Abstract:Copper-catalyzed radical transformations establish a powerful toolkit to construct versatile complex organic compounds. The copper-mediated bond formation step of radicals plays a critical role in controlling chemo- and stereoselectivity of copper-catalyzed radical transformation reactions. This bond formation step involves three possible pathways: ion-type bond formation, radical substitution, and reductive elimination. This review highlights the recent advances in theoretical studies on mechanisms and controlling models on the selectivity of Cu-mediated radical-involved bond formation, providing a general mechanistic comprehension of this key elementary step in copper catalysis.

Abstract:Radicals, with their unpaired electrons, exhibit unique chemical and physical properties that have long intrigued chemists. Despite early skepticism about their stability, the discovery of persistent radicals has opened new possibilities for molecular interactions. This review examines the mechanisms and applications of radically driven self-assembly, focusing on key motifs such as naphthalene diimides, tetrathiafulvalenes, and viologens, which serve as models for radical assembly. The potential of radical interactions in the development of artificial molecular machines (AMMs) are also discussed. These AMMs, powered by radical-radical interactions, represent significant advancements in non-equilibrium chemistry, mimicking the functionalities of biological systems. From molecular switches to ratchets and pumps, the versatility and unique properties of radically powered AMMs are highlighted. Additionally, the applications of radical assembly in materials science are explored, particularly in creating smart materials with redox-responsive properties. The review concludes by comparing AMMs to biological molecular machines, offering insights into future directions. This overview underscores the impact of radical chemistry on molecular assembly and its promising applications in both synthetic and biological systems.

Abstract:Minisci-type reactions have been widely known as reactions that involve the addition of carbon-centered radicals to basic heteroarenes followed by formal hydrogen atom loss. While the originally developed protocols for radical generation remain in active use today, in recent years, the new array of radical generation strategies have allowed the use of a wider variety of radical precursors that often operate under milder and more benign conditions. New transformations based on free radical reactivity are now available to a synthetic chemist, to utilize a Minisci-type reaction. Radical-generation methods based on photoredox catalysis and electrochemistry, which utilize thermal cleavage or the in situ generation of reactive radical precursors, have become popular approaches. Our review will cover the remarkable literature that has been reported on this topic in recent 5 years, from 2015-01 to 2020-01, in an attempt to provide guidance to the synthetic chemist on both the challenges that need to be overcome and the applications in organic synthesis.

Abstract:Propargylic alcohols are regarded as a class of readily available and versatile synthons in organic synthesis. Due to their unique bifunctional character, the transformation of propargylic alcohols has been widely utilized as a powerful strategy for the construction of various functionalized frameworks over the past few decades. Recently, the radical transformation of propargylic alcohols has attracted considerable attention in synthetic chemistry. This review summarises recent progress in the radical transformation of propargylic alcohols. Based on the different mechanisms of radical formation underlying these transformations, this review has been divided into four parts: (1) transition‐metal catalyzed radical transformation; (2) photo‐induced radical transformation; (3) electroc‐induced radical transformation; and (4) metal‐free or oxidant‐mediated radical transformation of propargylic alcohols.

Abstract:Pyridinium salts are valuable building blocks, which have been widely applied in various organic transformations during the past few decades. In particular, N-functionalized pyridinium salts have been explored as convenient radical precursors, which would go through reductive single-electron transfer. As a result, the chemistry of such pyridinium compounds for generating carbon-, nitrogen-, and oxygen-centered radicals has been witnessed, and a remarkable progress has been achieved, making it a hot topic over the last five years. This Review describes recent advances in the area of pyridinium salts as radical precursors, concerning the development of radical reactions involving pyridinium salts in organic synthesis.

Abstract:Kinetic, spectroscopic, and computational studies of radical C-H arylations highlight the interplay between chemical and physical rate processes in these multiphase reactions. Anomalous concentration dependences observed here may be reconciled by considering the role of phase transfer processes that mediate concentrations in each phase. In addition, understanding interactions through phase boundaries enables their use in optimization of reaction performance.

Abstract:while N-heterocyclic carbene (NHC) catalyzed electron-pair-transfer processes have been developed into an important tool for synthetically important bond formations during the past decades, the corresponding radical reactions via NHC catalysis have only received growing attention in the past six years. Taking into account the advantages NHC-catalyzed radical reactions might bring, such as creating new activation modes that were previously unobtainable, it is worthwhile to provide a conceptual understanding of this emerging area. Therefore, herein we give an overview of NHC-catalyzed radical reactions via different synthetic techniques.

Abstract:The many synthetic possibilities that arise when using radical intermediates, in place of their polar counterparts, make contemporary radical chemistry research an exhilarating field. The introduction of photocatalysis has helped tame aryl radicals, leading to a resurgence of interest in their chemistry, and an expansion of viable coupling partners and attainable transformations. These methods are more selective and safer than classical approaches, and they utilize new radical precursors. Given the importance of sustainability in current organic synthesis and our interest in light-assisted metal-free transformations, this Review focuses on recent advances in the use of aryl radicals in photoinduced cross-couplings that do not rely on metals for the crucial bond-forming event, and it is structured according to the key step that the aryl radicals engage in.

