Abstract:Imaging and tracking of near-surface three-dimensional volumetric nanoscale dynamic processes of live cells remains a challenging problem. In this paper, we propose a multi-color live-cell near-surface-volume super-resolution microscopy method that combines total internal reflection fluorescence structured illumination microscopy with multi-angle evanescent light illumination. We demonstrate that our approach of multi-angle interference microscopy is perfectly adapted to studying subcellular dynamics of mitochondria and microtubule architectures during cell migration.

Abstract:Super-resolution microscopy techniques are versatile and powerful tools for visualizing organelle structures, interactions, and protein functions in biomedical research, and its resolution ability to break the optical diffraction limit provides new analytical frameworks for cell biology on the nanoscale, which is indispensable to life science related fields. However, due to the effect of the diffraction limit, the axial resolution of a super-resolution microscope is more arduous to improve than the lateral resolution, which hinders the realization of sub-hundred-nanometer resolution three-dimensional imaging of cellular structures. Therefore, based on the two main techniques, stimulated emission depletion microscopy and single-molecule localization microscopy, the present paper introduces the principles and characteristics of a variety of existing three-dimensional imaging techniques, and finally discusses the future of that. Finally, we briefly discuss the research trend of the two techniques in the three-dimensional imaging area.

Abstract:The resolution of conventional optical equipment is always restricted by the diffraction limit, and improving on this was previously considered improbable. Optical super-resolution imaging, which has recently experienced rapid growth and attracted increasing global interest, will result in applications in many domains, benefiting fields such as biology, medicine and material research. This review discusses the contributions of different researchers who identified the diffractive barrier and attempted to realize optical super-resolution. This is followed by a personal viewpoint of the development of optical nanoscopy in recent decades and the road towards the next generation of optical nanoscopy.

Abstract:Achieving nanometer-scale resolution remains challenging in expansion microscopy due to photon loss. To address this concern, here we develop a multi-color expansion stimulated emission depletion technique based on small-molecule probes to realize high labeling density and intensity. Our method substantially lowers the barrier to visualizing diverse intracellular proteins and their interactions in three dimensions. It enables us to achieve sub-10-nm resolution in structures such as microfilaments, lysosomes, and mitochondria, providing new insights into cell biology.

Abstract:Fluorescence super-resolution microscopy has undergone several rounds of innovation and development since the 1990s. The spatial resolution of fluorescence super-resolution microscopy exceeds the diffraction limit by a wide margin, achieving a lateral resolution below to similar to 20 nm and enabling imaging and tracking at the molecular level. The latest generation of ultrahigh-resolution microscopy is the product of extensive development and innovative combination of conventional super-resolution techniques. This review aimed to introduce the new generation of fluorescence super-resolution microscopy at a sub-20-nm scale lateral resolution, and we also elucidated their similarities and differences compared with the conventional super-resolution techniques. Additionally, this review explored how emerging technologies optimize optical systems, scanning strategies, and sample preparation to improve resolution. Finally, we discussed the potential applications and development of high-resolution fluorescence microscopy in biomedical research.

Abstract:Optical microscopy is non-invasive, sample-friendly, and fast, rendering itself the major approach for human beings to explore the microcosmic world. However, the diffraction limit has hindered the resolution of all optical imaging systems to approximately half the wavelength of visible light for over a century, until it was fundamentally broken by the development of super-resolution optical fluorescence microscopy. This technology bridges the gap between the electron microscopes (1 nm) and the ordinary optical microscopes (200 nm to 250 nm), but it is essentially useless for most samples, especially those non-fluorescent-labeled. In recent years, inspired by the synthetic aperture imaging, we have developed the optical super-resolution imaging based on frequency shift, providing a new approach to optical super-resolution imaging. As the technique is not limited by the nonlinear effects of fluorescence, it is applicable for both non-fluorescent-labeled and fluorescent-labeled samples. Besides, due to such advantages as fast imaging, high sample universality, and low phototoxicity, the technique shows good prospects in multiple fields, including materials science, biology, and medicine. In this paper, we deliver a detailed overview on both the principles and methods of optical super-resolution imaging based on frequency shift, as well as our prospects to the future development direction of this technique.

Abstract:The investigation of the cell biology with fluorescence microscopy remains challenging because of the requirement of both high temporal and spatial resolution. In this paper, parallelized fluorescence emission difference microscopy (pFED) is presented to provide 2-fold increasing imaging speed than conventional FED and achieve high spatial resolution (0.2 λ–0.3 λ) with a novel system design. The super-resolution images are obtained by aligning and subtracting two images, that are acquired simultaneously using parallelized scanning solid- and donut-shaped excitation beams with a lateral offset between two foci. Herein, we demonstrate the general applicability of pFED with time-lapse imaging of various subcellular dynamics in live cells over an extended period.

