Abstract:A facile hydrogel fabrication strategy based on a simultaneous dual-photo-coupling reaction (i.e., photoinduced S-nitrosylation and Schiff base reaction) was reported. This strategy allowed a strengthened three-arm crosslinking network to form in one step and the hydrogels obtained displayed rapid gelation, excellent mechanical strength and biocompatibility for cell encapsulated-3D printing in real time. Our hydrogel fabrication strategy will likely foster advances in biomaterials and the extreme speed and reinforced mechanical strength should significantly benefit 3D printing and related applications.

Abstract:Owing to flexible structural design and rapid prototyping manufacturing, 3D printable hydrogels with solid-like and fluid-like characters have emerged as a promising candidate for biomedical engineering, soft robotics, and flexible electronics. However, existing 3D printing of hydrogel place substantial constraints on hydrogel high-fidelity of the printed hydrogel complex structures due to the shape deformation and collapse caused by water evaporation. Herein, we propose a mixed solvent printing strategy followed with water dialysis to achieve vat photopolymerization 3D printing of high-fidelity hydrogel structure by restraining water evaporation. Concretely, dimethyl sulfoxide (DMSO) is introduced into the hydrogel ink as co-solvent to improve the anti-dehydration performance during 3D printing. The as-prepared gels are then transformed into hydrogels with good fidelity and excellent performance through solvent exchange and water dialysis. Enabled by these characteristics, various functional devices with complex structures are demonstrated with the present high-fidelity 3D printing of diverse hydrogels. The solvent assisted high-fidelity hydrogel 3D printing protocol combining with the compatibility to various high-performance hydrogels is believed to be promising for wearable devices, wound dressings, electronic skin, and intelligent robots.

Abstract:Three-dimensional (3D) hydrogel printing enables production of volumetric architectures containing desired structures using programmed automation processes. Our study reports a unique method of resolution enhancement purely relying on post-printing treatment of hydrogel constructs. By immersing a 3D-printed patterned hydrogel consisting of a hydrophilic polyionic polymer network in a solution of polyions of the opposite net charge, shrinking can rapidly occur resulting in various degrees of reduced dimensions comparing to the original pattern. This phenomenon, caused by complex coacervation and water expulsion, enables us to reduce linear dimensions of printed constructs while maintaining cytocompatible conditions in a cell type-dependent manner. We anticipate our shrinking printing technology to find widespread applications in promoting the current 3D printing capacities for generating higher-resolution hydrogel-based structures without necessarily having to involve complex hardware upgrades or other printing parameter alterations.

Abstract:The rapid development of additive manufacturing techniques in the field of tissue regeneration offers unprecedented success for artificial tissue and organ fabrication. However, some limitations still remain for current bioinks, such as the compromised cell viability after printing, the low cross-linking efficiency leading to poor printing resolution and speed due to the relatively slow gelation rate, and the requirement of external stimuli for gelation. To address these problems, herein, a biocompatible and printable instant gelation hydrogel system has been developed based on a designed hyperbranched poly(ethylene glycol) (PEG)-based multihydrazide macro-cross-linker (HB-PEG-HDZ) and an aldehyde-functionalized hyaluronic acid (HA-CHO). HB-PEG-HDZ is prepared by the postfunctionalization of hyperbranched PEG-based multivinyl macromer via thiol-ene chemistry. Owing to the high functional group density of HB-PEG-HDZ, the hydrogel can be formed instantly upon mixing the solutions of two components. The reversible cross-linking mechanism between the hydrazide and aldehyde groups endows the hydrogel with shear-thinning and self-healing properties. The minimally toxic components and cross-linking chemistry allow the resulting hydrogel to be a biocompatible niche. Moreover, the fast sol-to-gel transition of the hydrogel, combining all of the advanced characteristics of this platform, protects the cells during the printing procedure, avoids their damage during extrusion, and improves the transplanted cell survival.

