Abstract:Transition metal oxide (TMO)-based nanomaterials are effectively utilized to fabricate clinically useful ultra-sensitive sensors. Different nanostructured nanomaterials of TMO have attracted a lot of interest from researchers for diverse applications. Herein, we utilized a hydrothermal method to develop porous nanosheets of cobalt oxide. This synthesis method is simple and low temperature-based. The morphology of the porous nanosheets like cobalt oxide was investigated in detail using FESEM and TEM. The morphological investigation confirmed the successful formation of the porous nanosheet-like nanostructure. The crystal characteristic of porous cobalt oxide nanosheets was evaluated by XRD analysis, which confirmed the crystallinity of as-synthesized cobalt oxide nanosheets. The uric acid sensor fabrication involves the fixing of porous cobalt oxide nanosheets onto the GCE (glassy carbon electrode). The non-enzymatic electrochemical sensing was measured using CV and DPV analysis. The application of DPV technique during electrochemical testing for uric acid resulted in ultra-high sensitivity (3566.5 µAmM-1cm-2), which is ~7.58 times better than CV-based sensitivity (470.4 µAmM-1cm-2). Additionally, uric acid sensors were tested for their selectivity and storage ability. The applicability of the uric acid sensors was tested in the serum sample through standard addition and recovery of known uric acid concentration. This ultrasensitive nature of porous cobalt oxide nanosheets could be utilized to realize the sensing of other biomolecules.

Abstract:Porous anodic alumina (PAA) membranes have attracted considerable interest in biosensor research because they are simple to prepare and the size of the nanochannels can be controlled. However, traditional methods only utilize the porous layer as the barrier layer is removed before use. In this study, we propose a new biosensor for the detection of uric acid based on ion channels in the barrier layer of a PAA membrane. This exhibits a wide linear range (1 nM-0.1 mM) and has excellent selectivity in the presence of interferents (D-(+)-glucose, L-ascorbic acid or dopamine). It is hoped that this new biosensor will prove useful in biological research and clinical medicine.
Palladium-Coated Submicron Electrospun Polymeric Fibers with Immobilized Uricase for Uric Acid Determination in Body Fluids

Abstract:This work describes the development of a flexible uric acid (UA) biosensor based on palladium-coated submicrometer electrospun poly(methylmethacrylate) (PMMA) fibers metalized with gold and attached to polyethylene terephthalate substrate (Pd/Au/PMMA/PET). The morphological characterization conducted by scanning electron microscopy revealed nanoscale Pd dendritic structures. Electrochemical investigations in the absence and in the presence of redox probes demonstrated that these Pd nanostructures are responsible for a six-fold increase in the electroactive area and enhanced electron transfer kinetics when compared to the gold-coated electrospun fibers. The UA biosensor obtained by immobilizing the uricase enzyme (UrOx) onto the Pd/Au/PMMA/PET electrode surface, allowed UA detection with a sensitivity of 431 μA cm–2 mM–1 and a limit of detection of 12 μM. Investigation of the redox reactions of hydrogen peroxide (a product of the enzymatic oxidation of UA by UrOx) at the Pd/Au/PMMA/PET electrode demonstrated that the working principle of the biosensor is based on the reduction of PdO produced at the electrode surface during the spontaneous reduction of hydrogen peroxide on Pd. This allows a biosensor operating potential of −0.05 V (vs Ag/AgCl) with high selectivity. The UrOx/Pd/Au/PMMA/PET biosensor was applied for UA detection in body fluids (sweat, urine, and blood serum) with recovery values between 98 and 105%, which were validated by high-performance liquid chromatography analysis. The stability of the device was evaluated over a period of 3 months, retaining 78% of the initial sensitivity, and reproducibility with RSD = 4.9% was achieved. The analytical performance of the biosensor under harsh mechanical deformations and at physiological temperatures demonstrated the potential applications of the device to wearable sensing platforms.

