Abstract:In this study, Chinese yam polysaccharide (CYP) was isolated from yam by hydroextraction and alcoholic precipitation. Subsequently, the chlorosulfate-pyridine (CSA-Pyr) method was used to obtain the sulfated Chinese yam polysaccharide derivative (S-CYP) to evaluate its immunomodulatory activity in RAW 264.7 cells and to investigate its mechanism of action. The results revealed that the sulfated modification altered the physicochemical properties of CYP but had no impact on the main chain structure. S-CYP demonstrated excellent immunomodulatory activity by increasing the viability of RAW 264.7 macrophage cells and stimulating the production of reactive oxygen species (ROS), nitric oxide (NO), tumor necrosis factor-alpha (TNF-alpha) and interleukin (IL)-6. Moreover, signal transduction experiments showed that S-CYP induced the activation of mitogen-activated protein kinase (MAPK) and nuclear factor-kappa B (NF-kappa B) pathways through toll-like receptor 4 (TLR4), dramatically increasing p-ERK, p-JNK and p-p38 proteins. Meanwhile, immunofluorescence results showed that S-CYP could significantly promote the entry of NF-kappa B p65 into the nucleus, which is essential for triggering the NF-kappa B pathway. Furthermore, blocking antibody experiments revealed that specific inhibitors of TLR4, MAPKs, and NF-kappa B suppressed the generation of TNF-alpha and IL-6 in RAW 264.7 cells. These findings suggested that both CYP and S-CYP could be used as immunomodulatory agents and may have potential application prospects in the food and pharmaceutical industries.

Abstract:It is well known that yam is an important industrial crop resource. Our previous study has demonstrated that sulfated modifications (S-CYP) can improve the immunomodulatory effect of polysaccharide from Chinese yam by activating and maturating RAW264.7 cells, while its mechanism is still unknown. In this study, the underlying mechanisms of S-CYP-induced immunomodulatory effect were examined based on RNA sequencing (RNA-seq), and a co-culture system with Caco-2 cells and RAW264.7 cells. RNA-seq analysis indicated that 1764 expressed genes differentially between the control and S-CYP groups. 13 cellular component terms, 20 biological process terms, and 10 molecular function terms were identified by GO. KEGG analysis showed that MAPK signaling pathways belong to the 20 most significantly affected pathways. Furthermore, S-CYP indirectly affects the immune function of RAW264.7 in co-culture model by increasing secretion of cytokines (IL-1β, TNF-α). These results provide a theoretical basis for exploring S-CYP as an immunomodulatory agent.

Abstract:Our previous study has demonstrated that sulfated Chinese yam polysaccharide (SCYP) can improve immunomodulatory activity in Raw 264.7 cells. However, its anti-inflammatory is little known. In this study, the anti-inflammatory effects of SCYP were systematically investigated via the Lipopolysaccharides (LPS)-induced Raw264.7 cell model, Caco-2/Raw264.7 co-culture system, and acute inflammation mice model. The results suggested SCYP promoted the cell proliferation and have no toxicity in Raw264.7 and Caco-2 cells at the concentration of 200 µg/mL. Moreover, when treated with SCYP, the production of pro-inflammatory cytokines (interleukin [IL]-1β, IL-6, and tumor necrosis factor-α) reduced significantly in Raw264.7 via the MAPK/NF-κB pathway. In the Caco-2/Raw264.7 co-cultured system, SCYP could regulate inflammation reaction by improving intestinal barrier, which might prevent systemic inflammation. Further, systemic inflammation was alleviated by SCYP in LPS-induced acute inflammation mice through MAPK/NF-κB pathway. PRACTICAL APPLICATION: These results supported that SCYP may be used as an anti-inflammation agent in the functional food field.