Abstract:Composite hydrogels have gained great attention as three-dimensional (3D) printing biomaterials because of their enhanced intrinsic mechanical strength and bioactivity compared to pure hydrogels. In most conventional printing methods for composite hydrogels, particles are preloaded in ink before printing, which often reduces the printability of composite ink with little mechanical improvement due to poor particle-hydrogel interaction of physical mixing. In contrast, the in situ incorporation of nanoparticles into a hydrogel during 3D printing achieves uniform distribution of particles with remarkable mechanical reinforcement, while precursors dissolved in inks do not influence the printing process. Herein, we introduced a "printing in liquid" technique coupled with a hybridization process, which allows 3D freeform printing of nanoparticle-reinforced composite hydrogels. A viscoplastic matrix for this printing system provides not only support for printed hydrogel filaments but also chemical reactants to induce various reactions in printed objects for in situ modification. Nanocomposite hydrogel scaffolds were successfully fabricated through this 3D freeform printing of hyaluronic acid (HAc)-alginate (Alg) hydrogel inks through a two-step crosslinking strategy. The first ionic crosslinking of Alg provided structural stability during printing, while the secondary crosslinking of photo-curable HAc improved the mechanical and physiological stability of the nanocomposite hydrogels. For in situ precipitation during 3D printing, phosphate ions were dissolved in the hydrogel ink and calcium ions were added to the viscoplastic matrix. The composite hydrogels demonstrated a significant improvement in mechanical strength, biostability, as well as biological performance compared to pure HAc. Moreover, the multi-material printing of composites with different calcium phosphate contents was achieved by adjusting the ionic concentration of inks. Our method greatly accelerates the 3D printing of various functional or hybridized materials with complex geometries through the design and modification of printing materials coupled with in situ post-printing functionalization and hybridization in reactive viscoplastic matrices.

Abstract:3D printing hydrogels with a good resolution, diversified geometries, and a tunable mechanical performance is still a big challenge, which is mainly due to the low photo-reactivity of the high-water containing precursor and the low modulus of the photo-cured hydrogel. In the present work, an acryloylmorpholine - acrylic acid - PEG diacrylate drygel was firstly printed with a good fidelity and subsequently post- treated with an aqueous solution containing specific ions, during which hydration of the hydrophilic network and coordination between the carboxyl and ions occurred simultaneously. The obtained dual-crosslinked hydrogel combined a complex geometry and a good stretchability. This approach decoupled the printability and mechanical properties of the hydrogels. To furtherly show this advantage over conventional direct hydrogel 3D printing, ultrasoft hydrogel lattice structures which cannot be self-supported and purely ionic crosslinked hydrogels with a good self-weldability were demonstrated.

Abstract:Cephalopods can change their color and patterns by activating the skin chromatophores for camouflage. However, in the man-made soft material systems, it is greatly challenging to fabricate the color-change structure in the desired patterns and shapes. Herein, we employ a multi-material microgel direct ink writing (DIW) printing method to make mechanochromic double network hydrogels in arbitrary shapes. We prepare the microparticles by grinding the freeze-dried polyelectrolyte hydrogel and immobilize the microparticles in the precursor solution to produce the printing ink. The polyelectrolyte microgels contain mechanophores as the cross-linkers. We adjust the rheological and printing properties of the microgel ink by tailoring the grinding time of freeze-dried hydrogels and microgel concentration. The multi-material DIW 3D printing technique is utilized to fabricate various 3D hydrogel structures which could change into a colorful pattern in response to applied force. The microgel printing strategy shows great potential in the fabrication of the mechanochromic device with arbitrary patterns and shapes.

Abstract:Although 3D printing can easily control the macroscopic structure of printed subjects based on the digital modeling, the control of micro-structure of polymer network was still a challenge. Herein, we report the using of macroinitiator induced rapid photoATRP to regulate the 3D printed hydrogel network. This approach consists of synthesizing of macroinitiator through CDs catalyzed photoATRP, and photocuring in the same pot by subsequent adding of crosslinker. By varying the chain length of the macroinitiator, we demonstrated the effect of physical entanglements of on the formation of hydrogel, as well as the mechanical property and swelling ratio. All the 3D printing objects with different macroinitiators exhibited outstanding print resolution. With PNIPAM macroinitiator, the printed hydrogel shows the characteristics of temperature-sensitive response. Overall, the findings of this work enable an interesting platform for control the mechanical property and functional of 3D printing hydrogel.

