Abstract:AbstractLeaf functional traits have attracted the attention of ecologists for several decades, but few studies have systematically assessed leaf morphological traits (termed “economic traits”), stomatal (termed “hydraulic”), and anatomical traits of entire forest communities, thus it is unclear whether their relationships are consistent among trees, shrubs, and herbs, and which anatomical traits should be assigned to economical or hydraulic traits. In this study, we collected leaf samples of 106 plant species in temperate forests and 164 plant species in subtropical forests and determined nine key functional traits. We found that functional traits differed between temperate and subtropical forests. Leaf traits also differed between different plant functional groups, irrespective of forest type; dry matter content, stomatal density, and cell tense ratio followed the order trees > shrubs > herbs, whereas specific leaf area and sponginess ratio showed the opposite pattern. The correlations of leaf traits were not consistent among trees, shrubs, and herbs, which may reflect different adaptive strategies. Principal component analysis indicated that leaf economics and hydraulic traits were uncoupled in temperate and subtropical forests, and correlations of anatomical traits and economic and hydraulic traits were weak, indicating anatomical traits should be emphasized in future studies.

Abstract:Mounting evidence has shown strong linkage of root function with root branch order. However, it is not known whether this linkage is consistent in different species. Here, root anatomic traits of the first five branch order were examined in five species differing in plant phylogeny and growth form in tropical and subtropical forests of south China. In Paramichelia baillonii, one tree species in Magnoliaceae, the intact cortex as well as mycorrhizal colonization existed even in the fifth-order root suggesting the preservation of absorption function in the higher-order roots. In contrast, dramatic decreases of cortex thickness and mycorrhizal colonization were observed from lower- to higher-order roots in three other tree species, Cunninghamia lanceolata, Acacia auriculiformis and Gordonia axillaries, which indicate the loss of absorption function. In a fern, Dicranopteris dichotoma, there were several cortex layers with prominently thickened cell wall and no mycorrhizal colonization in the third- and fourth-order roots, also demonstrating the loss of absorptive function in higher-order roots. Cluster analysis using these anatomic traits showed a different classification of root branch order in P. baillonii from other four species. As for the conduit diameter-density relationship in higher-order roots, the mechanism underpinning this relationship in P. baillonii was different from that in other species. In lower-order roots, different patterns of coefficient of variance for conduit diameter and density provided further evidence for the two types of linkage of root function with root branch order. These linkages corresponding to two types of ephemeral root modules have important implication in the prediction of terrestrial carbon cycling, although we caution that this study was pseudo-replicated. Future studies by sampling more species can test the generality of these two types of linkage.
Leading dimensions in absorptive root trait variation across 96 subtropical forest species

Abstract:Absorptive root traits show remarkable cross-species variation, but major root trait dimensions across species have not been defined. We sampled first-order roots and measured 14 root traits for 96 angiosperm woody species from subtropical China, including root diameter, specific root length, stele diameter, cortex thickness, root vessel size and density, mycorrhizal colonization rate, root branching intensity, tissue density, and concentrations of carbon and nitrogen ([N]). Root traits differed in the degree of variation and phylogenetic conservatism, but showed predictable patterns of cross-trait coordination. Root diameter, cortex thickness and stele diameter displayed high variation across species (coefficient of variation (CV)=0.51-0.69), whereas the stele:root diameter ratio and [N] showed low variation (CV<0.32). Root diameter, cortex thickness and stele diameter showed a strong phylogenetic signal across species, whereas root branching traits did not, and these two sets of traits were segregated onto two nearly orthogonal (independent) principal component analysis (PCA) axes. Two major dimensions of root trait variation were found: a diameter-related dimension potentially integrating root construction, maintenance, and persistence with mycorrhizal colonization, and a branching architecture dimension expressing root plastic responses to the environment. These two dimensions may offer a promising path for better understanding root trait economics and root ecological strategies world-wide.