Abstract:A sulfated Chinese yam polysaccharide (S-CYP) was obtained from non-starch Chinese yam polysaccharide (CYP) by chlorosulfonic acid-pyridine (CSA-Pyr) method. The physicochemical characteristics and immunological activity of S-CYP were evaluated in this study. Results showed that sulfated modification changed the physicochemical properties of Chinese yam polysaccharide. Body weights and thymus index of the mice in polysaccharides group were increased compared with the cyclophosphamide (Cy)-induced immunosuppressed mice in model group, and could restore the splenomegaly that caused by Cy. It also showed that S-CYP could enhance the immunomodulatory activity on splenic lymphocytes since it could increase the proliferation of splenic lymphocytes and effectively cooperate with ConA to induce splenic lymphocytes differentiation into T lymphocytes. Meanwhile, the number of CD3+CD4+ and CD3+CD8+ T lymphocytes was elevated, the CD4+/CD8+ ratio was restored in polysaccharides group. The levels of splenic lymphocytes cytokines (interleukin (IL)-1β, tumor necrosis (TNF)-α), and the levels of immunoglobulin (IgG and IgM) secretion in serum were also increased by S-CYP significantly. These findings demonstrate that CYP and S-CYP can serve as a promising immunomodulator with potential application in functional food and nutraceutical industry.

Abstract:Background Nuoshanyao (NSY) is a special species of Chinese yam ( Dioscorea opposita Thunb.) and shows rich functionality and superior nutritional value. The chemical components and structural properties of Nuoshanyao polysaccharide (NSYP) were investigated in his study. Results The molecular weight (Mw) of the major component in the polysaccharide was 23 kDa. NSYP was mainly composed of glucose, mannose, and galactose, and the molar ratio was 36.5:1.63:1. There might be an ordered helical conformation in NSYP, and the surface of NSYP was granular. NSYP exhibited strong scavenging capacity on the DPPH radical and chelating capacity on the ferrous ion in vitro with a rate of 94.94% and 82.49% at 4 mg/mL. Conclusion NSYP with the appropriate molecular weight range and helix structure could possess great antioxidant activity, while monosaccharide compositions may affect the activity. This work supplied important information for the potential application of NSYP in the nutritional and food industries.
Sulfated Chinese Yam Polysaccharides Alleviate LPS-Induced Acute Inflammation in Mice Through Modulating Intestinal Microbiota.

Abstract:This study aimed to test the preventive anti-inflammatory properties of Chinese yam polysaccharides (CYP) and sulfated Chinese yam polysaccharides (SCYP) on LPS-induced systemic acute inflammation in mice and investigate their mechanisms of action. The results showed that SCYP can efficiently reduce plasma TNF-α and IL-6 levels, exhibiting an obvious anti-inflammation ability. Moreover, SCYP reduced hepatic TNF-α, IL-6, and IL-1β secretion more effectively than CYP, and significantly altered intestinal oxidative stress levels. In addition, a 16S rRNA gene sequencing analysis showed that CYP regulated the gut microbiota by decreasing Desulfovibrio and Sutterella and increasing Prevotella. SCYP changed the gut microbiota by decreasing Desulfovibrio and increasing Coprococcus, which reversed the microbiota dysbiosis caused by LPS. Linear discriminant analysis (LDA) effect size (LEfSe) revealed that treatment with CYP and SCYP can produce more biomarkers of the gut microbiome that can promote the proliferation of polysaccharide-degrading bacteria and facilitate the intestinal de-utilization of polysaccharides. These results suggest that SCYP can differentially regulate intestinal flora, and that they exhibit anti-inflammatory effects, thus providing a new reference to rationalize the exploitation of sulfated yam polysaccharides.