Abstract:A systematic formation theory of photocurable bio‐hydrogels is established, and the formation process of hydrogels is clarified. Based on the theory, the photocrosslinking behavior of hydrogels is predicted accurately. Besides, an original projection‐based 3D printing (PBP) strategy is proposed, which significantly improves the printing resolution, printing efficiency, mechanical properties, and biocompatibility. Bio‐ink has gradually transited from ionic‐crosslinking to photocrosslinking due to photocurable bio‐hydrogel having good formability and biocompatibility. It is very important to understand and quantify the crosslinking process of photocurable hydrogels, otherwise, bioprinting cannot be standardized and scalable. However, there are few studies on hydrogel formation process and its photocrosslinking behavior which cannot be accurately predicted. Herein, the photoinitiated radical polymerized bio‐hydrogels are taken as an example to establish the formation theory. Three typical crosslinking reactions are first distinguished. It is further proposed that not all double‐bonds consumed during crosslinking contributeequally to polymerization. Then the concept of effective double‐bond conversion (EDBC) is elicited. Deriving from EDBC, several important formation indices are defined. According to theory, it is predicted that slow crosslinking can improve the crosslinking degree. Furthermore, based on the slow crosslinking effect, a new strategy of projection‐based 3D printing (PBP) is proposed, which significantly improved printing quality and efficiency. Overall, this work will fill the gap in hydrogel's formation theory, making it possible to accurately quantify the formation process.

Abstract:To mimic biological tissues with high toughness such as cartilage, it is highly desired to fabricate stable and tough hydrogels with intricate shapes to act as a structural support. Extrusion-based 3D printing is a promising method to fabricate 3D scaffolds with various architectures; however, printing tough hydrogel structures with high fidelity and resolution is still a challenge. In this work, we adopt the fast sol-to-gel transition of κ-carrageenan in the solution of acrylamide upon cooling to fix the printed scaffolds and polymerize the precursor solution to form the second network. The printed constructs of κ-carrageenan/polyacrylamide double-network gels are toughened by soaking in an aqueous solution of zirconyl chloride to form coordination complexes between the Zr4+ ions and sulfate groups of κ-carrageenan. The obtained hydrogels are stable in water and possess good mechanical properties, with a tensile breaking stress of 1-2 MPa, breaking strain of 100-150%, and Young's modulus of 4-10 MPa. The printed grid can hold 150 times its own weight. 3D printed constructs with a high aspect ratio and shape fidelity are obtained by optimizing the printing parameters. Furthermore, a biomimetic strategy is applied to construct a hydrogel composite by filling the printed tough hydrogel scaffold with a cell-laden fibrin hydrogel as the soft substance. Chondrocytes in the hydrogel composite maintain high viability after cyclic compression, demonstrating the load-bearing capacity of the tough scaffold and favorable microenvironment for cells provided by the embedded soft fibrin gel. We envision that this printing strategy for hydrogel constructs with high toughness and good stability, as well as the method to form tough-soft hydrogel composites, can be extended to other systems to develop structural elements and scaffolds towards applications in biomedical devices and tissue engineering.

Built upon capillary origami, well-controlled hydrogel structures with programmable shapes and sizes are produced by crosslinking during folding process. The profiles of the structure are dependent on crosslinking time, properties of the membrane and liquid, and the membrane shape. This method holds great potential to be high-throughput and applicable to various materials.

