Abstract:The skin serves as the primary barrier between the human body and the external environment and is therefore susceptible to damage from various factors. Due to their high porosity, flexible and tunable structure, large surface area, abundant binding sites, and excellent chemical and thermal stability, metal-organic frameworks (MOFs) provide a new option for the treatment of skin wound healing. In the past decade, MOF-based therapeutics have made great achievements and therefore have a wide range of applications in biomedicine. This paper provides a comprehensive review of the recent advancements in metal-organic frameworks in wound healing and discusses their current research status and potential applications. We first summarize the mechanisms and latest applications of MOFs for wound healing. Specifically, the application of MOFs as antimicrobial agents by releasing antibacterial drugs to inhibit infections is discussed. Additionally, the potential of MOFs in promoting wound healing has been explored; for example, MOFs can serve as carriers for delivering therapeutic drugs and for promoting angiogenesis and synergistic antibacterial and anti-inflammatory effects in wound treatment. Then, we discuss the biosafety of MOFs and further explore the applications of MOFs. Finally, based on our understanding of the field, challenges and prospects are proposed, hoping to provide a basic understanding of the future development of MOFs.

Abstract:Wound healing is the most important restorative process for injury to the skin and other tissues. In the process of wound healing, bacterial infection can easily lead to serious tissue damage. There is an urgent need to discover new drugs or methods to accelerate wound healing. Metal-organic frameworks (MOFs), a type of porous hybrid materials containing metal ions connected by organic ligands, have great potential in improving wound healing. Firstly, this review discusses the pathophysiology of complex wounds, current approaches to achieve healing in patients and the limitations of traditional methods. Then the review emphasizes the application of MOF materials in improving wound healing. MOF materials-based metal ion sustained-release systems can show long-term antibacterial effects or promote angiogenesis in the degradation process of MOFs. As an enzyme-like active material, it can be used to convert hydrogen peroxide into highly toxic hydroxyl groups to improve antibacterial efficiency in wounds. As a controlled drug delivery system for wound healing, it is important for MOFs to load drugs (molecular or gas therapeutic drugs), effectively inhibiting the growth of microorganisms or enhancing angiogenesis in the wound site. Moreover, MOF materials-based photoexcited systems can provide the potential of combining photodynamic therapy and photothermal therapy for bacterial infections. Abstract:Various wounds caused by burns, trauma, surgery, and genetic irregularities, including acute and chronic wounds, pose a huge burden on global healthcare. Many therapeutic strategies, such as nanomedicine and regenerative medicine, have been applied to wound repair and skin regeneration. As a promising and exciting nanomaterial, metal–organic frameworks (MOFs) encompass many desirable properties, including crystalline porous structure, tunable size, good mechanical stability, large specific surface area, and exceptional chemical stability. Most importantly, the performance of MOFs can be modulated by changing synthesis steps and conditions. MOFs can release metal ions to modulate cellular behaviors and kill various microorganisms, which meets this dual agent release therapeutic strategy for wound healing and skin regeneration. Additionally, MOFs can act as carriers for delivering bioactive agents in a desired manner. Recently, an increasing number of researchers have conducted many studies on applying various MOFs in wound healing and skin regeneration. Based on this, we perform this study to report the current status and further perspective of MOFs-based therapeutic strategy. Furthermore, the molecular process of wound healing and the fabrication of MOFs were also discussed.

Abstract:Metal-organic frameworks (MOFs) are metal-organic skeleton compounds composed of self-assembled metal ions or clusters and organic ligands. MOF materials often have porous structures, high specific surface areas, uniform and adjustable pores, high surface activity and easy modification and have a wide range of prospects for application. MOFs have been widely used. In recent years, with the continuous expansion of MOF materials, they have also achieved remarkable results in the field of antimicrobial agents. In this review, the structural composition and synthetic modification of MOF materials are introduced in detail, and the antimicrobial mechanisms and applications of these materials in the healing of infected wounds are described. Moreover, the opportunities and challenges encountered in the development of MOF materials are presented, and we expect that additional MOF materials with high biosafety and efficient antimicrobial capacity will be developed in the future.