Abstract:The yam (Dioscorea opposita Thunb) is a well-known edible food and widely used as the traditional Chinese medicine. The present investigation was designed to evaluate the immunomodulatory activity of glycoprotein (DOT) from yam and explore its possible molecular mechanisms. Results showed that the DOT could improve the cell immunity, humoral immunity and phagocytic system function of the normal mice. The DOT could also increase the production of TNF-α, interleukin-6 and nitric oxide and enhance the pinocytosis function of macrophages. Furthermore, the DOT increased phosphor-p38, JNK, ERK1/2 and nuclear factor kappa B (NF-κB) p65 protein expression in peritoneal macrophages. Taken together, our data suggest that DOT could be used as a potential immunostimulant and exert its immunomodulatory activity via mitogen-activated protein kinases and NF-κB signal pathways. Abstract:Yam (Dioscorea opposite Thunb) polysaccharides have in vitro immune activities. The present study investigated the effects of thermal, acidic, and enzymatic treatments on immune activities of the water-soluble yam polysaccharides, via assaying their abilities to activate three immune-related cells (splenocytes, macrophages, and natural killer cells) and to mediate the secretion of eight immune molecules (IL-2, IFN-gamma, IL-4, IL-1 beta, IL-6, TNF-alpha, iNOS, and LZM). The results indicated that the yam polysaccharides heated at 100 degrees C for 30-60 min, or digested by simulated stomach acid for 1-2 h, or digested by two proteases and two amylases for 1-3 h showed unchanged immune activities (P > 0.05). However, hydrolysis of the yam polysaccharides by HCl solution of 3 mol/L for 3-6 h brought about lower activities significantly (P < 0.05). It is evidenced that the yam polysaccharides can keep their immune activities during thermal processing and body digestion, but stronger acid is potential to destroy their immune activities.
Molecular Weight-Dependent Immunostimulative Activity Of Low Molecular Weight Chitosan Via Regulating Nf-Kappa B And Ap-1 Signaling Pathways In Raw264.7 Macrophages

Abstract:Chitosan and its derivatives such as low molecular weight chitosans (LMWCs) have been found to possess many important biological properties, such as antioxidant and antitumor effects. In our previous study, LMWCs were found to elicit a strong immunomodulatory response in macrophages dependent on molecular weight. Herein we further investigated the molecular weight-dependent immunostimulative activity of LMWCs and elucidated its mechanism of action on RAW264.7 macrophages. LMWCs (3 kDa and 50 kDa of molecular weight) could significantly enhance the mRNA expression levels of COX-2, IL-10 and MCP-1 in a molecular weight and concentration-dependent manner. The results suggested that LMWCs elicited a significant immunomodulatory response, which was dependent on the dose and the molecular weight. Regarding the possible molecular mechanism of action, LMWCs promoted the expression of the genes of key molecules in NF-B and AP-1 pathways, including IKK, TRAF6 and JNK1, and induced the phosphorylation of protein IKB in RAW264.7 macrophage. Moreover, LMWCs increased nuclear translocation of p65 and activation of activator protein-1 (AP-1, C-Jun and C-Fos) in a molecular weight-dependent manner. Taken together, our findings suggested that LMWCs exert immunostimulative activity via activation of NF-B and AP-1 pathways in RAW264.7 macrophages in a molecular weight-dependent manner and that 3 kDa LMWC shows great potential as a novel agent for the treatment of immune suppression diseases and in future vaccines.
Sulfated Cyclocarya Paliurus Polysaccharides Exert Immunomodulatory Potential on Macrophages Via Toll-like Receptor 4 Mediated MAPK/NF-κB Signaling Pathways

Abstract:The biological activity of plant polysaccharides can be enhanced by sulfated modification. In this study, the immunomodulatory effect of sulfated Cyclocarya paliurus polysaccharides (SCP3) on macrophages RAW264.7 and its potential molecular mechanism were investigated. Results showed that SCP3 at 25−100 μg/mL increased viability and improved phagocytosis of RAW264.7 cells. Meanwhile, SCP3 could activate mitogen-activated protein kinase (MAPK) and nuclear factor kappa B (NF-κB) signaling pathways, which increased the phosphorylation of Erk1/2, JNK, p38 and NF-κB p65, promoting secretion of cytokines tumor necrosis factor α (TNF-α), interleukin 6 (IL-6) and nitric oxide (NO) as well as the production of reactive oxygen species (ROS). In addition, Toll-like receptor 4 (TLR4) receptor inhibitors were able to block the production of NO and TNF-α by SCP3-stimulated macrophages. Based on Western blot analysis and validation using specific inhibitors against MAPK and NF-κB signaling pathways, the results demonstrated that SCP3 induced macrophages activation and enhanced TNF-α and NO production via TLR4-mediated MAPK and NF-κB pathways. In summary, SCP3 has significant immunomodulatory potential. The underlying molecular mechanism was that SCP3 activates macrophages via TLR4 receptors to promote ROS production, which in turn activates the downstream MAPK/NF-κB signaling pathway and then increases the secretion levels of cytokines and NO.

