Abstract:Bioinspired nanostructures can be the ideal functional smart materials to bridge the fundamental biology, biomedicine and nanobiotechnology fields. Among them, short peptides are among the most preferred building blocks as they can self-assemble to form versatile supramolecular architectures displaying unique physical and chemical properties, including intriguing optical features. Herein, we discuss the progress made over the past few decades in the design and characterization of optical short peptide nanomaterials, focusing on their intrinsic photoluminescent and waveguiding performances, along with the diverse modulation strategies. We review the complicated optical properties and the advanced applications of photoactive short peptide self-assemblies, including photocatalysis, as well as photothermal and photodynamic therapy. The diverse advantages of photoactive short peptide self-assemblies, such as eco-friendliness, morphological and functional flexibility, and ease of preparation and modification, endow them with the capability to potentially serve as next-generation, bio-organic optical materials, allowing the bridging of the optics world and the nanobiotechnology field.

Abstract:Responsive polymeric assemblies and hybrid superstructures fabricated from stimuli-sensitive polymers and inorganic nanoparticles (NPs) have been the subject of extensive investigations during the past few decades due to their distinct advantages such as an improved water solubility, stimuli-responsiveness, excellent biocompatibility, and facile introduction of functional units. In addition, the chemical compositions of polymeric assemblies and corresponding hybrid superstructures can be modulated via the initial synthetic design to target desired functions, fabricate smart nanostructures, and explore morphology-dependent functional optimization. Promising applications in the field of imaging, sensing, drug/gene delivery, diagnostics, and nanoreactors are being extensively investigated. This perspective article focuses on recent developments, microstructural control, and biomedical applications of stimuli-responsive polymeric assemblies as well as responsive hybrid superstructures fabricated from responsive polymers and inorganic NP building blocks (gold NPs and magnetic iron oxide NPs), and highlights their current status and future developments with selected literature reports.

Abstract:Azobenzene-containing small molecules and polymers are functional photoswitchable molecules to form supramolecular nanomaterials for various applications. Recently, supramolecular nanomaterials have received enormous attention in material science because of their simple bottom-up synthesis approach, understandable mechanisms and structural features, and batch-to-batch reproducibility. Azobenzene is a light-responsive functional moiety in the molecular design of small molecules and polymers and is used to switch the photophysical properties of supramolecular nanomaterials. Herein, we review the latest literature on supramolecular nano- and micro-materials formed from azobenzene-containing small molecules and polymers through the combinatorial effect of weak molecular interactions. Different classes including complex coacervates, host-guest systems, co-assembled, and self-assembled supramolecular materials, where azobenzene is an essential moiety in small molecules, and photophysical properties are discussed. Afterward, azobenzene-containing polymers-based supramolecular photoresponsive materials formed through the host-guest approach, polymerization-induced self-assembly, and post-polymerization assembly techniques are highlighted. In addition to this, the applications of photoswitchable supramolecular materials in pH sensing, and CO2 capture are presented. In the end, the conclusion and future perspective of azobenzene-based supramolecular materials for molecular assembly design, and applications are given.

Abstract:The supramolecular self-assembly of peptides offers a promising avenue for both materials science and biological applications. Peptides have garnered significant attention in molecular self-assembly, forming diverse nanostructures with α-helix, β-sheet, and random coil conformations. These self-assembly processes are primarily driven by the amphiphilic nature of peptides and stabilized by non-covalent interactions, leading to complex nanoarchitectures responsive to environmental stimuli. While extensively studied in biomedical applications, including drug delivery and tissue engineering, their potential applications in the fields of piezoresponsive materials, conducting materials, catalysis and energy harvesting remain underexplored. This review comprehensively elucidates the diverse material characteristics and applications of self-assembled peptides. We discuss the multi-stimuli-responsiveness of peptide self-assemblies and their roles as energy harvesters, catalysts, liquid crystalline materials, glass materials and contributors to electrical conductivity. Additionally, we address the challenges and present future perspectives associated with peptide nanomaterials. This review aims to provide insights into the versatile applications of peptide self-assemblies while concisely summarizing their well-established biomedical roles that have previously been extensively reviewed by various research groups, including our group.