Abstract:Chronic wounds are characterized by delayed and dysregulated healing processes. As such, they have emerged as an increasingly significant threat. The associated morbidity and socioeconomic toll are clinically and financially challenging, necessitating novel approaches in the management of chronic wounds. Metal‐organic frameworks (MOFs) are an innovative type of porous coordination polymers, with low toxicity and high eco‐friendliness. Documented anti‐bacterial effects and pro‐angiogenic activity predestine these nanomaterials as promising systems for the treatment of chronic wounds. In this context, the therapeutic applicability and efficacy of MOFs remain to be elucidated. We, therefore, review structural‐functional properties of MOFs and their composite materials and discuss how their multifunctionality and customizability could be leveraged as a clinical therapy for chronic wounds. Enhanced Photocatalytic and Photothermal Properties of Ecofriendly Metal-Organic Framework Heterojunction for Rapid Sterilization

Abstract:Injured tissues are susceptible to infections that suppress wound healing. Rapid and safe sterilization is therefore urgent for bacteria-infected wounds, especially under harsh conditions without antibiotic availability. In this work, we developed a photoresponsive metal-organic framework (MOF) heterojunction that responds to 660 nm light irradiation. It is composed of two kinds of MOFs (Prussian blue [PB] and PCN-224), which exhibited an enhanced photocatalytic performance due to the accelerated charge transfer of the heterojunction and the fast separation of photogenerated electron-hole pairs between PB and PCN-224. The combination of the enhanced photocatalytic performance and the intrinsic photothermal effect of PB endowed the MOFs heterojunction with highly effective sterilizing rates of 99.84% and 99.3% against Staphylococcus aureus and its biofilm after 660 nm light irradiation for 15 min, respectively. The iron and zirconium ions released from PB-PCN-224 composites were biocompatible and their cytotoxicity was negligible. More importantly, in vivo experiments showed that PBPCN-224 can expedite wound healing.

Abstract:Recent advancements pertaining to the applications of MOFs and their composites for wound healing and dressings are deliberated, with the purpose of identifying knowledge gaps, evaluating challenges, and guiding future directions in the field. In recent years, there has been growing interest in developing innovative materials and therapeutic strategies to enhance wound healing outcomes, especially for chronic wounds and antimicrobial resistance. Metal–organic frameworks (MOFs) represent a promising class of materials for next‐generation wound healing and dressings. Their high surface area, pore structures, stimuli‐responsiveness, antibacterial properties, biocompatibility, and potential for combination therapies make them suitable for complex wound care challenges. MOF‐based composites promote cell proliferation, angiogenesis, and matrix synthesis, acting as carriers for bioactive molecules and promoting tissue regeneration. They also have stimuli‐responsivity, enabling photothermal therapies for skin cancer and infections. Herein, a critical analysis of the current state of research on MOFs and MOF‐based composites for wound healing and dressings is provided, offering valuable insights into the potential applications, challenges, and future directions in this field. This literature review has targeted the multifunctionality nature of MOFs in wound‐disease therapy and healing from different aspects and discussed the most recent advancements made in the field. In this context, the potential reader will find how the MOFs contributed to this field to yield more effective, functional, and innovative dressings and how they lead to the next generation of biomaterials for skin therapy and regeneration.

Abstract:Some infectious or malignant diseases such as cancers are seriously threatening the health of human beings all over the world. The commonly used antibiotic therapy cannot effectively treat these diseases within a short time, and also bring about adverse effects such as drug resistance and immune system damage during long-term systemic treatment. Phototherapy is an emerging antibiotic-free strategy to treat these diseases. Upon light irradiation, phototherapeutic agents can generate cytotoxic reactive oxygen species (ROS) or induce a temperature increase, which leads to the death of targeted cells. These two kinds of killing strategies are referred to as photodynamic therapy (PDT) and photothermal therapy (PTT), respectively. So far, many photo-responsive agents have been developed. Among them, the metal-organic framework (MOF) is becoming one of the most promising photo-responsive materials because its structure and chemical compositions can be easily modulated to achieve specific functions. MOFs can have intrinsic photodynamic or photothermal ability under the rational design of MOF construction, or serve as the carrier of therapeutic agents, owing to its tunable porosity. MOFs also provide feasibility for various combined therapies and targeting methods, which improves the efficiency of phototherapy. In this review, we firstly investigated the principles of phototherapy, and comprehensively summarized recent advances of MOF in PDT, PTT and synergistic therapy, from construction to modification. We expect that our demonstration will shed light on the future development of this field, and bring it one step closer to clinical trials.