Abstract:In this work, we report the development of a highly sensitive and stable uric acid sensor based on the synergic action of multiwalled carbon nanotubes (MWNTs) and ZnO nanoparticles. MWNTs were first cast on pyrolytic graphite (PG) wafers. ZnO nanoparticles were then decorated onto the negatively charged MWNTs via the Vapor Liquid Solid (VLS) growth. Uricase was immobilized on the ZnO nanoparticles surface because of their large differences in the isoelectric point (IEP). Last, a cationic polydiallyldimethylammonium chloride (PDDA) layer was coated onto the uricase-contained ZnO nanoparticle layer and resulted in the PDDA/uricase/ZnO/MWNTs multilayer structure. The unique multilayer structure provides a favorable microenvironment to keep the bioactivity of uricase, which led to rapid amperometric response toward uric acid. Amperometric detection of uric acid was carried out at 320mV (vs. SCE) in 0.05mol/L (M) phosphate buffer solution (pH 6.8). For the sensor, a wide linear response range of 5.0M to 1mM with a linear sensitivity of 393mA cm-2M-1, a detection limit of 2.0M (3), and a long-term stability of 160 days can be obtained by using differential pulse voltammetry (DPV). Testing results in human urine obtained from the sensors were also compared with the data obtained by spectrometry. For five samples with different concentrations of urine, the relative deviations between them were smaller than 3.8%. The recovery was between 96.5 and 104.0%.

Abstract:We describe the use of individual zinc oxide (ZnO) micro/nanowires in an electrochemical biosensor for uric acid. The wires were synthesized by chemical vapor deposition and possess uniform morphology and high crystallinity as revealed by scanning electron microscopy, X-ray diffraction, and photoluminescence studies. The enzyme uricase was then immobilized on the surface of the ZnO micro/nanowires by physical adsorption, and this was proven by Raman spectroscopy and fluorescence microscopy. The resulting uric acid biosensor undergoes fast electron transfer between the active site of the enzyme and the surface of the electrode. It displays high sensitivity (89.74 μA cm−2 mM−1) and a wide linear analytical range (between 0.1 mM and 0.59 mM concentrations of uric acid). This study also demonstrates the potential of the use of individual ZnO micro/nanowires for the construction of highly sensitive nano-sized biosensors.

Abstract:Abstract A uric acid (UA) electrochemical biosensor was constructed using ferrocene (Fc) decorated cuprous oxide (Cu 2 O) enhanced electro-active characteristics and covalently immobilized with uricase (UOx) on glassy carbon electrode (GCE). The electrochemical characteristics of the fabricated electrode was analysed by cyclic voltammetry, electrochemical impedance spectroscopy and differential pulse voltammetry (DPV). DPV studies revealed rapid response of fabricated electrode UOx/Fc/Cu 2 O/GCE towards UA in a wide concentration range of 0.1–1,000 μM with a sensitivity of 1.900 μA mM −1 cm −2 and very low detection limit of 0.0596 μM. A very low magnitude Michaelis–Menten constant (Km) value was evaluated as 34.7351 μM which indicated the chemical attraction of the enzyme towards the UA was much higher. The developed biosensor was successfully applied to detect UA in human urine samples. Moreover, reproducibility and stability studies demonstrated the fabricated UOx/Fc/Cu 2 O/GCE biosensor had high reproducibility with a RSD of 2.8% and good reusability with a RSD of 3.2%. Specificity studies results showed the fabricated biosensor had strong anti-interference ability. The improved sensor performance was attributed to the synergistic electronic properties of Cu 2 O and Fc that provided enhances delectrocatalytic activity and electron transfer. The present biosensor can be extended for use in clinical settings.

Abstract:An ultrasensitive electrochemical biosensor was developed to identify the low levels of uric acid (UA) in human serum. The gold/cobalt (Au/Co) bimetallic nanoparticles (NPs) decorated hollow nanoporous carbon framework (Au/Co@HNCF) was synthesized as a nanozyme by pyrolysis of the Au (III)-etching zeolitic imidazolate framework-67 (ZIF-67). The external Au NPs combined with internal Co NPs on the hollow carbon framework exhibited enhanced activity for UA oxidation, thereby generating superior signals. Accordingly, the Au/Co@HNCF biosensor presented ranking performances with a low detection limit of 0.023 mu M (S/N = 3), an ultrahigh sensitivity of 48.4 mu A mu M-1 cm(-2), and an extensive response in the linear region of 0.1-25 mu M and the logarithmic region of 25-2500 mu M. Owing to the ordered nanoporous framework and carbon interfacial features, the Au/Co@HNCF biosensor displayed adequate selectivity for UA sensing over a series of biomolecules. In addition, the Au/Co@HNCF biosensor was employed to quantify UA in human serum samples. The test results were basically consistent with those of a commercial apparatus, and thus demonstrated that the proposed Au/Co@HNCF biosensor was reliable for UA determination in clinical research.

