HCN2 Deficiency Causes Cognitive Impairments Via Inducing Hyperexcitability of Dca1 Pyramidal Neurons in a Mouse Model of Alzheimer's Disease

Abstract:Aberrant activity of hippocampal neurons may lead to dysfunction of neural circuits and then causes cognitive impairments in Alzheimer’s disease (AD). However, the mechanisms underlying the abnormal activity of hippocampal neurons in AD remain to be fully elucidated. Here we found that the function of the hyperpolarization-activated cyclic nucleotide-gated (HCN) ion channels was impaired and the expression of HCN2 was reduced in the hippocampus in hAPP-J20 mice, a mouse model of Alzheimer’s disease. Importantly, dysregulation of the HCN channels contributed to the hyperactivity of dorsal CA1 (dCA1) pyramidal neurons (PNs) in hAPP-J20 mice. Furthermore, stimulating HCN channels or restoring HCN2 attenuated the hyperexcitability of CA1 PNs and improved the cognitive functions of hAPP-J20 mice. Finally, direct inhibition of CA1 PNs ameliorated the cognitive deficits in hAPP-J20 mice.