Abstract:Wound healing is one of the most important issues in clinical and scientific research. The healing process is complex and requires many agents to overcome in a short duration. A recent class of porous materials called metal-organic frameworks (MOFs) has great potential towards improving wound healing. This is attributed to their well-designed structures with large surface areas amenable to cargo loading and adjustable pore size ready for biological implementations. MOFs are assembled by several metal centers and organic linkers. In particular, metal ions can be released from MOFs when they are degraded in the biological environment. This endows MOF-based systems with dual functions to typically shorten the healing duration. This work focuses on using MOFs with different metal centers such as copper (Cu), zinc (Zn), cobalt (Co), magnesium (Mg), and zirconium (Zr) for healing diabetic wounds as one of the most required clinical issues to be resolved. By figuring out the presented examples of this work, several potential research ideas can emerge to explore new porous materials or even new MOFs for more control over the healing process.

Abstract:Wound healing is a critical but complex biological process of skin tissue repair and regeneration resulting from various systems working together at the cellular and molecular levels. Quick wound healing and the problems associated with traditional wound repair techniques are being overcome with multifunctional materials. Over time, this research area has drawn significant attention. Metal-organic frameworks (MOFs), owning to their peculiar physicochemical characteristics, are now considered a promising class of well-suited porous materials for wound healing in addition to their other biological applications. This detailed literature review provides an overview of the latest developments in MOFs for wound healing applications. We have discussed the synthesis, essential biomedical properties, wound-healing mechanism, MOF-based dressing materials, and their wound-healing applications. The possible major challenges and limitations of MOFs have been discussed, along with conclusions and future perspectives. This overview of the literature review addresses MOFs-based wound healing from several angles and covers the most current developments in the subject. The readers may discover how the MOFs advanced this discipline by producing more inventive, useful, and successful dressings. It influences the development of future generations of biomaterials for the healing and regeneration of skin wounds.

Abstract:Cutaneous treatment possesses advantages, including the decrease of transient drug overdosage or systematic side effects. The evolution of biomedical materials with multifunctionalities to overcome the limitations of existing materials can bring revolutionary development in this field. A promising solution can turn to the emerging materials known as metal-organic frameworks (MOFs), featuring high porosity, easy modification, and multifunctionality. The work herein developed a systemic assay package to seek suitable MOFs for cutaneous applications. Versatile platforms based on MOFs were built, which integrated superb functions and features, including biocompatibilities, sebum adsorbability, selective adsorption, antimicrobial activity, controlled release, and skin permeability promotion. Such features endow MOFs as high-performance matrixes for cutaneous applications. This study paves a new avenue for the reformation of cosmetic and biomedicinal materials. Moreover, this comprehensive MOF screening system can be broadly applied for material evaluation in this field, which also provides valuable guidance for the design of advanced biomedical materials.

Abstract:Abstract The synergistic effect of antibacterial and anti-inflammatory is needed to overcome the problem of wound healing difficulties. Based on the favorable antibacterial and anti-inflammatory effect of zinc ions (Zn2+) and the physicochemical properties of metal organic frameworks (MOFs), we prepared nanosized zinc-based MOF: Zn-BTC with the ability to slowly release Zn2+. In cellular levels, Zn-BTC possessed lower toxicity to fibroblasts and enhanced capacity of cell proliferation and migration. It also had good bactericidal effect on multiple drug-resistant bacteria by reducing 41.4% MRSA and 47.2% Escherichia coli. In addition, Zn-BTC also displayed the ability of lowering the expression of antioxidant genes: superoxide dismutase 1, superoxide dismutase 2 and interleukin 6, and enhancing the expression of wound healing genes: transforming growth factors-β and type I collagen. Finally, it also demonstrated that Zn-BTC could effectively improve the skin wound healing of SD rats and had no toxicity on major organs. The favorable biocompatibility, antibacterial and anti-inflammatory properties of Zn-BTC gave a new insight of designing novel MOFs for promoting skin wound healing.

Abstract:The wide energy band gap and sluggish electron transfer efficiency of emerging metal-organic frameworks (MOFs) sonosensitizers severely impaired the therapeutic effectiveness of sonodynamic therapy (SDT). Herein, we prepare an engineered MOF composition consisting of multivariate MOFs and further in-situ surface decoration of Ag nanoparticles (Ag@UT) for enhanced SDT of bacterial infections in wound healing. By incorporating tetrakis(4-carboxyphenyl)porphyrin (TCPP) into zirconium-based MOFs and the surface decoration of Ag NPs, the energy band gap of the resultant multivariate MOF decreased from 3.45 eV to 2.11 eV, which greatly promote the generation of electron-hole pairs under ultrasound (US) excitation. Meanwhile, the Ag NPs co-catalysts accelerate the electron transfer and improve the efficiency of carrier utilization. Consequently, the Ag@UT presents a remarkable antibacterial effect in vitro and effectively promotes wound healing in vivo under US excitation, and the underlying Ag@UT-mediated bactericidal mechanism was firstly elucidated. This work provides insights into the design and optimization of MOFs component and structure via ingenious integration of ligands and surface decoration to achieve advanced SDT, paving the way for effective antibacterial treatments and other biomedical research.

