Abstract:Nitro-compounds are one of the cheapest and most readily available materials in the chemical industry and are commonly utilized as versatile building blocks. Previously, the synthesis of N-heterocycles was largely based on anilines. The utilization of nitroarenes and nitroalkenes for the synthesis of N-heterocyclic compounds can save at least one step, however, as compared to anilines. Thus, considerable attention has been paid to nitroarenes and nitroalkenes as new potential amino sources. Significant progress has been made in the reductive cyclization of nitroarenes or nitroalkenes to access various N-heterocycles in recent years. Herein, we comprehensively summarize the recent progress in the construction of N-heterocycles using nitroarenes and nitroalkenes as potential amino sources. The compatibility of the reaction substrate, its mechanism, applications, advantages, and limitations in this field are also discussed in detail.

Abstract:This study sheds light on the cleavage and reorganization of C(sp(3))-H and C= N bonds of carbodiimides in a three-component reaction of terminal alkynes, sulfur, and carbodiimides by a combination of methods including 1) isolation and X-ray analysis of sixmembered- ring lithium species 2-S, 2) trapping of the oxygen-analogues (B-O and D-O) of both four-membered- ring intermediate B-S and ringopening intermediate D-S, 3) deuterium labeling studies, and 4) theoretical studies. These results show that 1) the reaction rate-determining step is [2+ 2] cycloaddition, 2) the C= N bond cleavage takes place before C(sp(3))-H bond cleavage, 3) the hydrogen attached to C6 in 2-S originates from the carbodiimide, and 4) three types of new azaheterocycles, such as 1,2-dihydrothiopyrimidines, N-acyl 2,3-dihydropyrimidinthiones, and 1,2-dihydropyrimidinamino acids are constructed efficiently based on 2-S. All results strongly support the idea that the reaction proceeds through [2+ 2] cycloaddition/4p electrocyclic ring-opening/1,5-H shift/6p electrocyclic ring-closing as key steps. The research strategy on the synthesis, isolation, and reactivity investigation of important intermediates in metal-mediated reactions not only helps achieve an in-depth understanding of reaction mechanisms but also leads to the discovery of new synthetically useful reactions based on the important intermediates.

Abstract:A novel pattern of the cleavage and reorganization of CN bond in the multicomponent reaction (MCR) of terminal alkynes or haloalkynes, carbodiimides, and benzynes is achieved for the first time to construct efficiently 2-aminoaryl alkynyl imines. The selective formation and ring-opening of the azetine intermediate with the high ring strain is essential for this reaction. Further transformation of 2-aminoaryl alkynyl imines via the Cu-catalyzed cycloisomerization is explored to provide steroselectively the bi-, tri-, and tetracyclic fused pyrrolines.

Abstract:The cascade [4+2] annulation/aromatization reactions between carbodiimide anions and α,β-unsaturated imines are developed. This is the first report on regulating N-Ts cyanamides to participate in reactions absolutely via carbodiimide anions rather than previously reported cyanamide anions, affording a convenient method for synthesizing 2-aminopyrimidines. More importantly, this work expands the application of carbodiimides, broadening the coupling substrate scope to electrophilic partners and providing new strategies to construct six-membered N-heteroaromatic scaffolds. A stepwise mechanism, supported by DFT calculations, is invoked to explain the reaction selectivity.

Abstract:The catalytic transformation of ubiquitous but inert C-N bonds is highly appealing in synthetic chemistry, but the efficient cleaving inert C-N bond and simultaneous incorporation of both the cleaved C-moiety and N-moiety into the desired products has been a long-standing formidable challenge so far. Here, we developed a radical-addition triggered cyclization and C-N bond cleavage process enabled by the unique I2/Ni or benzyl halide/Ni-catalytic system, allowing the formal insertion of diene into the inert C-N bond. This reaction features high atom economy and enables an expedient annulative carboamination of aminodienes to diverse pyrrolidines, piperidines, and tetrahydroisoquinolines. Mechanistic studies have revealed that the reaction is initiated via the generation of a benzyl radical and the formation of quaternary ammonium salt is key for the C-N bond cleavage.

Abstract:Cross-coupling azide and isocyanide have recently gained recognition as ideal methods for efficiently synthesizing unsymmetric carbodiimides. This reaction exhibits high reaction rates, efficiency, and favorable atom/step/redox economy. It enables the nitrene-transfer process, facilitating the formation of C-N bonds and providing a direct and cost-effective synthetic strategy for generating diverse carbodiimides. These carbodiimides are highly reactive compounds that can undergo in-situ transformations into various functional groups and organic compounds, including heterocycles. Developing one-pot and tandem processes in this field has significantly contributed to advancements in organic chemistry. Moreover, the demonstrated utility of these architectural motifs extends to areas such as chemical biology and medicinal chemistry, further highlighting their potential in various scientific applications.

Abstract:Bicyclic diaziridines are important heterocyclic scaffolds, which can be cleaved into 1,3‐zwiterionic species to serve as a versatile precursor in fabricating biologically important N‐hetero‐ and spiro‐cyclic structural frameworks. Owing to their staple architecture, inherent ring strain and ease of synthesis, considerable advancements have been made by employing bicyclic diaziridines towards the construction of heterocyclic systems of biological importance. A significant surge with appreciable efforts in the adoption of such strategies has led to their implementation by the synthetic chemists. This article focuses on the developments in ring expansion of bicyclic diaziridines employed in N‐heterocycle synthesis.

