Diosmetin modulates lipogenesis and alleviates inflammatory response in nonalcoholic steatohepatitis through STAT1/CXCL10-dependent pathway

Abstract:Nonalcoholic steatohepatitis (NASH) is an inflammatory lipotoxic disorder, characterized by lipid accumulation and inflammation. This study investigated the effects of the flavonoid diosmetin (Dios) on lipogenesis and inflammatory response and the underlying molecular mechanisms in NASH. Here, high-fat diet (HFD) and palmitic acid (PA) were used to induced NASH model in mice and HepG2 cells, respectively. After Dios supplementation, the functional parameters, morphological changes, lipogenesis and inflammatory response in HepG2 cells and NASH mice were reduced. The liver RNA-Seq analysis revealed the key targets CXCL10 and STAT1 interfered by Dios. Interestingly, Dios prevented CXCL10- and STAT1-mediated lipogenesis and inflammatory response through siCXCL10, STAT1 inhibitor and STAT1 overexpression in HepG2 cells. In conclusion, Dios alleviated NASH through suppression of lipogenesis and inflammatory response via STAT1/CXCL10-dependent pathway.