Abstract:For decades, the spatial resolution of conventional far‐field optical imaging has been constrained due to the diffraction limit. The emergence of optical super‐resolution imaging has facilitated biological research in the nanoscale regime. However, the existing super‐resolution modalities are not feasible in many biological applications due to weaknesses, like complex implementation and high cost. Recently, various newly proposed techniques are advantageous in the enhancement of the system resolution, background suppression, and improvement of the hardware complexity so that the above‐mentioned issues could be addressed. Most of these techniques entail the modification of factors, like hardware, light path, fluorescent probe, and algorithm, based on conventional imaging systems. Particularly, subtraction technique is an easily implemented, cost‐effective, and flexible imaging tool which has been applied in widespread utilizations. In this review, the principles, characteristics, advances, and biological applications of these techniques are highlighted in optical super‐resolution modalities.

Abstract:In article number 1900126, Xu Liu, Qing Yang, and co-workers present a waveguide chip design that permits label-free superresolution imaging. The method captures all sample spatial frequencies over a large bandwidth, permitting artifact-free reconstruction. Two-dimensional sample structures are imaged without distortion and reconstructed over large field of views at sub-wavelength resolution. The results match well with data obtained via SEM and AFM imaging.

Abstract:Cell is the basic unit and functional unit of living body. The study of the internal structure and function of living cells is one of the foundations of mastering the essence of life. Therefore, the real-time observation of living cells is of great significance for the development of life sciences. Conventional optical microscopy is limited by the diffraction limits and can not observe the details of biological structures below 200 nm. In the past 20 years, with the rapid development of super-diffraction limit optical theory, technology, devices and fluorescent probes, super-resolution microscopy has become an important method for life science research. However, most super-resolution microscopic methods or measurements take a long time, or are likely to cause photobleaching/phototoxicity, and are severely limited in living cell studies. In this paper, based on the study of fast super-resolution microscopy, the photoactivated localization microscopy and stochastic optical reconstruction microscopy were introduced based on single molecule localization microscopy. The stimulated emission depletion microscopy based on fluorescence non -linear saturated light conversion and structured illumination microscopy based on structured light illumination was also introduced. Besides, the development and application of cell imaging were explored. Finally, an outlook of the future development trend of super-resolution microscopy in living cell imaging was provided.

Abstract:Fluorescence microscopy has become an essential tool for biological research because it can be minimally invasive, acquire data rapidly, and target molecules of interest with specific labeling strategies. However, the diffraction-limited spatial resolution, which is classically limited to about 200 nm in the lateral direction and about 500 nm in the axial direction, hampers its application to identify delicate details of subcellular structure. Extensive efforts have been made to break diffraction limit for obtaining high-resolution imaging of a biological specimen. Various methods capable of obtaining super-resolution images with a resolution of tens of nanometers are currently available. These super-resolution techniques can be generally divided into three primary classes: (1) patterned illumination-based super-resolution imaging, which employs spatially and temporally modulated illumination light to reconstruct sub-diffraction structures; (2) single-molecule localization-based super-resolution imaging, which localizes the profile center of each individual fluorophore at subdiffraction precision; (3) bleaching/blinking-based super-resolution imaging. These super-resolution techniques have been utilized in different biological fields and provide novel insights into several new aspects of life science. Given unique technical merits and commercial availability of super-resolution fluorescence microscope, increasing applications of this powerful technique in life science can be expected.

Abstract:Super resolution microscopy based on single-molecule localization relies on precise and accurate localization of large numbers of single-molecules. However, the necessity of accumulating large numbers of localization estimates limits the time resolution typically to seconds to minutes1,2. Two major limitations are the acquisition speed and the photon budget. Replacing the usually used EMCCD with a recently introduced sCMOS camera results in leaps in both acquisition speed and effective quantum efficiency. However, the intrinsic pixel-dependent Gaussian noise of the sCMOS cameras introduces localization artifacts and greatly reduces the reliability of the results. Here, we present a set of specially designed methods that characterize an sCMOS camera for the first time and allow for unbiased and precise localization analysis. Using this method we demonstrate Cramer-Rao lower bound-limited single-molecule localization with an sCMOS camera. Combining the novel algorithm with a recently developed multi-emitter fitting algorithm3, We shortens the typical acquisition time for fixed samples by up to two orders of magnitude without compromising the field of view. Furthermore, we demonstrate localization-based super-resolution microscopy in live cells by monitoring dynamics of protein clusters, vesicles and organelles at a temporal resolution from 2 to 30 frames per second4. These methods allowed us to investigate cytokinetic apparatus in live fission yeast at 20-30 nm resolution. In general, the significantly improved temporal resolution allows super resolution imaging of a large range of dynamic events in living cells. 1. Application of super-resolution optical microscopy in biology

Abstract:Background: A noninvasive, real-time far-field optical microscopy is needed to study the dynamic function inside cells and proteins. However, the resolution limit of traditional optical microscope is about 200 nm due to the diffraction limit of light. So, it's hard to directly observe the subcellular structures. Over the past several years of microscopy development, the diffraction limit of fluorescence microscopy has been overcome and its resolution limit is about tens of nanometers. Methods: To overcome the diffraction limit of light, many super-resolution fluoresce microcopies, including stimulated emission of depletion microscopy (STED), photoactivation localization microscopy (PALM) and stochastic optical reconstruction microscopy (STORM), have been developed. Conclusions: These methods have been applied in cell biology, microbiology and neurobiology, and the technology of super-resolution provides a new insight into the life science.