Abstract:A new amperometric uric acid biosensor developed by immobilizing of uric acid oxidase in carbon electrodes, in which, potassium ferricyanide is used to electron medium of chemistry response. The uric acid measured is oxidized and potassium ferricyanide is deacidized on the biosensor through catalysis of uric acid oxidase and supply of stationary 0. 3 volt. As a result, the peak of oxidation-reduction is produced on the biosensor. The concentration of uric acid in product is measured indirectly by amperometric. And the range of measuring uric acid is about 1 mg/dL-20 mg/dL. It has better linear for the results and fit coefficient arrives at 0. 9723. It reaches to steady value of currents in 10 seconds. At the same time, the interference of easily oxidizable species in most sample, such as ascorbic acid and so on, can be minimized at the biosensor in glucose measurements.

Abstract:Recently, photoelectrochemical (PEC)biosensing has received considerable attention with wide applications in biotechnology and biomedicine. Here we report a novel PEC biosensing platform constructed via the combined strategy of both PEC analysis and bioetching. Urchin-like ZnO-Au@CdS microspheres were obtained by depositing Au nanoparticles (NPs) and CdS NPs on the surface of urchin-like ZnO micro spheres. The introduction of intermediate electrotransfer of Au NPs to isolated ZnO and CdS systems significantly enhances the PEC property. CdS NPs can be irreversibly bioetched by an enzymatic reaction catalyzed by horseradish peroxidase (HRP) and H2O2. As a result, H2O2 detection is realized based on the decrease of the intensity of the PEC signal. This mediator-free and interference-free sensing platform has a linear range from 0.15 to 100 mu M with a detection limit of 0.04 OA H2O2 at S/N = 3. Furthermore, glucose oxidase (GOD) was successfully immobilized on ZnO-Au@CdS microspheres to construct a glucose biosensor with a detection limit of 0.18 OA at S/N = 3. The PEC biosensor shows good sensing performance with low detection limit, mediator-free and accuracy, strong anti-interference, promising its application in PEC biosensing. (C) 2017 Elsevier B.V. Abstract:A reagentless uric acid (UA) biosensor based on uricase immobilized on ZnO nanorods was developed. Direct electrochemistry and thermal stability of immobilized uricase were studied. The ZnO nanorods derived electrode retained the enzyme bioactivity and could enhance the electron transfer between the enzyme and the electrode. This sensor showed a high thermal stability up to 85°C and an electrocatalytic activity to the oxidation of uric acid without the presence of an electron mediator. The electrocatalytic response showed a linear dependence on the uric acid concentration ranging from 5.0 × 10−6 to 1.0 × 10−3molL−1 with a detection limit of 2.0 × 10−6molL−1 at 3σ. The apparent KMapp value for the uric acid sensor was estimated to be 0.238mM, showing a high affinity.

Abstract:Background: Uric acid (UA) is an important metabolic intermediate of the human body. Abnormally high levels of UA will cause diseases. However, UA monitoring with commercial products relies on invasive blood collection, which not only causes pain in patients but also risks bacterial infections and skin irritation. In recent years, new models of noninvasive detection through body surface penetration have raised higher expectations for the sensitivity of uric acid detection, and rapid, accurate and highly sensitive UA sensors will become powerful tools for the diagnosis of UA-related diseases. Objective: This study aimed to identify the differences in catalytic efficiency between regular PB from spray crystallization (RPB) and irregular PB from electrodeposition (EDPB), which is used for fabricate a high sensitive uric acid sensor. Methods: Regular Prussian blue nanocrystals (RPB) were grown on graphene oxide flakes (GO), on the surface of a custom screen-printed carbon electrode (SPCE), using a spray method assisted by a constant magnetic field (CMF). After immobilizing uricase, the uric acid biosensor Uricase/RPB/CMF-GO/SPCE was obtained. Results: The detection range of the sensor response to UA was 0.005 similar to 2.525 mM, and the detection limit was as low as 3.6 mu M. The cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS) results showed that compared to amorphous electrodeposited Prussian blue (EDPB), RPB more favorably accelerated electron transport. Conclusion: This novel uric acid biosensor exhibits high sensitivity over a wide concentration range, strong anti-interference ability, and good stability and reproducibility. Thus, it has good application prospects for determining uric acid in physiological samples.
One-pot Preparation of Uricase–poly(thiophene-3-boronic Acid)–ptnano Composites for High-Performance Amperometric Biosensing of Uric Acid