Abstract:Azobenzene derivatives have become one of the most famous photoresponsive chromophores in the past few decades for their reversible molecular switches upon the irradiation of actinic light. To meet the ever-increasing requirements for applications in materials science, biomedicine, and light-driven devices, it is usually necessary to adjust their photochemical property from the molecular level by changing the substituents on the benzene rings of azobenzene groups. Among the diverse azobenzene derivatives, azopyridine combines the photoresponsive feature of azobenzene groups and the supramolecular function of pyridyl moieties in one molecule. This unique feature provides pH-responsiveness and hydrogen/halogen/coordination binding sites in the same chromophore, paving a new way to prepare multi-functional responsive materials through non-covalent interactions and reversible chemical reactions. This review summarizes the photochemical and photophysical properties of azopyridine derivatives in supramolecular states (e.g., hydrogen/halogen bonding, coordination interactions, and quaternization reactions) and illustrates their applications from photoresponsive liquid crystals to light-driven devices. We hope this review can highlight azopyridine as one more versatile candidate molecule for designing novel photoresponsive materials towards light-driven applications.

Abstract:Amphiphilic polymers are well known for their characteristics to form various self-assembling structures under proper conditions. Amphiphilic azo polymers, which are amphiphilic polymers functionalized with aromatic azo moieties, can combine wealthy photoresponsive properties with the self-assembling feature. In recent years, a wide variety of amphiphilic azo polymers have been developed through different molecular design strategies and synthetic routes. This article reviews the background, recent development, current approaches, and outlook of this rapidly developing research area. The emphasis is placed on the synthesis, self-assembly, and photoresponsive properties of amphiphilic azo polymers. According to the molecular architecture, the amphiphilic azo polymers are classified as homopolymers, random copolymers, block copolymers, star-like and dendritic polymers. The polymers show strong tendency to form well-organized structures at surfaces or interfaces, in solutions or dispersion mediums, and through phase-separation in solid thin-films. Colloidal arrays and porous thin films of amphiphilic azo polymers can be obtained through the vertical deposition method and solvent-induced structure inversion. The amphiphilic azo polymers and their self-assembled structures exhibit a variety of photoresponsive properties triggered by the trans–cis photoisomerization of azo chromophores. Some interesting variations caused by the light irradiation, such as the surface wettability change, surface-relief-grating formation, domain orientation, colloid deformation, micelle dissociation, vesicle bursting and fusion, are described in this review in detail. Such properties are promising for applications in areas such as optical devices, sensors, and drug delivery.

Abstract:Conjugated polymers (CPs) have been developed quickly as an emerging functional material with applications in optical and electronic devices, owing to their highly electron-delocalized backbones and versatile side groups for facile processibility, high mechanical strength, and environmental stability. CPs exhibit multistimuli responsive behavior and fluorescence quenching properties by incorporating azobenzene functionality into their molecular structures. Over the past few decades, significant progress has been made in developing functional azobenzene-based conjugated polymers (azo-CPs), utilizing diverse molecular design strategies and synthetic pathways. This article comprehensively reviews the rapidly evolving research field of azo-CPs, focusing on the structural characteristics and synthesis methods of general azo-CPs, as well as the applications of charged azo-CPs, specifically azobenzene-based conjugated polyelectrolytes (azo-CPEs). Based on their molecular structures, azo-CPs can be broadly categorized into three primary types: linear CPs with azobenzene incorporated into the side chain, linear CPs with azobenzene integrated into the main chain, and branched CPs containing azobenzene moieties. These systems are promising for biomedical applications in biosensing, bioimaging, targeted protein degradation, and cellular apoptosis.