Abstract:<p>With a spatial resolution breaking the diffraction limit of light, superresolution imaging allows the visualization of detailed structures of organelles such as mitochondria, cytoskeleton, nucleus, and so on. With multi-dimensional imaging (x, y, z, t, λ), namely, multi-color 3D live imaging enables us fully understand the function of the cell. It is necessary to analyze structural changes or molecular interactions across a large volume in 3D with different labelled targets. To achieve this goal, scientists recently have expanded the original 2D superresolution microscopic tools into 3D imaging techniques. In this review, we will discuss recent development in superresolution microscopy for live imaging with minimal phototoxicity. We will focus our discussion on the cell nucleus where the genetic materials are stored and processed. Machine learning algorism will be introduced to improve the axial resolution of superresolution imaging.</p>

Abstract:Researchers can now identify dynamic activitiesliving cells at the nanoscale with remarkable temporal and spatial resolution because of the advancement in fluorescent super -resolution imaging. Traditional perresolution microscopy requires high --power lasers or numerous raw images to rebuild a single super --resolution image, limiting its applicadons in live cell dynamic imaging. In many ways, deep learning -driven super -resolution imaging approaches break the bottleneck of existing super resolutionimaging technology. In this review, we explain the theory of optical super-resolution imaging systems and discuss their limitations. Furthermore, we outline the most recent advances and applications of deep learning in the field of super --resolution imaging, as well as address challenging difficulties and future possibilities.

Abstract:Abstract Single-molecule localization microscopy (SMLM) can be used to resolve subcellular structures and achieve a tenfold improvement in spatial resolution compared to that obtained by conventional fluorescence microscopy. However, the separation of single-molecule fluorescence events that requires thousands of frames dramatically increases the image acquisition time and phototoxicity, impeding the observation of instantaneous intracellular dynamics. Here we develop a deep-learning based single-frame super-resolution microscopy (SFSRM) method which utilizes a subpixel edge map and a multicomponent optimization strategy to guide the neural network to reconstruct a super-resolution image from a single frame of a diffraction-limited image. Under a tolerable signal density and an affordable signal-to-noise ratio, SFSRM enables high-fidelity live-cell imaging with spatiotemporal resolutions of 30 nm and 10 ms, allowing for prolonged monitoring of subcellular dynamics such as interplays between mitochondria and endoplasmic reticulum, the vesicle transport along microtubules, and the endosome fusion and fission. Moreover, its adaptability to different microscopes and spectra makes it a useful tool for various imaging systems.

Abstract:Understanding the interactions between nanomaterials and biological systems plays an essential role in enhancing the efficacy of nanomedicines and deepening the understanding of the biological domain. Fluorescence microscopy is a powerful optical imaging technique that allows direct visualization of the behavior of fluorescent-labeled nanomaterials in the intracellular microenvironment. However, conventional fluorescence microscopy, such as confocal microscopy, has limited optical resolution due to the diffraction of light and therefore cannot provide the precise details of nanomaterials with diameters of less than similar to 250 nm. Fortunately, the development of super-resolution fluorescence microscopy has overcome the resolution limitation, enabling more comprehensive studies of nano-cell interactions. Herein, we have summarized the recent advances in nano-cell interactions investigated by a variety of super-resolution microscopic techniques, which may benefit researchers in this multi-disciplinary area by providing a guideline to select appropriate platforms for studying materiobiology.

Abstract:Super-resolution fluorescence microscopy is a powerful tool to visualize biomolecules and cellular structures at the nanometer scale. Employing these techniques in living cells has opened up the possibility to study dynamic processes with unprecedented spatial and temporal resolution. Different physical approaches to super-resolution microscopy have been introduced over the last years. A bottleneck to apply these approaches for live-cell imaging has become the availability of appropriate fluorescent probes that can be specifically attached to biomolecules. In this Perspective, we discuss the role of small-molecule fluorescent probes for live-cell super resolution microscopy and the challenges that need to be overcome for their generation. Recent trends in the development of labeling strategies are reviewed together with the required chemical and spectroscopic properties of the probes. Finally, selected examples of the use of small-molecule fluorescent probes in live-cell super resolution microscopy are given.

Abstract:Super-resolution microscopy is rapidly developed in recent years, allowing biologists to extract more quantitative information on subcellular processes in live cells that is usually not accessible with conventional techniques. However, super-resolution imaging is not fully exploited because of the lack of an appropriate and multifunctional experimental platform. As an important tool in life sciences, microfluidics is capable of cell manipulation and the regulation of the cellular environment because of its superior flexibility and biocompatibility. The combination of microfluidics and super-resolution microscopy revolutionizes the study of complex cellular properties and dynamics, providing valuable insights into cellular structure and biological functions at the single-molecule level. In this perspective, an overview of the main advantages of microfluidic technology that are essential to the performance of super-resolution microscopy are offered. The main benefits of performing super-resolution imaging with microfluidic devices are highlighted and perspectives on the diverse applications that are facilitated by combining these two powerful techniques are provided.
