Abstract:A wave of dierentiating osteoblasts in the perichondrium of growing long bones is coupled to graded expression of PTEN, a negative regulator of phosphatidylinositol 3 kinase (PI3K) pathway. To better define the role of PTEN and PI3K signaling during bone formation, we conditionally deleted PTEN in osteoprogenitor cells using Dermo1cre deleter and PTEN floxed mice. Mice lacking PTEN in osteoprogenitors showed a dramatic increase in perichondral bone formation and precocious osteoblast dierentiation. In the absence of PTEN, there was a substantial increase in proliferation of perichondrial osteoprogenitors. This coincided with an increase in FGF18 expression, a potent osteoblast mitogen, and diminished feedback inhibition of FGF signaling as demonstrated by decreased Sprouty2 and DUSP4 (dual specificity phophatase 4) expression. PI3K-dependent inhibition of FoxO transcription factors contributes to the reduction in Sprouty2 as well as increase in FGF18 expression. These findings implicate unrestricted mitogen activated protein kinase (MAPK) signaling, downstream of FGF receptor signaling as the principle eector of PTEN deletion. Indeed, we show that precocious dierentiation of perichondral osteoprogenitors is accompanied by increased levels of Gli2, a major inducer of osteoblast dierentiation. Moreover, we find that Gli2 activity and protein level is increased by FGF receptor and MAPK signaling. To verify the requirement for FGF signaling in these events, we dampened FGF signaling by removing one allele of FGF receptor2, this caused a marked reversal of the PTEN null phenotype with a reduction of perichondral bone development and osteoblast dierentiation