Abstract:The roots of the majority of tree species are associated with either arbuscular mycorrhizal (AM) or ectomycorrhizal (EM) fungi. The absorptive roots of tree species also vary widely in their diameter. The linkages between root thickness, mycorrhiza type and nutrient foraging are poorly understood. We conducted a large root ingrowth experiment in the field to investigate how absorptive roots of varying thickness and their associated fungi (AM vs. EM) exploit different nutrient patches (inorganic and organic) in a common garden. In nutrient-rich patches, thin-root tree species more effectively proliferated absorptive roots than thick-root tree species, whereas thick-root tree species proliferated more mycorrhizal fungal biomass than thin-root tree species. Moreover, nutrient patches enriched with organic materials resulted in greater root and mycorrhizal fungal proliferation compared to those enriched with inorganic nutrients. Irrespective of root morphology, AM tree species had higher root foraging precision than mycorrhizal hyphae foraging precision within organic patches, whereas EM tree species exhibited the opposite. Our findings that roots and mycorrhizal fungi are complementary in foraging within nutrient patches provide new insights into species coexistence and element cycling in terrestrial ecosystems.

Abstract:Root hypodermis passage cells (PCs) lack suberin and lignin deposition, responsible for nutrient absorption and arbuscular mycorrhizal colonization, which are crucial for root resource acquisition. Nevertheless, their quantitative variability across diverse woody species and their relationships with root morphology and anatomy, as well as arbuscular mycorrhizal colonization, are still not well understood. Herein, the number and proportion of PCs in the root cross-section, root morphology, anatomy, and arbuscular mycorrhizal colonization rate were quantified across 10 temperate and 12 tropical woody species. The objectives of this study were to determine how PCs vary across contrasting environmental conditions and to explore their relationships with arbuscular mycorrhizal colonization rate and other root functional traits. The results showed that tropical species possessed 56% more PCs than temperate species; by contrast, they had similar proportions of PCs. In both biomes, the number of PCs had a tightly positive correlation with arbuscular mycorrhizal colonization rate (R2 = 0.35–0.87), root diameter (R2 = 0.84–0.93), and cortex thickness (R2 = 0.87–0.89), but the proportion of PCs was mostly independent of root morphological and anatomical traits. Our results suggest that variation in passage cells could well explain the tight linkage between arbuscular mycorrhizal colonization and root diameter across species and biomes, which provides insight into the collaboration gradient between plant roots and mycorrhizal fungi that dominates the root economics space.

Abstract:Root anatomical traits play crucial roles in understanding root functions and root form-function linkages. However, the root anatomy and form-function linkages of monocotyledonous and dicotyledonous herbs remain largely unknown. We measured order-based anatomical traits and mycorrhizal colonization rates of 32 perennial herbs of monocotyledons and dicotyledons in a temperate steppe. For monocots, relative constant proportion of cortex and mycorrhizal colonization rates, but increased cell-wall thickening of the endodermis and proportion of stele were observed across root orders, indicating a slight reduction in absorption capacity and improvement in transportation capacity across orders. For dicots, the cortex and mycorrhizal colonization disappeared in the fourth-order and/or fifth-order roots, whereas the secondary vascular tissue increased markedly, suggesting significant transition of root functions from absorption to transportation across root orders. The allometric relationships between stele and cortex differed across root orders and plant groups, suggesting different strategies to coordinate the absorption and transportation functions among plant groups. In summary, our results revealed different functional transition patterns across root orders and distinct strategies for coordinating the absorption and transportation of root system between monocots and dicots. These findings will contribute to our understanding of the root form and functions in herbaceous species.

Abstract:Plant trait‐based approaches are frequently used to explore the linkages between aboveground plant communities and belowground ecosystem functions. However, the role of plant leaf litter and living root traits in driving soil microbial biomass, community composition, and enzyme activities has rarely been explored. Here, we measured the soil microbial biomass, community composition and enzyme activities related to carbon (C), nitrogen (N), and phosphorus (P) acquisition under three‐year‐old monocultures of 28 common subtropical tree species in China. We found that plant leaf litter and absorptive root traits, including leaf litter C content, litter water holding capacity, and root N content, were the three best predictors for soil microbial biomass and enzyme activities. In particular, resource‐exploitative tree species with higher root N contents were associated with microbial communities with lower fungi to bacteria ratios and lower C‐ and P‐acquisition enzyme activities. Tree species with higher leaf litter water holding capacity were associated with microbial resource acquisition strategies for C and P acquisition. Synthesis: Our findings highlighted that plant leaf litter and root traits are important for mechanistically understanding the ecological linkage between the plant community and ecosystem functions.