Abstract:Amphiphilic polymers can self-assemble into various nanostructures in solution, which can find applications in many fields such as nanotechnology, drug delivery, and template synthesis. Herein, we report the controlled self-assembly and dual stimuli-responsive nanostructure transition of a class of three-arm branched amphiphilic polymers (AzoFcPEO) containing ferrocene (Fc) and azobenzene (Azo) moieties in aqueous solution. These amphiphilic polymers were synthesized by an esterification reaction of a variety of polyethylene oxide methyl ethers (Me-PEO) with 3-(6-ferrocenyhexyloxyl)-5-(6-azobenzenehexyloxy) benzoic acid. Both the isomerization of Azo and redox of Fc moieties can respectively change the amphiphilicity of these polymers to different degrees. Consequently, these amphiphilic polymers in aqueous solution can self-assemble into various nanostructures, such as spherical micelle, worm-like micelle, spherical compound micelle, rod-like compound micelle and vesicle dependent on the PEO molecular weight, applied stimuli, and polymer concentration. This work can offer tremendous possibilities not only for the fundamental science of the controlled self-assembly but also for establishing a suitable method for regulating the nanostructures of amphiphilic polymers in aqueous solution.

Abstract:Mesoporous silica nanoparticles (MSNs) endowed with polymer coatings present a versatile platform, offering notable advantages such as targeted, pH-controlled, and stimuli-responsive drug delivery. Surface functionalization, particularly through amine and carboxyl modification, enhances their suitability for polymerization, thereby augmenting their versatility and applicability. This review delves into the diverse therapeutic realms benefiting from polymer-coated MSNs, including photodynamic therapy (PDT), photothermal therapy (PTT), chemotherapy, RNA delivery, wound healing, tissue engineering, food packaging, and neurodegenerative disorder treatment. The multifaceted potential of polymer-coated MSNs underscores their significance as a focal point for future research endeavors and clinical applications. A comprehensive analysis of various polymers and biopolymers, such as polydopamine, chitosan, polyethylene glycol, polycaprolactone, alginate, gelatin, albumin, and others, is conducted to elucidate their advantages, benefits, and utilization across biomedical disciplines. Furthermore, this review extends its scope beyond polymerization and biomedical applications to encompass topics such as surface functionalization, chemical modification of MSNs, recent patents in the MSN domain, and the toxicity associated with MSN polymerization. Additionally, a brief discourse on green polymers is also included in review, highlighting their potential for fostering a sustainable future.

Abstract:The synthesis of multiple stimuli‐responsive fluorescent hollow nano/microparticles by templating coordination is reported. With this strategy, bisphosphonate‐merocyanine ligands are successfully assembled to hollow bisphosphonate‐spiropyran (BPSP)‐Zn2+ microparticles that exhibit responsiveness to light, pH, and glutathione. These remarkable features render the hollow BPSP‐Zn2+ microparticles to show high promise in the field of stimulus‐triggered drug delivery. Supramolecular multifunctional metal–organic networks offer immense potential in drug delivery, bioimaging, catalysis, and therapeutics. However, the creation of multiple stimuli‐responsive fluorescent hollow nano/microparticles is a formidable challenge in this field. In this study, a novel strategy is reported for synthesizing photochromic hollow microparticles that exhibit responsiveness to light, pH, and glutathione (GSH). With the assistance of templating coordination from zeolitic imidazolate framework‐8 microparticles, bisphosphonate‐merocyanine ligands are successfully assembled to create hollow bisphosphonate‐spiropyran (BPSP)‐Zn2+ microparticles. The photochromic and pH responses stem from the reversible conversion between spiropyran and zwitterionic merocyanine components and between merocyanine and protonated merocyanine, respectively. The GSH response is attributed to the stronger affinity of Zn2+ to thiol groups than to phosphonate groups. These remarkable features render the hollow BPSP‐Zn2+ microparticles highly suitable for applications in stimulus‐triggered drug delivery, and these findings provide a platform for developing innovative materials that have the potential to transform the biomedical field.