Abstract:Cyclic compounds are widely present in natural products, pharmaceuticals, agrichemicals and materials. The development of novel, high efficient and high selective strategies for the synthesis of cyclic compounds has obtained great research interests in organic synthetic field. In past two decades, the free-radical chemistry has been greatly developed than other periods in history, in which the radical cascade cyclizations of 1,n-enynes with trifluoromethyl have emerged as a powerful strategy for the preparation of trifluoromethyl containing carbocycles and heterocycles. In this context, we review the recent advances in the radical cascade cyclization of 1,n-enynes with trifluoromethyl. Furthermore, the detailed reaction mechanisms have also been discussed.

Abstract:Radical reactions are powerful in the synthesis of diverse molecular scaffolds bearing functional groups. In previous review articles, we have presented 1,2-difunctionalizations, remote 1,3-, 1,4-, 1,5-, 1,6- and 1,7-difunctionalizations, and addition followed by cyclization reactions. Presented in this paper is radical cyclization followed by the second functionalization reaction. The second functionalization could be realized by atom transfer reactions, radical or transition metal-assisted coupling reactions, and reactions with neutral molecules, cationic and anionic species.

Abstract:Radical transformations are powerful in organic synthesis for the construction of molecular scaffolds and introduction of functional groups. In radical difunctionalization reactions, the radicals in the first functionalized intermediates can be relocated through resonance, hydrogen atom or group transfer, and ring opening. The resulting radical intermediates can undertake the following paths for the second functionalization: (1) couple with other radical groups, (2) oxidize to cations and then react with nucleophiles, (3) reduce to anions and then react with electrophiles, (4) couple with metal-complexes. The rearrangements of radicals provide the opportunity for the synthesis of 1,3-, 1,4-, 1,5-, 1,6-, and 1,7-difunctionalization products. Multiple ways to initiate the radical reaction coupling with intermediate radical rearrangements make the radical reactions good for difunctionalization at the remote positions. These reactions offer the advantages of synthetic efficiency, operation simplicity, and product diversity.

Abstract:Radical chemistry has gained its renaissence in the past decade and trendendous progresses have been witnessed in synthetic and material chemistry. However, mechanism studies are largely lagging behind comparing with rapid paces in the development of synthetic methodologies. On the other hand, the study of radical species remains a central theme in physial organic chemistry, and a large amount of thermodynamic and kinetic data on radicals have been generated over a century's reserch. Unfortuately, there has been no systematic compilation and curation of these quatitative data that are dispersedly distributed and buried in literature. As a result, most synthetic chemists are not acquainted with these valuable data. In this review, we aim in compiling and curating thermodynamic and kinetic parameters of radicals, that may hopefully provide a quantitative data basis for rational development and evolution of radical chemistry. The key parameters include radical stability energy (RSE), radical lifetime (tau), substituent constant (sigma) and electrophilic index (omega). Fundamental concepts such as radical stability, Class S/O radical substitution effect, persistent/transient radical effect will be discussed on the basis of data.

Abstract:Abstract: Allenamides are special allenes, and the unique reactivity, selectivity (both stereoselective and regionally selective) and stability of allenamides have been widely studied. In this review, the development of the free radical transformation of allenamides over the last few years will be summarized. This review discusses in detail in three parts: intermolecular radical addition to C- X (X = N, S, O, Se) bonds, metal salt mediated cyclization of allenamides, and photocatalytic cyclization of allenamides. In addition, reasonable details of the mechanisms are provided for the vast majority of these transformations.

Abstract:Radical chemistry has played an important role in organic synthetic chemistry in the past decades. Additionally, the combination of N-heterocyclic carbene (NHC) catalysis and photocatalysis involving radical processes has emerged as a powerful strategy for the synthesis of useful molecules. Thus, it has received significant attention and underwent significant development in recent years. In this review, we summarized the recent advances in this rapidly growing area of research from the perspective of reaction mechanisms through NHC-bound ketyl radicals generated via single-electron oxidation with an excited-state photocatalyst, single-electron reduction with a reduced-state photocatalyst or energy transfer (EnT) with an excited-state photocatalyst, followed by the SET process. In addition, the catalytic mechanisms, design and application of these reaction modes are discussed in detail.

Abstract:Visible-light photoredox catalysis has been esteemed as one sustainable and attractive synthetic tool. In the past four years, a new yet challenging trend, visible-light-driven redox-neutral radical C-C crosscoupling involving putative radical intermediates, has been booming rapidly. Its advent brings a powerful platform to achieve non-classical C-C connections, and should lead to fundamental changes in retrosynthetic analysis. In this tutorial review, we highlight the recent achievements of visible-light-mediated redox-neutral radical C(sp(3))-C(sp(2)), C(sp(3))-C(sp), and C(sp(3))-C(sp(3)) bond formation, opening a new window for C-C cross-coupling through the photoredox electron shuttling cycle between two coupling partners. While radical-radical coupling steered by the persistent radical effect was proposed as a rational explanation for the redox-neutral photoredox events, alternative kinetically driven chain propagation and radical addition pathways cannot be ruled out. This tutorial review aims to highlight the recent achievements of photoredox-neutral radical C-C coupling in synthetic chemistry.