Abstract:The root economics space (RES) is multidimensional and largely shaped by belowground biotic and abiotic influences. However, how root-fungal symbioses and edaphic fertility drive this complexity remains unclear. Here, we measured absorptive root traits of 112 tree species in temperate and subtropical forests of China, including traits linked to functional differences between arbuscular mycorrhizal (AM) and ectomycorrhizal (ECM) hosts. Our data, from known mycorrhizal tree species, revealed a 'fungal-symbiosis' dimension distinguishing AM from ECM species. This divergence likely resulted from the contrasting mycorrhizal evolutionary development of AM vs ECM associations. Increased root tissue cortical space facilitates AM symbiosis, whereas increased root branching favours ECM symbiosis. Irrespective of mycorrhizal type, a 'root-lifespan' dimension reflecting aspects of root construction cost and defence was controlled by variation in specific root length and root tissue density, which was fully independent of root nitrogen content. Within this function-based RES, we observed a substantial covariation of axes with soil phosphorus and nitrate levels, highlighting the role played by these two axes in nutrient acquisition and conservation. Overall, our findings demonstrate the importance of evolved mycorrhizal symbiosis pathway and edaphic fertility in framing the RES, and provide theoretical and mechanistic insights into the complexity of root economics.

Abstract:Revealing the relationship between plants and root-associated fungi is very important in understanding diversity maintenance and community assembly in ecosystems. However, the community assembly of root-associated fungi of focal plant species along a subtropical plant species diversity gradient is less documented. Here, we examined root-associated fungal communities associated with five ectomycorrhizal (EM) plant species (Betula luminifera, Castanea henryi, Castanopsis fargesii, C. sclerophylla, and Quercus serrate) in a Chinese subtropical woody plant species diversity (1, 2, 4, 8, 16 and 24 species) experiment, using paired-end Illumina MiSeq sequencing of the ITS2 region. In total, we detected 1933 root-associated fungal operational taxonomic units (OTUs) at a 97% sequence similarity level. Plant identity had a significant effect on total and saprotrophic fungal OTU richness, but plant species diversity level had a significant effect on saprotrophic and pathogenic fungal OTU richness. The community composition of total, saprotrophic and EM fungi was structured by plant identity and plant species diversity level. However, the community composition of pathogenic fungi was only shaped by plant identity. This study highlights that plant identity has a stronger effect on the root-associated fungal community than plant species diversity level in a diverse subtropical forest ecosystem.

Abstract:Traits are critical in predicting decomposition that fuels carbon and nutrient cycling in ecosystems. However, our understanding of root trait‐decomposition linkage and especially its dependence on mycorrhizal type and environmental context remains limited.We explored the control of morphological and chemical (carbon‐ and nutrient‐related) traits over decomposition of absorptive roots in 30 tree species associated with either arbuscular mycorrhizal (AM) or ectomycorrhizal (ECM) fungi in temperate and subtropical forests in China.Carbon‐related traits (acid‐unhydrolyzable residue (AUR) and cellulose concentrations) had a predominant control of root decomposition in AM species while nutrient‐related traits (magnesium concentration) predominately controlled that in ECM species. Thicker absorptive roots decomposed faster in AM species due to their lower AUR concentrations, but more slowly in ECM angiosperm species potentially due to their higher magnesium concentrations. Root decomposition was linked to root nutrient economy in both forests while root diameter‐decomposition coordination emerged only in the subtropical forest where root diameter and decomposition presented similar cross‐species variations.Our findings suggest that root trait‐decomposition linkages differ strongly with mycorrhizal type and environment, and that root diameter can predict decomposition but in opposing directions and with contrasting mechanisms for AM and ECM species.