Abstract:A sulfated polysaccharide (SMP) from the Mesona chinensis Benth was successfully obtained and its immunomodulatory effects were evaluated. Results showed that SMP exhibited the proliferation effects in splenocytes in vitro and ameliorated the impaired of splenocytes in Cy-treated mice. SMP induced the production of nitric oxide (NO), interleukin (IL) -2, interferon-gamma (IFN-gamma), increased the expression of antioxidant enzymes (SOD and CAT), and reduced the production of malondialdehyde (MDA) in Cy-treated mice, as well as improved the blood index, body weight and organ index. In addition, SMP enhanced the viability of RAW264.7 macrophages and increased the secretion of IL-6, IL-1 beta and tumor necrosis factor-alpha (TNF-alpha) and production of NO in RAW264.7 cells. Furthermore, SMP binds to TLR4 (Toll-like Receptor 4) to promote RAW264.7 to produce cytokines, thereby exerting immunoregulatory function. These data indicated that SMP act a pivotal part in immunomodulatory and could be used as functional foods.
Sargassum Fusiforme Polysaccharide SFP-F2 Activates the NF-κB Signaling Pathway via CD14/IKK and P38 Axes in RAW264.7 Cells

Abstract:Sargassum fusifrome is considered a “longevity vegetable” in Asia. Sargassum fusifrome polysaccharides exhibit numerous biological activities, specially, the modulation of immune response via the NF-κB signaling pathway. However, the precise mechanisms by which these polysaccharides modulate the immune response through the NF-κB signaling pathway have not been elucidated. In this study, we purified and characterized a novel fraction of Sargassum fusifrome polysaccharide and named it SFP-F2. SFP-F2 significantly upregulated the production of the cytokines TNF-α, IL-1β and IL-6 in RAW264.7 cells. It also activated the NF-κB signaling pathway. Data obtained from experiments carried out with specific inhibitors (PDTC, BAY 11-7082, IKK16 and SB203580) suggested that SFP-F2 activated the NF-κB signaling pathway via CD14/IKK and P38 axes. SFP-F2 could therefore potentially exert an immune-enhancement effect through inducing the CD14/IKK/NF-κB and P38/NF-κB signaling pathways.

Abstract:Abstract BACKGROUND: In order to develop a novel dietary additive for potential application in domestic animals and humans, a fraction of Chinese yam polysaccharide (CYP) was prepared and a study was conducted to determine the effects of dietary supplementation with CYP on the immune response and growth performance in weanling rats. RESULTS: Dietary supplementation with CYP increased the average daily feed intake and average daily gain but decreased the feed/gain ratio overall compared with the non‐supplemented group. CYP also significantly increased the serum contents of interleukin‐2 and tumour necrosis factor and the proliferation activity of peripheral lymphocytes but decreased the serum contents of low‐density lipoprotein, total cholesterol and triglyceride compared with the non‐supplemented group. CONCLUSION: The increase in cellular immunity in CYP‐supplemented weanling rats confers an important protective role in the non‐specific defence against infections. This herbal polysaccharide, as a natural green dietary additive for promoting the immune response and healthy growth, may offer an effective alternative to antibiotics for weanling animals. Abstract:Objective:to investigate the structure and in vitro immunomodulatory activity of polysaccharides from bamboo shavings (BSP).Methods:the structure was analyzed by the methods of methylation and nuclear magnetic resonance spectroscopy;the in vitro immunomodulatory activity was evaluated by the assays of mouse splenic lymphocyte proliferation,the phagocytosis of neutral red and NO secretion of RAW264.7 cell.Results:BSP was O-acetyl-(4-O-methylglucurono)-arabinoxylans consisting of a linear (l→4)-β-D-xylopyranosyl backbone decorated with branches at O-2 of 4-O-methylglucuronic acid units and acetyl groups or O-3 of α-L-arabinofuranosyl and acetyl groups.The structural element could be expressed as below:1,4-β-D-[2-O-Ac][3-O-Ac][4-O-Me-D-GlcpA-(1→2)][α-L-Araf-(1→3)]-xylp;BSP could evidently strengthen concanavalin A or lipopolysaccharide-induced splenic lymphocyte proliferation at 10-100μg/mL;BSP could significantly enhance the phagocytosis activity and NO production of RAW264.7 cell at 10-1 000 μg/mL in a dose-dependent manner.Conclusion:BSP exhibits excellent immunostimulatory activity in vitro,and can be considered as an ideal source of bio-active polysaccharides due to the abundance of its raw material.
In Vitro Immunomodulation of the Polysaccharides from Yam (dioscorea Opposita Thunb.) in Response to a Selenylation of Lower Extent.