Abstract:Chemical oxidation of glycoprotein-bound thiophene-3-boronic acid (TBA) monomer by relatively mild oxidant chloroplatinate in neutral aqueous solution containing uricase yielded uricase–PTBA–Ptnano bionanocomposite with high enzymatic specific activity on a platinized (Ptplate) Au electrode, which was then covered by an outer-layer chitosan (CS) film to fabricate a CS/uricase–PTBA–Ptnano/Ptplate/Au electrode. This electrode exhibited a linear amperometric response to uric acid concentration from 5μM to 1.2mM with a sensitivity of 134μAmM−1cm−2, a limit of detection (S/N=3) of 1μM, and excellent operation/storage stability, which also worked well in serum samples.

Abstract:Fe3O4@Au nanomaterials were prepared by the self‐assembly method. An enzyme‐free, ultrasensitive electrochemical detection of uric acid was achieved based on the peroxidase‐like activity of Fe3O4@Au. The proposed procedure has exhibited excellent catalytic activities and achieved significant enhancements of the current responses to uric acid. The results showed that uric acid showed a good linear relationship in the range of 0.1~10 mmol/L, and the limit of detection was 0.087 μmol/L. Moreover, the selectivity of this method was also acceptable, and the established approach was successfully applied in the detection of uric acid in real samples.

Abstract:This work develops and validates an electrochemical approach for uric acid (UA) determinations in both endogenous (cell lysate) and physiological (serum) samples. This approach is based on the electrocatalytic reduction of enzymatically generated H2O2 at the biosensor of uricase-thionine-single-walled carbon nanotube/glassy carbon (UOx-Th-SWNTs/GC) with the use of Th-SWNTs nanostructure as a mediator and an enzyme immobilization matrix. The biosensor, which was fabricated by immobilizing UOx on the surface of Th-SWNTs, exhibited a rapid response (ca. 2 s), a low detection limit (0.5 +/- 0.05 mu M), a wide linear range (2 mu M to 2 mM), high sensitivity (similar to 90 mu A mM(-1) cm(-2)), as well as good stability and repeatability. In addition, the common interfering species, such as ascorbic acid, 3,4-dihydroxyphenylacetic acid, 4-acetamidophenol, etc., did not cause any interference due to the use of a low operating potential (-400 mV vs saturated calomel elect-rode). Therefore, this work has demonstrated a simple and effective sensing platform for selective detection of UA in the physiological levels. In particular, the developed approach could be very important and useful to determine the relative role of endogenous and physiological UA in various conditions such as hypertension and cardiovascular disease.
A Nanostructured Sensor Based on Gold Nanoparticles and Nafion for Determination of Uric Acid

Abstract:The paper discusses the mechanism of uric acid (UA) electrooxidation occurring on the surface of gold nanoparticles. It has been shown that the electrode process is purely electrochemical, uncomplicated with catalytic stages. The nanoeffects observed as the reduction of overvoltage and increased current of UA oxidation have been described. These nanoeffects are determined by the size of particles and do not depend on the method of particle preparation (citrate and "green" synthesis). The findings of these studies have been used to select a modifier for carbon screen-printed electrode (CSPE). It has been stated that CSPE modified with gold nanoparticles (5 nm) and 2.5% Nafion (Nf) may serve as non-enzymatic sensor for UA determination. The combination of the properties of nanoparticles and Nafion as a molecular sieve at the selected pH 5 phosphate buffer solution has significantly improved the resolution of the sensor compared to unmodified CSPE. A nanostructured sensor has demonstrated good selectivity in determining UA in the presence of ascorbic acid. The detection limit of UA is 0.25 μM. A linear calibration curve has been obtained over a range of 0.5-600 μM. The 2.5%Nf/Au(5nm)/CSPE has been successfully applied to determining UA in blood serum and milk samples. The accuracy and reliability of the obtained results have been confirmed by a good correlation with the enzymatic spectrophotometric analysis (R² = 0.9938) and the "added-found" technique (recovery close to 100%).