Abstract:To adapt to the complex belowground environment, plants make trade-offs between root resource acquisition and defence ability. This includes forming partnerships with different types of root associating microorganisms, such as arbuscular mycorrhizal and ectomycorrhizal fungi. These trade-offs, by mediating root chemistry, exert legacy effects on nutrient release during decomposition, which may, in turn, affect the ability of new roots to re-acquire resources, thereby generating a feedback loop. However, the linkages at the basis of this potential feedback loop remain largely unquantified. Here, we propose a trait-based root 'acquisition-defence-decomposition' conceptual framework and test the strength of relevant linkages across 90 angiosperm tree species. We show that, at the plant species level, the root-fungal symbiosis gradient within the root economics space, root chemical defence (condensed tannins), and root decomposition rate are closely linked, providing support to this framework. Beyond the dichotomy between arbuscular mycorrhizal-dominated versus ectomycorrhizal-dominated systems, we suggest a continuous shift in feedback loops, from 'high arbuscular mycorrhizal symbiosis-low defence-fast decomposition-inorganic nutrition' by evolutionarily ancient taxa to 'high ectomycorrhizal symbiosis-high defence-slow decomposition-organic nutrition' by more modern taxa. This 'acquisition-defence-decomposition' framework provides a foundation for testable hypotheses on multidimensional linkages between species' belowground strategies and ecosystem nutrient cycling in an evolutionary context.

Abstract:Arbuscular mycorrhizal (AM) fungi influence interactions among plant species through enhancing nutrient uptake and possibly facilitating nutrient transport among plants. However, the effects of one plant species on coexisting plant species with regard to mycorrhizal colonization are not well understood. We examined root mycorrhizal colonization and phosphorus (P) acquisition of plants in a highly P-limiting soil in Lanxi city, Zhejiang, China from the year 2000 to 2002. Three dominant native plant species with different mycorrhizal properties, Digitaria ciliaris (poorly mycorrhizal species), Ixeris denticulate (moderately mycorrhizal species) and Kummerowia striata (highly mycorrhizal species), were planted in experimental plots. In the monocultures, K. striata was found to have the highest infection and D. ciliaris the lowest mycorrhizal infection, but shoot P-concentration was higher in both I. denticulate and D. ciliaris than that in K. striata. In the mixtures, D. ciliaris and I. denticulate did not significantly affect the mycorrhizal colonization, spore production and shoot P-concentration of K. striata plants, but K. striata and I. denticulate significantly increased root mycorrhizal colonization and shoot P-concentration of D. ciliaris. K. striata enhanced but D. ciliaris reduced mycorrhizal infection and shoot P-concentration of I. denticulate. These results suggested that highly mycorrhizal plant species may positively impact coexisting species with respect to mycorrhizal colonization and P acquisition, but the effects on poorly mycorrhizal species are less predictable.
Tree growth is better explained by absorptive fine root traits than by transport fine root traits

Abstract:Abstract 1.Quantifying plant trait variation yields insights into trade-offs inherent in the ecological strategies of plants and is the basis for a trait-based prediction of plant performance and ecosystem functioning. Although the interest in root traits has increased in recent years, we still have limited knowledge of i) whether functionally different fine roots±absorptive versus transport roots±have similar trait coordination and ii) how they help to explain plant performance, such as growth. 2.We measured traits of 25 European broadleaved tree species growing in a research arboretum to study i) the coordination within absorptive and transport fine root traits and ii) the degree of trait-tree growth relationships. To do so, we combined a suite of morphological (root diameter, specific root length and root tissue density) and anatomical (cortex to stele ratio and mycorrhizal colonization rate) traits for each of the absorptive and transport roots roots and also leaf traits (leaf mass per area, dry matter content and toughness). 3.Despite remarkable differences in average trait values between absorptive and transport roots, our study shows that trait coordination within absorptive and transport roots is relatively equivalent. Our results also show that, for the traits we studied, tree growth is better explained by absorptive root traits than by transport root traits and is higher in species with a thinner root diameter. This suggests that variation primarily in absorptive roots affects the uptake of soil-based resources like nutrients and water and directly influences tree growth. 4.The significant relationship between absorptive roots and tree growth and the lack of such a relationship for transport highlight that roots mostly involved with resource absorption are more important in explaining tree growth than roots involved in transport.