Abstract:Azobenzene (Azo) and its derivatives are versatile stimuli‐responsive molecules. Their reversible photoisomerization and susceptibility to reduction‐mediated cleavage make them valuable for various biomedical applications. Upon exposure to the UV light, Azo units undergo a thermodynamically stable trans‐to‐cis transition, which can be reversed by heating in the dark or irradiation with visible light. Additionally, the N=N bonds in azobenzenes can be cleaved under hypoxic conditions by azoreductase, making azobenzenes useful as hypoxia‐responsive linkers. The integration of azobenzenes into nanomedicines holds promise for enhancing therapeutic efficacy, particularly in tumor targeting and controllable drug release. In this Concept paper, recent advances in the design and applications of azobenzene‐based nanomedicines are updated, and future development opportunities are also summarized.

Abstract:Magnetic nanoparticle (MNP) assemblies have demonstrated great potential in biomedical applications due to their controllable magnetic properties and collective functions. Among the versatile approaches to obtaining MNP assemblies, polymer-assisted assembling methods offer unique advances, by which the assembled nanostructures could exert the merits of both the inorganic magnetic nanoparticles and organic polymeric materials to realize combined advantages for medical diagnosis and treatment. Precise control over the interactions among different building moieties and spatial organization of magnetic nanoparticles with the aid of polymers provides promising strategies in manipulating the physical, chemical, and biological properties of nanoassemblies for biomedical applications. In this review, we summarize the recent progress of polymer-assisted MNP assemblies, which include the mutual interactions between building blocks, architectural diversity of the assemblies, and their synthetic strategies along with biomedical applications. The current review provides a comprehensive insight into controlled and intelligent nanomedicines, which shall facilitate the development of next-generation high-performance theranostic agents based on MNP assemblies.

Abstract:Porphyrins and their derivatives have attracted great attention due to their unique properties and diverse functionalities which contribute to their wide applications such as catalysis, solar cells, biomedicine and environmental science. It is interesting to note that porphyrin-containing polymers have recently aroused great interest due to the simultaneous presence of both porphyrinic and polymeric characteristics, especially for porphyrin-containing polymers linked by covalent bonds giving well-defined macromolecular architectures, unique self-assembly morphologies and promising applications. Herein, we summarize the different synthetic strategies and self-assembly protocols adopted in recent years to explore various applications of porphyrin-based functional polymers followed by a brief discussion on the challenges and outlooks for future research. With the discovery of advanced polymerization techniques such as living/controlled radical polymerization and click chemistry, the synthesis of porphyrin-containing functional polymers with well-defined architectures has been greatly developed. Porphyrin molecules tend to form the aggregates based on the self-assembly driving force of pi-pi stacking. Much attention has been paid to the self-assembly behavior of porphyrin-containing functional polymers, and various nanostructures have been constructed including nano-sized or micron-sized spheres, vesicles, belts, wires, rings, tubes, and rods. Porphyrin-containing functional polymers, with special photochemical and photophysical properties, present significant applications in photovoltaics, catalysis, biological and biomedicine fields. Particularly, the porphyrin-based photosensitizers for photodynamic therapy, porphyrin-conjugated polymers for solar cell devices, porphyrin-based polymeric nanocomposites in catalysis and chemical adsorption and separation have been widely studied. Abstract:<p>Owing to the tremendous advantages and unique well‐defined nanoscale structure, polyhedral oligomeric silsesquioxanes (POSS) has received considerable interest in the design of novel organic‐inorganic hybrid nanomaterials with all manner of prominent capabilities, which is recognized as a new generation of promising materials for advanced applications of material science, engineering science and biomedical fields. Benefitting from the recent progress in combination of controlled/living polymerization and emerging click chemistry, POSS‐based hybrid materials with ingenious design, versatile topological structure and sophisticated multifunctionality have been successfully fabricated and developed into abundant well‐defined hybrid nanostructures with desired physicochemical properties. Tailor‐made amphiphilic molecular design and nanosized hybrid architecture provide opportunities for the self‐assembly of POSS‐based hybrid materials with unique hierarchical morphologies in selective solvents. Through the in‐depth understanding of structure‐properties relationship, POSS‐based hybrid materials can achieve the modulation of precise control of self‐assembling process and multi‐purpose applications with improved material performances. In this review, we summarize the recent advances of POSS‐based hybrid materials in molecular design, self‐assembly behavior in solutions and potential biomedical applications with main concerns on drug delivery, gene therapy, bioimaging and tissue engineering field. Abstract:Polymeric assemblies self-assembled from amphiphilic copolymers are the topic of intense research and have emerged as versatile drug nanocarriers in the past few decades. To enhance the bioavailability of drugs at the target disease site, the use of stimuli-responsive nanocarriers with triggered release properties is highly desired. Among all the available chemical and physical stimuli, photo has attracted much attention since it can be localized in time and space, and it can also be triggered from outside of the system. In this chapter, we highlight the recent progress of photo-responsive assemblies as drug nanocarriers. Different types of photo-responsive polymers were classified. A wide variety of photo-responsive moieties, including UV and NIR responsive polymers, as well as synthetic routes were introduced to the drug nanocarriers. Finally, we suggest possible future developments of photo-responsive polymeric nanocarriers for biomedical applications, especially for drug delivery.

