles (NPs) were successfully constructed. Owing to the porosity and acid-sensitivity of zeolitic imidazolate framework-8 (ZIF-8), FCZ NPs not only displayed an improved drug loading capacity compared to most of the polymeric nanocarriers, but also exhibited excellent pH-triggered release of doxorubicin (DOX) in vitro. Moreover, carbon dots (CDs) embedded in the porous carbon shell and superparamagnetic iron oxide nanocrystals could simultaneously function as intracellular fluorescence imaging and T₂*-weighted magnetic resonance imaging (MRI) contrast agents, respectively. The results obtained from the MTT assay demonstrated good biocompatibility of FCZ NPs. DOX release experiments showed pH regulation-dominated drug release kinetics: a weak acidic pH in tumor areas could trigger sustained drug release, suggesting that FCZ NPs are ideal drug delivery systems. Moreover, the remarkable inhibition of tumor growth without side effects was confirmed in vivo. These results provide convincing evidence establishing the multifunctional FCZ NPs as promising candidates for tumor therapy.

Abstract:Nanoscaled metal organic frameworks have emerged as a novel type of organic-inorganic hybrid coordination polymeric nanomaterials and showed great promise in biomedical area in recent decades. As a special subclass, zeolitic imidazolate frameworks (ZIFs), constructing with metal ions and imidazolate linkers, own unique advantages of high porosity, large surface area, good biocompatibility, pH sensitivity and etc. These render ZIFs as attractive and promising platforms to delivery various cargos of drugs, imaging probes and biomacromolecules for cancer diagnosis and therapy. And this review specially summaries the state-of-the-art research on ZIFs for biosensing, cancer imaging and phototheranostics within the latest 5 years. The multiply strategies of synthesis, characterization and functionalization of ZIFsbased nanosystems are firstly discussed. The vital roles of ZIFs in biosensors, including electrochemical, fluorescent and colorimetric sensors are analyzed. Then, the monomodal and/or multimodal cancer imaging techniques mediated by ZIFs are addressed. Particular emphasis is placed on the ZIFs-based cancer phototheranostics which provide a potential alternative avenue to greatly improve the therapeutic index of cancer. Finally, the future opportunities and challenges of ZIFs are tentatively proposed and discussed from the point of translational medicine. (c) 2021 Elsevier Ltd. All rights reserved.