Abstract:The immunomodulation of chemically selenylated polysaccharides has been attracting more attention recently, but the corresponding performance of the yam polysaccharides (YPS) with lower selenylation extent remains, thus far, unsolved. In this study, the YPS was selenylated with Na2SeO3 under acidic conditions generated by HNO3 to reach two lower selenylation extents, yielding two selenylated YPSs, namely SeYPS-1 and SeYPS-2 with selenium contents of 715 and 1545 mg/kg, respectively. The results indicated that YPS, SeYPS-1, and SeYPS-2 all had in vitro immuno-modulation when using RAW 264.7 macrophages and murine splenocytes as cell models. In detail, the three polysaccharide samples at dose levels of 5–160 μg/mL showed insignificant cytotoxicity to the macrophages and splenocytes with cell exposure times of 12–24 h, because of the measured values of cell viability larger than 100%. However, Na2SeO3 at dose levels of 1.3–3.25 μg/mL mostly caused obvious cytotoxic effects on the cells, resulting in reduced cell viability values or cell death, efficiently. The results demonstrated that, compared with YPS, both SeYPS-1 and SeYPS-2 at a lower dose level (5 μg/mL) were more active at promoting phagocytosis activity, increasing the CD4+/CD8+ ratio of the T-lymphocyte sub-population in the murine splenocyte, improving cytokine secretion, including interleukin-6 (IL-6), IL-1β, and tumor necrosis factor-α in the macrophages, or increasing interferon-γ secretion, but suppressing IL-4 production in the splenocytes. Consistently, SeYPS-2 has more potential than SeYPS-1 at exerting these assessed bioactivities in the cells. Thus, we conclude that a chemical modification of YPS using trace element Se at a lower selenylation extent could bring about higher immunomodulatory activity towards macrophages and splenocytes, while selenylation extent of YPS is a critical factor used to govern the assessed activity changes of YPS.

Abstract:Plant polysaccharides possess many health-promoting properties, which are closely related to the digestion, absorption and utilization of polysaccharides in the host. The characteristic changes in Chinese yam polysaccharide (CYP) during simulated gastrointestinal digestion and faecal fermentation, as well as the intestinal anti-inflammatory effects of CYP and fermented CYP, were studied. The molecular weight of CYP remained unchanged during gastrointestinal digestion. After 24 h of faecal fermentation, free glucose and mannose released from CYP were utilized by gut microbiota, and the production of short-chain fatty acids increased. Simultaneously, CYP improved the growth of Bifidobacterium and Megasphaera. Compared with CYP, faecal fermented CYP exhibited greater suppression of anti-inflammatory mediators and increased intestinal tight junction expression in LPS-stimulated coculture Caco-2/Raw264.7 cells. The obtained results reveal that the role of CYP in promoting gut health is closely related to the interactions between CYP and gut microbes, and CYP has potential as an anti-inflammatory adjuvant in enteritis therapy.

Abstract:As an important ingredient of Chinese yam, Chinese yam polysaccharides have received wide attention for their remarkable adjuvant activity. Pickering emulsion is an attractive platform for the delivery of vaccines. Our previous study has demonstrated that the Chinese yam polysaccharides PLGA-stabilized Pickering emulsion (CYP-PPAS) is a potentially safe and efficient adjuvant to improve the immune response. In this work, we further investigate the adjuvant activity of CYP-PPAS on cellular immunity. In vitro, the CYP-PPAS increased antigen uptake efficiency by DCs. In vivo, CYP-PPAS triggered the recruitment of DCs and macrophages and subsequently facilitated DCs maturation and antigen migration to lymph nodes. Furthermore, CYP-PPAS induced a robust humoral response and Th1/Th2 immune response, enhanced the activation of CD4+ and CD8+ T lymphocyte subpopulations, and also promoted the activation of cytotoxic T lymphocyte response. As a result, the CYP-PPAS serves as a promising vaccine delivery system to induce robust humoral and cellular immunities against diseases.