Abstract:Bacterial infections pose a huge threat to human health due to the inevitable emergency of drug resistance. Metal-organic frameworks (MOFs) consisting of metal ions and organic linkers, as emerging efficient antibacterial material, have the merits of structural flexibility and adjustable physicochemical property. With assistance of photosensitive agents as organic linkers, MOFs have great potential in antibacterial application through photocatalytic therapy by the generation of reactive oxygen species (ROS). However, the limited light use efficiency and short lifespan of ROS are two obstacles for their applications. Inspired by the semiconductor heterostructure in photocatalysis, we rationally design and precisely synthesize MOFs based heterostructures, in which the TiO 2 nanoclusters are filled into the pores of Cu-TCPP nanosheets (i.e. TiO 2 NCs@Cu-TCPP HSs). And the composite materials possess three-dimensional (3D) hierarchical architectures, which have advantages of large surface area, excellent light-absorbing ability and photocatalytic efficiency. Significantly, this novel material displays >99.99% antibacterial efficiency against E. coli and S. aureus within 30 min and preserves the excellent antibacterial ability during reusing three times, which is superior to recently reported photocatalystic-based antibacterial materials. Our study provides new insights into the energy band engineering for enhanced antibacterial performance, paving a way for designing advanced clinical wound dressings.

Abstract:Abstract Bismuth metal–organic frameworks (MOFs) as catalysts are scarce. Herein, the first MOF comprising meso‐ tetra(4‐carboxyphenyl) porphine (TCPP) as an organic ligand and Bi ion as a metal node, denoted as Bi‐TCPP, is synthesized by a convenient one‐step hydrothermal method. Compared to other 2D (Cu‐TCPP) or 3D (PCN‐222) MOFs with TCPP as a ligand, Bi‐TCPP has ultrafast singlet oxygen generation ability and a high photothermal conversion efficiency of 49.5% under the irradiation of 660 nm light. This is due to Bi‐TCPP's strong ligand‐to‐metal charge transfer (LMCT) events that form singlet oxygen ( 1 O 2 ). Further, when the electrons and holes above the bandgap relax to the band edge, the excess energy is converted into heat. In vitro and in vivo experiments show excellent antibacterial and repair effects. Hence, the synthesis of Bi‐TCPP provides new ideas for the study of rare bismuth‐based MOFs, and its excellent biocompatibility provides a possibility for further application.

Abstract:Diabetes mellitus is an exponentially growing chronic metabolic disease identified by prolonged hyperglycemia that leads to a plethora of health problems. It is well established that the skin of diabetic patients is more prone to injury, and hence, wound healing is an utmost critical restorative process for injured skin and other tissues. Diabetes patients have problems with wound healing at all stages, which ultimately results in delays in the healing process. Therefore, it is vital to find new medications or techniques to hasten the healing of wounds. Metal-organic frameworks (MOFs), an assorted class of porous hybrid materials comprising metal ions coordinated to organic ligands, can display great potential in accelerating diabetic wound healing due to their good physicochemical properties. The release of metal ions during the degradation of MOFs can promote the differentiation of fibroblasts into myofibroblasts and subsequently angiogenesis. Secondly, similar to enzyme-like active substances, they can eliminate reactive oxygen species (ROS) overproduction (secondary to the bio-load of wound bacteria), which is conducive to accelerating diabetic wound healing. Subsequently, MOFs can support the slow release of drugs (molecular or gas therapeutics) in diabetic wounds and promote wound healing by regulating pathological signaling pathways in the wound microenvironment or inhibiting the expression of inflammatory factors. In addition, the combination of photodynamic and photothermal therapies using photo-stimulated porphyrin-based MOF nanosystems has brought up a new idea for treating complicated diabetic wound microenvironments. In this review, recent advances affecting diabetic wound healing, current means of rapid diabetic wound healing, and the limitations of traditional approaches are discussed. Further, the diabetic wound healing applications of MOFs have been discussed followed by the future challenges and directions of MOF materials in diabetic wound healing.