Abstract:The sustainable preparation of N‐heterocycles is one of the most active research areas owing to their predominance as synthetics building blocks with extensive applications in organic, pharmaceutical, and material chemistry fields. Among the various catalytic protocols, the C–H bond functionalization with the concomitant C–N bond formation, so‐called C–H bond annulation, has become one of the most sustainable routes to access N‐heterocycles because it starts from low‐functionalized materials and generates a limited amount of waste, all respecting the concept of atom economy. Rhodium complexes often catalyze these reactions. This review focuses on the synthesis of 5‐ and 6‐membered ring N‐containing heterocycles such as indoles, pyrroles, indolines, (iso)quinolinones, dihydroquinolines, and pyrrolidones from readily available starting materials, with an emphasis on the novel C–H bond cascade synthetic methodologies via C–N/C–C bond formation, as well as on the mechanisms of these reactions, especially the oxidation steps. We hope this review will help researchers looking to prepare N‐heterocycles in a minimum of steps and those who want to develop new methodologies based on C–H bond activation/functionalizations.

Abstract:The synthesis of N-heterocycles is one of the most active areas due to their structures with the extensive applications in the fields of organic, pharmaceutical and material chemistry. It is an increasing challenging task to develop greener, milder and cheaper N-heterocycles synthetic strategies through C–N/C–C bond formation owing to requiring multistep reactions /or harsh reaction conditions in many cases. Herein, this review focuses on the synthesis of N-containing heterocycles such as pyrimidines, imidazoles, 1,2,4-triazoles, 1,3,5-triazines, benzimidazoles, isoquinolines, quinazolines, quinazolinones, 1,2,4-oxadiazoles, thiazoles, and thiadiazoles using amidines as readily available starting materials, with an emphasis on the novel green synthetic methodologies via C–N/C–C bond formation, as well as the mechanisms of these reactions. We hope this review will help to give references for researchers who are fascinated with the increasing applications in this prolific area.

Abstract:Diazo compounds and organic azides are widely used as reagents for accessing valuable molecules in multiple areas of fundamental and applied chemistry. Their capacity to undergo versatile chemical transformations arises from the reactive nature of an incipient dinitrogen molecule at the terminal position. In this work, we report the synthesis and characterization of an N-heterocyclic carbene (NHC)–stabilized diazoborane—a boron-centered analog of organic azides and diazoalkanes. The diazoborane displays a strong tendency to release dinitrogen, thus serving as a borylene source, in analogy to organic azides and diazoalkanes serving as nitrene and carbene sources, respectively. Also reminiscent of diazoalkane and organic azide reactivity, the diazoborane serves as a 1,3-dipole that undergoes uncatalyzed [3+2] cycloaddition with an unactivated terminal alkyne, affording a five-membered heterocycle after a two-step rearrangement.

Abstract:Transition-metal-catalyzed, directed intermolecular C-H bond functionalization is synthetically useful but heavily underexplored in multiheteroatom heterocycle synthesis. Herein we report a cobalt catalytic method for the formation of a three-nitrogen-bearing benzotriazine scaffold via the coupling of arylhydrazine and oxadiazolone. This synthetic protocol features a low-cost base metal catalyst, a maximum number of heteroatoms built into a heterocycle, a distinct synthetic logic for benzotriazines, a superior step economy, and a broad substrate scope.

Abstract:Metal-catalyzed cyclization reactions have become a powerful and efficient approach for the stereoselective construction of both carbocyclic and heterocyclic ring systems. Transition metal complexes, with their ability to activate and selectively functionalize organic substrates, have revolutionized various areas of synthetic chemistry. This review highlights recent advancements in metal-catalyzed cyclization reactions, especially in the synthesis of nitrogen-containing heterocycles like imidazoles, pyridines, pyrimidines, and indoles. These advancements have significantly impacted fields such as natural product synthesis, pharmaceuticals, functional materials, and organic electronics. Novel catalytic systems, ligand designs, and reaction conditions continue to expand the capabilities of these reactions, driving further the progress made in synthetic organic chemistry. This review provides a comprehensive overview of recent research.

Abstract:Nitrogen- and sulfur-containing heterocyclic compounds widely existed in bioactive natural products and drug molecules with unique bioactivity. The synthesis of nitrogen-and sulfur-containing hybrids had always been a hot topic in the field of synthetic chemistry, and chemists had been committed to developing convenient and efficient strategies to synthesize these compounds. Pyridinium zwitterions with a unique reactivity played a significant role in the construction of nitrogen- and sulfur-containing heterocyclic compounds. In recent years, pyridinium zwitterions had been extensively used in cyclization reactions for the synthesis of novel heterocyclic compounds. This review summarized the recent advances of pyridinium 1,n-zwitterions (n = 4 and 5) participating in cyclization reactions and highlighted the applications of pyridinium 1, n-zwitterions (n = 4 and 5) in the efficient construction of heterocycles.

Abstract:A convenient Ni(II)-catalyzed C-C and C-N cascade coupling reaction was developed to directly access various 2,4-disubstituted imidazoles. The reaction scope covers a variety of aryl and aliphatic substitutions, which demonstrate moderate-to-excellent yields. The tolerance of halogen and N-containing heterocyclic groups demonstrates the versatility of this method for further synthetic explorations.

Abstract:While remarkable progress has recently been made for the direct C‐H‐functionalization of azines, its application is still limited by a lack of accessible functional groups (primarily carbon‐based) and poor regioselectivity. In contrast, C2‐functionalized pyridines and quinolines can be easily synthesized by treating readily available N‐oxides with various reagents under appropriate activation conditions. This review seeks to comprehensively document the available synthetic methods for introducing functional groups at the C2 position of pyridines and quinolines. In this work, we highlight recent developments in the C2‐functionalization of pyridine and quinoline N‐oxides and address both the mechanisms and regioselectivity of the reactions. We also describe the pathways and reactive species involved in these processes and highlight a number of medically relevant nitrogen heteroaromatics.