Abstract:Luminescent polymer nanomaterials not only have the characteristics of various types of luminescent functional materials and a wide range of applications, but also have the characteristics of good biocompatibility and easy functionalization of polymer nanomaterials. They are widely used in biomedical fields such as bioimaging, biosensing, and drug delivery. Designing and constructing new controllable synthesis methods for multifunctional fluorescent polymer nanomaterials with good water solubility and excellent biocompatibility is of great significance. Exploring efficient functionalization methods for luminescent materials is still one of the core issues in the design and development of new fluorescent materials. With this in mind, this review first introduces the structures, properties, and synthetic methods regarding fluorescent polymeric nanomaterials. Then, the functionalization strategies of fluorescent polymer nanomaterials are summarized. In addition, the research progress of multifunctional fluorescent polymer nanomaterials for bioimaging is also discussed. Finally, the synthesis, development, and application fields of fluorescent polymeric nanomaterials, as well as the challenges and opportunities of structure–property correlations, are comprehensively summarized and the corresponding perspectives are well illustrated.

Abstract:This review focuses on smart nano-materials built of stimuli-responsive (SR) polymers and will discuss their numerous applications in the biomedical field. The authors will first provide an overview of different stimuli and their corresponding, responsive polymers. By introducing myriad functionalities, SR polymers present a wide range of possibilities in the design of stimuli-responsive devices, making use of virtually all types of polymer constructs, from self-assembled structures (micelles, vesicles) to surfaces (polymer brushes, films) as described in the second section of the review. In the last section of this review the authors report on some of the most promising applications of stimuli-responsive polymers in nanomedicine. In particular, we will discuss applications pertaining to diagnosis, where SR polymers are used to construct sensors capable of selective recognition and quantification of analytes and physical variables, as well as imaging devices. We will also highlight some examples of responsive systems used for therapeutic applications, including smart drug delivery systems (micelles, vesicles, dendrimers...) and surfaces for regenerative medicine.

Abstract:Smart nanomaterials, thanks to their stimuli-responsive properties, represent a promising class of nanostructures for biomedical applications. Their ability to undergo structural or functional changes in response to specific external cues has been exploited for the manipulation of cellular activities as a potential treatment for various health disorders. Despite their rather interesting properties and applications, most smart nanostructures are composed of inorganic materials, thus limiting their translatability in clinical applications due to potential toxicity and long-term accumulation concerns. Polydopamine nanostructures can overcome the limitations posed by inorganic smart nanomaterials thanks to their high biocompatibility, biodegradability, antioxidant effects, pH-responsiveness, tunability, surface reactivity, photothermal conversion properties, and the ability to act as photoacoustic contrast agents. In this review, we will present a general analysis of the properties of polydopamine nanostructures and of their biomedical applications. Our aim is to provide the reader with the state of the art concerning the use of polydopamine nanostructures as smart organic nanoplatforms in nanomedicine, also providing an analysis of the current limitations connected to the translation in clinical applications and the potential solutions to these challenges.