Abstract:The uricase-gold nanoparticles (AuNPs) composite nanomaterial was covalently immobilized on a biofilm, eggshell membrane. The uric acid biosensor was constructed by positioning the immobilized biofilm on the surface of a Clark dissolved oxygen electrode. The scanning electron micrograph revealed that the uricase-AuNPs composite nanomaterial was successfully immobilized. The effects of enzyme loading, pH, concentration of phosphate buffer and temperature on the biosensor response have been studied in detail. The results showed that response time of the biosensor was 60 s, the linear range for the detection of uric acid varied from 1.0 µM to ~1.0 mM with a detection limit of 0.8 µM ( S / N = 3) and relative standard deviation of 2.6% for 0.1 mM uric acid ( n = 6). Some common potential interferents in samples demonstrated no interferences. The biosensor has been successfully applied to determine the uric acid level in urine and serum samples.

Abstract:A uric acid biosensor has been fabricated through the immobilization of uricase on glassy carbon electrode modified by biomimetic titanate nanotubes of high specific surface area synthesized by hydrothermal decomposition. The so-constructed biosensor presents a high affinity to uric acid with a small apparent Michaelis-Menten constant of only 0.66 mM. The biosensor exhibits fairly good electrochemical properties such as the high sensitivity of 184.3 microAcm(-2)mM(-1), the fast response of less than 2 s, as well as the wide linear range from 1 microM to 5 mM. These performances indicate that titanate nanotubes could provide a favorable microenvironment for uricase immobilization, stabilize its biological activity, and function as an efficient electron conducting tunnel to facilitate the electron transfer. This suggests an important potential of titanate nanotubes in uric acid biosensors.
Electrochemical Sensor Using Cobalt Oxide-Modified Porous Carbon for Uric Acid Determination.

Abstract:An electrochemical sensor for the detection of uric acid was constructed using cobalt oxide-modified porous carbon@multi-walled carbon nanotube (MWCNTs) composite material for the modification of the electrode. Firstly, ZIF-67 is generated on carbon nanotubes using the surfactant cetylammonium bromide (CTAB) as template. The vesicles generated by CTAB act as nucleation sites for the in situ growth of ZIF-67. Then, cobalt oxide-modified porous carbon was obtained after high-temperature carbonization of ZIF-67, leading to the formation of cobalt oxide-modified porous carbon@MWCNT composite materials. Co-N and Co-O active sites on the composite material can improve the oxidation of uric acid on the electrode surface, leading to enhanced sensitivity and selectivity for uric acid detection. The sensor has a good linear range from 1 to 40 μM for uric acid detection with a detection limit of 0.09 μM. The sensor was utilized for determination of uric acid in actual serum samples.

Abstract:A uric acid biosensor based on the direct electron transfer of a hemoglobin-encapsulated chitosan-modified glassy carbon electrode was developed for a highly sensitive and selective analysis in urine samples. The modified electrode was prepared by the encapsulation of hemoglobin and uricase in a chitosan matrix. The hydrogen peroxide produced from the catalytic oxidation of uric acid by uricase was reduced electrocatalytically by immobilized hemoglobin and used to obtain a sensitive amperometric response to uric acid. The linear response of the uric acid concentrations ranged from 2.00 to 30.0 µM with a correlation of 0.9982, the detection limit of uric acid was estimated to be 0.85 µM at a signal/noise ratio of 3. The uric acid biosensor can efficiently exclude the interference of commonly coexisted ascorbic acid, dopamine, epinephrine, etc . The relative standard deviation was under 2.56% ( n = 5) for the determination of real samples. This biosensor is satisfactory for the determination of human urine samples compared with the HPLC-UV method.

Abstract:Uric acid (UA) is a kind of purine metabolism product and important in clinical diagnosis. In this work, we present a study of ZnS nanostructures-based electrochemical and photoelectrochemical biosensors for UA detection. Through a simple hydrothermal method and varying the ratio of reaction solvents, we obtained ZnS nanomaterials of one-dimensional to three-dimensional morphologies and they were characterized using field emission scanning electron microscopy (FESEM) and X-ray diffraction (XRD). To fabricate the UA biosensor and study the effect of material morphology on its performance, ZnS nanomaterials were deposited on indium tin oxide (ITO) conducting glass and then coated with uricase by physical absorption. Three kinds of working electrodes were characterized by cyclic voltammetry method. The effect of material morphology on performance of UA detection was investigated via amperometric response based electrochemical method based on enzymatic reaction. The ZnS urchin-like nanostructures electrode shows better sensitivity compared with those made of nanoparticles and nanoflakes because of its high surface-area-to-volume ratio. The photoelectrochemical method for detection of UA was also studied. The sensitivity was increased 5 times after irradiation of 300 nm UV light. These results indicate that ZnS nanostructures are good candidate materials for developing enzyme-based UA biosensors.