Variation in fine root traits with thinning intensity in a Chinese fir plantation insights from branching order and functional groups

Abstract:Abstract Thinning is a widely used practice in forest management, but the acclimation mechanisms of fine roots to forest thinning are still unclear. We examined the variations in fine root traits of different branching orders and functional groups along a thinning intensity gradient in a 26-year-old Chinese fir ( Cunninghamia lanceolata ) plantation. With increasing thinning intensity, the root C concentration (RCC), root N concentration (RNC), specific root area (SRA), and specific root length (SRL) of the absorptive roots (the first two orders) significantly decreased, while root abundance (root biomass and root length density) and root tissue density (RTD) significantly increased. Fifty-four percent of the variation in the absorptive root traits could be explained by the soil N concentration and the biomass and diversity of the understorey vegetation. Conversely, transport root (third- and higher-order) traits did not vary significantly among different thinning intensities. The covariation of absorptive root traits across thinning intensities regarding two dimensions was as follows: the first dimension (46% of the total variation) represented changes in root abundance and chemical traits (related to RCC, RNC), belonging to an extensive foraging strategy; the second dimension (41% of the total variation) represented variations in root morphological traits (related to RTD, SRL and SRA), which is an intensive foraging strategy (i.e., root economic spectrum). These results suggested that the absorptive roots of Chinese fir adopt two-dimensional strategies to acclimate to the altered surroundings after thinning.

Abstract:Introduction As a crucial factor in determining ecosystem functioning, interaction between plants and soil-borne fungal pathogens deserves considerable attention. However, little attention has been paid into the determinants of root-associated fungal pathogens in subtropical seedlings, especially the influence of different mycorrhizal plants. Methods Using high-throughput sequencing techniques, we analyzed the root-associated fungal pathogen community for 19 subtropical forest species, including 10 ectomycorrhizal plants and 9 arbuscular mycorrhizal plants. We identified the roles of different factors in determining the root-associated fungal pathogen community. Further, we identified the community assembly process at species and mycorrhizal level and managed to reveal the drivers underlying the community assembly. Results We found that plant species identity, plant habitat, and plant mycorrhizal type accounted for the variations in fungal pathogen community composition, with species identity and mycorrhizal type showing dominant effects. The relative importance of different community assembly processes, mainly, homogeneous selection and drift, varied with plant species identity. Interestingly, functional traits associated with acquisitive resource-use strategy tended to promote the relative importance of homogeneous selection, while traits associated with conservative resource-use strategy showed converse effect. Drift showed the opposite relationships with functional traits compared with homogeneous selection. Notably, the relative importance of different community assembly processes was not structured by plant phylogeny. Drift was stronger in the pathogen community for ectomycorrhizal plants with more conservative traits, suggesting the predominant role of stochastic gain and loss in the community assembly. Discussion Our work demonstrates the determinants of root-associated fungal pathogens, addressing the important roles of plant species identity and plant mycorrhizal type. Furthermore, we explored the community assembly mechanisms of root-associated pathogens and stressed the determinant roles of functional traits, especially leaf phosphorus content (LP), root nitrogen content (RN) and root tissue density (RTD), at species and mycorrhizal type levels, offering new perspectives on the microbial dynamics underlying ecosystem functioning.

Abstract:Leaves, stems and roots as the main plant organs have specific functions and together modulate survival, growth and reproduction. The relationships between these organs are high research priority, and there have been many hypotheses about the trade-offs between them. However, the results of these hypotheses are inconsistent and confusing. In this study, we examined 15 core traits of leaves, stems and woody roots of 27 dominant shrub species and further tested the hypotheses about the relationships between these organs. Measurements were made for shrubs across 9 sites including desert, steppe, temperate forest and subtropical forest in Shaanxi Province of China. Many significant correlations of different organ traits were found, e.g. nitrogen and phosphorus content showed a significant positive correlation, either within or across organs. Also, representatives of structural traits (carbon content and dry matter content) and mineral nutrient traits (nitrogen and phosphorus content) showed significant positive correlations among the leaves, stems and roots. The results of this study supported the hypotheses that there were significant correlations between leaf and root and between stem and root. Similarly, we found that trade-off between leaf and stem-plus-root showed a significant correlation. Thus, root traits, which are difficult to measure, are coordinated with those of the leaf and stem. We conclude that the leaf component of shrubs is a good proxy for the whole-plant in studying trade-offs and it could provide a convenient way to understand the whole-plant economic spectrum by focusing on the leaf economic spectrum.