Abstract:3D printing technology has been widely explored for the rapid design and fabrication of hydrogels, as required by complicated soft structures and devices. Here, a new 3D printing method is presented based on the rheology modifier of Carbomer for direct ink writing of various functional hydrogels. Carbomer is shown to be highly efficient in providing ideal rheological behaviors for multifunctional hydrogel inks, including double network hydrogels, magnetic hydrogels, temperature-sensitive hydrogels, and biogels, with a low dosage (at least 0.5% w/v) recorded. Besides the excellent printing performance, mechanical behaviors, and biocompatibility, the 3D printed multifunctional hydrogels enable various soft devices, including loadable webs, soft robots, 4D printed leaves, and hydrogel Petri dishes. Moreover, with its unprecedented capability, the Carbomer-based 3D printing method opens new avenues for bioprinting manufacturing and integrated hydrogel devices.

Abstract:3D printing of hydrogels with high intrinsic mechanical performance has significant applications in many fields yet has been proven to be a fundamental challenge. Here, 3D printing of ultrahigh strength hydrogels is achieved by constructing cross-linkingDPC networks based on poly(vinyl alcohol) (PVA) and chitosan (CS). The hybrid ink with moderate rheology for direct ink writing is employed to manufacture complex hydrogel structures, first. Then, the cyclic freezing–thawing followed by sodium citrate solution soaking realize the first network of PVA crystallization and the second one of CS ionic interaction between amino and carboxyl groups. The optimized DPC hydrogel displays a tensile strength of 12.71 ± 1.32 MPa at a strain of 302.27 ± 15.70%, Young's modulus of 14.01 ± 1.35 MPa, and work of extension at fracture Wext of 22.10 ± 2.36 MJ m–3 because of the dominant energy dissipation of the stiff CS ionic network. Moreover, the tearing test supports that this DPC hydrogel possesses a high toughness of 9.92 ± 1.05 kJ m–2. This protocol can readily realize not only the hydrogel lattice, honeycomb, and spring, but also secondary-shaping hydrogel objects including whale, octopus, and butterfly via a local DPC strategy. Abstract:Recent decades have seen intense developments of hydrogel applications for cell cultures, tissue engineering, soft robotics, and ionic devices. Advanced fabrication techniques for hydrogel structures are being developed to meet user-specified requirements. Existing hydrogel 3D printing techniques place substantial constraints on the physical and chemical properties of hydrogel precursors as well as the printed hydrogel structures. This study proposes a novel method for patterning liquids with a resolution of 100 μm by using the capacitor edge effect. We establish a complete hydrogel 3D printing system combining the patterning and stacking processes. This technique is applicable to a wide variety of hydrogels, overcoming the limitations of existing techniques. We demonstrate printed hydrogel structures including a hydrogel scaffold, a hydrogel composite that responds sensitively to temperature, and an ionic high-integrity hydrogel display device. The proposed technique offers great opportunities in rapid prototyping hydrogel devices using multiple compositions and complex geometries.

Abstract:Printing of biologically functional constructs is significant for applications in tissue engineering and regenerative medicine. Designing bio‐inks remains remarkably challenging due to the multifaceted requirements in terms of the physical, chemical and biochemical properties of the three‐dimensional matrix, such as cytocompatibility, printability and shape fidelity. In order to promote matrix and materials stiffness, while not sacrificing stress relaxation mechanisms which support cell spreading, migration and differentiation, we report an interpenetrating network bio‐ink design. Our approach makes use of a chemically defined network, combining physical and chemical crosslinking unites with a tunable composition and network density, as well as spatiotemporal control over post‐assembly material stiffening. To this end, star‐shaped polyethylene glycols functionalized with Phe‐Gly‐Gly tripeptide or photoactive stilbazolium are synthesized, and used to prepare three‐dimensional networks with cucurbit[8]uril (CB[8]) through supramolecular host‐guest‐complexation. The hydrogel obtained shows fast relaxation and thus supports the proliferation and differentiation of cells. Upon irradiation, the mechanical properties of the hydrogel can be rapidly adapted via selective photochemical dimerization of stilbazolium within CB[8], leading to interpenetrating networks with increased form stability while retaining the dynamic nature of the hydrogels. This modular approach opens new design opportunities for extrudable and cell‐friendly dynamic biomaterials for applications in 3D‐bioprinting. This article is protected by copyright.