Abstract:Immunomodulatory potentials of water-soluble yam (Dioscorea opposita Thunb) polysaccharides (WYPs) for the normal and immuno-suppressed mice were described in this study. For the normal mice, the WYPs at 500mgkg(-1) elevated spleen and thymus indices by 22-42%; promoted macrophages' phagocytosis; lymphocytes' proliferation and natural killer (NK) cell activity by 18-53%; enhanced IL-2 and IFN- levels in the splenocytes by 42-45%; raised IL-1, IL-6, TNF-, iNOS and lysozyme levels in the macrophages by 40-219%; and increased serum IgM, IgA and IgG levels by 44-51%. The WYPs could restore and improve immune status of the cyclophosphamide-treated mice, as they at 500mgkg(-1) elevated spleen and thymus indices by 85-172%; promoted macrophages' phagocytosis, lymphocytes' proliferation and NK cell activity by 24-98%; enhanced IL-2 and IFN- levels in the splenocytes by 44-109% or IL-1, IL-6, TNF-, iNOS and lysozyme levels in the macrophages by 53-287%; and increased the three immunoglobulins by 24-69%.

Abstract:Our previous study has demonstrated that sulfated yam polysaccharide (SCYP) had a stronger immunomodulatory activity than yam polysaccharide (CYP). In order to investigate how the sulfated modification influence the immunological activity of CYP, this research was mainly focused on the study of gut microbiotal. The results showed that SCYP treatment could increase the digestive enzyme activities of colon contents and restore the production of short-chain fatty acids (SCFAs) in mice that were decreased by cyclophosphamide (Cy) treatment. Furthermore, SCYP treatment could modulate the structure of the gut microbiota, which was mainly manifest in the result of an increased abundances of Lactobacillus, Bacteroidetes and Akkermansia, and the decrease of the proportion of Proteobacteria and Verrucomicrobia in the microbial community. Diversity and overall structure of microbial community were also improved by SCYP treatment based on alpha-diversity and beta-diversity analysis results. The biomarkers of the gut microbial that were regulated by SCYP have been identified by linear discriminant analysis (LDA) effect size (LEfSe). These findings further indicate that there is great potential for SCYP to be developed into prebiotics or functional foods.
6,7-Dihydroxy-2,4-Dimethoxyphenanthrene from Chinese Yam Peels Alleviates DSS-Induced Intestinal Mucosal Injury in Mice via Modulation of the NF-κB/COX-2 Signaling Pathway

Abstract:In this study, we evaluated the protective effect and molecular mechanism of a dominant phenanthrene, (6,7-dihydroxy-2,4-dimethoxyphenanthrene, CYP4), from Chinese yam peels on intestinal epithelial integrity. Three doses of Chinese yam phenolic extract (CYPE) and Chinese yam phenanthrene 4 (CYP4) were administered to BALB/c mice for 7 days before dextran sulfate sodium (DSS) treatment, with berberine hydrochloride as a positive control (PC). Results showed that both disease activity indexes (DAIs), histological damage score (HDS) and survival rate in DSS mice, were improved with preintervention of CYPE and CYP4, which exhibited better efficiency than PC. Further studies showed that administration of CYP4 downregulated the oxidative stress-associated factors, MPO and NO, and improved tight junction protein occludin. Besides, the CYP4 treatment substantially downregulated the caspase-3 expression and the apoptosis rate of intestinal epithelial cells. In addition, the CYP4 treatment ameliorated the production of inflammatory cytokines including TNF-α, IFN-γ, IL-10, and IL-23 in the colon. Furthermore, the protein expression of ERK1/2, NF-κB p65, pNF-κB, and COX-2 was suppressed in CYE4 groups as compared with that in model control (MC). These findings suggested that CHP4 could effectively inhibit the activation of NF-κB/COX-2 in an experimental UC model in vivo. It was demonstrated for the first time that CYPE and CYP4 protected intestinal mucosa from damage and prevented DSS-induced colitis in mice. CYP4 was one of the active principles obligatory for the biological effect of Chinese yam in protecting intestinal health. These findings indicated that CYP4 might be a promising and useful approach for treatment of UC in humans.