In this study, a metal-organic framework (MOF), Fe-MIL88B-NH2, was incorporated with photosensitizer indocyanine green (ICG) to construct composite nanoparticles (MOF@ICG NPs) with multiple antibacterial activities. Under mild near-infrared (NIR) irradiation, the photosensitizer ICG in the MOF@ICG NPs undergoes photothermal conversion (~45 °C) and photodynamic reactions to generate heat and singlet oxygen (1O2). In addition, the Fenton reaction of the NPs with Hydrogen peroxide (H2O2) in the bacterial infection microenvironment resulted in the generation of hydroxyl radicals (·OH), thus achieving the three-mode combination of low-temperature photothermal therapy (PTT)/photodynamic therapy (PDT)/chemodynamic therapy (CDT). The experimental results in vitro showed that MOF@ICG MPs had excellent antibacterial property and good cytocompatibility, which presented some ability to promote the migration of L-929 fibroblasts. Furthermore, under NIR irradiation, MOF@ICG NPs could significantly kill bacteria and promote skin wound healing according to the results of animal experiments. The wound healing rate reached 87.1% after 7 days of treatment. The research results break through the limitations of single-mode antibacterial technology, and provide certain theoretical guidance and technical support for the research and development of new antibacterial materials.

Abstract:Metal-organic frameworks (MOFs) have received extensive attention in tumor therapy because of their advantages, including large specific surface area, regular pore size, adjustable shape, and facial functionalization. MOFs are porous materials formed by the coordination bonding of metal clusters and organic ligands. This review summarized the most recent advancements in tumor treatment based on nMOFs. First, we discuss the classification of MOFs, which primarily include the series of Isoreticular MOF (IRMOF), Zeolitic imidazolate framework (ZIF), Coordination Pillared-Layer (CPL), Materials of Institute Lavoisier (MIL), Porous Coordination Network (PCN), University of Oslo (UiO) and Biological metal-organic frameworks (BioMOFs). Then, we discuss the use of nMOFs to anticancer therapy, including drug delivery strategies, photodynamic therapy (PDT), photothermal therapy (PTT) and combination therapy. Finally, the obstacles and opportunities in nMOFs are discussed.

Integrating Porphyrinic Metal-Organic Frameworks in Nanofibrous Carrier for Photodynamic Antimicrobial Application

Abstract:The rise and spread of antimicrobial resistance is creating an ever greater challenge in wound management. Nanofibrous membranes (NFMs) incorporated with antibiotics have been widely used to remedy bacterial wound infections owing to their versatile features. However, misuse of antibiotics has resulted in drug resistance, and it remains a significant challenge to achieve both high antibacterial efficiency and without causing bacterial resistance. Here, the ‘MOF-first’ strategy was adopted, the porphyrinic metal-organic frameworks nanoparticles (PCN−224 NPs) were pre-synthesized first, and then the composite antibacterial PCN−224 NPs @ poly (ε-caprolactone) (PM) NFMs were fabricated via a facile co-electrospinning technology. This strategy allows large amounts of effective MOFs to be integrated into nanofibers to effectively eliminate bacteria without bacterial resistance and to realize a relatively fast production rate. Upon visible light (630 nm) irradiation for 30 min, the PM−25 NFMs have the best 1O2 generation performance, triggering remarkable photodynamic antibacterial effects against both S. aureus, MRSA, and E. coli bacteria with survival rates of 0.13%, 1.91%, and 2.06% respectively. Considering the photodynamic antibacterial performance of the composite nanofibrous membranes functionalized by porphyrinic MOFs, this simple approach may provide a feasible way to use MOF materials and biological materials to construct wound dressing with the versatility to serve as an antibacterial strategy in order to prevent bacterial resistance.

Abstract:Diabetes-related chronic wounds are often accompanied by a poor wound-healing environment such as high glucose, recurrent infections, and inflammation, and standard wound treatments are fairly limited in their ability to heal these wounds. Metal–organic frameworks (MOFs) have been developed to improve therapeutic outcomes due to their ease of engineering, surface functionalization, and therapeutic properties. In this review, we summarize the different synthesis methods of MOFs and conduct a comprehensive review of the latest research progress of MOFs in the treatment of diabetes and its wounds. State-of-the-art in vivo oral hypoglycemic strategies and the in vitro diagnosis of diabetes are enumerated and different antimicrobial strategies (including physical contact, oxidative stress, photothermal, and related ions or ligands) and provascular strategies for the treatment of diabetic wounds are compared. It focuses on the connections and differences between different applications of MOFs as well as possible directions for improvement. Finally, the potential toxicity of MOFs is also an issue that we cannot ignore.