Abstract:1. Root respiration is essential for nutrient acquisition. The respiration rate of absorptive roots theoretically relates to the economics of carbon-nutrient exchange, but its empirical role remains largely unexplored in the trait space defining nutrient uptake strategies.2. Here, we measured the respiration rates of the distal, non-woody, absorptive roots of 252 woody and herbaceous species from subtropical and temperate climate zones, including both arbuscular mycorrhizal and ectomycorrhizal fungal hosts.3. We found a consistent and positive correlation between root respiration rate and specific root length (root length per dry weight), irrespective of growth form, mycorrhizal type and climate zone. Root respiration rate was also positively, but less strongly and less frequently correlated with root nitrogen concentration. Root morphology strongly explained the fast-slow gradient of root respiration in the root economics space.4. By quantifying the ratio of arbuscular mycorrhizal fungal DNA copy number and root tissue DNA copy number using qPCR, we found that the morphology-driven gradient did not explain the full variation in fungal collaboration; thick roots were consistently well colonised, but medium and thin roots displayed a wide range of colonisation intensity.5. Synthesis: These results advance our understanding of the fundamental trait relationships that underpin the root economics space. Our study also provides a physiological linkage to the frequently measured root morphological traits and relates the root economics space to root-derived carbon-nutrient cycling processes.
Patterns in spatial distribution and root trait syndromes for ecto and arbuscular mycorrhizal temperate trees in a mixed broadleaf forest

Abstract:Functional differences between trees with arbuscular (AM) or ectomycorrhizal (ECM) partnerships influence important ecological processes including nutrient cycling, community assembly, and biomass allocation patterns. Although most broadleaf temperate forests show both mycorrhizal types, relatively few studies have addressed functional difference among coexisting mycorrhizal tree species. The maintenance of ECM associations usually requires higher C investment than AM, leading to (A) lower root biomass and (B) more conservative root trait syndromes in ECM tree species compared to AM species. Here we quantified the representation and trait syndromes of 14 canopy tree species associated with either AM or ECM fungi in a natural forest community. Our results showed that, whereas species root abundance was proportional to basal area, some ECM tree roots were largely under-represented (up to ~ 33%). Most of the under-representation was due to lower than expected root abundance of Quercus rubra and Fagus grandifolia. Functional root traits in tree species were similar, with the exception of higher tissue density in ECM species. Moreover, closely related AM and ECM exhibited similar traits, suggesting inherited trait syndrome from a common ancestor. Thus, we found little evidence of divergent functional root trait syndromes between mycorrhizal types. Cores dominated by ECM species influenced trait distribution at the community level, but not total biomass, suggesting that mycorrhizal affiliation may have a stronger effect on the spatial distribution of traits but not on biomass stocks. Our results present an important step toward relating belowground carbon dynamics to species traits, including mycorrhizal type, in broadleaf temperate forests.

Abstract:In most cases, both roots and mycorrhizal fungi are needed for plant nutrient foraging. Frequently, the colonization of roots by arbuscular mycorrhizal (AM) fungi seems to be greater in species with thick and sparsely branched roots than in species with thin and densely branched roots. Yet, whether a complementarity exists between roots and mycorrhizal fungi across these two types of root system remains unclear. We measured traits related to nutrient foraging (root morphology, architecture and proliferation, AM colonization and extramatrical hyphal length) across 14 coexisting AM subtropical tree species following root pruning and nutrient addition treatments. After root pruning, species with thinner roots showed more root growth, but lower mycorrhizal colonization, than species with thicker roots. Under multi-nutrient (NPK) addition, root growth increased, but mycorrhizal colonization decreased significantly, whereas no significant changes were found under nitrogen or phosphate additions. Moreover, root length proliferation was mainly achieved by altering root architecture, but not root morphology. Thin-root species seem to forage nutrients mainly via roots, whereas thick-root species rely more on mycorrhizal fungi. In addition, the reliance on mycorrhizal fungi was reduced by nutrient additions across all species. These findings highlight complementary strategies for nutrient foraging across coexisting species with contrasting root traits.
